Abstract
Description
この実験は、血漿アルブミンおよびIgG含量を低下させ、それによって問題となっているその他のサンプルタンパク質の質量分光分析を強化するための手段として、磁性粒子ベースのアフィニティークロマトグラフィーを評価するために行った。この手順の概要を、以下の表Iにまとめる。
Claims (19)
- a)鉄、コバルト、およびニッケルからなる群から選択された金属;酸化鉄、硫化鉄、塩化鉄、水酸化第二鉄、および酸化第一第二鉄からなる群から選択された金属化合物;および有機金属化合物を含む少なくとも1つの常磁性粒子を、1種または複数の目標化合物を含むサンプルに添加するステップであって、少なくとも1つの常磁性粒子が1つまたは複数の目標化合物を有し、少なくとも1つの常磁性粒子が、サンプル中の目標化合物の1種または複数に対してアフィニティーを有する1つまたは複数のリガンドまたは官能基を有するものであるステップと、
b)1つまたは複数のリガンドまたは官能基を、1種または複数の目標化合物に接触させて、錯体を形成するステップと、
c)磁場を印加することによって錯体を固定化するステップと、
d)磁場によって固定化されていないサンプルの部分を除去するステップと、
e)磁場を除去して前記錯体を解放するステップと、
f)前記錯体から目標化合物を溶出するステップと、
g)常磁性粒子を固定化するステップと、
h)目標化合物を回収するステップと
を含むことを特徴とするサンプルから目標化合物を単離するための方法。 - 1つまたは複数のリガンドまたは官能基は、少なくとも1つの常磁性粒子と共有結合することを特徴とする請求項1に記載の方法。
- 1つまたは複数のリガンドまたは官能基は、抗体、抗原、ハプテン、受容体、酵素、ポリペプチド、タンパク質、およびポリヌクレオチドからなる群から選択された目標化合物に特異的であることを特徴とする請求項1に記載の方法。
- 1つまたは複数のリガンドまたは官能基は、生物学的結合によって結合されることを特徴とする請求項1に記載の方法。
- 生物学的結合は、ストレプトアビジン−ビオチン、アビジン−ビオチン、炭水化物−レクチン、および酵素−酵素阻害剤からなる群から選択されることを特徴とする請求項4に記載の方法。
- a)少なくとも1つの常磁性粒子を、1種または複数の目標化合物を含むサンプルに添加するステップであって、少なくとも1つの常磁性粒子が、そこに結合している1つまたは複数のリガンドまたは官能基を有し、1つまたは複数のリガンドまたは官能基が、前記サンプル中の問題になっていない目標化合物の1種または複数に対してアフィニティーを有するものであるステップと、
b)リガンドまたは官能基を、問題となっていない化合物に接触させて、錯体を形成するステップと、
c)磁場を印加することによって錯体を固定化するステップと、
d)磁場によって固定化されていないサンプルの部分を除去するステップであって、このように除去された前記サンプルが、1種または複数の目標化合物を含有するものであるステップと
を含むことを特徴とする1種または複数の目標化合物および問題となっていない化合物を含むサンプルから目標化合物を単離するための方法。 - 1つまたは複数のリガンドまたは官能基は、常磁性粒子と共有結合することを特徴とする請求項6に記載の方法。
- 前記1つまたは複数のリガンドまたは官能基は、抗体、抗原、ハプテン、受容体、酵素、ポリペプチド、タンパク質、およびポリヌクレオチドからなる群から選択された化合物に特異的であることを特徴とする請求項6に記載の方法。
- 少なくとも1つの常磁性粒子は、鉄、コバルト、およびニッケルからなる群から選択された金属であることを特徴とする請求項6に記載の方法。
- 少なくとも1つの常磁性粒子は、酸化鉄、硫化鉄、塩化鉄、水酸化第二鉄、および酸化第一第二鉄からなる群から選択された鉄化合物であることを特徴とする請求項6に記載の方法。
- 1つまたは複数のリガンドまたは官能基は、生物学的結合により結合されることを特徴とする請求項6に記載の方法。
- 生物学的結合は、ストレプトアビジン−ビオチン、アビジン−ビオチン、炭水化物−レクチン、および酵素−酵素阻害剤からなる群から選択されることを特徴とする請求項11に記載の方法。
- a)少なくとも1つの常磁性粒子を、1種または複数の目標化合物を含むサンプルに添加するステップであって、少なくとも1つの常磁性粒子および1種または複数の目標化合物は、電荷の差を有するものであるステップと、
b)1種または複数の目標化合物と少なくとも1つの常磁性粒子との間に錯体を生成するステップと、
c)磁場を印加することによって錯体を固定化するステップと、
d)サンプルから、磁場によって固定化された錯体を分離するステップと、
e)磁場を除去することによって錯体を解放するステップと、
f)前記目標化合物を錯体から溶出するステップと、
g)前記常磁性粒子を固定化するステップと、
h)前記目標化合物を回収するステップと
を含むことを特徴とするサンプルから化合物を単離するための方法。 - 少なくとも1つの常磁性粒子は、そこに結合している1つまたは複数の荷電官能基を有し、それによって少なくとも1つの常磁性粒子に全電荷がもたらされるようになされていることを特徴とする請求項13に記載の方法。
- 遠心分離し、濾過し、アフィニティークロマトグラフィーを介して精製し、錯体を分解した後に磁場を印加するステップをさらに含むことを特徴とする請求項13に記載の方法。
- サンプルは、サンプルのpHを変化させることによって改変することを特徴とする請求項13に記載の方法。
- サンプルは、イオン強度を変化させることによって改変することを特徴とする請求項13に記載の方法。
- 少なくとも1つの常磁性粒子は、鉄、コバルト、およびニッケルからなる群から選択された金属であることを特徴とする請求項14に記載の方法。
- 少なくとも1つの常磁性粒子は、酸化鉄、硫化鉄、塩化鉄、水酸化第二鉄、および酸化第一第二鉄からなる群から選択された鉄化合物であることを特徴とする請求項13に記載の方法。
