- 【特許請求の範囲】 1.生物学的に活性な成分とイムノベクターとの間のカップリング生成物であ って、前記カップリング生成物が、真核細胞内での前記の生物学的に活性な成分 のインターナリゼーションを可能にする能力によって、及び前記イムノベクター が、これらの細胞の核の間近または前記核内に前記の生物学的に活性な成分を導 入できる程度の、これらの細胞のDNAに対する前記イムノベクターの親和性に よってことを特徴づけられるカップリング生成物。 2.前記の生物学的に活性な成分と該イムノベクターとの間のカップリングが 共有結合または非共有結合を含むことを特徴とする、請求の範囲第1項に記載の 生成物。 3.前記生成物の組成物中に含まれるイムノベクターが、細胞透過試験であっ て、前記イムノベクターと結合しうる活性成分が進入しなければならない核にお いて、細胞の存在下で、関心のあるイムノベクターを第1インキュベーションに 付し、ついで、前記細胞を固定し透過可能とした後、標識された抗イムノベクタ ー抗体と共に更にインキュベートし、最後に、該核の間近または該核内において さえも、前記イムノベクターと前記抗イムノベクター抗体との抗原−抗体免疫反 応を検出することを具備した細胞透過試験により選択可能なものから選ばれるこ とを特徴とする請求の範囲第1項または第2項に記載の生成物。 4.該イムノベクターが、核酸に対する親和性を有する抗体または前記親和性 を保持するこの抗体の断片よりなることを特徴とする請求の範囲第1項から第3 項のいずれか1項に記載の生成物。 5.該イムノベクターが、モノクローナルIgG、(Fab')2または(Fab')断片、ま たは対応する核酸の認識に関与する抗体の部位に対応する何れかのポリペプチド から選ばれることを特徴とする請求の範囲第4項に記載の生成物。 6.前記イムノベクターが、正常な個体から得られ抗DNA活性を保持する免 疫グロブリン、特にIgGであることを特徴とする請求の範囲第4項または第5項 に記載の生成物。 7.前記イムノベクターが、抗DNA活性を保持する免疫グロブリン、特にIgG であり、自己免疫症候群、特に播種状エリテマトーデス症候群を現している個体 から得られることを特徴とする請求の範囲第4項または第5項に記載の生成物。 8.請求の範囲第4項から第7項のいずれか1項に記載の生成物であって、こ れが、一方では該DNAを、他方ではHIVレトロウイルスのTat、Revのようなタ ンパク質を、他方ではCD3、CD4、CD8、CD19、CD34のような表面マーカーを認識 する二重特異性抗体であることを特徴とする生成物。 9.該イムノベクターにカップリングされた活性成分が、核酸、タンパク質ま たはハプテンから選ばれる分子であることを特徴とする請求の範囲第1項から第 8項のいずれか1項に記載の生成物。 10.該イムノベクターにカップリングされた活性成分が、プラスミドであっ て、前記プラスミドに含まれる遺伝子にコードされるタンパク質の発現を可能に するものよりなることを特徴とする請求の範囲第1項から第8項のいずれか1項 に記載の生成物。 11.該イムノベクターが抗体または抗体の断片である場合、後者をまず、該 DNAに対する圧縮効果を誘導しうる物質とカップリングさせることを特徴とし 、該物質が好ましくはポリリジンである請求の範囲第10項に記載の生成物。 12.該活性成分が、標的細胞のゲノム中に、特に相同的組換えにより組込む ことを意図した遺伝子、特に、細菌または真核細胞、真菌細胞またはウイルスに 由来するポリペプチドであって、前記ポリペプチドがワクチン特性を有するもの をコードする核酸配列を含有する「裸」のDNAよりなることを特徴とする請求 の範囲第1項から第8項のいずれか1項に記載の生成物。 13.該活性成分が、選択された細胞型、特にマクロファージ、樹状細胞、B およびT細胞および造血細胞、特にヒト由来のものを不死化しうることを特徴と する請求の範囲第1項から第8項のいずれか1項に記載の生成物。 14.前記活性成分が、例えばHIVレトロウイルスに感染した細胞または腫瘍 細胞において、タンパク質またはヌクレオチドの合成を抑制しうるアンチセンス オリゴヌクレオチドであることを特徴とする請求の範囲第1項から第8項のいず れか1項に記載の生成物。 15.好ましくは核で活性な分子を含有する真核細胞であって、前記分子が前 記細胞の核内に天然では取込まれることがないか、または前記細胞中で低い発現 レベルを有し、該分子が、これらの細胞内でそれらの核の間近またはそれらの核 内で見出され、請求の範囲第1項から第12項のいずれか1項に記載の生成物に 対応するこれらの細胞のDNAに対する親和性により特徴づけられるイムノベク ターにカップリングされていることを特徴とする真核細胞。 16.請求の範囲第15項に記載の細胞であって、これらがウイルスまたは腫 瘍細胞に感染しうることを特徴とする細胞。 17.CNCMに1995年6月30日に受託番号第I-1605号、第I-1606号および第I-16 07号として寄託されているハイブリドーマ。 18.選択された真核細胞の核内に該活性成分をインビトロで導入する方法で あって、前記活性成分をイムノベクターにカップリングすることによって特徴づ けられ、該イムノベクターがこれらの真核細胞における前記の生物学的に活性な 成分のインターナリゼーションを可能にする能力を有し、かつ、前記イムノベク ターがこれらの細胞の核の間近または該核内に前記の生物学的に活性な成分を輸 送できる程度に該細胞のDNAに対して親和性を有する方法。 19.該生物学的に活性な成分が、該イムノベクターに共有結合でまたは非共 有結合でカップリングされていることを特徴とする請求の範囲第8項に記載の方 法。 20.該生成物の組成物中に含まれるイムノベクターが、前記イムノベクター と結合しうる活性成分が進入しなければならない核において、細胞の存在下で、 関心のあるイムノベクターを第1インキュベーションに付し、ついで、該細胞を 固定し透過可能とした後、標識された抗イムノベクター抗体と共に更にインキュ ベートし、最後に、該核の間近または該核内においてさえも、該イムノベクター と該抗イムノベクター抗体との抗原−抗体免疫反応を検出することを具備する細 胞透過試験により選択可能なものから選ばれることを特徴とする請求の範囲第1 8項または第19項に記載の方法。 21.該イムノベクターが、核酸に対する親和性を有する抗体または前記親和 性を保持するこの抗体の断片より形成されることを特徴とする請求の範囲第16 項に記載の方法。 22.前記イムノベクターが、モノクローナルIgG、(Fab')2または(Fab')断片 、または対応する核酸の認識に関与する抗体の部位に対応する何れかのポリペプ チドから選ばれることを特徴とする請求の範囲第19項に記載の方法。 23.生理学的に許容しうる賦形剤と組み合わせて請求の範囲第1項から第9 項のいずれか1項に記載の生成物を含有することを特徴とする薬学的組成物であ って、該生物学的に活性な成分が医薬またはワクチン活性成分であり、該イムノ ベクターが、該医薬が向けられた宿主生物と適合しうることを特徴とする薬学的 組成物。 24.受容細胞における、前記宿主のDNAに対して異種であるヌクレオチド 配列のインビトロでの発現のための、請求の範囲第1項に記載の生成物の使用。
