Abstract
【解決手段】リン酸カルシウム類を含有する繊維状シートを細胞培養用基材に用いてコラーゲン産生細胞の培養を行った後、前記繊維状シートごと加熱滅菌(オートクレーブ)処理を行う手順を少なくとも含むコラーゲン類生産方法を提供する。リン酸カルシウム類を含有する繊維状シートにコラーゲン産生細胞を付着させ、培養した後、繊維状シートごと加熱滅菌処理を行うことにより、加熱処理後も、コラーゲン類の三重らせん構造を維持させることができる。この方法により、加熱滅菌処理済のコラーゲンを取得できる。従って、この方法は、医療・美容分野、特に、人工真皮、止血材、再生医療などにおける足場部材などの提供に特に有用である。

Description
本発明に係るコラーゲン類は、ローマ数字で分類される各種コラーゲンのほか、アテロコラーゲン、プロコラーゲン、コラーゲンペプチド、加水分解コラーゲン、コラーゲン様物質(コラーゲンと近似の一次構造を有するタンパク質)などを広く包含する。これらの各コラーゲン類は、例えば、本発明に係るコラーゲン類生産方法により、または、その手順中やその手順の前後に公知技術に基づいて酵素処理・化学処理などを施すことにより、取得できる。
本発明に係るコラーゲン類生産方法には、(1)細胞培養手順、(2)加熱滅菌処理手順、(3)コラーゲン類の精製手順、などが含まれる。以下、順に説明する。
本発明では、リン酸カルシウム類を含有する繊維状シートを細胞培養用基材に用いてコラーゲン産生細胞の培養を行う。
a.この細胞培養方法を用いることにより、三次元的・立体的に細胞培養を行うことができ、細胞同士が強固かつ高密度に結合した状態にできる。
b.この細胞培養方法では、細胞の増殖性を維持した状態で、長期間連続培養を行うことができる。即ち、継代培養を行わず、定期的な培地交換のみで、長期間連続培養を行うことができる。
続いて、リン酸カルシウム類を含有する繊維状シートにコラーゲン産生細胞を付着させ、培養した後、繊維状シートごと加熱滅菌処理を行う。
続いて、コラーゲン産生細胞が強固に結合した繊維状シートからコラーゲン類を抽出・精製する。コラーゲン類の抽出に伴い、細胞も剥離する。
Claims (6)
- リン酸カルシウム類を含有する繊維状シートを細胞培養用基材に用いてコラーゲン産生細胞の培養を行った後、前記繊維状シートごと加熱滅菌処理を行う手順を少なくとも含むコラーゲン類生産方法。
- 前記繊維状シートは、不織布であることを特徴とする請求項1記載のコラーゲン生産類方法。
- 前記リン酸カルシウム類は、ハイドロキシアパタイトであることを特徴とする請求項1記載のコラーゲン生産類方法。
- グリセリン存在下で、前記加熱滅菌処理を行うことを特徴とする請求項1記載のコラーゲン類生産方法。
- 前記培養を無血清培地で行う段階を少なくとも含むことを特徴とする請求項1記載のコラーゲン類生産方法。
- 請求項1記載のコラーゲン類生産方法により取得した加熱滅菌処理済みのコラーゲン類。
