Abstract
【解決手段】 本発明は、抗菌性持続作用を有するインプラント、特に人工血管であって、インプラントの形態を決定し、実質的に非吸収性または緩慢吸収性ポリマー材料、及び吸収性材料のコーティングからなる基本構造を有し、金属銀の層が、ポリマー材料の上、コーティングの下にあるインプラントに関する。

Description
比較試験
実施例1の人工血管で、吸収性充満層を施していないものを、37℃のリン酸バッファー(pH7.4)中に置いた。リン酸バッファーは毎日取り替えて、前のリン酸バッファー試料中の銀含有量を測定した。試験は、365日間続けた。除去したリン酸バッファー中の銀含有量は、初めは35μg/Lであったが、その後急速に減少し、50日後には、ゆっくりとなり(15μg/L)、365日後には、約5μg/Lであった。
組織の反応
同様の方法で調製した人工血管で、1600Åと2500Åの銀層を有するものを、ラット、ウサギおよびブタに移植した。3ヶ月および6ヶ月後の外植では、良好な統合が見られた。インプラントは、全て異常な所見を示さなかった。初期組織も、異常所見を示さなかった。慢性の炎症反応の徴候もみられなかった。
人工感染
本発明のインプラントと、基本構造の上に銀層を有する代わりに、吸収性コーティングに酢酸銀を合わせ持つインプラントとの比較を行った。比較試料を、問題の細菌に人工的に感染させて、ウサギに移植した。7日後に外植した。比較試料を48時間、CASO培養液中で培養し、細菌数測定を行った。細菌のコロニー形成を36試料で、細菌学的に測定した。本発明のインプラントでは、22%、すなわち8のインプラントにだけ、少量の細菌がコロニー形成したが、吸収性コーティングに酢酸銀を有するインプラントでは、67%の23インプラントに感染が見られた。
Claims (17)
- 抗菌性持続作用を有するインプラント、特に人工血管であって、インプラントの形態を決定し、かつ、実質的に非吸収性または緩慢吸収性のポリマー材料と、吸収性の材料のコーティングからなる基本構造を有し、ポリマー材料の上で、かつコーティングの下に、金属銀の層が位置しているインプラント。
- 銀層が、ポリマー材料に固着しており、特にポリマー材料中に固定されている請求項1に記載のインプラト。
- 銀層をポリマー表面に蒸着させる請求項1または2に記載のインプラント。
- 銀層の銀原子が、基本構造のポリマー表面に押し付けられ、特にポリマー表面にアルゴンイオンを用いて衝撃を与えることにより押し込められている、先の請求項のいずれかに記載のインプラント。
- 銀層が、実質的に密閉された層であって、特に、生体内条件下で1年より長く、詳しくは2年より長い滞留時間を有し、この間銀イオンを放出するような厚さである、先の請求項のいずれかに記載のインプラント。
- 銀層が、体内で分解するにつれて、1年に最大約5〜10%、特に最大7〜8%の層が除かれるような層の厚みである、先の請求項のいずれかに記載のインプラント。
- 銀層が、2500〜1000Å、特に約1300Åの層厚を有する、先の請求項のいずれかに記載のインプラント。
- 銀層が、殆ど純銀からなる、先の請求項のいずれかに記載のインプラント。
- 基本構造が多孔性であって、銀層が孔を開いたままにし、吸収性層がインプラントの孔をシールするように充満している、先の請求項のいずれかに記載のインプラント。
- 吸収性層が、所望により架橋した生物性材料からなる、先の請求項のいずれかに記載のインプラント。
- 吸収性層が、吸収性であって、特に生体内条件下で分解性で、更に詳しくは少なくとも1つのヒドロキシ酸からなる、合成ポリマーおよびコポリマーから作製される、先の請求項のいずれかに記載のインプラント。
- 吸収性層の成分を、最長で4ヶ月後、詳しくは最長で40日後に吸収されるように選択する、先の請求項のいずれかに記載のインプラント。
- 吸収性材料のコーティングが、更に吸収性層の吸収の間に放出される活性物質を含む、先の請求項のいずれかに記載のインプラント。
- 基本構造が、織物材料、特に形成ループニットから作製される、先の請求項のいずれかに記載のインプラント。
- 織物基本構造の繊維が、少なくともインプラントの一面に向かい合う位置が銀でコートされており、好ましくは実質的に繊維の全表面が銀でコートされている、請求項14のインプラント。
- 基本構造が、焼結材料、特に発泡ポリテトラフルオロエチレンから作製される、請求項1から13のいずれかに記載のインプラント。
- 該インプラントが、中空器官の置き換え、特に人工血管用に設計されている、先の請求項のいずれかに記載のインプラント。
