Abstract
Description
本出願は、2003年12月12日に出願された米国仮出願第60/529,329号の優先権を主張するものであり、当該仮出願を参照してその全てを本明細書に取り込む。
本明細書において、本発明者らは、とりわけ、非VNTR領域の外側に存在し、癌治療における免疫療法にとって重要な、MUC−1の新規クラスI HLA−A2エピトープの同定について説明する。本発明者らは、IFN−γ産生によって測定したところ、これらのエピトープがヒトT細胞を活性化できることを明らかにしている。特に、アミノ酸第92〜101位がであるためP−92と命名された一つのエピトープATWGQDVTSV(配列番号1)が、HLA−A2にもっとも強いレベルで結合し、ヒトT細胞においてIFN−γを最も大量に誘導することが示された。本発明は、また、エピトープALWGQDVTSV(配列番号19;P−93Lと命名)の何れかに示すエンハンサーアゴニストエピトープの作成法も提供する。

*等級I〜IIの毒性が生じた場合、被験者は注入計画を続行することができる。
*等級IIIの毒性が生じた場合、毒性が等級I〜IIに低下するまで「薬剤」を留保し、その後注入を再開する。再開後、等級III又はIVの毒性が生じた場合、「薬剤]の注入を停止する。
*等級IVの毒性が生じた場合、被験者に等級IVの毒性が生じていることを記録し、次回注入はそれ以前の用量に減少させる。以前の用量で等級IVの毒性が生じた場合、「薬剤」を停止する。
*特定の投薬量で3人の被験者のうち1人に等級IVの毒性が生じた場合、さらに3人の被験者をその細胞用量レベルで参加させ、その用量レベルで、全部で6人の被験者としなければならない。ある細胞用量レベルで6被験者のうち2人が等級IVの毒性を表した場合、この用量は最大耐用量(MTD)と定義される。次の3被験者に前回細胞投与量の66%(3分の2)を投与する。用量増大を評価するため、各被験者に、6回の注入のうち少なくとも4回は、同じ用量レベルで投与しなければならない。
造血幹細胞及び前駆細胞を生体外で増殖させる方法は、参照されて本明細書に組み込まれる米国特許第5,199,942号に記載されている。その他にも適当な方法が当該技術分野公知である。簡単に言うと、生体外での培養及び増殖は、(1)CD34+造血幹細胞及び前駆細胞を患者の末梢血又は骨髄外植片から採集すること、そして(2)このような細胞を生体外で増殖させること、を含む。米国特許第5,199,942号に記載されている細胞増殖因子以外にも、flt3−L、IL−1、IL−3及びc−kitリガンドを用いることができる。
さまざまなサイトカインが、樹状細胞の生体外培養において役立つ。Flt3−Lは、参照されて本明細書に組み込まれるEP0627487A2及び国際公開第94/28391号に記載されているポリペプチドの属を意味する。ヒトflt3−LcDNAは、1993年8月6日に米国メリーランド州ロックビルにあるアメリカン・タイプ・カルチャー・コレクション(ATCC)に寄託され、ATCC69382という受託番号を付与された。IL−3は、参照されて本明細書に組み込まれる米国特許第5,108,910号に記載されているようなインターロイキン−3ポリペプチドの属を意味する。本発明において使用するのに適したヒトIL−3タンパク質をコードするDNA配列は、アメリカン・タイプ・カルチャー・コレクション(ATCC)から、受託番号ATCC 67747として公に入手できる。c−kitリガンドは、肥満細胞増殖因子(MGF)、造血幹細胞因子、又は幹細胞因子(SCF)とも呼ばれ、参照されて本明細書に組み込まれるEP 423,980に記載されている。その他有用なサイトカインには、インターロイキン−4(IL−4;Mosleyら、Cell 59:335(1989)、Idzerdaら、J.Exp.Med.171:861(1990)及びGalizziら、Intl.Immunol.2:669(1990)、これらは各々、参照されて本明細書に組み込まれる)及び顆粒球−マクロファージ・コロニー刺激因子(GM−CSF;米国特許第5,108,910号及び第5,229,496号に記載されており、これらは各々参照されて本明細書に組み込まれる)などがある。市販されているGM−CSF(サルグラモスチム、登録商標Leukine)は、ワシントン州、シアトルにあるImmunex社から入手可能である。さらに、GM−CSF/IL−3融合タンパク質(すなわち、GM−CSFとIL−3のC末端からN末端までの融合)も、樹状細胞の生体外培養において有用である。このような融合タンパク質は公知であり、米国特許第5,199,942号、第5,108,910号及び第5,073,627号に記載されており、これらは各々参照されて本明細書に組み込まれる)。好適な融合タンパク質は、米国特許第5,199,942号に記載されているPIXY321である。
MUC−1の非VNTRの一部をコードする核酸を改変するこ と、
改変した核酸を発現させて分子を産生させること、
樹状細胞を当該分子に接触させること、そして
T細胞を樹状細胞に接触させることとを含み、
T細胞によるIFN−γ産生の変調が、当該分子が免疫応答を生じさせる可能性があることを示す。
(実施例)
細胞培養物
ヒト乳房腺癌細胞株MCF−7(HLA−A2陽性及びMUC−1陽性)及びSK−Mel−24(HLA−A2陽性及びMUC−1陰性)をアメリカン・タイプ・カルチャー・コレクション(Manassas,バージニア州)から購入した。培養物は、マイコプラズマを含んでおらず、完全培地[10%のウシ胎児血清、2mMのグルタミン、100ユニット/mlのペニシリン及び100μg/mlのストレプトマイシン(Invitrogen Life Technologies,Inc)を添加したRPMI 1640(Invitrogen Life Technologies,Carlsbad,カリフォルニア州)]の中で維持した。C1R細胞株は、内在性のHLA−A又はHLA−B抗原を発現しないヒト血漿白血病細胞株である。C1R−A2細胞は、形質移入されたゲノムクローンであるHLA−A2.1を発現するC1R細胞である。これらの細胞は、Dr.William E.Biddison(国立神経疾患・脳卒中研究所(National Institute of Neurological Disorders and Stroke)、NIH,Bethesda,メリーランド州)から入手した。174CEM−T2細胞株(T2)の輸送欠失変異体は、Dr.Peter Cresswell(エール大学医学部(Yale University School of Medicine)、New Haven,コネチカット州)から提供された。C1R−A2細胞及びT2細胞はマイコプラズマを含まず、それぞれ、RPMI1640完全培地及びイスコフ改変ダルベッコ完全培地(Invitrogen Life Technologies)の中で維持した。
HLA−A2結合ペプチドの共通モチーフとの一致について、MUC−1のアミノ酸配列を調べた。Parker K.C.ら(J.Immunol.,152:163−175,1994)によって開発された、NIHのバイオインフォマティクス及び分子解析部門(BioInformatics and Molecule Analysis Section of NIH(BIMAS))のコンピューターアルゴリズムを用いたが、これは、ペプチド/MHC複合体の予測された解離半減期に従って、MHC結合ペプチドの可能性があるものをランク付けする。HLA−A2対立遺伝子が最も普通に発現するクラスI対立遺伝子であるため、それを選択した。非VNTR由来の10量体のペプチドが各共通モチーフに一致している場合には、それらを合成した。10量体のMUC−1ペプチドのパネル(表1)及びP−92MUC−1ペプチドのP1からP10の位置に一アミノ酸置換を有する類似体(図1参照)は純度>90%で、アメリカン・ペプチド・カンパニー社(American Peptide Company(Sunnyvale,カリフォルニア州))によって製造された。また、純度>96%のCEAペプチドであるCAP1−6D(28)は、アメリカン・ペプチド・カンパニー社(Sunnyvale,カリフォルニア州)によって製造された。
(i) 単色フローサイトメトリー解析
単色フローサイトメトリー解析法は、既述されている(Gaudagni F.ら、Cancer Res.:50:6248−6255,1990)。簡単に言えば、Ca2+及びMg2+を含まない冷たいDPBSで細胞を3回洗浄してから、HLA−A2(A2,28,One Lambda,Inc.,Canoga Park,カリフォルニア州)、CD3、CD4、CD8及びCD56(BD Biosciences,San Jose,カリフォルニア州)に対するmAbを1μg用いて、4℃で1時間染色した。ミネラルオイル形質細胞腫104E(Cappel/Organon Teknika Corp.,West Chester,ペンシルバニア州)をアイソタイプ対照として用いた。次いで、細胞を3回洗浄して、フルオレセインイソチオシアネート(FITC)標識したヤギ抗マウス免疫グロブリン(IgG)(Kirkegaard and Perry Laboratories,Gaithersburg,メリーランド州)の1:100の希釈液を用いてインキュベートした。488nmで15nWの励起を有するブルーレーザーを備えたBecton Dickinson FACScanを用いて、細胞を直ちに解析した。10,000個の生細胞からデータを収集し、保存して、結果を生成するために使用した。
2色フローサイトメトリー解析の手順は、以下の例外を除いては、単色フローサイトメトリー解析と同様であった。抗MHCクラスII FITC/抗CD11c PE、抗MHCクラスII FITC/抗CD80 PE、抗CD58 FITC/抗CD54 PE、抗MHCクラスI FITC/抗MHCクラスII PE及び抗IgG1 FITC/抗IgG2a PE(アイソタイプ対照)を用いて樹状細胞を解析し、>96%の樹状細胞がCD11c及びMHCクラスII陽性であった。
P−92及びP−92類似体のHLA−A2分子への結合を、フローサイトメトリーにより示される、T2A2細胞への結合によって評価した。本アッセイにおいて、ペプチド結合の結果、T2細胞の表面上にあるHLA−A2分子の安定性が増大(蓄積)することを、HLA−A2分子に対する抗体結合の増加により測定する。要約すれば、無血清イスコフ改変ダルベッコ完全培地中1×106個の細胞を、濃度50μg/mlのペプチドとともに、5%CO2中、24ウェル培養プレート内において、37℃でインキュベートした。T2細胞及び単色解析法を用いて、ペプチド結合についてフローサイトメトリーを行った。上記したように、細胞をDPBSで3回洗浄した後、細胞106個につきHLA−A2特異的なMAb(One Lambda,Inc.)の1:100希釈液を用いて1時間インキュベートした。アイソタイプ対照としてUPC−10(Cappel/Organon Teknika)を用いた。次いで、細胞を3回洗浄して、FITC標識した抗マウスIgG(BD Biosciences)の1:100希釈液を用いてインキュベートした。上記したように、FACScanで解析を行った。細胞の調製及び染色の全過程で、細胞を氷上に置いた。
ヘパリン処理した血液から、HLA−A2の正常なドナーであるPBMCを採取した。既述されているように(Boyum,A.Scand.J.Clin.Lab.Invest.97(Suppl):51−76,1968)、リンパ球分離用媒体勾配(Organon Teknika,Durham,ノースカロライナ州)を用いて、PBMCを分離した。Sallustoら(J.Exp.Med.,179:1109−1118,1994)に記載されている手順の変法を用いてDCを調製した。グルタミン2mM、ストレプトマイシン50μg/ml及びゲンタマイシン10μg/ml(Invitrogen Life Technologies,Inc.)を含むAIM−V培地にPBMC(1.5×108)を再懸濁して、T−150フラスコ(Corning Costar Corp.,Cambridge,マサチューセツ州)に付着させた。37℃で2時間置いた後、非付着細胞を静かに洗い流した。付着細胞は、100ng/mlの組換えヒトGM−CSF(rhGM−CSF)及び20ng/mlの組換えヒトIL−4(rhIL−4)を含むAIM−V培地の中で6〜7日間培養した。3日ごとに培養培地を補充した。
Jenkins S.ら、AIDS Res.Hum.Retroviruses,7:991−998,1991に記載された通りに、組換え鶏痘ウイルスを作成した。この組換えウイルスの母体ウイルスとして、鶏痘ウイルスのPOXVAC−TCワクチン株からプラーク精製した単離株を用いた。MUC−1、LFA−3、ICAM−1及びB7−1の配列を、鶏痘ウイルスゲノムのBamHI、J領域に挿入した。さらに、鶏痘C1プロモーターの制御下にlacZ遺伝子を含ませて,β−ガラクトシダーゼの発色アッセイ法を用いて、組換えウイルスを同定して単離した。鶏痘ウイルスに挿入したMUC−1遺伝子は、天然のMUC−1遺伝子から変異している。それは、30アミノ酸のシグナル配列、続いてMUC−1タンパク質の成熟型N末端配列の最初の38アミノ酸、20アミノ酸反復配列の10の同一コピー及びこのタンパク質のC末端部分をコードする。各反復配列中に、コードされたアミノ酸を変更しない多数の第3塩基変異を有するコドンを含む重複合成ヌクレオチドを用いて、600 bpの反復配列を作成した。これは、反復アミノ酸配列を維持する際に、ポックス・ウイルスで不安定な重複ヌクレオチド配列を最小限に抑えるために行われた。
感染したDCを、100ng/mlのrhGM−CSF、20ng/mlのrhIL−4及び20ng/mlのTNF−αを含む10mlの新鮮で温かいRPMI−1640完全培地中に懸濁し、24時間培養してから、APCとして用いた。
Tsang K.Y.ら、J.Natl.Cancer Inst.,87:982−990,1995に記載されたプロトコルの変法を用いて、MUC−1特異的CTLを作成した。T細胞株T−1191−P−93L及びT−1191−P−92を作成するために、rF−MUC−1/TRICOMに感染させた自己樹状細胞をAPCとして用いた。次いで、10:1のエフェクター対APCの比にして、自己の非接着性細胞をAPCに添加した。そして、CO2を5%含有する加湿した雰囲気中、37℃で3日間、培養物をインキュベートした。次いで、7日間、濃度20単位/mlの組換えヒトIL−2を培養物に添加し、IL−2含有培地を3日毎に補充した。ペプチドとともに3日間インキュベートすることと、7日間のIL−2補充とが、1回のIVSサイクルを構成した。11日目に、rF−MUC−1/TRICOMに感染させた自己DCによって、上記したように一次培養物を再刺激して、次のIVSサイクルを開始させた。rF−MUC−1/TRICOMに感染した自己DCを、3回のIVSサイクルでAPCとして用いた。3回目のIVSサイクルの後、(23,000ラド)照射した自己EBV形質転換B細胞をAPCとして用いた。EBV形質転換B細胞で再刺激するには、濃度50mg/mlのペプチドを用いて、エフェクター対APCの比1:3で再刺激のために自己EBV形質転換B細胞をパルスした。次いで、CO2を5%含んだ加湿雰囲気中、37℃で3日間培養物をインキュベートした。ペプチドを含んだ培地を除去してから、濃度20単位/mlの組換えヒトIL−2を培養液に数日間添加した。上記と同一の刺激プロトコルを用いて、P−92ペプチド又はP93Lペプチドでパルスされた自己DCによってPBMCを刺激することにより、患者18及び23からT細胞株を(T−18−P−92、T−18−P93L、T−23−P−92及びT−23−P93L)作成した。DCの解析及び同定に用いたマーカーは、CD11c、MHC−クラスII、CD80、CD54、CD58及びCD83であった。CD3も陰性マーカーとして用いた。
標的細胞(C1R−A2又は腫瘍細胞)を、50μCiの111インジウム標識したオキシキノリン(Medi−Physics Inc.,Arlington,イリノイ州)を用いて室温で15分間標識した。100μlのRPMI−1640完全培地中の標的細胞(0.3×104個)を、平底測定プレート(Corning Costar Corp)の96ウェルのそれぞれに添加した。標識したC1R−A2標的細胞を、エフェクター細胞を添加する前に、5%のCO2中において37℃で60分間、表示した濃度のペプチドとともにインキュベートした。標的として癌腫細胞を用いる場合には、ペプチドを用いなかった。エフェクター細胞を、10%のプールされたヒトAB型血清を添加したRPMI1640完全培地100μlに懸濁して、標的細胞に添加した。そして、プレートを、37℃で4時間又は16時間、5%のCO2中においてインキュベートした。採取器(harvester frames:Skatron,Inc.,Sterling,バージニア州)を用いて上清を回収して、ガンマ線計数を行った。測定は3回反復して行ない、標準偏差を計算した。以下の公式を用いて特異的溶解を計算した(値はすべてcpm)。

自発的放出量は、100μlのRPMI−1640完全培地を添加したウェルから測定した。放出可能な全放射活性量は、2.5%のTriton X−100で標的を処理して得られた。
さまざまなペプチド濃度でIL−2を含まない培地中において、ペプチドをパルスされた自己EBV形質転換B細胞に24時間曝露されたT細胞の上清を、ELISAキット(R&D Systems,Minneapolis,ミネソタ州)を用いて、INF−γの分泌についてスクリーニングした。結果をpg/mlで表示した。CBA装置(BD PharMingen,San Diego,カリフォルニア州)を用いて、特異T細胞による複数のサイトカインの分泌も測定した。CBA装置は、フローサイトメトリーによる蛍光検出を利用して、粒子による免疫アッセイ法において可溶性検体を測定する。BDのヒトTh1/Th2サイトカインCBAキットを用いて、単一サンプル中のIL−2、IL−4、IL−5、IL−10、TNF−aタンパク質のレベルを測定した。サイトカイン捕捉用ビーズを、PE結合検出抗体と混合してから、組換えサイトカイン標準、又は試験試料とともにインキュベートして、サンドウィッチ複合体を形成させた。BD PharMingen CBA解析ソフトウェアを用いて、グラフィック及び表形式で、サンプルの結果を作成した。結果はpg/mlで表示した。
両側ペアt検定(Stat View statistical software,Abacus Concepts,Berkeley,カリフォルニア州)を用いて、平均値間の差を統計的に解析した。

*特定の投薬量で3人の被検者のうち1人に等級IVの毒性が生じた場合、さらに3人の被検者をその細胞用量レベルで参加させ、その用量レベルで、全部で6人の被検者としなければならない。ある細胞用量レベルで6被検者のうち2人が等級IVの毒性を示した場合、この用量はMTDと定義される。次の3被検者に前回細胞投与量の66%(3分の2)を投与する。用量増大を評価するため、各被検者に、6回の注入のうち少なくとも4回は、同じ用量レベルで投与しなければならない。
ヒトMUC−1の一次アミノ酸配列を、新規HLA−A2結合ペプチドの共通モチーフについて解析した。12個の10量体ペプチドを同定し、その結果合成して、T2細胞結合アッセイにおいてHLA−A2分子との結合を調べた。アミノ酸配列及びこれらの10量体ペプチドの位置を表1に示す。CEA CAP1−6Dペプチド及びNCAペプチドを、それぞれ陽性対照及び陰性対照として用いた。12個のペプチドの予測される結合も表1に示す。これらのペプチドのうち3つ(P−92、P−94及びP−1108)が、T2アッセイにおいて最高レベルの結合を有することを示した。
次いで、外見上健常なドナーのPBMCからMCU−1特異的T細胞株を樹立することができるかを決定するために実験を行った。これを行うために、rF−MUC−1/TRICOMに感染した自己DCをAPCとして用いた。rF−MUC−1/TRICOMは、MUC−1及び三連構造ヒト共刺激分子(TRICOMと命名された、B7−1、ICAM−1及びLFA−3)に相当する導入遺伝子を含む複製欠損アビポックスベクターである。rF−MUC−1/TRICOMは、ヒトDCに効率的に感染して、MUC−1同様、各共刺激分子をDC表面上に過剰発現させることが示された(表2)。細胞の約96%がCD11c及びMHC−クラスII陽性であった。
P−92の2位及び10位における一次及び二次のHLA−A2アンカーアミノ酸残基の解析により、これらの位置におけるアミノ酸の改変が、ペプチドのHLA−A2分子への結合能力を強化しうることが明らかになった。従って、表4に示すように、P−92の6つの異なる類似体を合成し、天然のP−92ペプチドとともに、T2細胞への結合能力について試験を行った。CEAのCAP−7(HLA−A3結合ペプチド)は、HLA−A2に結合しないことが既に示されているので、陰性対照として用いた。表4に示したように、6つの類似体ペプチドのうち4つが、P−92ペプチドよりも高レベルでHLA−A2に結合した。類似体P−93L及びP−93Iは、もっとも効率的にHLA−A2に結合した。
ペプチドP−92(天然型)、P−93L及びP−93Iに関するペプチド−MHC複合体の安定性を調べた。ペプチドをT2細胞とともに一晩インキュベートし、未結合ペプチドを洗い落としてから、ブレフェルジン−Aとともにインキュベートして、新しいクラスI分子が細胞表面に送達されるのをブロックした。さまざまな時点で、ペプチド−HLA−A2複合体の存在について細胞を解析した。図2に示したように、P−93L−HLA−A2複合体及びP−93I−HLA−A2複合体は、8時間にわたる観察時間に於いて、P−92−HLA−A2複合体よりも安定していたが、P−93L−HLA−A2複合体の方が、同じ時間に於いて、P−93I−HLA−A2複合体よりわずかに安定していた。従って、P−93L及びP−93Iの方が、天然のP−92ペプチドより、ペプチドのMHCへの結合性及びペプチド−MHC複合体の安定性ともに勝っていることが示された。
次に、試験を行って、これら2つのT細胞株が、MUC−1陽性及びHLA−A2陽性の乳癌細胞株MCF−7を溶解することができるかを測定した。MUC−1陰性でHLA−A2陽性のSK−Mel−24メラノーマ細胞株を陰性対照として用いた。表5に示したように、MCF−7細胞は、T−1191−P−92細胞及びT−1191−P−93L細胞によって溶解された。SK−Mel−24細胞に対する溶解は観察されなかった。T−1191−P−93L細胞は、T−1191−P−92細胞株と比較して、MCF−7細胞をより大きく溶解することが示された。CEA CAP1−6D対照用ペプチドではなく、対応するMUC−1ペプチドでパルスされた非標識C1R−A2細胞を添加すると、両方のT細胞株の細胞傷害活性が低下して、溶解のMUC−1特異性を示した(表5)。抗HLA−A2抗体を添加すると溶解が阻害され、対照用抗体UPC−10では阻害されなかったことから明らかなように、MCF−7細胞に対するこれらのT細胞株の細胞傷害活性も、HLA−A2拘束性であることが示された(表6)。
次に、癌患者23から得たT細胞株が、MUC−1陽性HLA−A2陽性の癌細胞を溶解することができるか否かを決定するために実験を行った。SK−Mel−24細胞株(MUC−1陰性及びHLA−A2陽性)を、特異性についての陰性対照として用いた。表9及び10に見られるように、T−23−P−93L細胞株、T−23−P−92細胞株、T−18−P−93L細胞株、及びT−18−P−92細胞株は、2つの異なるE:T比でMCF−7癌細胞の溶解を示したが、メラノーマ細胞株の溶解は認められなかった。上記の結果に従って、アゴニストペプチドを用いて得られたT細胞株(T−23−P−93L、T−18−P−93L)は、天然ペプチドを用いて得られたT細胞株(T−23−P−92、T−18−P−92)よりも優れた腫瘍細胞溶解を示した。これは、2つの異なったE:T比で見られた。
解析した2つのウイルスベクターは、複製可能な組換えワクシニアウイルス(rV−)及びアビポックスベクターである鶏痘(rF−)であり、後者は哺乳動物細胞において複製不能である。各ベクターは、3種類のヒト共刺激分子(TRICOMと名づけられたB7−1、ICAM−1、LFA−3)に対する導入遺伝子をコードし、CEA及びMUC−1の両導入遺伝子は、アゴニスト・エピトープも含んでいる。ベクターをrV−CEA/MUC/TRICOM及びrF−CEA/MUC/TRICOMと命名した。
ヒト乳房腺癌細胞株MCF−7(HLA−A2陽性、CEA陰性及びMUC−1陽性)、結腸直腸癌細胞株SW1463(HLA−A2陽性、CEA陽性及びMUC−1陽性)及びメラノーマ細胞株SK−Mel−24(HLA−A2陽性、CEA陰性及びMUC−1陰性)をアメリカン・タイプ・カルチャー・コレクション(Manassas,バージニア州)から購入した。培養物は、マイコプラズマを含んでおらず、完全培地[10%のウシ胎児血清、2mMのグルタミン、100ユニット/mlのペニシリン及び100μg/mlのストレプトマイシン(Invitrogen Life Technologies)を添加したRPMI 1640(Invitrogen Life Technologies,Carlsbad,カリフォルニア州)]の中で維持した。C1R細胞株は、内在性のHLA−A又はHLA−B抗原を発現しないヒト血漿白血病細胞株である。C1R−A2細胞は、形質移入されたゲノムクローンであるHLA−A2.1を発現するC1R細胞である。これらの細胞は、Dr.William E.Biddison(国立神経疾患・脳卒中研究所(National Institute of Neurological Disorders and Stroke)、NIH,Bethesda,メリーランド州)から入手した。174CEM−T2細胞株(T2)の輸送欠失変異体は、Dr.Peter Cresswell(エール大学医学部(Yale University School of Medicine)、New Haven,コネチカット州)から提供された。C1R−A2細胞及びT2細胞はマイコプラズマを含まず、それぞれ、RPMI1640完全培地及びイスコフ改変ダルベッコ完全培地(Invitrogen Life Technologies)の中で維持した。V8T細胞株は、CEAのCAP−1エピトープに対するCD8+CTL細胞株である。T−1191−P93L細胞株は、MUC−1ペプチドを用いてインビトロにおいて刺激された健常なドナー由来の末梢血単核細胞(PBMC)から作成されたCD8+MUC−1特異的CTL細胞株である。V8T細胞株及びT−1191−P93L細胞株は両方とも、既述したとおりに培養した。
使用したHLA−A2 結合ペプチドは、以下のものを含んでいた。すなわち、(a)CEAペプチドと命名されたCEAアゴニストペプチドCAP1−6D(YLSGADLNL)、(b)MUC−1ペプチドと命名されたMUC−1アゴニストペプチドP−93L(ALWGQDVTSV)、(c)前立腺特異的抗原(PSA)ペプチドPSA−3(VISNDVCAQV)。すべてのペプチドは96%よりも高い純度で、アメリカン・ペプチド・カンパニー社(American Peptide Company,Inc,(Sunnyvale,カリフォルニア州))によって製造された。
HLA−A2の正常なドナーPBMCを、ヘパリン化血液から得た。既述されているとおりに(51)、(Boyum A.「ヒト血液から顆粒球及びリンパ球を単離するための一段階処理法。1gの重力場における白血球細胞の一般的な沈降特性(A one−stage procedure for isolation of granulocytes and lymphocytes from human blood.General sedimentation properties of white blood cells in 1 g gravity field)、 Scand J Clin Lab Invest 1968;97(Suppl):51−76)、リンパ球分離媒体勾配(Organon Teknika,Durham,ノースカロライナ州)を用いてPBMCを分離した。Sallustoら(Sallusto F,Lanzavecchia A.、培養ヒト樹状細胞による可溶性抗原の効率的な提示は、顆粒球/マクロファージ・コロニー刺激因子とインターロイキン−4によって維持され、腫瘍壊死因子αによって下方制御される(Efficient presentation of soluble antigen by cultured human dendritic cells is maintained by granulocyte/macrophage colony stimulating factor plus interleukin−4 and down−regulated by tumor necrosis factor alpha)、J.Exp.Med.,179:1109−1118,1994))によって説明されている手順の変法を用いてDCを調製した。2mMのグルタミン、50μg/mlのストレプトマイシン及び10μg/mlのゲンタマイシン(Invitrogen Life Technologies,Inc.)を含むAIM−V培地にPBMC(1.5×108)を再懸濁して、T−150フラスコ(Corning Costar Corp.,Cambridge,マサチューセツ州)に付着させた。37℃で2時間経過後、非付着細胞を静かに洗い流した。100ng/mlの組換えヒトGM−CSF(rhGM−CSF)及び20ng/mlの組換えヒトIL−4(rhIL−4)を含むAIM−V培地の中で6〜7日間、付着細胞を培養した。3日ごとに培養基を補充した。
rV−CEA/MUC/TRICOM及びrF−CEA/MUC/TRICOMは、6Dの変更を含むヒトCEA遺伝子、93Lの変更を含むヒトMUC−1遺伝子並びにヒト共刺激分子B7−1、ICAM−1及びLFA−3をコードする(図5)(Zaremba S,Barzaga E,Zhu Mら、ヒト癌胎児性抗原由来のエンハンサーアゴニストである細胞傷害性Tリンパ球ペプチドの同定(Identification of an enhancer agonist cytotoxic T lymphocyte peptide from human carcinoembryonic antigen)、Cancer Res 1997;57:4570−7,及びTsang KY,Palena C,Gulley J,Arlen P,Schlom J.ヒト細胞傷害性Tリンパ球エピトープ、及びそのMUC−1の非VNTR由来のアゴニストエピトープ(A human cytotoxic T−lymphocyte epitope and its agonist epitope from the non−variable number of tandem repeat sequence of MUC−1)、 Clin Cancer Res 2004;10:2139−49))。(Hodge JW,McLaughlin JP,Kantor JA,Schlom J.;Diversified prime and boost protocols using recombinant vaccinia virus and recombinant nonreplicating avian pox virus to enhance T−cell immunity and antitumor responses;Vaccine 1997;16:759−68)に記載されているように、相同的組換えによって組換えベクターを作成した。DC(1×106個)を、1mlのOpti−MEM培地(Invitrogen Life Technologies)中で、rF−CEA/MUC/TRICOM、rV−CEA/MUC/TRICOM、対照のアビポックスウイルスベクター(FP−WT)又は対照のワクシニアベクター(V−WT)とともに37℃でインキュベートした。滴定実験によって、40pfu/細胞の感染多重度(MOI)に等しい、4×104プラーク形成単位(pfu)/mlのrF−CEA/MUC/TRICOMを2時間DCに感染させると、約60%の感染DCの中で導入遺伝子の発現を定常的に誘導できることが実証された。同様の滴定実験によって、5pfu/細胞のMOIに等しい、0.5×107pfu/mlのrV−CEA/MUC/TRICOMを1時間DCに感染させると、約35%の感染DCの中で導入遺伝子の発現を定常的に誘導できることが実証された。rF−CEA/MUC/TRICOMについては50%〜65%の感染効率、rV−CEA/MUC/TRICOMについては30%〜59%の感染効率で、rF−CEA/MUC/TRICOM及びrV−CEA/MUC/TRICOMを感染させるのに、さまざまなドナー由来のDCを用いた。100ng/mlのrhGM−CSF及び20ng/mlのrhIL−4を含む新鮮で温かいRPMI−1640完全培地10mlに感染したDCを懸濁し、24時間培養してから、APCとして用いた。
以下の抗体の組み合わせを用いてT細胞株上で2色フローサイトメトリー解析を行った。抗CD56−FITC/抗CD8−PE、抗CD8−FITC/抗CD45RA−FITC及び抗CD8−FITC/抗CD27−PE。抗体はすべてBDバイオサイエンス社(San Jose,カリフォルニア州)から購入した。4℃で1時間、同時に染色を行ってから、Ca2+及びMg2+を含まない冷たいリン酸緩衝生理食塩水(PBS)で細胞を洗浄し,同じ緩衝液に再懸濁して、FACScan及びCELLQuestプログラム(BD Biosciences)を用いて直ちに解析した。10,000個の生細胞からデータを収集し、保存し、結果を作成するために用いた。DCを解析する手順は、上記した手順と同じであった。以下の抗体の組み合わせを用いた。すなわち、抗MHC−クラスII−FITC/抗CD80−PE、抗CD58−FITC/抗CD54−PE、抗MHCクラスI−FITC/抗MHCクラスII−PE及び抗IgG1−FITC/抗IgG2a−PE(アイソタイプ対照)。MHCクラスI及びクラスIIに対する抗体は、Serotec社(オックスフォード,イギリス)から購入し、他の抗体はBD Bioscience社から購入した。抗CEAモノクローナル抗体COL−1及び抗MUC−1抗体(DF−3及びDF−3P)も用いた(Muraro R,Wunderlich D,Thor A.ら、ヒト結腸癌腫対正常な成人組織で異なって発現される癌胎児性抗原上にある同じレパートリーのエピトープのモノクローナル抗体による定義(Definition by monoclonal antibodies of a repertoire of epitopes on carcinoembryonic antigen differentially expressed in human colon carcinoma versus normal adult tissues) Cancer Res 1985;45:5769−80)。MOPC−104E(IgM)(Cappel/Organon Teknika Corp.,West Chester,ペンシルバニア州)を陰性対照として用いた。
MPER哺乳動物タンパク質抽出用試薬(Pierce,Rockford,イリノイ州)を用いて、非感染DC、40MOIのrF−CEA/MUC/TRICOMベクター、rF−CEA(6D)−TRICOMベクター、又はrF−MUC−1/TRICOMベクターに感染させたDC及び5MOIのrV−CEA/MUC/TRICOMベクターに感染させたDCを溶解した。MicroBCAタンパク質アッセイ用キット(Pierce)を用いて、溶解物のタンパク質濃度を測定し、Bio−Dot精密濾過装置(BioRad Laboratories,Hercules,カリフォルニア州)を製造業者の指示に従って用いて、1サンプル当たり20μgのタンパク質画分をPVDF膜12の上にブロットした。ブロットした後、BSA(Biosource Internationa,Camarillo,カリフォルニア州)を5%含むPBSを用いて、室温で1時間、膜をブロックした。次いで、0.25%のTween−20を含むPBSを用いて膜を3回洗浄してから、1μg/mlのCOL−1抗体、DF−3抗体、又はDF3−P抗体の溶液を用いて、室温で2時間インキュベートした。そして、上記したように膜を3回洗浄してから、HRPに結合した抗マウスIgG(Kirkegaard & Perry Laboratories)の1:3000希釈液を用いて室温で1時間インキュベートした。CEA及びMUC−1タンパク質の免疫検出には、Super Signal West Pico Chemiluminescent Substrate(Pierce)を用いた。
Tsangらに記載されたプロトコルの変法を用いて、CEA及び/又はMUC−1に特異的なCTLを作成した(Tsang KY,Zaremba S,Nieroda CAら.組換えワクシニアCEAワクチンによって免疫された患者由来のヒト癌胎児性抗原に特異的なヒト細胞傷害性T細胞の作成(Generation of human cytotoxic T cells specific for human carcinoembryonic antigen epitopes from patients immunized with recombinant vaccinia−CEA vaccine)J Natl Cancer Inst 1995;87:982−90)。T細胞株T−rV及びT−rFを作成するために、それぞれrV−CEA/MUC/TRICOM又はrF−CEA/MUC/TRICOMに感染させた自己DCをAPCとして用いた。10:1のエフェクター対APCの比にして、自己の非接着性細胞をAPCに添加した。そして、CO2を5%含有する加湿した雰囲気中、37℃で3日間、培養物をインキュベートした。次いで、7日間、濃度20単位/mlの組換えヒトIL−2を培養物に添加し、IL−2含有培地を3日毎に補充した。ペプチドとともに3日間インキュベートすることと、7日間のIL−2補充とが、1回のインビトロ刺激(IVS)サイクルを構成した。上記したように、rV−CEA/MUC/TRICOM又はrF−CEA/MUC/TRICOMに感染した自己DCで、それぞれT−rV及びT−rFを再刺激し、11日目に次のIVSサイクルを開始した。rV−CEA/MUC/TRICOM及びrF−CEA/MUC/TRICOMに感染した自己DCを、3回のIVSサイクルの間APCとして用いた。T−rF(CEA)細胞株及びT−rF(MUC)細胞株を作成するために、1回のIVSの間、rF−CEA/MUC/TRICOMを感染させた自己DCでT細胞を刺激してから、さらに2回のIVSの間、それぞれCAP1−6Dペプチド又はP−93Lペプチドでそれぞれぞれパルスした非感染の自己DCを用いて再刺激した。3回目のIVSの後、(23,000ラド)照射した自己EBV形質転換B細胞をAPCとして用いた。EBV形質転換B細胞を25μg/mlのペプチドでパルスして、エフェクター対APCの比率1:3で再刺激に用いた。
標的細胞(C1R−A2又は腫瘍細胞)を、50μCiの111インジウム標識したオキシキノリン(Medi−Physics Inc.,Arlington,イリノイ州)を用いて室温で15分間標識した。100μlのRPMI−1640完全培地中の標的細胞(0.3×104個)を、平底測定プレート(Corning Costar Corp)の96ウェルのそれぞれに添加した。標識したC1R−A2標的細胞を、エフェクター細胞を添加する前に、5%のCO2中において37℃で60分間、表示した濃度のペプチドとともにインキュベートした。標的として癌腫細胞を用いる場合には、ペプチドを用いなかった。エフェクター細胞を、10%のプールされたヒトAB型血清を添加したRPMI1640完全培地100μlに懸濁して、標的細胞に添加した。そして、プレートを、6時間又は16時間、5%のCO2中においてインキュベートした。採取器(Skatron,Inc.,Sterling,バージニア州)を用いて上清を回収して、ガンマ線計数を行った。測定は3回反復して行ない、標準偏差を計算した。以下の公式を用いて特異的溶解を計算した(値はすべてcpm)。

自発的放出量は、100μlのRPMI−1640完全培地を添加したウェルから測定した。放出可能な全放射活性量は、2.5%のTritonX−100で標的を処理して得られた。
さまざまなペプチド濃度でIL−2を含まない培地中において、ペプチドをパルスされた自己EBV形質転換B細胞に24時間曝露されたT細胞の上清を、ELISAキット(Biosource International)を用いて、INF−αの分泌についてスクリーニングした。結果をpg/mlで表示した。
両側ペアt検定(Stat View statistical software,Abacus Concepts,Berkeley,カリフォルニア州)を用いて、平均値間の差を統計的に解析した。
これは、インビトロで培養した場合、MUC−1が、ヒトのDC及び単核球由来のDC上で発現することを示した、Wykesらの報告(Wykes M,MacDonald KP,Tran Mら.MUC1上皮ムチン(CD227)は、活性化樹状細胞によって発現される(MUC1 epithelial mucin(CD227) is expressed by activated dendritic cells)、J Leukoc Biol 2002;72:692−701)と一致している。
上記のように、DCは低レベルのMUC−1を発現したが、rF−MUC−1−TRICOM、rV−CEA/MUC/TRICOM、及びrF−CEA/MUC/TRICOMに感染したDCでは、MUC−1発現の大きな増加が明確に見られ、一方、rF−CEA(6D) TRICOMに感染したCDではこの増加は見られなかった(図8)。

- 単離された核酸分子が、配列番号1〜19の何れかに示すアミノ酸配列に少なくとも約90%の相同性を有するポリペプチドをコードする、請求項31に記載の方法。
- 単離された核酸分子が、配列番号1〜19の何れかに示すアミノ酸配列に少なくとも約99.9%の相同性を有するポリペプチドをコードする、請求項31に記載の方法。
- 単離された核酸分子が、配列番号1〜19の何れかに示すアミノ酸配列の一つ又はそれ以上をコードするベクターを含有してなる、請求項31に記載の方法。
- 単離された核酸分子が、配列番号19に示すポリペプチドをコードするベクターを含有して成る、請求項31に記載の方法。
- MUC−1腫瘍に対して免疫応答が惹起される、請求項37に記載の方法。
- 免疫応答が細胞傷害性T細胞応答である、請求項31に記載の方法。
- 配列番号1〜19の何れか一つ又はそれ以上に示すポリペプチドをコードする一つ又はそれ以上の、誘導型プロモーターに作動可能に連結された核酸配列を含有して成る、核酸ベクター。
- ベクターがウイルスベクターである、請求項40に記載の核酸ベクター。
- ベクターがプラスミドである、請求項40に記載の核酸ベクター。
- 誘導型プロモーターが組織特異的である、請求項40に記載の核酸ベクター。
- 配列番号1〜19に示すポリペプチドの何れか一つをコードする核酸配列を含有してなる、組換えベクター。
- 請求項40〜44の何れか一項に記載のベクターを含有してなる、宿主細胞。
- 配列番号1〜19に示すペプチドの何れか一つ又はそれ以上を患者に投与することを含有してなる、MUC−1腫瘍を患っているか、又はそれに罹患する恐れのある患者を治療する方法。
- 配列番号1〜19に示するアミノ酸配列の何れか一つ又はそれ以上に、少なくとも約60%の相同性を有するペプチドを投与することにより患者を治療する、請求項46に記載の方法。
- 配列番号1〜19に示すアミノ酸配列の何れか一つ又はそれ以上に、少なくとも約80%の相同性を有するペプチドを投与することにより患者を治療する、請求項46に記載の方法。
- 配列番号1〜19に示すアミノ酸配列の何れか一つ又はそれ以上に、少なくとも約90%の相同性を有するペプチドを投与することにより患者を治療する、請求項46に記載の方法。

- 癌を患っている患者から樹状細胞を単離し、
配列番号1〜19に示す一つ又はそれ以上のポリペプチドで樹状細胞を処理し、そして
処理した樹状細胞を患者に投与する、
ことを含有して成る、MUC−1腫瘍を患っているか、又はそれに罹患する恐れのある患者を治療する方法。 - 配列番号1〜19の何れかに示すアミノ酸配列に、少なくとも約60%の相同性を有する一つ又はそれ以上のポリペプチドで樹状細胞を処理する、請求項51に記載の方法。
- 配列番号1〜19の何れかに示すアミノ酸配列に、少なくとも約80%の相同性を有する一つ又はそれ以上のポリペプチドで樹状細胞を処理する、請求項51に記載の方法。
- 配列番号1〜19の何れかに示すアミノ酸配列に、少なくとも約90%の相同性を有する一つ又はそれ以上のポリペプチドで樹状細胞を処理する、請求項51に記載の方法。
- 配列番号1〜19の何れかに示すアミノ酸配列に、少なくとも約99.9%の相同性を有する一つ又はそれ以上のポリペプチドで樹状細胞を処理する、請求項51に記載の方法。
- 弱い免疫原を結合したHLAに高い親和力を示すポリペプチドを患者に投与することを含有して成る、弱い免疫原性の抗原に対する免疫応答を惹起させる方法。
- 弱い免疫原が分化抗原である、請求項56に記載の方法。
- 弱い免疫原が腫瘍抗原である、請求項56に記載の方法。
- ポリペプチドが、配列番号19に示すHLAが結合している断片を含有して成る、請求項56に記載の方法。

- 配列番号19に示すHLAが結合している断片がウイルス抗原を結合する、請求項59に記載の方法。

- 配列番号1又は3〜18の何れかに示すアミノ酸配列に対応する核酸配列又はそれらの断片若しくは変異体を含有して成る、免疫応答を刺激する、MUC−1の非VNTR領域由来のアゴニストポリペプチド抗原をコードする単離された核酸分子。
- MUC−1の非VNTRの一部をコードする核酸を改変すること、
改変した核酸を発現させて分子を産生し、
樹状細胞を前記分子と接触させ、そして
T細胞を前記樹状細胞と接触させ、
T細胞に於けるIFN−γ産生の変調が、当該分子が免疫応答を起こすことが可能であることを示す、
ことを含有して成る、MUC−1腫瘍抗原に対する免疫応答を惹起させる分子をスクリーニングする方法。 - 樹状細胞が、癌と診断された患者由来のものである、請求項64に記載の方法。
- 樹状細胞を、前記分子で処理した後に末梢血単核細胞と接触させる、請求項64に記載の方法。
- 癌を患っている患者から樹状細胞を単離し、
樹状細胞を配列番号1〜19の何れかに示すポリペプチドで処理し、
処理した樹状細胞で末梢血単核細胞を活性化し、そして
活性化されたPBMC細胞を患者に投与する、
ことを含有してなる、MUC−1腫瘍を患っているか、又はそれに罹患する恐れのある患者を治療する方法。 - 樹状細胞を配列番号1〜19の何れかに示すアミノ酸配列のいずれか一つに対して少なくとも約60%の相同性を有する一つ以上のポリペプチドで処理する、請求項67に記載の方法。
- 樹状細胞を配列番号1〜19の何れかに示すアミノ酸配列に、少なくとも約80%の相同性を有する一つ又はそれ以上のポリペプチドで処理する、請求項67に記載の方法。
- 樹状細胞を配列番号1〜19の何れかに示すアミノ酸配列に、少なくとも約90%の相同性を有する一つ又はそれ以上のポリペプチドで処理する、請求項67に記載の方法。
- 樹状細胞を配列番号1〜19の何れかに示すアミノ酸配列に、少なくとも約99.9%の相同性を有する一つ又はそれ以上のポリペプチドで処理する、請求項67に記載の方法。
- 配列番号20〜37の何れかに示す単離された核酸分子の、細胞性免疫応答を生じさせるのに十分な治療有効量を投与することを含んで成る、MUC−1腫瘍抗原に対する免疫応答を惹起させる方法。
- 単離された核酸分子が、配列番号20〜37の何れかに示すアミノ酸配列に、少なくとも約60%の相同性を有するポリペプチドをコードする、請求項72に記載の方法。
- 単離された核酸分子が、配列番号20〜37の何れかに示すアミノ酸配列に、少なくとも約80%の相同性を有するポリペプチドをコードする、請求項72に記載の方法。
- 単離核酸分子が、配列番号20〜37の何れかに示すアミノ酸配列に、少なくとも約90%の相同性を有するポリペプチドをコードする、請求項72に記載の方法。
- 単離された核酸分子が、配列番号20〜37の何れかに示すアミノ酸配列に、少なくとも約99.9%の相同性を有するポリペプチドをコードする、請求項72に記載の方法。
- 単離された核酸分子が、配列番号20〜37の何れかに示す配列を含有して成る、請求項72に記載の方法。
