Abstract

本発明は、能動免疫療法による癌の治療における治療標的としての、癌細胞及び新生血管系により発現される多数の分子の使用を意図する。このような分子としては、癌細胞それ自体により発現されるか、又は腫瘍の非癌性構成成分、例えば腫瘍関連新生血管系又はその他の間質と関連づけられる抗原である腫瘍関連抗原(TuAA)が挙げられる。TuAA発現プロファイルの確定は、患者の癌状態又は型を、適切な免疫療法薬又はレジメンと一致させる助けとなり得る。特定の実施形態では、腫瘍関連抗原PRAME及びPSMAのエピトープは、このようなプラスミドが癌療法薬として投与される被験体における強力な免疫応答を引き出し得る二価プラスミドを設計するのに用いられる。PRAME及びPSMAの交差反応性類似体も、本発明の実施形態において意図される。

二価免疫応答を誘発することが可能なPSMA及びPRAMEの免疫原性エピトープを含有するプラスミドを設計するに際して本発明の好ましい実施形態を実証するために、以下の実施例を包含する。以下の実施例に開示される方法は、本発明の実行において良好に機能するために本発明者等により発見された方法を表す、と当業者により理解されるべきであり、したがってその実行のための好ましい方式を構成すると考えられ得る。しかしながら、本発明の開示の点から見て、開示されている特定の実施形態において多数の変更が成され得るし、そしてさらに本発明の精神及び範囲を逸脱しない限り、同様の又は類似の結果を得ることができる、と当業者は理解する。
プラスミドP2及びR2(pCTLR2とも呼ばれる)は、それぞれPSMA(広範囲の癌腫の新生血管系上で、又は前立腺癌腫細胞により発現される)及びPRAME(癌性細胞により発現される)からの因子を含有する(図1)。それぞれ挿入物は、抗原の配列の断片を、標的細胞により発現され、そして免疫媒介性攻撃により取組み可能な多コピーのエピトープと共に包含する。これらのエピトープに隣接するのは、細胞性区画中のエピトープペプチドのプロセシング及び生成を促すことが既知であるアミノ酸をコードする配列である。さらに、プラスミドRP5は、P2及びR2の両方からの因子を、互いに接合
された発現免疫原と共に包含する。
同一発現ベクター中での異なる抗原からの因子を操作する戦略がそれらの因子の間に優勢/亜優勢階層を作り出すか否かを査定するために、試験を実行した。
日後に、脾臓細胞をPRAME425-433又はPSMA288-297ペプチドでex vivo刺激して、種々のエフェクター対標的細胞比(E:T比)で、51Cr標識ペプチド被覆T2細胞に対して試験した。
PRAME及びPSMAエピトープ(RP5の状況での優勢及び亜優勢)の両方に対するより均衡の取れた免疫を生じる発現ベクターを設計するために、以下の3つの方法の種々の組合せを用いることにより、一組の免疫原を設計し、同一プラスミド主鎖内に組入れた:
1)PRAME425-433(優勢)エピトープのコピー数及びPSMA288-297(亜優勢)エピトープのコピー数間の比を、後者に有利となるように調整した。
2)プロテアソーム・プロセシングと無関係に適正C末端を有するようにC末端位置に低優勢エピトープを配置した。
3)低優勢エピトープ(PSMA)を変異させ(発現挿入物内に1又は多コピー)、固有の免疫原特性、例えばクラスI MHCとの結合及びそれについての半減期を改善した。
上記の実施例3に記載したように設計されたプラスミドを試験して、PRAME425-433及びPSMA288-297腫瘍関連抗原に対する二価免疫応答を初回刺激するそれらの能力を確定した。
物を保有するプラスミド(RP8、RP9、RP10、RP11、RP12又はRP13)で免疫化した。最終プラスミド注射の7日後、即ち11日目に、免疫化動物全てを、5つのナイーブ対照を含めて、屠殺した。下記のように、IFN−γ産生スポット形成コロニー(SFC)の頻度を測定することにより、ELISPOT分析を実行した。

6つのプラスミド(RP8、RP9、RP10、RP11、RP12及びRP13)のうちの4つの実施例における比較に基づいて、さらなる分析のために、PRAME425-433及びPSMA288-297の両方に対する強固な二価免疫応答を初回刺激する唯一のプラスミドであったため、RP12を選択した。
プラスミドRP12及びRP8による免疫化が、PSMA288-297I297V類似体によるペプチド追加免疫後に、腫瘍関連抗原Prame425-433及びPSMA288-297に対する二価応答を誘導し得たか否かを判定するために、免疫化動物の四量体分析を実行した。
プラスミドRP12及びRP8による免疫化が、PSMA288-297I297V類似体による一次ペプチド追加免疫及びPrame425-433L426Nva、L433Nleペプ

を包含する、癌を有する個体の治療方法。 - 前記個体が、癌細胞上でPRAMEを、且つ腫瘍関連血管系細胞上でPSMAを発現する癌を有する、請求項22に記載の方法。
- 前記個体が、新生物細胞又は腫瘍関連新生血管系細胞においてPRAME、PSMA又はその両方を発現する癌を有する、請求項22に記載の方法。
- 癌を有する個体の治療のための薬剤の調製における、請求項1〜17のいずれか一項に記載の核酸構築物の使用。
- 前記薬剤が、節内に該薬剤を投与することによって癌を有する個体を治療するためのものである、請求項26に記載の使用。
- 前記個体が、新生物細胞又は腫瘍関連新生血管系細胞においてPRAME、PSMA又はその両方を発現する癌を有する、請求項26又は27に記載の使用。
- 癌を有する個体の治療のための薬剤の調製における、請求項18に記載の免疫原性組成物の使用。
- 前記薬剤が、節内に該薬剤を投与することによって癌を有する個体を治療するためのものである、請求項28に記載の使用。
- 前記個体が、新生物細胞又は腫瘍関連新生血管系細胞においてPRAME、PSMA又はその両方を発現する癌を有する、請求項28又は29に記載の使用。
- 腫瘍関連新生血管系の免疫応答標的化を誘導するのに用いるための薬剤の調製における、請求項1〜17のいずれか一項に記載の核酸構築物の使用。
- 前記腫瘍関連血管系細胞がPRAMEを表示する、請求項31に記載の使用。
