Abstract
Description
本発明は、α−イミノエステルへの末端アルキンの付加の方法に関する。
エナンチオマーα−アミノ酸、特に、非タンパク質由来アミノ酸はタンパク質工学およびペプチドベース薬剤発見の重要な手段として格別であり、急速に人気が増加している。精力的な研究はエナンチオマー的に富化された人工α−アミノ酸の製造に集中してきた。いくつかの手法、例えば、エンアミドの生物分解ルートならびにロジウム触媒不斉水素化が非常に有望と示されてきた。しかし、未だ人工アミノ酸誘導体の異なる型の便利な合成のための効率よく、技術的に可能な方法の必要性がある。
本発明は、式
本発明は、α−イミノエステルの触媒不斉アルキニル化のための方法に関する。
R1は所望により置換されていてもよいアルキル、シクロアルキル、アリールまたはヘテロアリールであり;そして
Yは水素または窒素保護基である〕を有する化合物を意味する。
本明細書で使用される“アルコキシ”なる用語はアルキル−O−を意味する。
本明細書で使用される“シクロアルキル”なる用語は1個またはそれ以上の置換基、例えば、アルキルまたはアルコキシにより置換されていてもよい3から6個の炭素原子の所望により置換されていてもよい単環式脂肪族炭化水素基を意味する。
化合物Iのそれぞれのアルキニル化反応(下記)は下記の一般法にしたがって実施される:
すべての反応は窒素雰囲気下で実施される。すべての化学物質および溶媒、例えば、有機溶媒は特に記載のない限りさらなる精製なしに受け取ったまま使用する。有機溶媒であるCH2Cl2はCaH2から蒸留する。化合物Iは当業者に既知の方法にしたがって合成される。例えば、Andrew Taggi et al., “A-imino esters: versatile substrates for the catalytic, asymmetric synthesis of α and β-amino acids and β-lactams,” 36 ACC. CHEM. RES. 10 -19 (2003)参照。

以下に実施例3の表3の生成物、すなわち、3a−3fの分析条件を記載する。
本発明の方法の生成物、すなわち不斉アルキニル化α−イミノエステルは光学的に活性な人工α−アミノ酸誘導体を合成するために使用され得る。この例は下記スキーム1に示され、これは成長ホルモン生成物で使用される薬学的に興味のあるペプチドの重要な中間体であるビスホモフェニルアラニン誘導体またはスキーム1の(R)−12を得るための実施例3の生成物3bの修飾である。
