Abstract
【解決手段】式I:
で示されるジヒドロキシモノマー化合物が開示される。(式中、R1及びR2は、独立に、水素及び18個までの炭素原子を有する直鎖又は分岐アルキル基からなる群から選択され、R3は、−CH=CH−及び(−CH2−)k(kは0と6との間にある)からなる群から選択され、各Zは、独立に、臭素又はヨウ素であり、d及びnは、独立に、0、1又は2であり、Xは、水素であるか、カルボキシ側鎖基である。)このジヒドロキシモノマー化合物から調製されたポリ(アミドカーボネート)及びポリ(エステルアミド)が開示される。

Description
米国特許第5,099,060号は3-(4-ヒドロキシフェニル)プロピオン酸とL-チロシンアルキルエステル(デスアミノチロシル−チロシンアルキルエステル)に基づくジフェノールモノマーを開示している。その後の関連特許にはこの基本モノマー構造の変異体が含まれている。これらのモノマーは多くの応用例において有用であるがいくつかの制限がある:
この要求は本発明によって満たされる。本発明は、新規なクラスの非毒性の脂肪族−芳香族ジヒドロキシモノマー及びそれらから誘導される生体吸収性ポリマーを提供する。このモノマーはα−、β−、γ−ヒドロキシ酸と天然のアミノ酸L-チロシンの誘導体から調製される。
ポリ(ヒドロキシ酸)、例えばPGA及びPLAは、最も成功した合成バイオマテリアルである。しかしながら、その分解生成物の酸性度、物理機械的性質の制限範囲、及び生物学的リガンド、薬剤又は架橋剤に対する化学的接点を提供しない単純な化学構造に問題がある。従って、ヒドロキシ酸を広範な他の成分と共重合して、最適な性質を達成しようと試みた。
本発明のポリマーの分解速度(インビトロ)及びいくつかの基本的性質は、ポリ(D,L−乳酸)及びポリ(DTEアジペート)についての研究との比較において評価した。ポリ(D,L−乳酸)は、非常に水溶性であるモノマー(乳酸)を含有する。多くの応用において、ポリ(D,L−乳酸)はあまりに速く分解する。限定的な水溶性を有する脂肪族−芳香族ジヒドロキシモノマーの利用は、対応するポリマーの分解速度を減少することができる。さらに、先行技術の実質的に水に不溶性のモノマー、デスアミノチロシル−チロシンアルキルエステルと比較して、限定的な水溶性を有する脂肪族−芳香族ジヒドロキシモノマーの利用は対応するポリマーの分解速度を促進し得る。従って、新規な脂肪族−芳香族ジヒドロキシモノマーから得られるポリマーは、ポリ(D,L−乳酸)、及びデスアミノチロシル−チロシンポリカーボネート及びポリアリーレートの分解及び再吸収速度の中間の速度を有する。
物質:L−チロシン、グリコール酸、L−(+)−乳酸、コハク酸、アジピン酸、スベリン酸、セバシン酸、チオニルクロリド、エタノール、ヒドロキシベンゾトリアゾール水和物(HOBt)、ジイソプロピルカルボジイミド(DIPC)、ジメチルアミノピリジン(DMAP)、及びp−トルエンスルホン酸をアルドリッチより購入した。エチル−3−(3−ジメチルアミノ)プロピルカルボジイミド塩酸塩(EDCI・HCl)をJBL Scientificより入手した。ポリ(D,L−乳酸) (Mw=1.0×105ダルトン)をMEDISORBより入手した。全ての溶媒はHPLCグレードのものを受理したまま使用した。
本発明の新規ジヒドロキシ化合物、GATE及びLATEは、脂肪族ヒドロキシ酸とアミノ酸−L−チロシンから作られたモノマーの最初の例である。これらの脂肪族−芳香族ジヒドロキシモノマーを使用して新規な分解性バイオマテリアルを開発した。最初の4つのGATE誘導交互コポリエステルは、ポリマー主鎖のメチレン基の数が異なることを除いて化学構造が似ている。
本発明の新規ポリマーは、組織工学用新規骨格材料及び薬剤放出システムを含む医用用途に有用である。
