Abstract
【解決手段】 前記ポリペプチドは膜結合タンパク質として細胞の表面に圧搾され、膜糖タンパク質の一点または数点において解糖化される。膜糖タンパク質は腫瘍性の細胞で、特に抗原としてヒト単クローン抗体PM−2に結合して、特に「ガン」細胞の枯死に寄与する。

Description
「アポトーシス(細胞の枯死)」
アポトーシスとは細胞の自己死、すなわちDNAの細分による細胞の自殺のことを言うのであって、細胞の細分と膜小胞の形成、いわゆる細胞枯死体の形成を伴う。最も多くの場合には真核細胞の死を招ねき、胚形成、変態、そして組織萎縮を発生する。細胞の枯死は、細胞死の生理学的な形式として、壊死(ネクローシス)の場合のように、炎症作用あるいは細胞の損傷などを初期に生ずることなく、必要としない細胞を極めて迅速に、きれいに除くものである。異常なコンディションの場合には、細胞の枯死は、また、「ガン」前駆細胞などの悪性の細胞を除去する働きをする。細胞の枯死は、腫瘍壊死因子は細胞毒性T−リンパ球や腫瘍壊死因子、グリココルチコルド及び抗体などのサイトカインによって広く阻害される。
膜グリコシレーションは、その細胞外の場所に糖が残留する(グリコカリックス)で、これはアスパラギン側鎖(N−結合)のアミドニトロゲンか、あるいはセリンの酸素原子か、またはトレオニン側鎖(O−結合)の酸素原子のいずれかに結合されている。側鎖にじかに関連している糖は通常、N−アセチルグルコミン酸か、あるいはN−アセチルガラクトサミンである。炭水化物は極めて多様な構造を形成することができる。まず第一に、数多くの単糖類は1種もしくは数種のOH類を介して互に連鎖されることができる。第二として、C−1原子にとりついた連鎖はαまたはβ配置になることができる。これらの数多の結合を利用して、グリコメンブラン・タンパク質が少糖類(オリゴ糖類)を含有する拡大した領域を占めることができる。
新生物つまり腫瘍は異常な細胞集団であって(増殖因子が独立し)、同調しておらず、これという特別な目的のあるものでもなく、進行性のものである。腫瘍は2種類の成分から成っている。腫瘍性の細胞として知られていて、非腫瘍状支質、すなわち、連結組織と血管も、実質細胞である。この発明による抗原の腫瘍性の細胞も細胞を表わすものである。そして、これは抑制されない細胞分裂あるいは細胞枯死の機序をもっていない無制御の細胞である。この発明において、腫瘍性の細胞とは、障害を具備すると共に、正常な細胞周期とは掛け離れた細胞周期を備えていることを特徴とするものである。

細胞培養:
「ガン」細胞株BXPC−3(ATCC番号CRL1687)を受容体を得るために用いた。比較研究、たとえば、ウエスタンブロット分析のために、良く知られている胃腺腫株23132/87(DSMZ受入れ番号DSM201)[ヘンセル(Hensel)氏外、1999,Int.J.Cancer 81:229−235)を用いた。これらの細胞は10%FCSを補充して、RPMI−1640(PAA,ウィーン,オーストリア)の80%と、ペニシリン/ストレプトマイシン(両者とも1%)を合流して培養された。前述した研究によって、細胞がトリプシン/EDTAで除かれ、リン酸塩緩衝生理的食塩水(PBS)で2度洗浄された。
腫瘍細胞から膜タンパク質を分離するのにヘンセル氏外(Hensel et al., 1999,Int.J.Cancer 81:229−235)に記載されている方法で細胞株BXPC−3を使用して行った。これを簡単に述べると、可干渉性の腫瘍細胞をPBSを用いて2度洗浄し、細胞剥離器で除外し、遠心分離をして、低浸透緩衝液(20mM HEPES,3mM KCl,3mM MgCl2)に懸濁した。15分後に氷と超
。上澄みをスイングロータにて、100,000gで30分間遠心分離し、それによって膜をペレット状にした。ペレットを低浸透的緩衝液(50mM HEPES pH7.4,0.1mM EDTA,10%グリセロールと1%トリトンX−100)に再び懸濁させた。タンパク分解酵素阻害薬(ボエルンゲル,マンハイム,ドイツ)をすべての溶液に添加した。
抗原の精製には薬剤(フライブルク,ドイツ)FPLCユニットを使用するカラムクロマトグラフィーで行なわれた。サイズに無関係なクロマトグラフィーについては、ファーマジア・スーパーデックス(Pharmazia Superdex200)(XK16/60)カラムに膜調合前5mgを加えて、緩衝剤A(100mM tris/Cl,pH7.5,2mM EDTA,40mM NaCl,1%トリトンX−100)で効果をあらわした。次で、溶出液をウエスタンブロット分析によって抗体PM−2に反応させて、分割させ調べた。陽性の留分を緩衝剤Aを用いてモノQ(5/5列)に加えた。結合したタンパク質を緩衝剤B(100mM, tris/Cl, pH7.5,1M NaCl,2mM EDTA,1M NaCl, 1% トリトンX−100)を用いてライナーグラディエントの手段で洗浄し、クーマシー染色SDS−PAGEとウエスタンブロット分析で分割して調べた。
10%SDS−PAGEゲルとタンパク質のウエスタンブロッティングによる分離を、この発明に関してではなく、ほかの場合にヘンス氏外(Hense et al., 1999,Int.J.Cancer 81:229−235)による記載のような標準的な状態で遂行した。簡単に述べると、ブロッテッドされたニトロセルロース膜は、2%脱脂粉乳を含有するPBSで遮断され、次で10pg/ml精製抗体103/51で1時間の培養がなされた。PBS+0.05%トゥイーン20で3回洗液した後に、第二の抗体(ペルオキシダーゼ連結ハーレ抗ヒトIgM抗体(ディアノーバ,ハンブルク,ドイツ)が培養された。その反応はスーパーシグナル化学発光装置によってピアース(KMF,セント,オースチン,ドイツ)から表示された。
SDSゲルから分離されているタンパク結合は凡そ1mm×1mmの小片に切断される。そのゲル片はDTTで洗浄され、還元され、ヨードアセトアミドによってS−アルキレートされ、各種の文献に記載されている[シエブチエンコ(Shevchenko)氏外、1996b Anal.Chem. 68:850−858)ようにトリプシンで処理される。その溶液の0.3μlを37℃の温度で、3時間の消化後に、下流摘出手段を備えているブラッカーリフレックスMALDI−TOFの手段で質量分光分析(MALIDI)を行った(ブラッカー,フランツェン,ブレーメン,ドイツ)。薄層塗抹標本テクニックを標本の作成に利用した[ジエンセン氏外,1996ラピッド,コムン マス,スペクトロム(Jensen et al., 1996 Rapid Commun.Mass Spectrom)10:1371−1378]。トリプシン消化ペプチド質量をハウスで発生されたペプチド探索プログラムによって少数タンパク配列データを探索するのに使用した。
RNA分離、cDNA合成とPCR(ポリメラーゼ鎖反応)とは、他に記載[ヘンセル氏外,(Hensel et at.,1999 Int.J.Cancer 81:229−235)]に記載されている通りに行なわれる。それを簡略に述べると、p80(受入れ番号AJ131720号)のヌクレオチド配列の位置181から位置681に至るPCRによって、断片の拡大のために、次のプライマーを用いた。すなわち、
P80−Rev3’5’ CTGTTCCATACGATTTTCATGC 3’
P80−Rev5’5’ TCGAACTGGTCTATCATCCAA 3’
a)細胞株BXPC−3が対照ベクトル(p−HOOK−2)を貫通し、その他の部分 はp80アンチセンス・ベクターを通した。
b)導入後48時間、細胞のサイトスピン標本を調製した。
c)サイトスピン調製物をPM−2抗体と対照抗体(専ら二次性抗体のみ)とによって 染色した。
d)p80アンチセンス仲介物を以て貫通されている細胞は抗体PM−2の結合によっ て、明確な還元を示している。
使用した細胞はトリプシン/EDTAによりその培養びんの基質から解離され、次でRPMI−1640媒質+10%FCSを4℃において1時間かけて膜タンパク質を再生するために培養された。次で、サイトスピンが細胞で作られた。サイトスピンはRTにて夜通し乾燥された。乾燥後に、細胞は100%アセトンで10分間固定され、PBSで3度洗浄された。それから、固定された細胞は5mU/ml N−グリコサイダーゼ(100μlリン酸塩緩衝液、pH7.0)にて恒温器内で37℃の温度で3時間消化された。次で、サイトスピン調合製剤はPBSで3回に亘って洗浄され、免疫組織化学染色を異なる抗体について行なわせた。負の調節として、単にリン酸塩緩衝で培養されただけのサイトスピンが負の調節として用いられ、また、グリコシダーゼ処理をすることなく通常の免疫組織化学の染色が行なわれた。最終の染色は、その後に、顕微鏡で判定され、その結果は写真およびオリンパス顕微鏡とを利用して記録された。
ここにおいて、再び、細胞をトリプシンで除去し、培養液に於て1時間氷に再構成した。サイトスピンの調製とそれに続く固定の後に、細胞を3時間37℃の温度において、20μl/ml O−グリコシダーゼ(100μlリン酸塩緩衝液pH6.8)を用いて培養した。対照として、サイトスピンをリン酸塩緩衝液だけで培養し、または培養せずに極く普通に染色した。免疫組織化学の染色については、次に述べるように行った。
生存力のある細胞を染色するために、細胞を解離し、洗浄し、細胞1ml当り1×106に希釈した。1mlの細胞溶液を5分間かけて1500gに遠心分離した。完全RPMIで40μg/mlに希釈した抗体を最終容積1mlの量にして、90分間氷を用いて保った。次で、細胞を5分間かけて小球状にし、500μl PRMIで再度解離した。サイトスピン標本を細胞溶液200μlで作成して、30分間乾燥をした。細胞は30分間アセトンに固定された。これをTris/NaClで3回洗浄した。HRP−連結ヘーア抗ヒトIgM(DAKO)をPBS/BSA(0.1%)に1:50に希釈し、室温で30分間保温した。3回洗浄の後に、前に述べたように染色を行った。
消化作用:
図1は膵臓「ガン」細胞BXPC−3の細胞の表面にPM−2の抗体の結合に関するグリコシダーゼ消化作用の影響を示す。消化した後に、サイトスピンが陽性対照CAMケラチン(A,C,E)とPM−2(B,D,F)とによって免疫組織化学的に染色された。
図2は抗体PM−2と生きている細胞染色(200倍に拡大)の効果を示す。
図3はBXPC−3細胞と23132/87細胞として示される抗原のPM−2抗体の助けをかりて行なわれる免疫特別エビデンスを示す。
図4aと4b、そして5aと5bとに示されているグリコシレーション部位は[「UKMRCヒト・ゲノム・マッピング・プロジェクト」(UK MRC Human Genome Mapping Project)](http://www.hgmp.mrc.ac.uk/Genome Web/protanal.html)のデータベースのソフトウェアによって決定されたのである。
図6はPM−2抗体によってSDSゲルから選択されたタンパク質帯域の質量分光器分析の結果を示す。質量スペクトロメータを利用して、ペプチド区分No.2,No.3,No.4及びNo.6の比較はp80タンパク質あるいはREV1タンパク質と同族関係の配列を示している。
Claims (21)
- 膜結合タンパク質として細胞の表面に圧搾され一以上の点(膜糖タンパク質)であって、そのアミノ酸配列がインテグリン結合タンパク質p80(受入れ番号AJ131720)またはREV1(受入れ番号AF206019)であり、前記膜糖タンパク質を、腫瘍性の細胞によって表現し、非腫瘍性の細胞によっては表現することなく、抗原としてヒト単クローン抗体PM−2(DSM受入れ番号:DSM ACC2600)とし、N−グリコシド的とO−グリコシド的とすることを特徴とするポリペプチド。
- 前記ヒト単クローン抗体PM−2(DSM受入れ番号:DSM ACC2600)を少くとも一つのN−グリコシル化炭化物残基(=糖残基)により媒介したことを特徴とする前記請求項1に記載のポリペプチド。
- N−グリコシレーションをP−80抗原の部分列(受入れ番号AJ131720)のアミノ酸位置333,353,450及び568のうちの少くとも一つの部位に存在するものとしたことを特徴とする前記請求項1と2とのいずれかの一つによるポリペプチド。
- N−グリコシレーションをREV1抗原の部分列(受入れ番号AF206019)のアミノ酸位置810,830,927及び1045のうちの少くとも一つの部位に存在するものとしたことを特徴とする前記請求項1と2のいずれかの一つによるポリペプチド。
- 膜グリコプロティンをモノマーとするか、或は少くとも一つの同一のサブユニットまたは他のタンパク質と連合されるものから構成されるものとすることを特徴とする前記請求項第1項乃至第4項のいずれかの一項によるポリペプチド。
- 膜グリコプロティンの分子量を凡そ80乃至160KDAとすることを特徴とする前記第1項乃至第5項のいずれかの一項に記載のポリペプチド。
- 膜グリコプロティンを大腸、膵臓、前立腺、子宮、卵管、副腎および/または肺および食道または肺、胃がん、乳房の管癌および非腫瘍性の細胞による腫瘍性の細胞によって示されるものとすることを特徴とする前記第1項乃至第6項のいずれかの1項に記載するポリペプチド。
- 膜グリコプロティンを膵臓がん細胞株BXPC−3(ATCC番号CRL1687)の表面に発現されたものであることを特徴とする前記第1項乃至第7項のいずれかの1項に記載するポリペプチド。
- 膜グリコプロティンを腫瘍性の上皮細胞の表面に露出されたものとすることを特徴とする前記請求項のいずれかの1項に記載されたポリペプチド。
- PM−2抗体(DSM受入れ番号:DSM ACC2600)と腫瘍性の細胞に関する膜糖タンパク質とを結合した後に、非腫瘍性の細胞でなく、前記腫瘍性の細胞に枯死を招ねかせるようにしたことを特徴とする前記各請求項のいずれかの1項に記載のポリペプチド。
- PM−2抗体(DSM受入れ番号DSM ACC2600)と腫瘍性の細胞とを結合した後に、細胞の増殖を前記細胞の膜糖タンパク質を介して抑制するようにしたことを特徴とする前記各請求項のいずれかの1項に記載のポリペプチド。
- 前記膜糖タンパク質を膵臓がん細胞株BXPC−3の膜抽出物から、溶解化と、粒度除外クロマトグラフィーとを利用して隔離することを特徴とする前記請求項第1項乃至第11項のうちのいずれかの1項に記載するポリペプチド。
- 前記粒度除外クロマトグラフィーに陰イオン交換クロマトグラフィーの動作を追従させることを特徴とする請求項12によるポリペプチドを得る方法。
- ビボ(vivo)投与後に免疫化により腫瘍と戦う役目を果す通常の薬学の付形剤とキャリアとを用いる薬剤の製造に関して前記の総ての請求項の内のいずれかの1項によるポリペプチドの製法。
- 腫瘍性の細胞について細胞枯死および/または細胞増殖抑制効果のある結合配位あるいは接着ペプチドを特に開発するため、前記のすべての請求項のいずれかの1項によるポリペプチドの用法。
- 放射性ヌクレオチド、蛍光標識、細胞毒、そして配位子または癒着性ペプチドに連結される細胞毒、配位子または癒着性ペプチドに結合される細胞毒あるいは成長抑制物質を特に開発するため、前記のすべての請求項のいずれかの1項によるポリペプチドの用法。
- 処理能力が優秀な選別法(HTS)において作用物質を選別するようにする前記のすべての請求項のいずれかの1項によるポリペプチドの用法。
- 腫瘍マーカーとして前記のすべての請求項のいずれかの1項によるポリペプチドの用法。
- 抗原の存在、抗原の集積および/または抗原の量の証拠をPM−2抗体(DSM受入れ番号:DSM ACC2600)に膜糖タンパク質を特に結合して実行することを特徴とする前記のすべての請求項のいずれかの1項によるポリペプチドの用法。
- ベクトルのアミノ酸配列の一部がSEQ NO ID:1に指示されている配列に該当することを特徴とするアンチセンスベクトル。
- ベクトルの核状物質の一部がSEQ NO ID:2に示されている配列に該当することを特徴とするアンチセンスベクトル。
