Abstract
Description
K−252aは、式I中に示される立体化学を有するインドロカルバゾール骨格[特開昭60−41489号(米国第4555402号)]を有する化合物である。
パーキンソン病は、黒色線条体経路のドーパミン作動性ニューロンの進行性および選択的損失を含む神経変性障害である(Agid、Lancet:337:1991)。マウスへの 1-メチル-4-フェニル-1,2,3,6-テトラヒドロピリジン(MPTP)の投与は、ドーパミン作動性ニューロン変性に導き、パーキンソン病で観察されるドーパミン作動性ニューロン損失および行動欠損の動物モデルを供する。MPTPの末梢投与は、ヒト、サルおよびマウスにおける黒色線条体ドーパミン作動性ニューロン系の高度に選択的な変性に導く(Heikkilaら、Science224;1451〜1453、1984;Burnsら、Proc.Natl.Acad.Sci.USA 80;4546〜4550、1983)。
末梢性神経障害とは、大抵しばしば、運動、感覚、感覚運動性または自律神経機能不全の1つまたは組合せとして現れる、一般的に末梢神経に影響する障害をいう。末梢性神経障害によって表される広範な各種の形態は、各々に独特の、等しく広範な各種の原因にあるとできる。例えば、末梢性神経障害は、遺伝学的に獲得でき、全身疾患に起因でき、あるいは毒性薬品によって誘導できる。神経毒性を引き起こすいくつかの毒性薬品は、治療剤、抗腫瘍剤、食物または医薬品中の汚染物品、ならびに環境および産業汚染物である。
腫瘍壊死因子α(TNF-α)およびインターロイキン-1β(IL-1β)は、多数の炎症および代謝過程に関与することが知られているポリペプチドである。敗血症性ショックを含む炎症疾患におけるTNF-αの役割に関係する総説については、Ann.Rev.Immunol.7:625(1980)、およびClinical Trials for the Treatment of Sepsis、Sibbald、W.J.およびVincent,J.−L(Eds.)、Springer−Verlag Berlin Heidelberg1995を参照。TNF-αの過剰産生または不適当な産生は、敗血症性ショックを含むいくつかの病的な状態(Spoonerら、Clinical Immunology and immunopathology、62:p.S11(1992))および限定されるものではないが慢性関節リウマチ、変形性関節症、喘息、気管支炎、慢性閉塞性気道疾患、乾癬、アレルギー性鼻炎、皮膚炎および炎症性腸疾患、および他の自己免疫疾患を含めた、各種の他のアレルギー性および炎症性状態または疾患に関与することが一般的に受け入れられている。Immunol. 本明細書で用いるごとく、「有害な」とは、損傷するおよび/または有害なおよび/または否定的なを意味する。
本明細書で用いるごとく、「GABA作動性ニューロン」は、神経伝達物質としてγ-アミノ酪酸を使用するニューロンを意味する。
本明細書で用いるごとく、「nbm」は、基底核大型細胞を意味する。
図1は、化合物AはモノアミンオキシダーゼA阻害剤ではないことを示すデータのグラフである。クロルグリシンのIC50は、21nmであった。正立した三角形、クロルグリシン;逆三角形、化合物A;四角形、L-デプレニル。
図2は、化合物AはモノアミンオキシダーゼB阻害剤ではないことを示すデータのグラフである。クロルグリシンのIC50は、21nmであった。正立した三角形、クロルグリシン;逆三角形、化合物A;四角形、L-デプレニル。
図3は、ビヒクルのみを受けた障害動物(ダーク・バー)に比較して、化合物Aを受けた動物(ライト・バー)は、障害されたnbmの吻側部分(障害の中点かた400μmにわたって)において有意により多くのフルオロ-金標識ニューロンを有したことを示す捧グラフである。Newman‐Keuls検定による*=P<0.05。
図5は、アクリルアミド誘導末梢性神経障害に対する化合物Aの効果を示す。
本発明は、哺乳類の中枢神経系の特殊な型のニューロンの機能および/または生存を増強することによって、かかる疾患および障害のために死ぬ危険性においてニューロンの機能および/または生存に否定的に影響する病気、障害および疾患の有害な影響を治療する方法を提供する。さらに詳細には、本発明は、以下の化学構造:
化合物Aは、当業者によって有効と考えられるように投与でき;投与のより好ましい方法は皮下注射である。
化合物Aで有効に治療できる好ましい神経障害は、全身病、例えば、ポストポリオ症候群に関連する神経障害;遺伝的に獲得した神経障害、例えば、シャルコー・マリー・ツース病:および毒性薬品、例えばアクリルアミド、または化学療法剤、例えば、ビンクリスチンによってひき起こされた神経障害を含む。
本発明は、以下の実施例によってさらに示されるであろう。これらの実施例は、本発明の範囲およびここに添付された請求の範囲を限定として解釈するものではない。
ドーパミン作動性ニューロンに対する薬理学的活性
実験は、マウスのMPTP障害ドーパミン作動性ニューロン(MPTPマウスモデル)において化合物Aを用いて行った。C57黒色マウスに供されたMPTPの単回s.c.用量(20mg/kg)は、線条体チロシンヒドロキシラーゼ活性のおおよそ60%喪失を生じた。0.01ないし10mg/kg(s.c.)の範囲の日用量にて、かかるMPTP処置マウスに対して化合物Aを投与すると、線条体チロシンヒドロキシラーゼ活性の喪失を減少させた。これらのデータを表2に示す。
MPTPマウスドーパミン作動性障害モデルにおける化合物Aの効力を示すこれらのデータは、パーキンソン病のごとき神経変性障害の治療において化合物Aの有用性を示す。
GABA作動性ニューロンに対する薬理学的活性
よく確立されたイボテン酸(ibotenic acid)障害モデルを用いて、基底核大型細胞中のGABA作動性ニューロンの涸渇(例えば、生存を高める)を予防する能力について、化合物Aを試験した。興奮毒性であるイボテン酸は、nbm領域においてGABA発現ニューロンの数を減少させることが知られている(Lindeforsら、Neurosci.Lett.135:262〜264、1992:Shaugnessyら、Brain Res.、637:15〜16、1994)
成体雄性スプレーグ・ドーリーラットは、nbmの片側にイボテン酸(5.0g)の注射を受けた。次いで、ラットには、障害18時間後に開始して皮下注射を介して化合物A(0.03mg/kg)が投与され、障害後18日まで1日4回にて続けた。次いで、組織切片を、nbmの吻側−尾側にわたり集め、加工してGABAの生合成に必要とされる酵素であるグルタミン酸デカルボキシラーゼを検出した。次いで、基底核大型細胞中の吻側−尾側にわたりグルタミン酸デカルボキシラーゼ発現ニューロン数をカウントし、この領域では、nbmのグルタミン酸デカルボキシラーゼ発現ニューロンが隣接した構造(淡蒼球に対して内側および腹側の白色物)中のグルタミン酸デカルボキシラーゼ発現ニューロンと容易に区別される。結果は、反対の非障害側の数に対して、障害側での百分率グルタミン酸デカルボキシラーゼ発現ニューロンとして表した。また、結果は、吻側nbm、中間nbmおよび尾側nbmについて、個別に計算した。
これらの結果は、化合物Aが興奮毒性障害から起因するGABA作動性ニューロン喪失に対して保護する、従ってそのニューロン集団に対して神経親和性効果を有することを示す。
nbmニューロンに対する薬理学的活性
興奮毒性ニューロン死を予防する化合物Aの能力を直接的に試験するために、成体雄スプレーグ・ドーリーラットのnbmニューロンを、まず、長持続性マーカーで標識した。これは、神経末端に取り込まれ、細胞体に逆輸送されるニューロントレーサーであるフルオロ−金(FG)を、前頭皮質および壁側皮質中のnbmニューロンの標的領域内に注入することによってなされた(Bookら、J.Neuropath.Exp. Neurology、53:95〜102、1994)。7〜10日後、動物にはイボテン酸の5μgを用いて、nbmの障害を片側に受けさせた。この障害の18時間後に開始し、動物には皮下注射を介して、障害後18日間まで一日おきに、化合物A(0.03mg/kg)を受けさせた。組織切片は、脳の両側においてnbmの全吻側−尾側にわたり集め、FG標識を有するnbmニューロン数をカウントした。カウントは、標準的手順を用いてサイズ差について修正し、結果を脳の反対側の非障害側でのnbm中の標識ニューロン数に対する各動物の障害領域のnbm中の標識ニューロンの百分率として表した。
これらの結果は、化合物Aが興奮毒性損傷に起因するnbm中の予め標識したニューロンの喪失を予防できることを直接的に示す。
化合物Aを用いた短期間投与に対する長持続性機能改善
成体雄スプレーグ・ドーリーラットを、まず、標準的T迷路変更課題で応答を変更するために慣らした(Heplerら、J.Neurosci.、5:866〜873、1985)。次いで動物には、T迷路課題中に効率を損なうように基底核大型細胞の両側障害を受けさせた(例えば、Salamoneら、Behav.Brain Res.、13:63〜70、1984)。障害後18時間に開始して、障害後12日まで一日おきに、皮下注射(0.1mg/kg)を介して化合物Aを動物に摂取させた。最終注射の24時間後に全動物は、それらの改変効率が所定のレベルに達するまで、概して3ないし10日間かかるが、1日1回T迷路に入れた。基準に達した後、動物は8から10日週間さらに試験しなかった。その時点で、所定の効率に達するまで1日1回動物を再度T迷路に入れた。この試験の間、動物が犯した誤りの総回数を図4に示す。ビヒクルのみを受けた障害動物は、非手術の正常または偽手術のいずれよりも有意に多い誤りを犯したが、先に化合物Aを10〜12週間摂取した障害動物は、正常なものと違いは無かった。
これらの結果は、化合物Aが注意または記憶の改善を反映する行動の長期の改善を、投薬を中止した後数カ月、生じることを示す。
内側嗅領皮質障害モデルにおける化合物Aの効力
内側嗅領皮質の第2層におけるグルタミン酸含有ニューロンは、歯状回顆粒細胞の樹状突起上のシナプスを形成する歯状回の分子層に突き出す。障害は、興奮毒性 N-メチル-D-アスパラギン酸(NMDA)の定位の注入によって内側嗅領皮質の第2層において引き起せた。ペントバルビタールナトリウム麻酔下、ラットには内側嗅領皮質中の各2部位でNMDA(15nmols/位置)の注入を受けさせた。2週間後、50mM硫化ナトリウム溶液を用いラットを犠牲にし、潅流した。脳を取り出し、40μmの厚さの水平面で薄片に切り、クレシルバイオレットまたはTimm’sの染液のいずれかで染色した。内側嗅領皮質の第2層中のニューロンの生存を、クレシルバイオレット染色切片中で評価し、一方歯状回の分子層中のそれらの軸索末端の完全さは、Timm’sの染液で染色された別の切片で測定した。
アクリルアミド誘導末梢性神経障害に対する化合物Aの効果
アクリルアミドは、ヒトおよび動物において「dying back」中枢−末梢性遠位神経障害を生じる。該障害は、ヒトの薄弱、不安感および失調によって特徴付けられる混合した感覚/運動の神経障害である。動物において、アクリルアミドは、習性(感覚、運動および固有受容)、組織病理学、電気生理学および体重減少に変化を生じる。末梢の大直径の長い軸索において、Ab繊維は、アクリルアミドによって優先的に影響される。ラットにおいて、アクリルアミド投与は、固有受容の尺度であるlanding foot spread(LFS)によって測定されるごとくに軸索障害を生じる。
これらのデータは、化合物Aが末梢性神経障害の治療に有用であることを示す。
化合物Aはin vitroにおいてプロテインキナーゼCを阻害しないこと
プロテインキナーゼCアッセイおよびK−252aによるプロテインキナーゼCの阻害は、米国特許第4,923,986号およびKase、Hら、(cds.)Japan Scientics Press、Tokyo、pp293〜296(1988)に開示されている。本質的に同様なアッセイ手段によって、K−252aはプロテインキナーゼCを0.028μMのIC50で阻害し、一方、化合物AのIC50は16μMであり、すなわち570倍低い活性であることが見出された。化合物Aは、TNF−αの産生の阻害剤として139nMのIC50およびIL−1βの産生の阻害剤として261nMのIC50値(化合物AがプロテインキナーゼCの阻害剤として不活性である濃度)を有する。
化合物AによるTNF−αおよびIL−1β産生のin vitro阻害
TNF−αの誘導を阻害する化合物Aの能力を以下に記載した標準的in vitro薬理学的試験手順の使用によって示した。
100%DMSO中の4mMの化合物Aよりなる貯蔵溶液を4℃で保存した。細胞培養基RPMI 1640(Media Tech、Herndon、Va)および胎仔ウシ血清(Hyclone、Logan、UT)は、試験培養基を構成した。トリクロロ酢酸で抽出されたE.Coli血清型0111:B4からのリポ多糖類(LPS)(Sigma、St.Louis、MO)を使用した。LPSの貯蔵溶液を調製し、リン酸緩衝生理的食塩水(PBS)中で4℃にて保存した。腫瘍壊死因子アルファ(TNF−α)およびIL−1βのアッセイ用ELISAキットはBoehringer−mannheim(Indianapolis、IN)から購入し、製造業者の使用説明書に従って使用した。ヒト単球由来細胞ラインであるTHP−1細胞は、American Type Culture Collection(ATCC TIB 202)から得た。37℃にて5%CO2:95%空気の湿潤雰囲気で、細胞を10%胎仔ウシ血清を含有するRPMI 1640(培養基)中で増殖させた。実験は、培養基の1ml中に5×105THP−1細胞を有する24ウェル培養プレート(Nunc)中で行った。細胞は、TNF−αを誘導するために2μg/mlのLPSにてインキュベートした。3時間のインキュベーションの後、細胞および培養基を5分間1000×gにて遠心し、得られた上清液を直ちにTNF−αについてアッセイするかまたは−70℃にて保存した後にアッセイした。培養基試料は、ElisaアッセイによってTNF−αおよびIL−1βの含量につき測定した。
マウス血清中でのLPS誘導TNF−αおよびIL−1βの阻害
60匹の雌C57BLマウス(4〜6週齢)を各10匹の6群に分けた。3群(1、3、5)は、腹腔内(ip)投与を介して200μl/マウスでビヒクル(0.1%TEA(テトラエチルアンモニウム塩酸塩)を含有するPBS)のみを受けた。他の3群(2、4、6)は、ビヒクル(12-ヒドロキシステアリン酸のエチルエステル:ソルトールの10%を含有するPBS)の化合物A(200μl中に30mg/kg、ip)を受けた。2時間後、動物はip投与によって、LPSの0.1mg/kg(群1および2)、LPSの0.5mg/kg(群3および4)およびLPSの1.0mg/kg(群5および6)を受けた。LPS投与2時間後、全ての動物を犠牲にし、血漿試料を得、それらのTNF−αおよびIL−1β含量につきアッセイした。アッセイは、実施例8に記載のごとく行った。
データを表4にまとめた。( )中の数は、前記のごとき群番号である。Nは、血漿測定がなされた動物数を示す。ND=実施せず。
マウスにおけるLPS誘導死の予防
40匹のマウスを10匹のマウスよりなる2群および20匹のマウスよりなる1群の3群に分けた。ビヒクル、LPSおよび化合物Aの投与は、腹腔内であった。第1群(n=10)マウスは、ゼロ時点にてビヒクル(LPSのリン酸緩衝生理的食塩水、200μl/マウス)および2時間後に化合物Aのビヒクル(200μl/マウス)を受けた。第2群(n=10)マウスは、ゼロ時点にて化合物Aのビヒクル(200μl/マウス)および2時間後にLPS(200μl/マウス中に2mg)を受けた。第3群(n=20)は、ゼロ時点にて化合物A(ビヒクルの200μl中に30mg/kg)および2時間後にLPS(200μl/マウス中に2mg)を受けた。

- 該有害な効果が、敗血性ショック、慢性関節リウマチ、変形性関節症、喘息、気管支炎、慢性閉塞性気道疾患、乾癬、アレルギー性鼻炎、皮膚炎および炎症性腸疾患よりなる群から選択される請求項2記載の使用。
- 該有害な効果が、自己免疫疾患である請求項2記載の使用。
- 該有害な効果が、敗血性ショック、慢性関節リウマチ、変形性関節症、喘息、気管支炎、慢性閉塞性気道疾患、乾癬、アレルギー性鼻炎、皮膚炎および炎症性腸疾患よりなる群から選択される請求項8記載の医薬組成物。
- 該有害な効果が、自己免疫疾患である請求項8記載の医薬組成物。
