Abstract
【解決手段】イムノベクターとカップリングするための生成物であって、該イムノベクターが生物学的に活性な成分に、真核細胞にインターナリゼーションする能力を付与でき、前記イムノベクターが細胞核内又は細胞核の間近に該生物学的に活性な成分を導入することができる程度に、細胞のDNAに対して親和性を有することを特徴とする生成物。

Description
1.イムノベクターの調製
A)ポリクローナルイムノベクター
まず、播種状エリテマトーデスに罹患した個体から−、または狼瘡マウス(NZB×NZW)F1から−得た血清の混合物を、セファロース上に固定化されたプロテインA上に通すことにより、ヒトまたはネズミIgGを単離する(14)。
狼瘡マウス(NZB×NZW)F1から得られた脾細胞を、KohlerおよびMilsteinの方法により、X63メラノーマと融合させる。得られたハイブリドーマを、IgGの分泌および抗DNA活性に関してELISAにより試験する。抗DNA IgGを分泌するハイブリドーマを少なくとも2回サブクローニングし、依然として二重陽性(IgG+抗DNA)のクローンをバルク培養するか、又はそのほかの方法として、マウスにおいてこれらのクローンから腹水を調製する。典型的な実験では、マウス(NZB×NZW)F1の脾臓から出発して、IgGを分泌する約300個の陽性ウェルを得た。そのうちの60個は、DNAと反応する能力を有していた。クローニングの後、20個のクローンが、DNAを認識するIgGを分泌した。これら20個のクローンのうちの約半数が、該細胞の核内に浸透する能力を有する抗体を分泌したが、残りはこの能力を有していなかった(C:「イムノベクターの核内浸透試験」を参照)。45%硫酸アンモニウムで沈殿させ、ついで透析した後、セファロース上に固定化されたプロテインA上に通すことにより、培養上清または腹水から該モノクローナルIgGを単離する。溶出したIgGを中和した後、該調製物を透析し、濃縮し、使用するまで−20℃で保存する。
1)イムノベクターの核内浸透試験
2つの繊維芽細胞系(カンガルーネズミの腎臓から得たPtK2およびハムスターの腎臓から得たGMA-32)を主として使用した。24時間前に2×104細胞/mlで接種し、RPMI 1640またはMEM培地(10%ウシ胎児血清、2mM L−グルタミンおよび1%ピルビン酸ナトリウムを含有するもの)中で培養した指数増殖期の繊維芽細胞を保持するスライドを、選択された量のイムノベクター(1から70μg/ml)を含有する新たに取り換えられた培地中37℃でインキュベートする。2から4時間インキュベートした後、該細胞をPBSで洗浄し、エタノールで−20℃で10分間またはPBS中0.2%グルタルアルデヒドおよび2%ホルムアルデヒドで20分間固定する。PBSで3回洗浄した後、0.2%ウシ血清アルブミンおよび0.5%サポニンを含有するPBS中で20分間、該細胞を透過可能とした。
該細胞膜との反応を確認するために、106個のマウス胸腺細胞または脾細胞を、0.2%アジ化ナトリウムを含有する0.1%のウシアルブミン溶液中に希釈した種々のモノクローナル抗体の0.1mlと低温で45分間インキュベートした。洗浄後、蛍光抗マウスIgG抗体と共に該細胞を低温で45分間インキュベートする。洗浄後、該細胞をFACSにより調べ、各集団内の陽性細胞の数を測定する。
A)イムノベクターのF(ab’)2およびFab’断片の調製
ペプシンでタンパク質分解し、ついでシステインで還元してFab’断片を得ることを含む、開示されている方法により(14)、イムノベクターのF(ab’)2断片を調製する。ついで、イムノベクター5mgを含有する0.1Mクエン酸塩−クエン酸緩衝液(pH3.5)の5mlに、150μgのペプシンを加え、該混合物を37℃で2時間インキュベートする。該培地をpH8に調整し、該調製物をプロテインA−セファロースカラム上で濾過して、未消化のIgGを除去する。PBSに対して透析した後、使用するまでこのF(ab’)2調製物を−20℃で保存する。Fab’断片を得るために、該F(ab’)2調製物にシステインを最終濃度が0.02Mになるまで加える。37℃で10分間インキュベートした後、0.04Mヨードアセトアミドを加え、該混合物を30分間インキュベートする。このFab’調製物をPBSに対して透析し、使用するまで−20℃で保存する。
ビオチンへのカップリング
ジメチルホルムアミド中のd−ビオチン−N−ヒドロキシスクシンイミドエステルの0.1M溶液2μl(ジメチルホルムアミドの30μl中の活性エステルの1mg)を、抗体1mgを含有する0.1Mリン酸緩衝液(pH7)0.5mlに加える。該溶液を室温で1時間放置し、PBSに対して+4℃で一晩透析する。
抗体1mgを含有する炭酸ナトリウムの0.1M溶液1mlに、ジメチルスルホキシド中のフルオレセインイソチオシアネートの溶液(10mg/ml)20μlを加える。該溶液を実験室温度で3時間放置し、PBSに対して+4℃で透析する。
ペルオキシダーゼとのカップリング
0.1Mリン酸塩緩衝液(pH6.8)中の1%グルタルアルデヒド0.2mlに10ミリグラムのペルオキシダーゼを溶解する。実験室温度で18時間インキュベートした後、0.15M NaClで平衡化させたセファデックスG25カラム(0.9×60cm)上で該溶液を濾過して、過剰なグルタルアルデヒドを除去する。この活性化されたペルオキシダーゼ溶液に、抗体5mgを含有する0.15M NaCl溶液1mlおよび1M炭酸塩−炭酸水素塩緩衝液(pH9.5)0.2mlを加える。該溶液を+4℃で24時間保存し、ついでリジンで最終濃度が0.1Mになるまで補足し、ついでPBSに対して4℃で透析する。
ポリヌクレオチド
使用するポリヌクレオチドは15個のヌクレオチドよりなり、5’側にフルオレセインを、3’側に遊離NH2基を保持していた。これは、核酸合成の通常の方法により調製した。このヌクレオチドを、p−ベンゾキノンを介してイムノベクターにカップリングさせた(14)。p−ベンゾキノン3mgを含有するエタノール0.1mlを、イムノベクター(全分子、F(ab’)2またはFab’)1mgを含有する0.1Mリン酸塩緩衝液(pH6)0.4mlに加える。実験室温度で1時間インキュベートした後、該調製物をセファデックスG-25カラム上で濾過する。活性化されたイムノベクターを含有する画分を、ポリヌクレオチド4分子に対してイムノベクター1分子の割合にてポリヌクレオチドで補足し、該溶液を炭酸塩−炭酸水素塩緩衝液でpH9.2に調整する。実験室温度で18時間インキュベーションした後、リジンを最終濃度が0.1Mになるまで加えることにより該反応を停止させ、ついでPBSに対して透析する。この調製物を、使用するまで+4℃で保存する。
2つのプラスミドを試験した。第1プラスミドは、SV40 T、t抗原をコードする遺伝子の上流にビメンチンプロモーターを保持し(pHuVim 830 T, t)(21)、第2プラスミドは、サイトメガロウイルスプロモーターの制御下でルシフェラーゼ遺伝子(22)を保持する(pCMV-Luc)(5)。
A)フルオレセインのインビトロでの導入
ガラスカバーグラス上の培養中のGM A-32系の繊維芽細胞を、フルオレセインで標識した次第に増加する量のモノクローナルイムノベクター(J-20.8抗体またはFab’2断片)を含有するRPMI培地中、37℃で2から4時間インキュベートする。この時間の終了時に、IC1に記載されているとおりに、該細胞を洗浄し、固定する。それらを、Mowiol培地に加えた後、蛍光顕微鏡下で検査する。事実上、該繊維芽細胞のすべての核が蛍光標識を示す。一方、核へ浸透しない抗DNA活性を有さない対照モノクローナル抗体Ig 2aと共にインキュベートした繊維芽細胞の核は、蛍光を全く示さない(図1および2)。
1mgのイムノベクター(モノクローナル抗体C-2.1またはF-4.1)または対照抗体(モノクローナル抗体G-14)(フルオレセインで標識されているもの)を、0.2mlの量にて静脈内に、0.3mlの量にて腹腔内に2匹のマウスに注射する。5時間後、該マウスを出血させ犠牲にし、循環血リンパ球をFACSにより分析する。イムノベクターを注射したマウスから得た末梢血リンパ球の60%が蛍光性であり、一方、対照動物からのリンパ球はいずれも蛍光性でないことが認められる(図3)。顕微鏡検査は、該細胞の大多数において核のレベルで蛍光を示す。
前もって24時間培養したPtK2系の繊維芽細胞(105/ml)を、ビオチンで標識された次第に増加する量のヒト抗DNAポリクローナルIgG(5〜100μg)と共に完全RPMI培地中でインキュベートする。3時間後、ICに記載されているとおりに、該細胞を洗浄し、固定し、透過可能とする。ついで該細胞を、ペルオキシダーゼで標識されたストレプトアビジン1μg/mlを含有するRPMIと共にインキュベートする。1時間後、該細胞をPBSで3回洗浄し、DAB+H2O2培地を用いて、該細胞に結合したペルオキシダーゼを表示させた。該調製物をMowiolに加え、光学顕微鏡下で検査した。抗DNA IgGと共にインキュベートした繊維芽細胞の多数の核は陽性であり、一方、正常な個体から得、ビオチンで標識したIgGと共にインキュベートした細胞は陰性である。
第IIIC節で定義した条件下、ペルオキシダーゼで標識した次第に増加する量のイムノベクター(抗体J-20.8)のFab’断片と共に、PtK2繊維芽細胞をインキュベートする。3時間後、該細胞をPBSで3回洗浄し、PBS中の0.2%グルタルアルデヒドおよび2%ホルムアルデヒドで20分間固定する。洗浄後、着色DAB+H2O2試験により該ペルオキシダーゼ活性を表示させ、該調製物を光学顕微鏡下で検査する。J-20.8抗体のFab’断片と共にインキュベートした繊維芽細胞の核の大部分はペルオキシダーゼに関して陽性であり、一方、対照抗体48.9と共にインキュベートしたものは陰性である。
BALB/cマウス脾臓から調製した3×106個の脾細胞を、イムノベクター(J-20.8)またはそのFab’断片(ポリヌクレオチドに共有結合でカップリングしているもの)の40μg/mlを含有するRPMIの1ml中でインキュベートする。37℃で3時間インキュベートした後、該細胞をPBSで洗浄し、4%パラホルムアルデヒド中で固定し、顕微鏡下で検査する。8から10%の細胞が、核内蛍光を示す(図4)。
ペルオキシダーゼにカップリングした抗T抗原抗体の助けによりこれらの遺伝子にコードされるタンパク質の合成を確認することにより、あるいはルシフェラーゼの基質(ルシフェリン)上のルシフェラーゼの活性の発光測定アッセイにより、トランスフェクション効率を評価した。
前日にHep2細胞を、24ウェルプレートの1ウェル当たり完全培地0.5ml中2×104細胞の量で接種する。トランスフェクションのために、該培地を除去し、20μgの抗体−ポリ−L−リジンおよび2μgのプラスミド、または20μgの天然抗体および2μgのプラスミド、または2μgのプラスミドのみを含有する0.3mlの完全培地と取り換える。6時間後に培地を変え、必要であれば、2日毎に培地を変え細胞を2つに細分することにより、該培養を続ける。種々の時点でトランスフェクション効率を調べる。
前日にGMA-32細胞を、完全培地の24ウェルプレートの1ウェル当たり7から10×104細胞/完全培地0.5mlの量で接種する。トランスフェクションのために、該培地を除去し、8μgのJ-20.8/ポリリジンまたはF-14.6/ポリリジン、または20μgのJ-20.8ポリクローナルIgGおよび2μgのプラスミド、または2μgのプラスミドのみを含有する0.5mlの完全培地と取り換える。6時間後に培地を変える。トランスフェクション開始の24時間後に、トランスフェクション効率を調べる。
プラスミドpHuVim 830 T,t
トランスフェクションされた細胞の核内でのT抗原の合成を、免疫細胞化学的方法により確認する。該細胞をPBSで3回洗浄し、ついでメタノール中、−20℃で10分間固定する。ついで、ペルオキシダーゼにカップリングされた抗T抗原抗体と共にそれらを1時間インキュベートする。洗浄後、該ペルオキシダーゼを、DAB+H2O2混合物で表示させる。J-20.8ポリリジンおよびプラスミドコンプレックスと共にインキュベートしたウェルでは、単離された細胞および細胞の数個のクラスターが、48時間後および2週間後に褐色に強く着色された核を有する。天然抗体による対照または該プラスミド単独の場合は、陰性である。
トランスフェクションされた細胞の溶解物中で検出可能なルシフェラーゼ合成により、トランスフェクション効率を確認する。この酵素は、ルシフェリンの酸化を触媒し、それにより、ルミノメーターで検出可能な生成物が生じる。培養後、該細胞をPBS中で洗浄し、ついで8mM MgCl2、1mM DTT、1%トリトンX100、1%BSAおよび15%グリセロールを含有する25mMトリス−リン酸塩緩衝液(pH7.8)中で溶解する。ルシフェリン(0.25mM)およびATP(1mM)の溶液の自動添加により、該溶解物をルミノメーター中でアッセイする。同じ溶解物のアリコートを、Coomassie(Bio-Rad Protein Assay)試薬を用いて、そのタンパク質濃度に関してアッセイする。その結果を、タンパク質1mg当たりの単位(RLU)で表す。表に示されているとおり、抗体調製物J-20.8/ポリリジンおよびF-14.6/ポリリジンの存在下では遺伝子導入が起こるが、IgG/ポリリジン調製物は何ら影響を及ぼさない。

Claims (35)
- 生物学的に活性な成分とイムノベクターとの間のカップリング生成物であって、前記カップリング生成物が、真核細胞内での前記の生物学的に活性な成分のインターナリゼーションを可能にする能力によって、及び前記イムノベクターが、これらの細胞の核の間近または前記核内に前記の生物学的に活性な成分を導入できる程度の、これらの細胞のDNAに対する前記イムノベクターの親和性によって特徴づけられるカップリング生成物。
- 前記の生物学的に活性な成分と該イムノベクターとの間のカップリングが共有結合または非共有結合を含むことを特徴とする、請求の範囲第1項に記載の生成物。
- 前記生成物の組成物中に含まれるイムノベクターが、細胞透過試験であって、前記イムノベクターと結合しうる活性成分が進入しなければならない核において、細胞の存在下で、関心のあるイムノベクターを第1インキュベーションに付し、ついで、前記細胞を固定し透過可能とした後、標識された抗イムノベクター抗体と共に更にインキュベートし、最後に、該核の間近または該核内においてさえも、前記イムノベクターと前記抗イムノベクター抗体との抗原−抗体免疫反応を検出することを具備した細胞透過試験により選択可能なものから選ばれることを特徴とする請求の範囲第1項または第2項に記載の生成物。
- 該イムノベクターが、核酸に対する親和性を有する抗体または前記親和性を保持するこの抗体の断片よりなることを特徴とする請求の範囲第1項から第3項のいずれか1項に記載の生成物。
- 該イムノベクターが、モノクローナルIgG、(Fab’)2または(Fab’)断片、または対応する核酸の認識に関与する抗体の部位に対応する何れかのポリペプチドから選ばれることを特徴とする請求の範囲第4項に記載の生成物。
- 前記イムノベクターが、免疫グロブリンであることを特徴とする請求の範囲第4項または第5項に記載の生成物。
- 前記イムノベクターが、正常な個体から得られた抗DNA活性を保持するIgG免疫グロブリンであることを特徴とする請求の範囲第4項から第6項のいずれか1項に記載の生成物。
- 前記イムノベクターが、抗DNA活性を保持するIgG免疫グロブリンであり、自己免疫症候群を現している個体から得られることを特徴とする請求の範囲第4項から第6項のいずれか1項に記載の生成物。
- 前記自己免疫症候群が、播種状エリテマトーデス症候群である請求の範囲第8項に記載の生成物。
- 請求の範囲第4項から第9項のいずれか1項に記載の生成物であって、前記抗体が、一方では該DNAを、他方ではHIVレトロウイルスのTat、Revのようなタンパク質を、他方ではCD3、CD4、CD8、CD19、CD34のような表面マーカーを認識する二重特異性抗体であることを特徴とする生成物。
- 該イムノベクターにカップリングされた活性成分が、核酸、タンパク質またはハプテンから選ばれる分子であることを特徴とする請求の範囲第1項から第10項のいずれか1項に記載の生成物。
- 該イムノベクターにカップリングされた活性成分が、プラスミドであって、前記プラスミドに含まれる遺伝子にコードされるタンパク質の発現を可能にするものよりなることを特徴とする請求の範囲第1項から第10項のいずれか1項に記載の生成物。
- 該イムノベクターが抗体または抗体の断片である場合、後者をまず、該DNAに対する圧縮効果を誘導しうる物質とカップリングさせることを特徴とする請求の範囲第12項に記載の生成物。
- 該DNAに対する圧縮効果を誘導しうる物質が、ポリリジンである請求の範囲第13項に記載の生成物。
- 該活性成分が、標的細胞のゲノム中に組込むことを意図した遺伝子よりなることを特徴とする請求の範囲第1項から第10項のいずれか1項に記載の生成物。
- 該組込みが、相同的組換えによって行われる請求の範囲第15項に記載の生成物。
- 該遺伝子が、細菌または真核細胞、真菌細胞またはウイルスに由来するポリペプチドについてコードする核酸配列を含有する「裸」のDNAよりなる請求の範囲第15項に記載の生成物。
- 該ポリペプチドが、ワクチン特性を有する請求の範囲第17項に記載の生成物。
- 該活性成分が、アンチセンスオリゴヌクレオチド、生物学的活性を有するハプテン、ホルモン、医薬および抗ウイルス剤から選択されることを特徴とする請求の範囲第1項から第10項のいずれか1項に記載の生成物。
- 該活性成分が、選択された細胞タイプの不死化を可能にすることを特徴とする請求の範囲第1項から第10項のいずれか1項に記載の生成物。
- 該選択された細胞が、マクロファージ、樹状細胞、BおよびT細胞、または造血細胞である請求の範囲第20項に記載の生成物。
- 該細胞が、ヒト由来のものである請求の範囲第20項または第21項に記載の生成物。
- 該活性成分が、タンパク質またはヌクレオチドの合成を抑制しうるアンチセンスオリゴヌクレオチドであることを特徴とする請求の範囲第1項から第10項のいずれか1項に記載の生成物。
- 該タンパク質またはヌクレオチドの合成の抑制が、HIVレトロウイルスに感染した細胞または腫瘍細胞において得られる請求の範囲第23項に記載の生成物。
- 活性な分子を含有する真核細胞であって、前記分子が前記細胞の核内に天然では取込まれることがないか、または前記細胞中で低い発現レベルを有し、該分子が、これらの細胞内でそれらの核の間近またはそれらの核内で見出され、請求の範囲第1項から第19項のいずれか1項に記載の生成物に対応するこれらの細胞のDNAに対する親和性により特徴づけられるイムノベクターにカップリングされていることを特徴とする真核細胞。
- 該活性な分子が核内で活性を示す請求の範囲第25項に記載の真核細胞。
- 請求の範囲第25項または第26項に記載の細胞であって、該細胞が、ウイルスまたは腫瘍細胞に感染しうることを特徴とする細胞。
- CNCMに1995年6月30日に受託番号第I-1605号、第I-1606号および第I-1607号として寄託されているハイブリドーマ。
- 選択された真核細胞の核内に該活性成分をインビトロで導入する方法であって、前記活性成分をイムノベクターにカップリングすることによって特徴づけられ、該イムノベクターがこれらの真核細胞における前記の生物学的に活性な成分のインターナリゼーションを可能にする能力を有し、かつ、前記イムノベクターがこれらの細胞の核の間近または該核内に前記の生物学的に活性な成分を輸送できる程度に該細胞のDNAに対して親和性を有する方法。
- 該生物学的に活性な成分が、該イムノベクターに共有結合でまたは非共有結合でカップリングされていることを特徴とする請求の範囲第29項に記載の方法。
- 該生成物の組成物中に含まれるイムノベクターが、前記イムノベクターと結合しうる活性成分が進入しなければならない核において、細胞の存在下で、関心のあるイムノベクターを第1インキュベーションに付し、ついで、該細胞を固定し透過可能とした後、標識された抗イムノベクター抗体と共に更にインキュベートし、最後に、該核の間近または該核内においてさえも、該イムノベクターと該抗イムノベクター抗体との抗原−抗体免疫反応を検出することを具備する細胞透過試験により選択可能なものから選ばれることを特徴とする請求の範囲第29項または第30項に記載の方法。
- 該イムノベクターが、核酸に対する親和性を有する抗体または前記親和性を保持するこの抗体の断片より形成されることを特徴とする請求の範囲第29項に記載の方法。
- 前記イムノベクターが、モノクローナルIgG、(Fab’)2または(Fab’)断片、または対応する核酸の認識に関与する抗体の部位に対応する何れかのポリペプチドから選ばれることを特徴とする請求の範囲第29項に記載の方法。
- 生理学的に許容しうる賦形剤と組み合わせて請求の範囲第1項から第11項のいずれか1項に記載の生成物を含有することを特徴とする薬学的組成物であって、該生物学的に活性な成分が医薬またはワクチン活性成分であり、該イムノベクターが、該医薬が向けられた宿主生物と適合しうることを特徴とする薬学的組成物。
- 受容細胞における、前記宿主のDNAに対して異種であるヌクレオチド配列のインビトロでの発現のための、請求の範囲第1項に記載の生成物の使用。
