該樹状細胞上の受容体に対するリガンドであって、該リガンド上の金属親和性標識を介して該金属キレート基に連結される該リガンドと、を含み、上記の抗原を含む小胞またはリポソームが免疫調節因子を含む、該組成物が提供される。
i)抗原を含む膜小胞または抗原を含むリポソームを調製する工程;
ii)少なくとも1つの免疫調節因子の取込みによって上記の抗原を含む膜小胞または抗原を含むリポソームを修飾する工程;
iii)両親媒性分子の取り込みによって前記抗原を含む膜小胞または抗原を含むリポソームを修飾する工程、ここに両親媒性分子は、前記抗原を含む膜小胞または抗原を含むリポソームに取り込まれた場合にそれらの表面上に位置するキレート基を含むものであり;次いで

試作品の試験において、本発明者らは、キレーター−脂質(NTA)3−DTDAを用いることで、DCの標的化のために腫瘍由来の細胞膜小胞(PMV)上もしくは腫瘍抗原を含むステルスリポソーム上にHisで標識したScFvが固着できることを見出している。この方法によるAgのDCに対する直接的な標的化が強力な抗腫瘍応答を誘発した。
試薬
[3H]−チミジンおよび51Cr(Na51CrO4)をアマシャム(Amersham)(バッキンガムシャー州、英国)から入手した。パルミトイル−オレオイル−ホスファチジル−コリン(POPC)、OVA(Grade II、FPLCにより精製)、LPS(大腸菌(Escherichia coli)血清型0111:B4由来)、Isopaque、Ficollおよびβ−メルカプトエタノールは、シグマ・アルドリッチ(Sigma−Aldrich)(キャッスルヒル、ニューサウスウェールズ州、オーストラリア)により供給された。ホスファチジルエタノールアミン−ポリエチレングリコール−2000(PE−PEG2000)をアバンティ・ポーラー・リピッヅ(Avanti Polar Lipids Inc.)(アラバスター)から入手した。2−(4,4−ジフルオロ−5オクチル−4−ボラ−3a,4a−ジアザ−s−インダセン−3−ペンタノイル)−1−ヘキサデカノイル−sn−グリセロ3−ホスホコリン(PC−BODIPY)および5−(および−6)−カルボキシフルオレセインジアセテート、サクシニミジルエステル、混合イソマー(CFSE)をモレキュラー・プローブス(Molecular Probes)(ユージーン、オレゴン州)から購入した。2つのジテトラデシルアミン(DTDA)鎖に共有結合した3つのニトリロ三酢酸(NTA)頭部基からなるキレーター−脂質(NTA)3−DTDAを記載27のように本質的に合成したが、NTA−DTDAの各カルボキシル基の上にNTA基を共有結合させる更なる工程とともに(NTA)3−DTDAを生成した。Ni2+の緩衝液へのあらゆる添加にはNiSO4を用いた。
マウスCD56(クローン42.18、ラットIgG2a)mAbは、6thHuman NK CellWorkshopから、およびマウスCD3 mAb(クローン145−2C11、ArmenianハムスターIgG)はファーミンゲン(PharMingen)(サンディエゴ、カリフォルニア州)から購入した。組換えマウスのIFN−γおよびGM−CSFは、ペプロテック(PeproTech Inc)(ロッキーヒル、ニュージャージー州)によって供給された。組換えScFv抗体N418(抗CD11c)およびNLDC145(抗DEC−205)は、それぞれカルボキシ末端にヘキサヒスチジン(6H)標識を有し、それぞれCD11c−ScFvおよびDEC−205−ScFvを示し、これらをバキュロウイルスタンパク質発現系を用いて産生し、記載16,28のように精製した。ペプチドをBiomolecular Resource Facility、John Curtin School of Medical Research(JCSMR)、ANU、キャンベラによって合成した。血漿タンパク質のヒスチジンに富む糖タンパク質内に認められる10個のアミノ酸配列であるL2ペプチド(GHHPHGHHPH)は、高い結合活性を有するNi−(NTA)3−DTDAと結合しその細胞に対する非特異的結合を遮断できることから、これを通常用いて対照のPMVおよびSLを移植した。ペプチドAgをDCへ送達するのに、H−2bマウス内のOVAの免疫優勢CTLエピトープ(OVA残基257〜264)を示すペプチドSIINFEKL−6Hを付着させたヘキサヒスチジン標識とともに用いた。
6〜8週齢の雌または雄のC57BL6マウス(H−2b)はAnimal Breeding Establishment(JCSMR、ANU)から供給され、かつC57BL6エオタキシンノックアウトマウス(H−2b)(エオタキシン−/−)はDrポール・フォスター(Paul Foster)、Division of Biochemistry and Molecular Biology(JCSMR)からの贈呈であり、そして同マウスを用いることでin vitroにおけるアッセイおよびin vivoにおける腫瘍成長試験においてリンパ球様細胞を得た。10%ウシ胎仔血清(FCS、トレースバイオサイエンシーズ(TraceBiosciences)、ノーブルパーク、ビクトリア州、オーストラリア)および0.5mg/mL Geneticin(インヴィトロジェン(Invitrogen))を含有するRPMI1640培地(Gibco−BRL、インヴィトロジェン(Invitrogen)、メルボルン、オーストラリア)内で、OVAを分泌する腫瘍細胞系である高転移性を有するマウスB16−OVAメラノーマ[C57BL6(H−2b)]を5%CO2の大気中で37℃で培養した。マウス胎児皮膚樹状細胞(Murine Foetal Skin Dendritic Cells)(FSDC)[C57BL6−DBA/2JF1(H−2b/d)]を同じ培地内で培養したが、ジェネチシンを含まなかった。記載29のように単離して培養したマウス長期培養樹状細胞(Murine Long Term culture Dendritic Cells)(LTC−DC)[B10.A(2R)(H−2k/b)]は、Dr H.オニール(Dr H.O’Neill)(School of Biochemistry and Molecular Biology、ANU)からの贈呈であった。
マウスのDCおよびT細胞をC57BL/6マウスの脾臓から単離した。つまり、脾細胞をコラゲナーゼIV(ベーリンガー・マンハイム(Boerhringer Mannheim))を用いた消化によって単離してから、Isopaque−Ficoll勾配を用いて密度勾配遠心分離によって低密度の脾細胞を単離した。DCを記載31のようにプラスチック付着によって単離し、次いで10%FCS、5×10−5Mのβ−メルカプトエタノール、100IU/mlのペニシリン、100μg/mlのネオマイシン、および10mMのHEPESを含有する完全RPMI1640成長培地内で懸濁した。T細胞の単離のために、脾臓を単一細胞懸濁液中に溶解し、低張による溶解(hypotonic lysis)によって赤血球を取り出した後、ナイロンウールカラムを用いてT細胞を単離した32。
培養細胞由来のPMVをショ糖勾配遠心分離によって調製し30、本質的には概説したように修飾した16,17。PMVの修飾に用いるリポソームを以下のように調製した。すなわち、POPC、(NTA)3−DTDA、LPSおよびPC−BODIPY(モル比94:2:2:2);またはPOPC、(NTA)3−DTDAおよびPC−BODIPY(モル比96:2:2)のエタノール溶液を混合し、N2の流れの下で乾燥させ、次いで60μMのNi2+を含有するPBS100μl中で再水和した。LPSの代わりとして必要な場合、IFN−γもしくはGM−CSF(50ng)を再水和用緩衝液中に含有させた。TOSCO 100Wの超音波破砕機(メジャリング・アンド・サイエンティフィック(Measuring and Scientific Ltd.)、ロンドン、英国)を最大振幅で用いて水和混合物を超音波で破砕した(3回、15秒バースト)。15%PEG400を添加してPBSで10倍に希釈する前に、リポソーム(100μl)をB16−OVA細胞由来のPMV(1×108細胞当量)100μlと混合させた。適切なScFvを移植する前に、サイズ排除クロマトグラフィーにより(NTA)3−DTDAおよびサイトカインを含むPMVを精製した17。
試験の追跡において高度に蛍光性のPMVを得るために、PMVをフルオレセイン−イソチオシアネート(FITC、モレキュラー・プローブス(Molecular Probes))と反応させ、PMVにL2もしくはScFvを移植し、次いでマウスの後足蹠(hind footpad)内にPMVを注射した。16時間後、各動物の流出鼡径リンパ節(draining lymph node)を採取し、いずれかをビオチン化CD11c mAbおよびストレプトアビジン−フィコエリトリン(ストレプトアビジン−PE)による染色後の2色フローサイトメトリー分析または共焦点蛍光イメージングのためのリンパ節細胞の単離用に用いた。イメージング用にリンパ節を10%のホルマリンに固定し、次いでパラフィン包埋し、そして切片に切断した。次いで同切片をスライドに付着させ、脱ワックスした。PBS−ヤギ血清中でCD11cに対するmAb N418とともに室温で1時間インキュベートする前に、室温で30分間のPBS+20%ヤギ血清(PBS−ヤギ血清)を用いたインキュベーションによってスライドをブロッキングした。次いでスライドを水中で広範に洗浄し、PBS−ヤギ血清中でストレプトアビジン−ローダミンで染色した。さらに洗浄した後、Radiance 2000蛍光共焦点顕微鏡(バイオ・ラッド(Bio−Rad)、リッチモンド、カリフォルニア州)を用いてフルオレセインおよびローダミン蛍光についてスライドを分析した。30回連続するレーザー走査を平均化するKalmanを介して画像を取得し、Bio−Rad画像ソフトウェアを用いて画像処理を行った。
完全培地内でDCを修飾したPMVまたはSLとともに37℃で4時間インキュベートし、次いで洗浄することで結合していないPMVまたはSLを除去し、γ照射(5000rad)し、そして成長培地(2×104細胞/200μl/ウェル)内で96ウェル平底プレートに分注した。[3H]−チミジンの取込みの測定によって増殖を評価する前に、同系T細胞を添加し(2×104/ウェル)、同細胞を4日間共培養した14。記載33のようにDCとの共培養に先立ってT細胞をCFSE(5μM)で標識することにより、Ag提示アッセイにおける増殖CD4+とCD8+T細胞の割合を評価した。4日後、共培養細胞を洗浄し、抗マウスCD4(クローンL3T4)−Cy−クロム(10μg/ml)、および抗マウスCD8(クローンLy−2)−PE(10μg/ml)で染色し、そしてフローサイトメトリーによってCFSE−、Cy−クロム−、およびPE−蛍光について分析した。
記載34のアッセイと同様に、Agに特異的なCTLアッセイを実施した。同系C57BL6マウスをPBS(対照)、またはScFvを移植したB16−OVA細胞由来のPMVもしくはAgを担持するSL(上記)で静脈内(i.v.)に免疫した。免疫の14日目に、脾臓を摘出し、Tリンパ球(エフェクターT細胞)を上記の如く単離した。次いで完全な成長培地でT細胞を懸濁し、1×105細胞/ウェルの濃度で24ウェル平板プレートに分注し(アイシーエヌ・バイオメディカルズ(ICN Biomedicals))、1×105のγ照射(5000rad)されたB16−OVA細胞と共培養した。記載16のように、共培養の5日後、標準51Cr−放出アッセイにおいてT細胞の細胞溶解活性を評価した。
週単位に行う3回のi.v.尾静脈注射によってPBS(対照)、またはScFvを移植したB16−OVA細胞由来のPMV(2×105細胞当量)、もしくは関連Ag(〜0.2μgのOVAもしくは〜0.8ngのSIINFEKL−6H)を担持するSL(〜0.16μgの総脂質)でマウスを免疫し、それぞれを容量200μlのPBSで懸濁した。最終回の注射の2週間後、3×105のB16−OVA細胞のi.v.注射を介してマウスに負荷した。16日目に両肺を摘出し、解剖顕微鏡下で腫瘍病巣数を視覚的に計数した。あるいは、1.5×105のB16−OVA細胞をi.v.注射した3、6および9日後に、マウスをScFvを移植したB16−OVA PMVで免疫した。
リポソームを用いることでin vitroおよびin vivoの両方においてDCに対して腫瘍抗原を標的化できる。
2種類のリポソーム調製物を用いることで腫瘍AgをDCに対して標的化した(下記図1参照)。第1はDCを標的化するScFvの移植によって修飾された腫瘍細胞由来PMVの粗調製物の使用を伴うものであり、第2はAgを含み、DCを標的化するScFvも移植したステルスリポソームの調製物であった。ステルスリポソーム(SL)は、PE−PEG2000などの脂質を含むことによって立体的に安定化されている合成脂質構造であり、細網内皮系による非特異的除去を回避する能力によって、その静脈内投与後の数日間、血液循環内に残存できる14。T細胞共刺激分子の移植において腫瘍細胞および腫瘍細胞由来PMVを修飾するためのキレーター脂質であるNTA−DTDAの使用について記載している15,16。発明者らは最近、NTA−DTDAに関与する新規脂質の(NTA)3−DTDAを得ているが(図1A)、より高い局部密度のNTA頭部を得ることによって両PMV上およびSL上でのヒスチジンで標識したタンパク質のより安定した固着が可能になる(図示せず)。ゆえに、CD11cおよびDEC−205などのDCマーカーに対する、ヒスチジンで標識したScFvにおける(NTA)3−DTDAを介したリポソームの付着は、リポソームのDCに対する有効な標的化を可能にする(図1B)。
腫瘍抗原の樹状細胞に対するリポソーム介在性標的化がin vitroおよびin vivoの両方において強力な腫瘍特異的免疫を誘発する
DCに対して標的化されたAgを担持するPMVおよびSLが機能的AgのT細胞への提示を誘発できるか否かを判定するために、発明者らはまず、Ag提示アッセイにおいてScFvを移植したPMVおよびSLのT細胞増殖に対する刺激能について試験した。C57BL/6マウスから単離した脾臓DCに、いずれかの対照のヒスチジンで標識したペプチド(L2)もしくはCD11cおよびDEC−205に対してScFvを移植した、B16−OVA−PMV、SIINFEKL−6Hを担持するSL、またはOVAを担持するSLで別々にパルスした。インキュベーション後、同細胞を精製した同系T細胞と共培養し、次いで[3H]−チミジンでパルスすることでT細胞増殖の速度を評価した。対照培養物と比較し、CD11c−ScFvを移植したPMVまたはSL(SIINFEKL−6HもしくはOVAを担持)に暴露したDCは、実質的により高レベルのT細胞増殖を誘発した。T細胞をDEC−205 ScFvを移植したPMVまたはSLに暴露したDCと共培養すると、さらにより高い増殖速度を認めた(図4A)。したがって、ScFvを移植した、Agを担持するPMVおよびSLは、AgをDCに有効に送達してT細胞増殖を刺激することができる。
DCを標的化するリポソームに基づくワクチンは腫瘍に対する防御免疫を誘発する
様々なB16−OVA調製物で免疫したマウスのB16−OVA腫瘍細胞のi.v.負荷に対する耐性能について試験し、それに伴い腫瘍細胞への注射の16日後に肺転移を定量した。ScFvを移植し、LPSもしくはIFN−γを含むPMVまたはOVAを担持するSLで免疫したマウスでは、対照マウスと比較して転移数の著しい低下を観察した(図6)。PMVまたはOVAを担持するSLにScFvを移植ぜずにLPSもしくはIFN−γを含まなかった場合、腫瘍細胞への負荷に対する防御はほとんど検出されなかった。それとは全く対照的に、一部のワクチンが強力なCTL活性の誘発を構築するにもかかわらず(図5)、SLを含むSIINFEKLは腫瘍負荷に対してマウスを防御できなかった(図6B)。これらのデータは、CD8+CTLによるクリアランスに耐性を示すB16−OVAメラノーマに一致する(参考文献14)。
病原菌に関連する抗原の樹状細胞に対する標的化による病原菌に対する免疫の増強
本実施例では、発明者らは本発明を用いることでDCに対して病原菌の抗原を標的化できることを実証する。BCGは、ヒトにおける結核の原因である病原体Mycobacterium tuberculiにも存在する多数の抗原を含むマイコバクテリウムである。本明細書において記載される実施例では、BCGマイコバクテリアをMycobacterium tuberculiの代わりに用いた。修飾を行ってDCに対する標的化を可能にする前に、BCGマイコバクテリアを培養物中で成長させ、熱殺菌し、そして[トレーサー6(フルオレセイン−5(および−6)−カルボキシアミド)ヘキサン酸サクシニミジルエステルとの反応を介して]標識することで、追跡できるようにした。したがって、熱殺菌したBCGを適切な量のNi−(NTA)3−DTDAと混合し、そして短時間、超音波破砕することでBCG抗原を含むBCG膜小胞内へのキレーター脂質の取込みが可能になった。次いで、BCG膜内へのNi−(NTA)3−DTDAの取込みにより、CD11cもしくはDEC−205に対するScFvの移植によってそれぞれのDC上のCD11cおよびDEC−205マーカーに対する特異的標的化が可能になった。

Claims (30)
- in vivoにおいて抗原を樹状細胞に対して標的化することによって免疫を調節するための組成物であって、
複数の金属キレート基を有する前記抗原を表面上に含む膜小胞または抗原を含むリポソームの調製物;および
前記樹状細胞上の受容体に対するリガンドであって、前記リガンド上の金属親和性標識を介して前記金属キレート基に連結されるリガンド
を含み、前記抗原を含む小胞またはリポソームが免疫調節因子を含む、組成物。 - 前記抗原を含む膜小胞が、腫瘍由来の細胞膜小胞、リンパ球由来の細胞膜小胞、白血球由来の細胞膜小胞、および細菌、原虫、ウイルスまたは真菌の膜調製物からなる群から選択される請求項1に記載の組成物。
- 前記抗原を含むリポソームがステルスリポソームである請求項1に記載の組成物。
- 前記抗原を含む膜小胞またはリポソームの抗原が複数の異なる抗原を含む請求項1に記載の組成物。
- 前記リガンドが抗体、抗体断片およびドメイン抗体からなる群から選択される請求項1に記載の組成物。
- 前記抗体断片が一本鎖抗体断片である請求項5に記載の組成物。
- 前記リガンド上の前記金属親和性標識がヘキサヒスチジンである請求項1に記載の組成物。
- 前記免疫調節因子が危険シグナル、サイトカイン、ケモカイン、ホルモンまたは成長因子様分子、および上記分子のいずれかをコードするDNAからなる群から選択される請求項1に記載の組成物。
- 前記危険シグナルが細菌性リポ多糖である請求項8に記載の組成物。
- 前記サイトカインがインターフェロン−γ、インターロイキン−2、インターロイキン−4、インターロイキン−10、インターロイキン−12および形質転換成長因子−βからなる群から選択される請求項8に記載の組成物。
- in vivoにおいて抗原を樹状細胞に対して標的化することによって免疫応答を調節するための組成物を調製するための方法であって、
i)抗原を含む膜小胞または抗原を含むリポソームを調製する工程;
ii)少なくとも1つの免疫調節因子の取込みによって前記抗原を含む膜小胞または抗原を含むリポソームを修飾する工程;
iii)両親媒性分子の取り込みによって前記抗原を含む膜小胞または抗原を含むリポソームを修飾する工程、ここに両親媒性分子は、前記抗原を含む膜小胞または抗原を含むリポソームに取り込まれた場合にそれらの表面上に位置するキレート基を含むものであり;次いで
iv)前記樹状細胞上の受容体に対する、前記キレート基に結合するための金属親和性標識を含むリガンドに工程(iii)の前記産物を接触させる工程
を含む、方法。 - 工程(i)において調製される前記抗原を含む膜小胞が、腫瘍由来の細胞膜小胞、リンパ球由来の細胞膜小胞、白血球由来の細胞膜小胞、および細菌、原虫、ウイルスもしくは真菌の膜調製物からなる群から選択される請求項11に記載の方法。
- 工程(i)において調製される前記抗原を含むリポソームがステルスリポソームである請求項11に記載の方法。
- 前記抗原を含む膜小胞および抗原を含むリポソームの前記抗原が、タンパク質、糖タンパク質、ペプチド、多糖、および先のいずれかをコードするDNAからなる群から選択される請求項11に記載の方法。
- 工程(ii)において取込まれる前記免疫調節因子が、危険シグナル、サイトカイン、ケモカイン、ホルモンまたは成長因子様分子、および上記分子のいずれかをコードするDNAからなる群から選択される請求項11に記載の方法。
- 前記危険シグナルが細菌性リポ多糖である請求項15に記載の方法。
- 前記サイトカインが、インターフェロン−γ、インターロイキン−2、インターロイキン−4、インターロイキン−10、インターロイキン−12および形質転換成長因子−βからなる群から選択される請求項15に記載の方法。
- 工程(iii)において取込まれる前記両親媒性分子が、ニトリロ三酢酸ジテトラデシルアミン、トリ(ニトリロ三酢酸)ジテトラデシルアミン、またはニトリロ三酢酸ホスファチジルエタノールアミンからなる群から選択される請求項11に記載の方法。
- 工程(iii)の前記産物と接触する前記リガンドが、抗体、抗体断片およびドメイン抗体からなる群から選択される請求項11に記載の方法。
- 前記抗体断片が一本鎖抗体断片である請求項19に記載の方法。
- 前記リガンドが、CD11c、DEC−205(CD205)、DC−SIGN(CD209)、CD206およびCD207からなる群から選択される受容体に対する請求項11に記載の方法。
- 前記リガンド上の金属親和性標識がヘキサヒスチジンである請求項11に記載の方法。
- 対象における免疫応答を調節する方法であって、請求項1に記載の組成物を前記対象に投与することを含む、方法。
- 免疫応答の前記調節が移植拒絶もしくは自己免疫疾患の予防または治療を目的とする請求項23に記載の方法。
- 前記自己免疫疾患がI型糖尿病、関節リウマチ、全身性エリテマトーデスまたは多発性硬化症である請求項24に記載の方法。
- 請求項1に記載の組成物を前記対象に投与することを含む、対象における腫瘍を予防または治療する方法であって、前記抗原を含む膜小胞または抗原を含むリポソームに含まれる前記抗原が腫瘍抗原である、方法。
- 前記腫瘍がメラノーマ、または前立腺、腸、乳房または肺における癌である請求項26に記載の方法。
- 第1の実施形態に記載の組成物を前記対象に投与することを含む、対象における感染症を予防または治療する方法であって、前記抗原を含む膜小胞または抗原を含むリポソームに含まれる前記抗原が前記感染症を引き起こす作用物質由来の抗原である、方法。
- 前記感染症の前記原因物質が細菌、マイコバクテリウム、ウイルス、または菌類である請求項28に記載の方法。
- 前記対象がヒト対象である請求項23〜29のいずれか一項に記載の方法。
