C4及びAリンカーペプチドを含む発現ベクターの構築及び使用
1.細菌菌株及びプラスミド
大腸菌菌株DH5α−T1は、Invitrogen(USA)から市販されている。大腸菌菌株B121(DE3)及びNovaBlueは、Novagen(USA)から市販されている。pET29a(+)細菌発現ベクターは、Novagen(USA)から得た。121アミノ酸残基イソ型のヒトVEGFをコードするDNA配列を含むpLen−121プラスミドは、その全容が参照として本明細書に組み込まれる米国特許第5,219,739号中に記載されており、Dr.J.Abraham(Scios Nova、Inc.、USA)から得た。ヒトアネキシンVをコードするDNA配列を含むpPAP1−1.6プラスミドは、Dr.J.Tait(University of Washington School of Medicine、シアトル、WA)から得た。炭そ菌致死性因子(LFn)の非毒性N末端断片をコードするDNA配列を含むpGEX−KG LF254プラスミドは、Dr.S.Leppla(National Institute of Allergy and Infectious Diseases、NIH、ベセスダ、MD)から得た。
本明細書で使用する制限及び修飾酵素は米国内で市販されており、製造者の教示書に従い使用した。コンピテント細胞の調製、形質転換、及び細菌培地はSambrookら(J.Sambrook、E.F.Fritsch及びT.Maniatis.(1989)「分子クローニング(Molecular Cloning)」:「研究室マニュアル(A Laboratory Manual)」、Cold Spring Harbor Laboratory Press、Cold Spring Harbor、NY)に従ったか、又は製造者の教示書に従った。プラスミドの精製は、製造者の教示書に従いWizard Plus SV Minipreps又はMaxipreps DNA精製システム(Promega、USA)を使用して行った。アガロースゲルからのDNA断片の精製は、製造者の教示書に従いGeneclean Spinキット(Bio101、USA)を使用して行った。
(G4S)3リンカーによってタンパク質のORFと融合したN末端Hu−タグを有する組換えタンパク質の発現用に、pET/Hu(G4S)3ベクターを記載されたように構築した(Backerら、2004)。Gene−Tailor部位特異的突然変異誘発キット(Invitrogen)を使用する部位特異的突然変異誘発によって、pET/Hu(G4S)3ベクターのHu−タグ中にR4Cアミノ酸置換を導入した。プライマー(配列番号23及び配列番号24)は、R4C突然変異を導入するために使用した。生成したベクターはシークエンシングによって確認し、pET/Hu−C4(G4S)3と明示した。
pET/Hu−C4(G4S)3及びpET/Hu−C4−G4SベクターのDNAを、NcoIを用いて別々に切断し、脱リン酸化した。前述のベクターにおいてクローニングする全DNA断片を、増幅DNA断片の両端にNcoI部位を導入したプライマーを使用してPCRによって増幅させた。121−aaイソ型のヒトVEGFをコードするDNAは、プライマー(配列番号27及び配列番号28)を使用してpLen−121プラスミドDNAから増幅させた。炭そ菌致死性因子(LFn)の1〜254−aa長のN末端断片をコードするDNAは、プライマー(配列番号29及び配列番号30)を使用してpGEX−KG LF254プラスミドDNAから増幅させた。ヒトアネキシンVをコードするDNAは、プライマー(配列番号31及び配列番号32)を使用してpPAP1−1.6プラスミドから増幅させた。それぞれの増幅させたDNA断片はNcoIを用いて切断し、精製し、ベクターpET/Hu−C4(G4S)3及びpET/Hu−C4−G4SのNcoI部位にクローニングした。正しい方向のクローニングORFを有するクローンを、T7プロモーター系センスプライマーを使用するPCRによって選択し、シークエンシングによって確認した。
全てのタンパク質が、LB培地(Q−Biogen、カールズバッド、CA)中で増殖させたBL21(DE3)大腸菌において発現した。600nmにおいて0.5〜0.7単位の光学濃度で、1mMのIPTGによって発現を誘導した。誘導後、300rpmで振盪させながら37℃で2.5〜3時間、培養物を増殖させ、次いで遠心分離によって採取し、EmulsiFlex−C5(Avestin、オタワ、カナダ)を使用して破壊した。
6.1.ウシRNaseAの1〜29aa断片及びヒトRNaseIの30〜127aa断片を含む、キメラ1−29B/30−127H−RNase(BH−RNase)を、記載されたのと同様に(Gaynutdinovら、2003)構築し、発現させ精製した。Gene−Tailor部位特異的突然変異誘発キット(Invitrogen)及びプライマー(配列番号37及び配列番号38)を使用する部位特異的突然変異誘発によって、V118C突然変異をpET29/1−29B/30−127H−RNaseプラスミドDNA中に導入した。VC118の置換はシークエンシングによって確認し、このタンパク質はBH−RNase(C118)と明示した。BH−RNase(C118)は、野生型BH−RNaseに関して記載されたのと同様に(Gaynutdinovら、2003)発現させ精製した。反応性チオール基の存在は、チオール試薬N−(1−ピレン)−マレイミド(Molecular Probes、Eugene、OR)との反応、次に記載されたのと同様のRP HPLC分析(Backerら、2002)によって試験した。HuS(C118)アダプタータンパク質は、BH−RNaseに関して記載されたのと同様に(Gaynutdinovら、2003)、サブチリジン(Sigma)を用いたBH−RNase(C118)の制限酵素による消化によって得た。
プライマー(配列番号39及び配列番号40)を使用する部位特異的突然変異誘発によって、pET29/1−29B/30−127H−RNase(V118C)プラスミドDNA中にN88C突然変異を導入した。N88Cの置換はシークエンシングによって確認し、このタンパク質はBH−RNase(C88、C118)と明示した。BH−RNase(C88、C118)は、BH−RNase(C118)に関して前に記載したのと同様に発現させ、精製し、サブチリジンによって消化してHuS(C88、C118)アダプタータンパク質を得た。
Q11P及びF8A突然変異を、それぞれプライマー配列番号41〜44を使用する部位特異的突然変異誘発によって、pET29/1−29B/30−127H−RNase(V118C)プラスミドDNA中に必然的に導入した。両方の置換をシークエンシングによって確認し、このタンパク質はBH−RNase(A8、P11、C118)と明示した。BH−RNase(A8、P11、C118)は、BH−RNase(C118)に関して前に記載したのと同様に発現させ、精製した。
7.1.SLT−VEGF。
N末端S−タグ及びHis−タグを有するSLT−VEGF融合タンパク質をコードするpET/VEGF121−SLT/Lプラスミドが以前に記載された(Backer及びBacker、2001)。SLT−VEGF DNAコード配列は、NdeI部位を導入したプライマー(配列番号45)及びXhoI部位を導入したプライマー(配列番号46)を使用して、pET/VEGF121−SLT/LプラスミドDNAからPCRによって増幅させた。精製したPCR産物はNdeI及びXhoI制限酵素を用いて切断し、精製し、pET29a(+)ベクター(Novagen)のNdeI−XhoI部位にクローニングした。生成したプラスミドはシークエンシングによって確認し、pET29/SLT−VEGFと明示した。C4含有タンパク質に関して前に記載したのと同様に、SLT−VEGFをBL21(DE3)中で発現させ、以下のように精製した:封入体を8Mの尿素、20mMのトリス−HCl pH8.0、150mMのNaCl、80mMのNa2SO3、10mMのNa2S4O6、10mMのDTT中に溶かし、攪拌しながら4℃で6〜8時間インキュベートし、次いで5mMのトリス[2カルボキシエチル]ホスフィン(Pierce)を補い、4℃で16〜18時間インキュベートした。溶解したタンパク質は、最初に、20mMのトリス−HCl pH8.0、2Mの尿素、0.5Mのアルギニン、1mMの還元型グルタチオン、0.4mMの酸化型グルタチオンの10体積中において4℃で8〜10時間、第2に、20mMのトリス−HCl pH8.0、150mMのNaCl、0.01%のNP−40の100体積中において4℃で24時間、及び第3に、20mMのトリス−HCl pH8.0、20mMのNaClの100体積中において4℃で24時間の3ステップの透析によってリフォールディングさせた。透析後、溶液を遠心分離(15,000×g、20分間)によって浄化し、HiTrap Qセファロース(1ml、予め充填したカラム、Amersham)上でのクロマトグラフィーによってSLT−VEGFを精製した。
N末端野生型Hu−タグを有するHu−VEGF121融合タンパク質を、Backerら、2003中に記載されたのと同様に構築した。Hu−VEGF121を発現させ、以下の修飾でC4−VEGFに関して前に記載したのと同様に、封入体からリフォールディングさせた:封入体からのリフォールディングは2ステップの透析によって行った。最初に、20mMのトリス−HCl pH8.0、2Mの尿素、0.5Mのアルギニン、1mMの還元型グルタチオン、0.4mMの酸化型グルタチオンの10体積中において4℃で8〜10時間、第2に、20mMのトリス−HCl pH8.0の50体積中において4℃で24時間。さらに、最終的なHu−VEGF121の精製のために、HiTrap SPセファロースFF、次にヘパリンHPセファロースクロマトグラフィー(1ml、予め充填したカラム、Amersham)を実施した。
A.VEGFの活性。VEGF及びVEGF系複合体の機能活性を2つのアッセイにおいて試験した。第1のアッセイ、VEGFR−2自己リン酸化の刺激は以下のように実施した:一晩の飢餓状態(DMEM/0.5%FBS)の後にほぼ融合状態の293/KDR細胞を、37℃において20分間0.5mMのバナジン酸ナトリウムを含む無血清DMEMに移し、次いで37℃において10分間VEGFで刺激し、溶解させ、抗ホスホチロシンRC20:HRPO複合体(BD Transduction Labs、USA)を使用してウエスタンブロッティングによって分析した。第2のアッセイ、SLT−VEGFの細胞毒性効果からの293/KDR細胞の保護は以下のように実施した。293/KDR細胞は、1000個の細胞/ウエルで、実験の20時間前に96ウエルプレート上に平板培養した。様々な量のVEGF又はVEGF系複合体を完全培養培地中でSLT−VEGFと混合させ、1nMのSLT−VEGFの最終濃度で三連のウエルにおいて細胞に加えた。生存細胞数を、CellTiter96(登録商標)AQueous One Solution細胞増殖アッセイキット(Promega)によって96時間後に定量化した。
HEK293ヒト形質転換胚腎細胞(CRL−1573)及びRAW264.7マウス単核(TIB−71)は、American Type Culture Collection(ATCC、Rockville、MD)からのものであった。2.5×106個のVEGFR−2/細胞を発現する293/KDR細胞は、SibTech、Inc.(Newington、CT;Backer及びBacker、2001a)において開発されてきた。全ての細胞は、10%のウシ胎児血清(Gemini、USA)、2mMのグルタミン(Life Technologies、USA)、及びペニシリン−ストレプトマイシン(Life Technologies、USA)を補ったDMEM(Life Technologies、USA)中に37℃、5%CO2で保った。
C4タグタンパク質とアダプタータンパク質HuS(C118)の部位特異的結合。
このプロトコルは、C4タグタンパク質とアダプタータンパク質HuS(C118)の部位特異的結合、及び組織培養中の複合体の機能活性の試験を含んでいた。
C4−VEGF及びC4−scVEGFと放射性核種キレート化剤、5−マレイミド−2−ヒドラジニウムピリジン塩酸塩の部位特異的結合。
このプロトコルは、C4−VEGF及びC4−scVEGFと化学的に活性がある放射性核種キレート化剤5−マレイミド−2−ヒドラジニウムピリジン塩酸塩(Solulink、サンディエゴ、CA)の部位特異的結合、及び組織培養中の複合体の試験を含んでいた。
C4−VEGFとポリエチレングリコールの部位特異的結合。
このプロトコルは、C4−VEGF110と化学的に活性があるマレイミド基で官能性を与えたポリエチレングリコール(Nektar Therapeutics)の部位特異的結合、及び組織培養中の複合体の試験を含んでいた。
シアニン色素Cy5.5とHuS−C4−VEGF複合体の部位特異的結合。
このプロトコルはHuS(C88、C118)の調製、C4−VEGFとのその結合、生成したHus−C4−VEGF複合体の精製、及びCy5.5−Hus−C4−VEGFという名称の複合体を生成する前記複合体とシアニン色素Cy5.5の結合を含んでいた(図6A)。比較目的で、C4−VEGFをアミノ基においてNHS−Cy5.5で1:1の比にランダムに修飾し、又はC4タグ中のC4残基においてマレイミド−Cy5.5で修飾した。全てのCy5.5含有複合体中のVEGF部分の機能活性を、前に記載した2つの組織培養アッセイ、VEGFR−2チロシン自己リン酸化の誘導(図6B)、及びSLT−VEGFの細胞毒性活性からの293/KDR細胞の保護(図6C)において試験した。両方のアッセイにおいて、Cy5.5−Hus−C4−VEGF複合体の活性のみが非修飾C4−VEGFの活性に匹敵し、タンパク質の誘導体化のためにC4タグと結合することができるアダプタータンパク質の使用の有効性が根底にあった。
標的型薬剤送達用の薬剤充填リポソームを含むVEGF誘導型複合体を構築するための、C4タグ及びこのタグと共有結合したアダプタータンパク質の使用。
このプロトコルはHuS(C118)の調製、PEG−脂質−マレイミドとのその結合、Lip/HuS(C118)という名称の標準的な構築体をもたらすドキソルビシン充填リポソーム中への脂質化HuS(C118)の挿入、及びLip/Hus−C4−VEGFをもたらしたこの構築体とC4−VEGFの結合を含んでいた(図7A)。Lip/Hus−C4−VEGF複合体中のVEGF部分の機能活性を、前に記載した組織培養アッセイ、293/KDR細胞におけるVEGFR−2チロシンリン酸化の誘導において試験した(図7B)。このアッセイにおいて、Lip/Hus−C4−VEGFリポソームのVEGFの活性は非修飾C4−VEGFの活性に匹敵し、タンパク質の誘導体化のためにC4タグと結合することができるアダプタータンパク質の使用の有効性が根底にあった。
標的型薬剤送達用の薬剤充填リポソームを含むScvegf誘導型複合体を構築するためのC4タグの使用。
このプロトコルはC4−scVEGFの調製、PEG−脂質−マレイミドとのその結合、及びドキソルビシン充填リポソーム(「DOXIL」)中への脂質化C4−scVEGFの挿入を含んでいた(図8A)。Lip/C4−scVEGF複合体中のVEGF部分の機能活性を、前に記載した組織培養アッセイ、293/KDR細胞におけるVEGFR−2チロシンリン酸化の誘導において試験した(図8B)。このアッセイにおいて、Lip/C4−scVEGFリポソームのVEGFの活性は非修飾C4−VEGFの活性に匹敵し、C4−scVEGF使用の有効性が根底にあった。
