Abstract
Description
米国癌学会は、毎年、百万を超える人々が癌にかかり、米国人男性の約2人に1人及び米国人女性の約3人に1人が、生涯の或る時点で何らかの種類の癌を有するであろうと推定している。

〔実施例〕
動物
ヒトT細胞エピトープに対する免疫応答は、免疫の固有のMHC制限のために、元の非臨床モデルで試験され得ないため、ヒトA*0201遺伝子(Pascolo et al., 1997)(これはヒト集団で頻繁に発現される)を発現する遺伝子操作マウスモデルを選択した。免疫欠損マウス(異種移植片モデルのための基礎)に対比して、A*0201トランスジェニックモデル(HHD)は免疫能を有し、したがって能動免疫療法戦略の評価を可能にする。
腫瘍関連抗原メラン−A26-35(配列番号9)及びチロシナーゼ369-377(配列番号10)をコードする二価pSEMプラスミド(非複製組換えDNA)、並びに腫瘍関連抗原SSX−241-49(配列番号1)及びNY−ESO−1157-165(配列番号2)をコードするpBPLの二価のプラスミドを、実施例2〜6におけるTc1(ガンマインターフェロン産生)免疫応答を初回刺激する能力に関して評価した。メラン−A26-35(A27Nva;ENvaAGIGILTV(配列番号11))ペプチド類似体、チロシナーゼ369-377(V377Nva;YMDGTMSQNva(配列番号12))ペプチド類似体、NY−ESO−1157-165(L158Nva、C165V;S(Nva)LMWITQV(配列番号6))ペプチド類似体及びSSX−241-49(A42V;KVSEKIFY(配列番号5))ペプチド類似体を、その後の追加免疫のために用いた。
試験の1、4、15、18、29、32、49及び53日目に、両側の鼠径部リンパ節内注射により、用量調製物を投与した。イソフランの吸入によりマウスを麻酔し、そして無菌条件下で手術を実行した。外科手術の準備後、長さ0.5〜1cmの切開を鼠径部襞に作製して、鼠径部リンパ節を曝露した。0.5mLインスリン注射器を用いて、最大容積25μL(1mg/mL溶液中25μg)のプラスミド又はペプチドを、右及び左の鼠径部リンパ節に直接注射した。滅菌した6.0ナイロン皮膚縫合糸で切り口を閉じた。
両側の鼠径部リンパ節に対してpSEM/pBPL(100μg/日)の混合物を用いて、上記のように、雌HHD動物の3群に免疫付与した。群1(n=10匹のマウス)には、1、4、15、18、28、32、49及び53日目にプラスミド注射を施した;群2及び群3(n=25匹のマウス/群)には、表2(下記)に示すように、それぞれ、1、4、15及び18日目にプラスミド注射を施した。
CD8+抗原特異的T細胞の計数は、クラスI MHC/ペプチド複合体によるT細胞受容体(TCR)の同族(cognate)認識を要する。これは、それぞれ特定のペプチドと結合され、そして蛍光タンパク質と接合される4つのHLA MHCクラスI分子の複合体から成るクラスI MHCテトラマーを用いて成し遂げられ得る。したがってテトラマー検定は、特定のMHC分子中に複合される所定のペプチドに特異的な総T細胞集団の定量を可能にする。フローサイトメトリーは、適切なT細胞受容体を有する細胞と標識化テトラマーとの結合を定量するのに用いる。さらに、結合は機能的経路によらないため、この集団は、機能的状態にかかわらず、すべての特異的CD8+T細胞を包含する。

細胞障害活性を測定する代わりに、ELISPOT検定において特定のエフェクター細胞によるIFN−γ生産を測定することによって、CD8+CTL応答を評価することができる。この検定では、抗原提示細胞(APC)が、マイクロタイターウェルのプラスチック表面上に固定され、エフェクター細胞が、様々なエフェクター(標的の比率)で添加される。抗原特異的エフェクター細胞によるAPCの結合は、エフェクター細胞によるIFN−γを含むサイトカインの生産を引き起こす。細胞は染色されて、細胞内IFN−γの存在、及び顕微鏡下でカウントされる陽性染色病巣(スポット)の数を検出することができる。
ソフトウェアバージョン3.2.3を用いて、AID国際プレート読取器における、展開から24時間後に免疫付与動物におけるCTL応答を測定した。
クロム放出検定は、CTL活性を評価するための既知の検定である。手短にいうと、それらの表面に抗原を発現する標的細胞を、クロム(51Cr)の放射性同位体で標識する。次に患者細胞を標的細胞と混合し、数時間インキュベートする。抗原発現細胞の溶解は、培地中に51Crを放出する。患者エフェクター細胞の存在下又は非存在下で、当該抗原又は対照抗原を発現する標的細胞の溶解を比較することにより、細胞特異的溶解が算定され、通常は、特異的溶解%として表す。

免疫付与マウスにおけるDNA初回刺激及びペプチド追加免疫並びに1回のin vitro刺激(IVS)後の51Cr細胞傷害活性検定を用いたPRAME425-433(配列番号3)、PSMA288-297(配列番号4)、NY−ESO−1157-165(配列番号2)及びSSX−241-49(配列番号1)に対するCTL応答を査定した。ペプチド免疫付与レジメン完了の22日後に免疫付与マウス(N=6)から採取した脾臓細胞のex vivo
刺激により、CTLを発生させた。

Claims (48)
- 1つ又は複数の核酸組成物及び1つ又は複数のペプチド組成物を含む複数の組成物を含む免疫原性生成物であって、前記1つ又は複数の核酸組成物が、SSX−1エピトープ、NY−ESO−1エピトープ、PRAMEエピトープ、PSMAエピトープ、チロシナーゼエピトープ及びメラン−Aエピトープからなる群から選択される、1つ又は複数のクラスI MHC拘束エピトープ又はその類似体を発現することができ;前記1つ又は複数のペプチド組成物が、SSX−1エピトープ、NY−ESO−1エピトープ、PRAMEエピトープ、PSMAエピトープ、チロシナーゼエピトープ及びメラン−Aエピトープからなる群から選択される前記1つ又は複数のクラスI MHC拘束エピトープ又はその類似体から本質的になり;そして前記1つ又は複数のペプチドが選択された前記核酸により発現される前記エピトープに対応する免疫原性生成物。
- 前記1つ又は複数の核酸組成物がpSEM、pBPL及びpRP12からなる群から選択されるプラスミドを含む、請求項1に記載の免疫原性生成物。
- 前記1つ又は複数のペプチド組成物が、SSX−241-49(配列番号1)、その類似体KVSEKIFYV(配列番号5);NY−ESO−1157-I65(配列番号2)、その類似体SNvaLMWITQV(配列番号6);PRAME425-433(配列番号3)、その類似体S(Nva)LQHLIG(Nle)(配列番号7);PSMA288-297(配列番号4)、その類似体GLPSIPVHPV(配列番号8);メランA26-35(配列番号9)、その類似体ENvaAGIGILTV(配列番号11);チロシナーゼ369-377(配列番号10)、及びその類似体YMDGTMSQNva(配列番号12)からなる群から選択されるペプチドを含む、請求項1に記載の免疫原性生成物。
- 前記複数の組成物が以下の:

を含む、請求項1に記載の免疫原性生成物。 - 前記a)及びb)の核酸分子が同一組成物の一部である、請求項4に記載の生成物。
- 前記核酸分子が同一物である、請求項5に記載の生成物。
- 前記核酸分子がpBPLの遊離配列をコードする配列(配列番号13)を含む、請求項6に記載の生成物。
- 前記核酸がpBPLの免疫原性ポリペプチドをコードする配列(配列番号16)を含む、請求項7に記載の生成物。
- 前記核酸分子がpBPL(配列番号20)である、請求項8に記載の生成物。

- 請求項1に記載の生成物をそれを必要とする患者に投与することを含む、癌の治療方法。
- 前記癌が乳癌、卵巣癌、膵臓癌、前立腺癌、結腸癌、膀胱癌、肺癌、肝臓癌、胃癌、精巣癌、子宮癌、脳腫瘍、リンパ癌、皮膚癌、骨肉腫、腎臓癌、直腸癌、黒色腫、膠芽細胞腫又は肉腫である、請求項23に記載の方法。
- 複数の組成物をそれを必要とする患者に投与する工程を含む、癌腫の治療方法であって、該複数の組成物が:
i)SSX−2クラスI MHC拘束エピトープ又はその類似体を発現することができる核酸分子;
ii)NY−ESO−1クラスI MHC拘束エピトープ又はその類似体を発現する
ことができる核酸分子;及び
iii)PRAMEクラスI MHC拘束エピトープ又はその類似体を発現することができる核酸分子;
vi)PSMAクラスI MHC拘束エピトープ又はその類似体を発現することができる核酸分子
を含む、癌腫の治療方法。 - 本質的にSSX−2、NY−ESO−1、PRAME及びPSMAからなるエピトープ又は類似体の群から選択される1つ又は複数のペプチドを投与する工程をさらに含む、請求項25に記載の方法。
- 前記複数の組成物が以下の:
i)メラン−AクラスI MHC拘束エピトープ又はその類似体を発現することができる核酸分子;

- 複数の組成物をそれを必要とする患者に投与する工程を含む、癌の治療方法であって、該複数の組成物が:

を含む方法。 - 前記癌が膠芽細胞腫である、請求項38に記載の方法。
- 前記癌が黒色腫である、請求項38に記載の方法。
- i)SSX−2クラスI MHC拘束エピトープ又はその類似体を発現することができる核酸分子;
ii)NY−ESO−1クラスI MHC拘束エピトープ又はその類似体を発現することができる核酸分子;
iii)本質的に前記NY−ESO−1エピトープ又はその類似体からなるペプチド;及び
vi)本質的に前記SSX−2エピトープ又はその類似体からなるペプチド
を含む組成物をそれを必要とする患者に投与する工程をさらに含む、請求項38に記載の方法。 - 前記複数の組成物が以下の:
