Claims (15)
- T細胞レセプター(TCR)が、
(i)少なくとも1つの相補性決定領域中又は少なくとも1つの可変領域フレームワーク領域中で天然型MEL TCRのα鎖可変領域(配列番号1)及び/又はβ鎖可変領域(配列番号2)に対して変異しており、
(ii)3μMより少ないか等しいAAGIGILTV−HLA-A*0201複合体に対するKD及び/又は1×10-3 S-1かそれより遅い解離速度(koff)を有することを特徴とするAAGIGILTV−HLA-A*0201への結合特性を有し且つ少なくとも1つのTCRα鎖可変ドメイン及び/又は少なくとも1つのTCRβ鎖可変ドメインを含有するTCR。 - α鎖可変ドメイン及びTCRβ鎖可変ドメインの両方を含有する請求項1に記載のTCR。
- 配列番号1に示される番号付けを用いてα鎖可変領域アミノ酸28D、29R、30G、31S、49M、51I、53S、54N、72Y、94V、95A、96G、97K、98S及び99Tの1又はそれより多くが変異している請求項1又は2に記載のTCR。
- 配列番号2に示される番号付けを用いてβ鎖可変領域アミノ酸45L、51S、52V、53G、54I、76I、100G、101T、102G、103E、104L及び105Fの1又はそれより多くが変異している請求項1〜3のいずれか1項に記載のTCR。
- 配列番号11〜24又は47〜53のα鎖可変領域アミノ酸配列の1つを含有する請求項1又は2に記載のTCR。
- 配列番号54〜67のβ鎖可変領域アミノ酸配列の1つを含有する請求項1、2及び5のいずれか1項に記載のTCR。
- 配列番号25に示されるα鎖定常領域アミノ酸配列及び/又は配列番号26及び27に示されるβ鎖アミノ酸定常領域配列の一方を更に含有する請求項1〜7のいずれか1項に記載のTCR。
- 二量体T細胞レセプター(dTCR)又は単鎖T細胞レセプター(scTCR)である請求項1〜8のいずれか1項に記載のTCR。
- TCRα鎖可変領域配列に相当する配列がTCRα鎖定常ドメイン細胞外配列に相当する配列のN末端に融合した第1のポリペプチドと、
TCRβ鎖可変領域配列に相当する配列がTCRβ鎖定常ドメイン細胞外配列に相当する配列のN末端に融合した第2のポリペプチドと
を含有し、該第1のポリペプチド及び該第2のポリペプチドが、天然型αβT細胞レセプター中に等価物を有さないジスルフィド結合により連結し、TRAC * 01のエキソン1のThr 48及びTRBC1 * 01若しくはTRBC2 * 01のエキソン1のSer 57、又はこれらの非ヒト等価物から置換されたシステイン残基同士間にあるdTCRである請求項9に記載のTCR。 - 治療剤又は検出可能部分と組み合わされている請求項1〜10のいずれか1項に記載のTCR。
- 前記治療剤が
(i)免疫エフェクター分子、
(ii)サイトカイン若しくは放射性核種、又は
(iii)細胞毒性剤
である請求項11に記載のTCR。 - 請求項1〜12のいずれかに記載の少なくとも2つのTCRを含有する多価TCR複合体。
- 請求項1〜10のいずれか1項に記載のTCRを提示する単離細胞。
- 請求項1〜12のいずれか1項に記載のTCR若しくは請求項13に記載の多価TCR複合体、又は請求項14に記載の複数の細胞を、医薬的に許容されるキャリアと共に含有する医薬組成物。
