Abstract
Description

本明細書において使用されるように、「多能性の」という用語は、複数の異なる型の細胞(例えば、最終分化細胞)へと分化する細胞の能力を意味する。例えば、多能性細胞には、三つの主な胚葉:内胚葉、外胚葉、および中胚葉へと分化し得るものが含まれる。
中枢神経系は、脳および脊髄を含む。体内のその他の神経は全て、末梢神経系を構成する。遠心性神経は、中枢神経系から身体の全ての部分(末梢)へとメッセージを伝え、求心性神経は、末梢から中枢神経系へと疼痛強度のような情報を伝える。遠心性神経には、二つの型が存在する:骨格筋に通じる体性神経、ならびに平滑筋、腺、および心臓に通じる自律神経。メッセージは、電気活動の形態で、神経繊維または軸索に沿って伝導される。軸索の終末と、その神経が制御する(神経支配する)筋肉または腺との間には、シナプスまたはシナプス間隙と呼ばれる隙間が存在する。伝導された電気的インパルス(活動電位)は、神経終末に到達すると、神経伝達物質と呼ばれる化学物質の放出を誘発する。これらの化学物質はシナプス間隙一帯に拡散し、接合部後膜上の固有の構造(受容体)と反応する。そのとき、受容体は、活性化される、または興奮すると言われ、その活性化は、筋収縮のような生物学的応答を最終的にもたらす一連の化学的事象の引き金を引く。神経伝達物質の放出、拡散、および受容体活性化を含む過程は、集合的に、伝達と呼ばれる。多くの型の伝達が存在し、それらは、含まれている特異的な神経伝達物質にちなんで命名されている。従って、コリン作動性伝達には、神経伝達物質アセチルコリンの放出、およびそれによるシナプス後受容体の活性化が含まれる。受容体に結合し、それを活性化するものは、アゴニストと呼ばれる。従って、アセチルコリンは、全てのコリン作動性受容体の内因性アゴニストである。
本発明において使用されるペプチド両親媒性物質(PA)組成物は、当業者に周知の調製技術を使用して、好ましくは、アルキル化または疎水性モエティによるペプチド要素のN末端のその他の修飾を含む標準的な固相化学により、合成され得、ペプチドのNまたはC末端に付着したモノまたはジアルキル部分は、合成系および天然系の両方において水中での凝集および二次構造に影響を及ぼし得る。ベータ鎖コンフォメーションの優先性を有しているイオン性ペプチドにカップリングされた十分な数の炭素原子を有する疎水性炭化水素および/またはアルキル・テール要素が、ある種の態様において、ナノファイバー構造へと(例えば、インビボで)集合する両親媒性物質を作出するために使用され得る。両親媒性物質の全体的な円錐状の形も、そのような集合に対して効果を及ぼし得る。自己集合は、体液(例えば、脳脊髄液)によって引き金を引かれ得る。
本発明の開発の過程で実施された実験は、神経前駆細胞が、本発明の方法を使用して、ニューロンへと効率的に分化し得ることを証明した(例えば、実施例1〜8参照)。細胞は、有意な量のアストロサイトの形成なしに、分化を示した(例えば、実施例6および11参照)。さらに、実験は、損傷脊髄を有する対象への本発明のPAを含む組成物の投与(例えば、損傷脊髄への注射)が、損傷の部位におけるアストログリオーシスを低下させ、感覚および運動線維の実質的な再生を促進し、行動学的回復(例えば、そのような処置の前には麻痺していた四肢の可動性)を有意に増強することを証明した(例えば、実施例1、9〜13参照)。

以下の実施例は、本発明のある種の好ましい態様および局面を例証し、さらに例示するために提供され、その範囲を制限するものと解釈されるべきではない。
材料および方法
細胞培養およびIKVAV-PAナノ-ネットワークへのインビトロ封入。神経前駆細胞(NPC)は、以前に記載されたようにして培養された(例えば、Zhu et al.,J.Neurosci.Res.59,312(2000)参照)。簡単に説明すると、E13マウス胚の皮質が解剖され、bFGF(10ng/ml)が補足されたDMEM/F12培地で未処理のペトリ皿に播種された。4日後に、機械的かつ酵素的に解離させられたNPCおよび未解離のニューロスフェア(例えば、未解離のNPC凝集物)が、適切な基質へと播種された(例えば、IKVAV-ペプチド両親媒性物質(PA)、EQS-PA、もしくはアルギン酸ゲルに封入されるか、またはラミニン、ポリD-リジン、もしくはIKVAVペプチドでコーティングされたカバーガラスにおいて培養された)。全ての場合に、これを、インビトロ0日目とした。
細胞生存率/細胞障害性は、Molecular Probes LIVE/DEAD細胞アッセイ(Molecular Probes)を使用することにより査定した。NPCのため最適化されたエチジウム・ホモダイマー-1(EthD)およびカルセインの作業濃度を、Molecular Probesの指示通りに決定し、それぞれ0.5および8μMであることを決定した。培養培地をウェルから除去し、PAゲルの液浸(submersion)を保証するために十分なPBS中のEthD/カルセイン溶液をウェルに添加した。培養プレートを20分間インキュベーターに戻し、次いで、EthD/カルセイン溶液を除去し、細胞をPBSで1回洗浄した。EthDおよびカルセインの蛍光を、Nikon TE-2000蛍光顕微鏡で、それぞれFITCおよびTRITCフィルターを使用して画像化した。
封入されたNPCから培養培地を除去し、固定液にPA-ゲル全体を浸すことにより、封入された細胞を、室温で20分間、4%パラホルムアルデヒドで固定した。0.2%トリトン-X(tオクチルフェノキシポリエトキシエタノール(toctylphenoxyplyethoxyethanol))との5分間のインキュベーションの前に、PBSで2回洗浄した。これに続いて、さらに2回PBSで洗浄し、4℃で一夜、5%ヤギまたはウマ血清を含有しているPBS中で一次抗体(1:400の抗β-チューブリンIII IgGまたは1:400の抗GFAP、Sigma)をインキュベートした。PBSで3回洗浄した後、室温で2時間、5%ヤギまたはウマ血清を含有しているPBS中のTRITC-またはFITC-接合二次抗体と共に細胞をインキュベートした。さらに3回PBSで洗浄した後、β-チューブリンおよびGFAP陰性の細胞を可視化するため、全ての核を、室温で10分間、Hoescht's Stain(1:5000、Sigma)で染色した。細胞画像化は、PC実行MetaView画像化ソフトウェアとインターフェースで接続されたNikon TE-2000蛍光顕微鏡に取り付けられた高分解能Cool Snapカメラ、またはPC実行AxioVision画像化ソフトウェアとインターフェースで接続されたZeiss Axiovert 200蛍光顕微鏡に取り付けられたAxiocamカメラにより行った。
異なる実験条件についてランダムに選択された視野を画像化し、ImageJ(Scion Corporation)形態測定分析ソフトウェアを使用して細胞を計数した。フィルター間の蛍光の漏出(bleed-through)がなかったことを確認するために画像をチェックし、ImageJを使用して半自動的に細胞を計数した。特に、全細胞数の自動的な連続計数を維持するマーキング・ツール(marking tool)を使用して、手動で細胞を選択することにより、所定の領域内の全細胞数を計数した。細胞数の定量的および統計的な分析は、Matlab(Math works)および/またはExcel(Microsoft)を使用して行った。
45mg/kgペントバルビタール(NEMBUTAL)を使用して、ラットに麻酔をかけた。脊髄分節T13を露出させるために椎弓切除を実施し、32ゲージ注射針に取り付けられたハミルトン注射器を有する定位マイクロマニピュレーター(Kopf Instruments)を使用して、等浸透圧グルコース(媒体)またはペプチド両親媒性物質6μlを、333nl/秒で、1.5mmの深さで脊髄のT10へ注射した。IKVAV-PAが妨害なしにゲル化することを可能にするため、各注射の後に2分間、針を注射部位の内部に維持した。ペプチド両親媒性物質を注射された動物は、歩行運動行動または全身状態の変化を示さず、そのことから、ペプチド両親媒性物質の注射が毒性効果を有していないことが示された。
全ての実験を、Association for Research in Vision and Ophthalmology (ARVO)およびAnimal Care and Use Committee (ACUC) of Northwestern Universityの規則に従って行った。成体Sprague-Dawleyラット(200〜250g)をペントバルビタール(Pentabarbital)ナトリウムの過量投与またはCO2過量投与により屠殺し、直ちに外科的に眼球除去した。25ゲージ針を有する100μlハミルトン注射器に、IKVAV-PA溶液80〜100μlをあらかじめ負荷し、眼球除去された目を、Nikon SMZ-1000手術用実体顕微鏡のプラットフォームに置いた。ほぼ網膜下または硝子体(vitreal)の空間へと斜角で実体顕微鏡下で眼窩の背部へIKVAV-PA溶液を手動で注射し、MetaView画像化ソフトウェアを使用して、Cool Snap高解像度カメラとインターフェースで接続された実体顕微鏡を使用して画像化した。
固液界面におけるタンパク質の吸着は、典型的には、1μg/cm2の付近にある(例えば、Ratner,Biomaterials Science:An introduction to materials in medicine(Academic Press,San Diego,1996)参照)。この値を使用し、ラミニンの分子量が800kDaであると仮定し(例えば、Tunggal et al.,Microsc.Res.Tech.51,214(2000)参照)、かつネイティブ・ラミニン-1分子上のIKVAVエピトープの数が1であると仮定すると、カバーガラスまたは培養プレートのような二次元表面において、表面上のIKVAVエピトープの密度は、
であると計算された。
これらの二つの数は、二次元表面上のものに対するナノファイバー上のIKVAVエピトープの比率を見出すために割られ、密接にパックされたラミニン分子の二次元表面に対するナノファイバー上のIKVAVエピトープの増幅定数を与える:
IKVAVペプチド実験のため、三次元実験のために使用されたのと同一の12mmカバーグラスを、親水性を強めるためにエタノールに浸し、次いで、1mg/mL IKVAVペプチド溶液50μLをスピン・コーティングした。IKVAV-PA実験のためのカバーグラスは、(例えば、吸着を強めるため)PDLでコーティングし、続いてIKVAV-PA溶液でコーティングした。いずれの場合にも、カバーガラスを一夜乾燥させ、次いで、弱く接着性の材料を除去するために蒸留水で3回洗浄した後、細胞懸濁物を添加した。1 DIV後のβ-チューブリン染色の結果は、図5に示される。
全ての動物ケアおよび外科的介入が、Public Health Service Policy on Humane Care and Use of Laboratory Animals,Guide for the Care and Use of Laboratory Animals(Institute of Laboratory Animal Resources,National Research Council,1996)に厳密に従い実施された。The Institutional Animal Care and Use Committeeは、全ての作業手技を承認した。雌成体129SvJマウス(10週齢;Jackson Labs,USA)に、腹腔内でアバーチン(avertin)を使用して麻酔をかけた。椎弓切除を実施し、1分間、修飾型Kerr-Lougheed動脈瘤クリップの硬膜外適用により、T10で背腹側に脊髄を圧迫した(EEJOTA mouse clip,University Health Network,Canada)。AUTOCLIP(9mm、Becton Dickinson)を使用して、皮膚を縫合した。術後、体温を維持するために、ヒート-ランプの下で動物を維持した。損傷後、最初の1週間、生理食塩水を1.0cc皮下注射することを毎日繰り返した。損傷から24時間後に何らかの後肢運動を示したマウスは、研究から排除した。不快の事象においては、ブプレネックス(buprenex)(2mg/kg SC、1日2回)を投与した。血尿症(20mg/kg)の事象においては、ゲンタマイシンを、5日間、1日1回、皮下に投与した。
索の非損傷部分における基線レベルに対する、病変部周囲のGFAPレベルの増加倍率を推定するため、免疫染色の後、GFAP免疫反応性の蛍光強度を測定した。各動物について、等しい内外方向の深さの切片を分析のために使用した。次いで、切片を、Zeiss UVLSM-Meta共焦点顕微鏡(Carl Zeiss,Inc.,Thornwood,NY)で画像化した。各共焦点スキャンを、同一のレーザー出力、利得、およびオフセット値を使用して実施した。これらの値は、損傷区域の画像の中のピクセルが飽和しないよう設定された。スキャンのZスタックを、LSM画像ブラウザー(Carl Zeiss)を使用して再構築した。全Zスタックを単色(.tif)画像に変換し、続いて、各ピクセルのグレイレベルを測定することにより、蛍光定量化を実施した。各ピクセルは、0〜255の範囲のグレイスケールを有する。各スタックの全ピクセル強度を、MetaMorph 2.6ソフトウェアを使用して積分した。個々の各切片についての損傷区域における強度値を、増加したGFAP免疫反応性の区域の端から(吻側にも尾側にも)500μm>遠位として定義された非損傷部分で得られたスキャンに由来する基線値に対して規準化した。各切片について、損傷区域における四つの部位(病変部中心点から吻側二つおよび尾側二つ)および(灰白質および白質の両方にかかる)非損傷区域における三つの部位をスキャンし、全強度値を各群について平均化した。そのような様式で、各動物について少なくとも四つの切片を分析した。最終的な蛍光値を、個々の切片について基線(非損傷区域)値に対する増加倍率として表し、次いで、それを、ゲル注射群と媒体注射群との間の比較のため、各動物について分類した。
損傷後1日目または9週目に、マウスにアバーチン麻酔をかけ、プルド(pulled)ガラス・マイクロピペットが取り付けられた10μlハミルトン・マイクロシリンジを使用して、mini-ruby接合BDA(Molecular Probes,Eugene OR)を注射した。後柱標識のため、2μlをL5後根神経節に注射した。皮質脊髄路は、正中に対して1.0mm外側、ブレグマに対して0.5mm前側、0.5mm後側、1.0mm後側、皮質表面から0.5mmの深さでなされた三つの注射(各0.5μl)を通して標識した。14日後にCO2吸入を使用して動物を屠殺し、灌流した。
浮遊連続切片を収集し、1×PBSおよび0.1%トリトンX-100で3回洗浄し、アビジンおよびビオチン化西洋ワサビペルオキシダーゼ(Vectastain ABC Kit,Vector,Burlingame,CA)と共に4℃で一夜インキュベートし、再び1×PBSで3回洗浄し、次いで、50mMトリス緩衝液、pH7.6、0.024%過酸化水素、および0.5%塩化ニッケルの中でDABと反応させた。次いで、切片をPBSに移し、顕微鏡スライドに連続する順序でマウントし、Neurolucidaソフトウェア(MicroBrightField,Inc.)を使用して路を追跡した。
体重150〜200gのAdult Long Evans Hooded雌ラットにペントバルビタール麻酔を使用して麻酔をかけた。椎弓切除を実施し、MASCISインパクター(最大重度の損傷を生じる10gm重量/50mm落下)により脊髄分節T13において脊髄に挫傷を与えた。上記のように、体温および水和状態を維持した。動物を、各ケージに1匹ずつ収容した。ゲル注射のため、27ゲージ針を使用し、両親媒性物質を上記のように希釈した。挫傷の24時間後、ラットにペントバルビタール麻酔を使用して再度麻酔をかけた。損傷部位を露出させた後、希釈された両親媒性物質5μlを、1.5mmの深さで、病変部中心点に対して0.5mm吻側および尾側に、1μl/分の速度で注射した。注射の終了時、さらに2分間索に針を残し、その後、引き抜き、創傷を閉じた。その他の動物は、媒体(グルコース溶液)の類似した注射を受容した。第三の群(偽注射)においては、創傷を再切開し、次いで、注射なしで再び閉じた。
CO2吸入を使用して動物を屠殺し、リン酸緩衝生理食塩水(PBS)中の4%パラホルムアルデヒドで経心的に灌流した。脊髄を解剖し、4%PFA中の30%ショ糖で一夜固定した。次いで、脊髄をTissue-Tek包埋化合物で凍結させ、Leica CM3050Sクリオスタットで切片化した。20μmの厚い縦方向切片を採取した。切片をPBSで2回濯ぎ、次いで、室温で1時間抗GFAP[1:250](Sigma、マウス・モノクローナルIgG1)と共にインキュベートした。これに続いて、切片をPBSで3回濯ぎ、室温で1時間、alexa-fluor接合抗マウスIgG1二次抗体[1:500]Molecular Probes)と共にインキュベートした。切片を最後にPBSで3回濯ぎ、次いで、室温で10分間、Hoechst核染色と共にインキュベートした。最後にPBSで濯いだ後、Prolong Gold褪色防止試薬(Molecular Probes)を使用してマウントし、Zeiss UVLSM-Meta共焦点顕微鏡(Carl Zeiss,Inc.,Thornwood,NY)を使用して画像化した。
P1(生後1日目)マウスの脳室下帯を解剖し、EGF(20ng/ml)、N2およびB27サプリメント、ヘパリン、ペニシリン、ストレプトマイシン、ならびにL-グルタミンが補足されたDMEM/F12培地で増殖させ、浮遊スフェアを形成させた。細胞を1回継代し、得られた二次スフェアを分析のために使用した。細胞を解離させ、EGF(5ng/ml)が補足されたDMEM/F12培地で、適切な基質へと播種した(例えば、IKVAV-PAへの封入またはポリd-リジン/ラミニン上での培養)。全ての場合に、これをインビトロ0日目とした。IKVAV-PAネットワークへの前駆細胞の封入は、まず、24穴培養プレート内の12mmカバーガラスへPA溶液100μlを分注して、独立した液滴を形成させることにより達成した。次いで、細胞懸濁物が導入される間、ピペット・チップを穏やかに回転させながら、培養培地中の細胞懸濁物100μlをPA溶液の液滴へとピペットで移し、PAゲルを形成させた。ゲルを、(37℃および5%CO2、95%湿度の)インキュベーター内に>2時間静置し、その後、培養培地300μlをウェルに添加し、PAゲルを部分的に浸した。次いで、プレートをインキュベーターに戻した。対照培養のための、12mmカバーガラスを、1時間ポリD-リジンでコーティングし、それに続き、蒸留水で洗浄し、次いでラミニン(Sigma、1mg/100ml DMEM)で一夜コーティングした。細胞を含む培養培地500μlを、5×104細胞/mlの播種密度でウェルに添加した。免疫細胞化学のため、封入された細胞から培養培地を除去し、PAゲル全体を固定液に浸すことにより、IKVAV-PAに封入された細胞を、室温で20分間、4%パラホルムアルデヒドで固定した。ラミニン上に播種された細胞については、カバーグラスを固定液中に置いた。0.2%トリトン-Xとの5分間のインキュベーションの前に、PBSで2回洗浄した。これに続き、PBSでさらに2回洗浄し、4℃で一夜、5%ヤギ血清を含有しているPBS中での一次抗体(1:400抗β-チューブリンIII IgG2aまたは1:400抗GFAP IgG1、Sigma)インキュベーションを行った。PBSで3回洗浄した後、室温で1時間、PBS中でTRITC-またはFITC-接合二次抗体と共に細胞をインキュベートした。PBSでさらに3回洗浄した後、β-チューブリンおよびGFAP陰性細胞を含む全ての細胞の核を可視化するため、室温で10分間、Hoechst染色(1:5000、Sigma)により染色した。細胞画像化は、PC実行AxioVision画像化ソフトウェア(Zeiss)とインターフェースで接続されたZeiss Axiovert 200蛍光顕微鏡に取り付けられたAxiocamカメラを用いて実施された。
測定は、25mm平行プレート構成を有するPaar Physica Modular Compact Rheometerを使用してなされた。3%ひずみで、各PAについて、0.1〜100Hzの周波数掃引(Frequency sweeps)を得た。
自己集合性足場の作製
細胞分化を指図するための自己集合性人工足場の使用をインビトロで研究するため、マウス神経前駆細胞(NPC)を使用した。NPCは、(例えば、変性性または外傷性の傷害の後の)失われた中枢神経系細胞の補充において有用である(例えば、Okano,J.Neurosci.Res.69,698(2002);Storch and Schwarz,Curr.Opin.Invest.Drugs 3,774(2002);Mehler and Kessler,Arch.Neurol.56,780(1999);Pincus et al.,Neurosurgery 42,858(1998)参照)。足場の分子設計は、ラミニンに見出され、神経突起発芽を促進し、かつ神経突起成長を指図することが公知であるペンタペプチド・エピトープ、イソロイシン(isolucine)-リジン-バリン-アラニン-バリン(IKVAV)を組み入れた(例えば、Kam et al.,Biomaterials 22,1049(2001);Matsuzawa et al.,Int.J.Dev.Neurosci.14,283(1996);Powell et al.,J.Neurosci.Res.61,302(2000);Cornish et al.,Mol.Cell.Neurosci.20,140(2002);Chang et al.,Biosens.Bioelectron.16,527(2001);Wheeler et al.,J.Biomech.Eng.121,73(1999);Lauer et al.,Biomaterials 23,3123(2002);Thiebaud et al.,Biosens.Bioelectron.17,87(2002).Yeung et al.,Neurosci.Lett.301,147(2001)参照)。生理活性に関する対照として、それを非生理学的配列グルタミン酸-グルタミン-セリン(EQS)に交換して、天然エピトープを欠く類似した分子を合成した。これらの分子は、自己集合により物理的に類似した足場を形成するが、EQSゲル内に封入された細胞は、神経突起を発芽せず、形態学的にも組織学的にも分化しなかった。
ナノファイバー足場の特徴決定
1重量%(wt%)ペプチド両親媒性物質水性溶液を、1:1の容積比で媒体または生理学的液体の中のNPC懸濁物と混合すると、図1Cおよび1Dに示される透明なゲル様固体が数秒以内に入手された。この固体は、封入された解離したNPC、またはニューロスフェアとして公知の細胞のクラスターを含有していた。細胞は、自己集合過程に耐え、観察期間中(22日)生存可能なままであった(図2A〜2D参照)。ポリ(D-リジン)(PDL、多くの細胞型を培養するために使用されている標準的な基質)上で培養された細胞と、ナノファイバー・ネットワークに封入された細胞との間に、生存率の有意な差は存在しなかった(図2D参照)。従って、本発明は、これらの高度水和ネットワークを通した栄養素、生理活性因子、および酸素の拡散が、より長い期間の多数の細胞の生存にとって十分であることを証明している。自己集合性分子により形成された人工足場は、99.5wt%の水を含有しており、メカニズムの理解は本発明を実施するのには必要でなく、本発明はいかなる特定の作用機序にも制限されないが、ナノファイバーの高い縦横比が、ペプチド両親媒性物質のそのような低い濃度での、機械的に支持性のマトリックスの形成を可能にすることが企図される。従って、人工細胞外マトリックスは、細胞のための機械的支持を提供するのみならず、可溶性因子の拡散および細胞の遊走が起こり得る媒体としても役立つ。
足場に曝された(例えば、足場に封入された)神経前駆細胞の増強された分化
生理活性足場において、免疫細胞化学により決定されるようなニューロンに分化したNPCの細胞体面積および神経突起長は、PDLまたはラミニンでコーティングされた基質上で培養された細胞と比較して統計的に有意な差を示した。ナノファイバー・ネットワーク内のニューロンは、対照培養物中のニューロンより顕著に大きかった。ネットワークに封入された前駆細胞の平均細胞体面積は、1および7日後に有意に大きかった(図2E参照)。ナノファイバー足場への封入は、わずか1日後に大きな神経突起の形成に至ったが(約57±26μm、平均値±SD)、PDLおよびラミニンで培養された細胞は、この初期の時点で神経突起を発達させていなかった。7日後にも、足場内のニューロンは、PDL基質で培養された細胞と比較して、有意に長い突起を有していた(P<0.01)。7日後、PA足場で培養された細胞と、ラミニンでカバーされた基質で培養された細胞との間に、神経突起長の統計的な差は存在しなかった。7日間生理活性足場に封入されていたNPCの透過型電子顕微鏡検(TEM)は、横断面全体に可視の豊富な突起を含む、健康で正常な超微細構造的形態学を示した(図2F参照)。
ナノファイバー足場内の細胞遊走
ナノファイバー足場内の細胞遊走の可能性を査定するために、三つの封入されたニューロスフェアを14日間追跡した(図3Aおよび3B参照)。構成細胞が外側へ遊走するにつれ、三つのニューロスフェア全てが中心から拡散した(図3B参照)。各ニューロスフェアの中心と外辺部の細胞体との間の距離を複数回測定することにより、この効果を定量化し(例えば、Zhu et al,J.Neurosci.Res.59,312(2000)参照)、個々の細胞が、細胞塊の中心から離れて遊走するのが見られた。ナノファイバー・マトリックス内の細胞の遊走は、時間の関数として統計的に有意であった(P<0.05)(図3A参照)。対照的に、より密で、より強固なネットワーク(99.5%水に対して98%)に封入されたNPCは、生存しなかった。生理活性IKVAV配列の代わりにEQS配列を含むナノファイバーを含有している非生理活性足場においては、細胞は、生存可能なままではあったが、ニューロスフェアから離れて遊走しなかった。IKVAV-PAにおいては、非生理活性EQS-PAと比較して、より大きな神経突起伸長の程度も観察された(図3C参照)。
神経前駆細胞の分化
免疫細胞化学を使用して、培養1および7日後の前駆細胞のインビトロの分化を確立した。それぞれニューロンおよびアストロサイト(中枢神経系(CNS)グリアのサブクラス)のためのβ-チューブリンIIIおよびグリア線維性酸性タンパク質(GFAP)マーカー(図4A〜4E参照)。免疫細胞化学により示されたように、表面上にIKVAVを提示するナノファイバーを含むネットワークに封入されたNPCは、ニューロンへと迅速に分化し、わずか1日後に全細胞の約35%がβ-チューブリン染色陽性となった。対照的に、7日後ですら、GFAP+アストロサイト分化はほとんど存在しなかった(<5%)。アストロサイト増殖の阻害は、CNS外傷後の軸索延長の公知の障壁であるグリア性瘢痕の防止において重要であると考えられる(例えば、Rabchevsky and Smith,Arch.Neurol.58,721(2001);Chen et al.,Mol.Cell.Neurosci.20,125(2002);Costa et al.,Glia 37,105(2002)参照)。
二次元(2D)基質におけるNPC分化
IKVAV-PAナノファイバーでコーティングされた二次元(2D)基質におけるNPC分化を調査した。PA分子は、乾燥により表面上で自己集合するが(例えば、Hartgerink et al.,Science 294,1684(2001);Hartgerink et al.,Proc.Natl.Acad.Sci.U.S.A.99,5133 (2002)参照)、そのことをTEMにより確証した。細胞をこれらの表面に1日間播種し、免疫細胞化学により示されるように、2D表面は、ニューロンへの分化を誘導する効果が同等であった。実験誤差内で、2D基質においてニューロンに分化した細胞の百分率は、3D足場と比べて、同一であった(図5参照)。IKVAV可溶性ペプチドまたはラミニンでコーティングされた基質(図4参照)は、同一の期間にIKVAV-PAナノファイバーで観察された有意なニューロン分化に至らなかった。IKVAVペプチドでコーティングされた基質で培養された前駆細胞は、観察期間中、ほぼ定量不可能な量のβ-チューブリンIIIおよび/またはGFAPを発現した。従って、本発明は、ナノファイバーが、2D培養においても3D培養においても、分化を促進する高密度の利用可能なエピトープを細胞に提示することを証明している。さらに(Futhermore)、本発明は、エピトープ提示の次元というよりむしろ密度が、迅速かつ選択的な細胞のニューロンへの分化において重要な役割を果たすことを提供する。ネットワーク中の平均サイズのナノファイバーは、推定7.1×1014IKVAVエピトープ/cm2を含有している。固体基質上の二次元格子内に密接にパックされたラミニン・タンパク質分子は、推定7.5×1011IKVAVエピトープ/cm2を有する。従って、本発明のIKVAVを含むナノファイバーは、ラミニン単層に比べてエピトープ密度をおよそ103倍増幅する。
組織における足場の自己集合
足場の自己集合は、ペプチド両親媒性物質溶液の組織への注射によっても誘発され得る。1wt%ペプチド両親媒性物質溶液10〜80μlを、新鮮に眼球除去されたラット目調製物、および索を露出させるための椎弓切除の後のインビボのラット脊髄へと注射した。このように、これらのペプチド両親媒性物質溶液は、組織との接触により固体の足場へと変換され得る。この過程は、組織にネットワークを局在化し、注射部位の中心点からの分子の受動拡散を防止する。さらに、脊髄へのペプチド両親媒性物質溶液の注射の後、より長い期間、動物が生存することが公知である。
本発明の組成物の特徴決定のためのインビボ・モデル
本発明の開発の過程で作製され特徴決定されたペプチド両親媒性物質のいくつかの態様の模式図が、図6および7に提示される。上記実施例1〜4に記述されたように、これらのバイオマテリアルは、水性溶液から、生理活性であるよう設計された明確な超分子ナノファイバーから構築された3次元マトリックスへと自己集合する。個々のナノファイバーは、6〜8ナノメーターの範囲の明確な直径を有する円柱状の形を有し、繊維軸に垂直な生理活性ペプチド配列を提示することができる(図9a〜9bおよび図7a参照)。上記のバイオマテリアルの一例は、ラミニンの向神経活性ペンタペプチド・エピトープ、イソロイシン(isolucine)-リジン-バリン-アラニン-バリン(IKVAV)が組み入れられたナノファイバーを形成する。これらのナノファイバー・マトリックスは、培養されたニューロンからの突起の伸長を促進し、培養された胚性および出生後の(実施例1〜4および図8を参照)神経前駆細胞のアストロサイト分化を抑制することが見出された。従って、神経損傷(例えば、脊髄損傷)後の(例えば、(例えば、水性溶液に製剤化された)ペプチド両親媒性物質を含む)本発明の組成物の、傷害を受けた神経組織の部位への注射が、ニューロン軸索伸長を促進し、かつ/または脊髄損傷後の軸索再生に対する主要な障壁であるアストログリオーシスおよびグリア性瘢痕の形成を低下させ得るか否かを決定するための実験が、本発明の開発の過程で実施された。
ペプチド両親媒性物質のインビボ特徴決定
損傷脊髄における生分解性ペプチド両親媒性物質の安定性を評価するため、UV範囲の光による励起による索内の足場の可視化を可能にするための蛍光性の誘導体を合成した(図6a参照)。インビボ注射の間に、よりゆっくりとゲル化する系を作出するためにも、両親媒性物質を修飾した(図6および7参照)。等張グルコース中の1%ペプチド両親媒性物質溶液を、ガラス・キャピラリー・マイクロピペットを使用して、損傷の24時間後に、損傷部位に注射した。図9c(注射の24時間後に実施された画像化)に示されるように、蛍光性のゲルが索内に形成され、注射の5週間後にも依然として、損傷を受けた索にレムナントが見られた(図9d参照)。注射の5週間後、蛍光性のゲルは、注射の部位から索内を拡散し始めた(例えば、メカニズムの理解は本発明を実施するのには必要でなく、本発明はいかなる特定の作用機序にも制限されないが、これは、ペプチドおよび脂質に基づくマトリックスの生分解を反映しているのかもしれない)。従って、本発明は、インビボのナノファイバー足場の寿命が、ほぼ数週間であることを証明している。
アストログリオーシスに対するナノファイバー足場の効果
神経損傷後のアストログリオーシスは、初期の肥大応答(細胞サイズの増加)および後期の過形成応答(細胞数の増加)を含む(例えば、Faulkner,J.R.et al.,J Neurosci 24,2143-55(2004);Steward et al.,J Comp Neurol 459,1-8(2003);および図10参照)。アストログリオーシスに対するゲルの効果を分析するために、脊髄損傷の24時間後に、損傷部位に、グルコースで1:1に希釈されたペプチド両親媒性物質または媒体(グルコース、実施例1参照)のいずれかを注射した。処置は、所見を臨床的に関連するようにするため(例えば、ヒト対象において起こり得る事象を表すため)、損傷の24時間後まで遅らせた。媒体注射群は、偽操作動物と比較してわずかに増強された回復を有していることが実験で証明されたため、媒体注射動物を対照群として使用した(図10b参照)。これおよび全てのその後の実験について、実験者は、動物の同一性を知らされないままであった。
損傷を受けた運動および感覚軸索の復元を促進する、ナノファイバー・ゲルのインビボ投与
本発明の組成物の投与が、損傷を受けた運動および感覚軸索の実際の再生を促進し得るか否かを、次に決定した。下行性皮質脊髄運動線維(CST)の分析のため、ビオチン化デキストラン・アミン(BDA)を、損傷の9週間後に感覚運動皮質に注射した(方法論に関しては実施例1を参照のこと)。2週間後、動物を屠殺し、連続する20μm厚さの縦方向切片の分析のため各群3匹の動物から組織を加工した。切片から切片まで個々の軸索を追跡することができるように、連続切片を収集し、病変部に対して吻側500μmの距離内で、標識された各軸索についてNeurolucida画像化ソフトウェアを使用してコースを追跡した。ゲル注射動物および媒体注射動物からの代表的な軌跡は、(11週で分析された)損傷の9週間後にBDA注射を受容した処理群と対照群との間の著しい差を示す(図11参照)。ナノファイバー・ゲル群においては、標識された軸索のほぼ80%が病変部に入ったのに対し、対照動物においては繊維の約50%であった(図11参照)。対照動物においては、病変部を通る進路の25%まで、繊維が検出されなかった。対照的に、ナノファイバー・ゲル処理群においては、繊維のおよそ50%が、病変部を通る進路の半分まで侵入し、繊維の約45%がその距離の4分の3まで侵入した。驚くべきことに、繊維の約35%が、実際に病変部を通って増殖し、損傷に対して尾側の脊髄に入った。使用された損傷の重度では、軸索が保存されていた可能性は低い。
行動学的回復に関連している解剖学的改善
観察された解剖学的改善が、行動学的回復に関連しているか否かを、マウスのために修飾されたBasso、Beattie、およびBresnahan(BBB)の歩行運動スケールを使用して決定した(例えば、Joshi and Fehlings,J Neurotrauma 19,175-190(2002);Bresnahan et al.,Exp Neurol 95,548-70(1987);Basso et al.,Exp Neurol 139,244-56(1996)参照)。行動学的試験は、処置後9週間、週1回、実施された(図13a)。最初の5週間は、対照と自己集合性分子が注射された群との間に区別可能な差は存在しなかったが、5週目以降は、ゲル注射群は、対照群と比較して有意な行動学的改善を示した。9週目、対照群についての平均BBBスコアは7.03+0.8であり、ゲル注射群についての平均スコアは9.2+0.5であった(p<0.04)。7というスコアは、後肢における三つの関節全てにおける広範囲の運動にもかかわらず機能的な運動がないことを意味し、9というスコアは、動物が歩行運動中に足の背側に歩く背側歩行(dorsal stepping)(例えば、後肢運動は機能的な使用を有する)を示すため、これは有意な機能的回復を表す。とりわけ、群間の差は、損傷後の後期の段階において明白であり、このことは、保護効果より再生応答と一致している。
