Abstract
【解決手段】本発明は、金属イオンをキレート化することが可能な化合物に関する。特に、本発明は、鉄イオンを含めた金属イオンをキレート化することが可能な(チオ)セミカルバゾン化合物および(チオ)ヒドラジン化合物に関する。また、かかる化合物および/またはそれらの金属イオン錯体の、細胞増殖に関連する疾病の治療法を含めた治療的使用も開示される。

(細胞機能の修飾)
結果としてG1−S細胞停止を生じてよい。Ndrg1の過剰発現はまた、腫瘍細胞の増殖を遅らせ、腫瘍細胞の転移を阻害することが示されている4−7。
(疾病の治療および/または予防)

(製薬および/または治療用の製剤)
本文において「PCIH」および「QCIH」類似体と呼ばれる比較用の2−ピリジル−およびキノリル−アロイルヒドラゾン類似体は、各々、2−ピリジルカルボキサルデヒドまたは2−キノリンカルボキサルデヒドと、適当な酸ヒドラジンとのシッフ塩基縮合により合成された。
代表的なPKIH類似体は、バッキ(Bacchi)ら(1996)の方法に従って、2−ジ−ピリジルケトンを適当な酸ヒドラジンと縮合することにより合成された13。適当な溶媒は、エタノール、メタノール、エタノール/水、メタノール/水、アセトン、ベンゼン、トルエンを含む。代表的なPKIH類似体は、図2Bに示されている。

DpT類似体は、2−ジ−ピリジルケトンと、各々のチオセミカルバジドまたは酸ヒドラジドとのシッフ塩基縮合により、標準的な方法(ジョンソン(Johnson)ら、1982年)14を用いて合成された。反応を行なうための適当溶媒は、エタノール、メタノール、エタノール/水、メタノール/水、アセトン、ベンゼン、トルエンを含む。
PKIH類似体の鉄錯体は、リチャードソン&バーンハート(Richardson & Bernhardt)、1999年3により記述された技術を用いて調製された。一般に、1モル当量のFe(III)塩は、2モル当量のPKIH類似体とともに、熱エタノール中で1時間灌流された。暗緑/黒色の錯体の固体が形成され、減圧下の濾過により母液から単離された。その錯体はそれからエタノールで洗われ、空気乾燥された。
親油性は、平均logP値(n−オクタノール−水の分配計数)として、すべてのキレート化剤について測定された。logP値は、ブロートの方法(ブロート(Broto)ら、1984年)15、クリッペンのフラグメンテ−ション法(ゴース&クリッペン(Ghose & Crippen)、1987年)16、またはビシュバナダハンのフラグメンテ−ション法(ビシュバナダハン(Viswanadhan)ら、1987年)17に従って、Chem Draw Pro(v.4.5、ケンブリッジ・ソフトウエア(Cambridge Software)、1997年)を用いて概算された(表1)。これらの値の平均値(平均logP計算値;表1)は、次にIC50値か、またはキレート化剤が59Feの動員を誘導するかまたは59Feの取込みを妨げる能力の、いずれかに対してプロットされた。これらのプロットは、親油性と、試験化合物の抗増殖活性またはFeキレート化効率との間に強い相関関係がないことを示している(すべての相関:r<0.52)(データは示さず)。
(インビボおよびインビトロの研究)
DFOはノバルティス(Novartis)(スイス、バーゼル)から購入された。
311は標準的な方法3に従って調製された。
Triapine(登録商標)(3−アミノピリジン−2− カルボキサルデヒド・チオセミカルバゾン;図1B)は、バイオン・ファーマスーティカルズ(Vion Pharmaceuticals)米国、ニュージャジー州、からの寄贈であった。
化合物はすべて、実験の直前に10mMの保存溶液としてジメチルスルホキシド(DMSO)中に溶解され、次いで10%ウシ胎児血清(FCS;コモンウェルス・シーラム・ラボラトリーズ(Commonwealth Serum Laboratories)、オーストラリア、メルボルン)中に、最終DMSO濃度が0.5%(v/v)以下になるように希釈された。
ヒトSK−N−MC神経上皮細胞腫細胞系、SK−Mel−28メラノーマ細胞、およびMCF−7乳癌細胞は、アメリカン・タイプ・カルチャー・コレクション(American Type Culture Collection)(ATCC)、米国、メリ−ランド州、ロックビル、から購入された。正常な細胞タイプ、MRC−5線維芽細胞およびL8ラット骨格筋細胞もまた、アメリカン・タイプ・カルチャー・コレクション(ATCC)、米国、メリ−ランド州、ロックビル、から購入された。細胞は、10%FCS(コモンウェルス・ラボラトリーズ、オーストラリア、メルボルン)、1%(v/v)非必須アミノ酸(ギブコ(Gibco))、2mM L−グルタミン(シグマ・ケミカル社(Sigma Chemical Co.)、米国、ミズーリ州、セントルイス)、100μg/mlのストレプトマイシン(ギブコ)、100U/mlのペニシリン(ギブコ)、および0.28μg/mlのフンギゾン(スクイブ・ファーマスーティカルズ(Squibb Pharmaceuticals)、カナダ、モントリオール)を含有するイーグル変法最少必須培地(MEM;ギブコ BRL、オーストラリア、シドニー)中で増殖された。この増殖培地は、後に「完全培地」と呼ばれる。細胞はインキュベーター(フォーマ・サイエンティフィック(Forma Scienttific)、米国、オハイオ州)中で、37℃において、5%CO2/95%空気の加湿雰囲気中で増殖され、継代培養された。細胞の増殖および生存率は、位相差顕微鏡およびトリパンブルー染色を用いて慎重に査定された。
細胞増殖は、MTT(3−(4,5−ジメチルチアゾール−2−イル)−2,5−ジフェニルテトラゾリウム)アッセイを用いて、本質的には先に記述されたものと同様の方法で調べられた18。細胞は96穴マイクロタイタープレート中に、上記の2.2節において記述されたようなすべての補足物(完全培地)および1.25μMのヒトのジフェリックトランスフェリンも含有する0.1mLのMEM培地中に、15000細胞/ウエルに播種された。この播種濃度は、細胞の指数増殖をアッセイの期間にわたって生じる結果となる。細胞は一晩増殖され、次いで、試験されるべき化合物が、1.25μMのジフェリックトランスフェリンを含有する0.1mLの完全培地において添加された。化合物の最終濃度は0.39〜50μMであった。対照サンプルは、完全培地および1.25μMのジフェリックTfを含んでいた。細胞は、化合物とともに、95%空気および5%CO2を含有する加湿雰囲気中で、37℃において90時間インキュベートされた。このインキュベーションの後、0.01mLのMTTが各ウエルに添加され、プレートは37℃において2時間インキュベートされた。細胞は次いで、0.1mLの、0.01M HCl中の10%SDS−50%イソブタノールの添加により可溶化され、プレートは次に、走査型マルチウエル分光光度計(タイターテック・マルチスキャン(Titertek Multiscan);ベックマン・インスツルメンツ・インク(Beckman Instruments Inc)、カリフォルニア州)で、570nmにおいて読取られた。MTTの発色は、生細胞の数に正比例していた。MTTアッセイの結果は、対照値のパーセントとして表される。
実験データは、スチューデントのペアードt検定を用いて比較された。結果は、p<0.05であるとき、統計学的に有意であると見なされた。結果は、2〜5の別個の実験についての平均±標準偏差(SD)として表される。
59Feを用いたトランスフェリンの標識化
アポトランスフェリン(シグマ・ケミカル社、米国、セントルイス)が用意され、59Fe(0.1M HCl中の塩化第二鉄として、デュポン(Dupont)NEN、米国、マサチューセッツ州)を用いて、リチャードソン&ベーカー(Richardson & Baker)、1992年、により記述された標準的な方法を用いて標識化され、59Fe−トランスフェリン(59Fe−Tf)を生成した。
方法論:鉄動員アッセイ
プレラベルされた細胞からの59Feの放出に対するキレート化剤の影響は、標準的な方法を用いて研究された18。細胞は59Fe−トランスフェリン(0.75μM)を用いて、37℃において3時間にわたってプレラベルされた。次いで細胞は氷上に置かれ、氷冷されたBSSで4回洗浄され、次に培地のみ(対照)か、あるいはDFO(100μM)または他の化合物(50μM)を含有する培地の存在下に、37℃において3時間にわたって再びインキュベートされた。次いで重層された培地は除去され、γ−計数管に入れられた。細胞は、プラスチックスパチュラを用いてペトリ皿から1mlのBSS中に除去され、別のセットのγ−計数管へ移された。
方法論:鉄取込みアッセイ
PKIH類似体の、59Fe−Tfからの59Fe取込みの、細胞の取込みを阻害する能力が、標準的な方法を用いて調べられた18。SK−N−MC細胞は、59Fe−Tf(0.75μM)および、DFO(100μM)か、またはPCIH、QCIH、またはPKIHシリーズ(50μM)のうちの(のうちの)一つの化合物のいずれかを含有する培地中で、37℃において3時間インキュベートされた。細胞は次に氷上に置かれ、氷冷されたハンクスの平衡塩類溶液(BSS)で4回洗浄され、非特異的に結合した59Fe−Tfを除去した。次に細胞は一般的なタンパク質分解酵素、プロナーゼ(1mg/ml)とともに4℃において30分間インキュベートされ、膜結合性の59FeおよびTfを除去した。
方法論:鉄動員アッセイ
59Fe動員に対するPKIH類似体の効力が、化合物濃度の関数として査定された(図5A)。細胞は、59Fe−Tf(0.75μM)を用いて、37℃において3時間にわたってプレラベルされ、洗浄され、次いで濃度範囲(0.5〜50μM)のPKIH化合物と、37℃で3時間、再度インキュベートされた。
方法論:鉄動員アッセイ
アポトランスフェリン(シグマ)は、59Fe(デュポン−NEN、米国、マサチューセッツ州)により、標準的な方法を用いて標識化され18、59Feトランスフェリン(59Fe−Tf)を産生した。細胞は59Fe−Tf(0.75μM)を用いて、37℃において3時間にわたってプレラベルされ、洗浄され、次いでキレート化剤(25μM;図11A)とともに、37℃において3時間、再度インキュベートされた。DFOおよび311は、内部標準として使用された。
方法論:鉄取込みアッセイ
DpT類似体の、59Fe−Tfからの細胞の59Feの取込みを阻害する能力は、標準的な方法18を用いて調べられた。細胞によって内在化される(internalized)59Feの量は、一般的なタンパク質分解酵素プロナーゼ(1mg/ml)とともに4℃において30分間インキュベートして膜結合性の59FeおよびTfを除去することにより測定された。
マウスのM109肺癌細胞の増殖を阻害する、化合物Dp44mTの能力が査定された。研究は、培養にあるM109癌細胞を用いて行なわれ、これらの細胞から59Feを動員すること(図11C)および59Fe−Tfからの59Fe取込みを阻害すること(図11D)に対するDp44mTの影響を測定した。内部対照として、細胞はDFOおよび311とともにインキュベートされた。M109細胞から59Fe動員を増大することに対する化合物濃度の影響は、細胞を59Fe−Tf(0.75mM)で37℃において3時間標識化すること、続いて洗浄すること、および各々の化合物(0.2〜25μM)とともに、37℃で3時間再度インキュベートすることにより調べられた(図11C)。DFOは調べた化合物の中で最も効力がなく、25μMにおいて、細胞からわずか12±1%の59Feを放出したに過ぎず、一方対照培地は、8±1%を放出した(図11C)。対照的に、311は細胞の59Fe放出を、8±1%(対照培地)から、5μMにおいて60±1%まで増大させた(図11C)。Dp44mTに関しては、59Fe放出は2μMにおいて横ばい状態になり、結果として48±2%の59Feの流出を生じた(図11C)。
方法論:細胞増殖アッセイ
PCIH、QCIH、およびPKIHシリーズの化合物の、SK−N−MC神経上皮腫細胞系の増殖に対する影響が研究された(表2)。SK−N−MC神経上皮腫細胞の増殖に対する化合物の影響は、MTTアッセイ(前文に記述)を用いて調べられた。MTT発色は、トリパンブルー染色によって測定された生細胞の数に正比例した。すべての実験において、化合物311およびDFOは、関連する内部対照として使用された。
実験は、PKIH類似体がFe(III)と複合した場合、SK−N−MC細胞の増殖の阻害において有効であるかどうかを測定するべく行なわれた。キレート化剤対Feのモル比1:1および2:1において、すべてのPKIH−Fe錯体は、遊離の配位子に関して見られたものと同様のレベルの抗増殖活性を示した(データは示されず)。同一の研究における関連の対照として、311のFe錯体のための1:1および2:1の化合物もまた調製され、査定された。311のFe錯体は、50μMまでの濃度において、増殖に対し何ら有意の影響をもたなかった(データは示されず)。
SK−N−MC神経上皮腫細胞およびMRC−5線維芽細胞の増殖に対するPKIH、PKTH、PKBH、およびPKBBHの影響が調べられた(図6)。MRC−5細胞タイプは、正常なヒト肺線維芽細胞の細胞系であり、インビトロでの多数の継代の後は致死性および老化性であるSK−N−MC神経上皮腫細胞タイプは不死である。増殖を阻害することにおけるPKIH類似体の影響は、線維芽細胞よりもSK−N−MC細胞において増大した。たとえばPKIH類似体は、SK−N−MC細胞では1〜3μMのIC50値を有していたのに対し、線維芽細胞におけるIC50値は、PKBBH、PKBH、PKTH、およびPKIHについて各々8、11、16、および>25μMであった。
DpT類似体(図2A)の、SK−N−MC神経上皮腫細胞増殖を阻害する能力が査定された(図10)。化合物(0〜25μM)は、細胞とともに72時間インキュベートされ、このインキュベーション期間の終わりに、上述のように細胞密度がMTTアッセイにより測定された。Dp2mTはネガティブ対照として役割を果たした。MTTアッセイを用いて得られたデータは、光学顕微鏡によって概算された細胞数と正比例した。
化合物311、DFO、DOX、PKIH、PKTH、Dp44mT、Dp4eT、Dp4pT(0〜50μM)は、細胞とともに72時間インキュベートされた。インキュベーション時間の最後に、細胞密度がMTTアッセイ(上記)により測定された。結果は表4に示されている。
一般方法論
CD21ハイブリッドマウスにおける最大耐量の測定
BALB/cおよびDBA/2マウスは、アニマル・リソーセズ・センター(Animal Resources Center)(ニューサウスウェールズ大学(UNSW)、オーストラリア、シドニー)から入手され、交配されてCD2F1動物を生じた(用いたすべての方法は、動物倫理委員会(Animal Ethics Committee, UNSW)により承認された)。8匹の雌のCD2F1マウス(8〜10週齢)の群は、自由に餌および水が与えられ、2週間にわたり毎週5日、6時間間隔で一日2回、0.9%の無菌食塩水中の30%プロピレングリゴール中の調査される化合物を用いて、尾静脈から注射された。最大耐量(MTD)は、コホート(同齢集団)の30%が死んだかまたは10%を超えて体重が減少した用量として定義された19。
8〜10週齢の雌のCD2F1マウスは、1x105個のM109細胞が皮下移植された。腫瘍細胞は、腫瘍からブライを調製すること、およびそれを皮下に注射することにより、CD2F1マウスにおいて継代された20。Dp44mTは、食塩水中の30%(v/v)プロピレングリコール中に溶解され、腫瘍移植の4日後に、一日2回、5日間にわたって静注され、その後に動物が注射されない2日間の「休止期間」が続けられた。腫瘍移植後12日目に、マウスはメトキシフルオランを用いて犠牲にされ、心臓穿刺が行なわれた。血球数および赤血球指数は、シスメックス・ブラッド・カウンタ(Sysmex Blood Counter)(プリンス・オブ・ウェールズ病院(Prnce of Wales Hospital)、オーストラリア、シドニー)を用いて測定された。処置期間の間のマウスの体重変化が記録された。腫瘍は秤量され、組織学検査用に固定された。実験群は、対照および各処置についてマウス8匹で構成された(表5)。
方法論:3H−チミジン、3H−ロイシン、および3H−ウリジンの取込みアッセイ
SK−N−MC細胞の、類似体の選択物との20時間のインキュベーションの後、3H−チミジン、3H−ロイシン、および3H−ウリジン(1μCi/ml)が、37℃において2時間にわたり添加された。細胞は、Ca(II)/Mg(II)なしのPBS中の1mM EDTAを用いて96穴プレートから剥離され、セル・ハーベスタ(トムテック・ハーベスタ(Tomtc Harvester)96、リンブルック、オーストラリア、クイーンズランド)を用いて収集および洗浄された。放射能は、β−シンチレーションカウンタ(LKBウォレス(Wallace)、フィンランド、トゥールク(Turku))で測定された。
3H−チミジン取込みに対するPKIHシリーズの化合物の影響は、対応するPCIHおよびQCIH類似体と、また関連する対照としてDFOおよび311とも比較された(表6)。
3H−チミジン取込みに対するPKIH群の類似体の著しい効果を考慮して、これらの化合物の、3H−ロイシン(表7)および3H−ウリジン(表8)の取込みに対する影響も査定された。得られた結果は、高いFeキレート化活性を示すが、低い抗増殖効果を示すPCIHシリーズの類似体と比較された。
方法論:ノーザンブロット分析
ノーザンブロット分析は、トータルRNAアイソレーション・リエイジェント(Total RNA Isolation Reagent)(アドバンスド・バイオテクノロジーズ社(AdvancedBiotechnologies Ltd)、英国、サーリー)を用いて全RNAを単離することによって行なわれた。膜は、ヒトWAF1、GADD45、およびβ−アクチンに特異的なプローブとハイブリダイズされた。WAF1プローブは、pSXVからの1kbフラグメントから構成された(ATCC;Cat.No.79928)。GADD45プローブは、pHu145B2(アルバート・フォルナーチェ(Albert Fornace)博士、国立癌研究所、NIH、メリーランド州、のご厚意により提供された)へクローニングされた、ヒトGADD45cDNAからの760bpのフラグメントから構成された。β−アクチンプローブは、pBluescript SK−へクローニングされた、ヒトβ−アクチンcDNAからの1.4kbフラグメントから構成された(ATCC;Cat.No.37997)。
MCF−7細胞は、ある範囲の金属イオンキレート化剤とともに24時間インキュベートされた。細胞膜不透過性のキレート化剤、EDTA(250μM)は、抗増殖活性を示さず(IC50>250μM)、より透過性のある、各々低いおよび高い抗増殖活性を示すキレート化剤と比較された。MCF−7細胞において低い抗増殖活性をもつ化合物は、DFO(IC50=20μM)、L1(デフェリプロン、IC50=180μM)、およびジピリジル(IC50>250μM)を含んでおり、これらは25および250μMにおいて査定された。化合物311およびdp44mTは、MCF−7細胞に対して高い抗腫瘍活性を示し(IC50=各々0.6μMおよび0.1μM)、25μMにおいて査定された(図16) ペニシラミンは周知の銅キレート化剤であり、比較のため査定された。Dp2mTは、ネガティブ対照として用いられた。
方法論:ゲル遅延アッセイ
ゲル遅延アッセイは、鉄調節タンパク質(IRP)と鉄反応エレメント(IRE)
の間の相互作用を測定するべく、確立された技術21を用いて使用された。細胞はDFO(100μM)、FAC(100μg/ml)、およびPKIH類似体(25μM)とともに、37℃において20時間インキュベートされ、細胞ライゼートが調製された。次いでIRP−RNA−結合活性が、ゲル遅延アッセイにより査定された。細胞ライゼートの、2μgのタンパク質を含有するアリコートは、32P−標識された0.1ng(約1x105cpm)のリボプローブとともに、室温において30分間インキュベートされた。このプロトコールを用いる場合、添加されたRNAレベルは飽和していた。保護されていないプローブは、1UのRNアーゼT1との、室温で10分間のインキュベーションにより分解された。次いでヘパリン(5mg/ml)がさらに10分間にわたって添加され、非特異的な結合が除外された。IRE−タンパク質複合体は、6%の未変性ポリアクリルアミドゲルで分析された。平行実験では、サンプルはIREプローブの添加に先立ち、フェリシアニド(FeCN、5mM)および5%β−メルカプトエタノール(β−ME)で処理された。この方法はIRP1を、自発性に活性があるか否かにかかわらず、活性のあるIRE−結合型へ転換する結果を生じる22。

フローサイトメトリーを用いたアポトーシスアッセイ
アポトーシス細胞は、フルオレセイン・イソチオシアネート(FITC)標識されたアネキシン(Annexin)Vの、トランスロケートした形質膜ホスファチジルセリン(PS)への結合を用いて23、フローサイトメーター(FACSCalibur、ベクトン・ディッキンソン(Becton Dickinson)、米国、カリフォルニア州)により検出された。ヨウ化プロピオジウム(PI)が添加され、細胞膜の完全性の低下が確認されたが、それは壊死細胞の指標である。
全般的な方法論
抗体
ウサギ抗活性カスパーゼ−3抗体、ウサギ抗活性カスパーゼ−8抗体、およびアネキシンVフルオレセイン・イソチオシアエネート(FITC)は、BDファーミンジェン(PharMingen)(サンディエゴ、米国、カリフォルニア州)からであった。抗h−cytcMoAbは、R&Dシステムズ(米国、ミネソタ州、ミネアポリス)からであった。ウサギ抗活性カスパーゼ−9抗体、マウス抗Bcl−2、およびマウス抗Bax抗体は、サンタクルズ(Santa Cruz)(米国、カリフォルニア州、サンタクルズ)から入手された。マウス抗マウスβ−アクチン抗体、ホースラディッシュペルオキシダーゼ結合抗ウサギ、および抗マウスIgGは、シグマ・ケミカル社(Sigma Chemical Co.)(米国、ミズーリ州、セントルイス)からであった。
培養M109細胞は、Dp44mT(1μM)を用いて、または用いずに、37℃において0〜48時間処理された。細胞は溶解され27、カスパーゼ活性は、BD ApolertTMカスパーゼ・アッセイ・プレート(BDバイオサンエンセズ(Biosciences)、米国、カリフォルニア州)を用いて測定された。細胞透過性のパン−カスパーゼ−3、−8、および−9阻害剤(BDバイオサンエンセズ)は、阻害性の対照として含まれた。
アポトーシスの誘導時の、ミトコンドリアからのh−cyt放出の重要性のため8、このプロセスにおけるDp44mTの影響についてさらに研究調査された(図15A)。サイトゾルおよびミトコンドリア分画の調製、およびウェスタンブロッティングは、標準的な技術を用いて行なわれた27。対照のM109細胞(すなわち、0時間)のサイトゾル分画においては、h−cytcの発現は非常にわずかであり、最大量のh−cytcは、この時間にはミトコンドリア中に見出された(図15A)。しかしながら、Dp44mT(1μM)への2時間の暴露の後、h−cytcはM109細胞のサイトゾル分画中に見出され、一方ミトコンドリアのh−cytcレベルには、随伴する減少があった(図15A)。サイトゾルへのh−cytcの放出は、Dp44mTとの4時間のインキュベーションの後に横ばい状態となり、次いで24および48時間後に対照細胞において見られるものまで減少した。サイトゾルのh−cytcにおけるこの増加は、ミトコンドリアからの消失と相関していた(図15A)。24および48時間におけるh−cytcの、サイトゾルからのほぼ完全な消失は、細胞からのその放出か、またはその分解によるのかもしれない28(図15A)。
アポトーシスのミトコンドリア経路におけるBcl−2およびBaxの役割、およびh−cytc放出におけるそれらの役割を考慮して8、11、12、これらの分子のタンパク質レベルに対するDp44mT(1μM)の影響が査定された(図15B)。Dp44mT(1μM)とのインキュベーション後、抗アポトーシス分子、Bcl−2は時間の関数として徐々に減少し、2および48時間のインキュベーションの後では、各々0時間の時点での69%および37%まで減少した(図15B)。反対に、Bax発現は、Dp44mTとの48時間のインキュベーションの後にのみ著しく増加し、0時間の時点で見られるものの500%に達した(図15B)。
細胞内の活性酸素種(ROS)アッセイ
細胞内のROS発生は、細胞透過性色素、2’,7’−ジクロロフルオレセイン−ジアセテート(DCF−DA)(シグマ)を用いて査定された。DCF−DAは容易に細胞内へ拡散し、O2・ ‐、H2O2、またはOH−によって酸化されると高度に蛍光性になる29。細胞の蛍光強度は、細胞内ROSのレベルに正比例する29。M109細胞(1x105細胞/ウエル)は、1μMのDp44mTを用いるかまたは用いずに0〜48時間にわたり、あるいは50μMのH2O2(ポジティブ対照)とともに37℃において10分間インキュベートされた。細胞は次に、DCF−DA(5μM)とともに37℃において20分間インキュベートされ、2回洗浄され、培養プレートから剥離された。細胞内で酸化されたDCFの蛍光は、フローサイトメトリーを用いて測定された29。

- 条件(i)および(ii)を満たす若しくは満たさない、治療上有効な量の、式1、2、3、または4の少なくとも一つの化合物か、あるいは条件(i)および(ii)を満たす若しくは満たさない式1、2、3、または4の少なくとも一つの化合物を製薬上許容される希釈剤、補助剤、または賦形剤と一緒に含む製薬組成物を、前記哺乳類へ投与することを含む、哺乳類における鉄キレート化療法。
- 治療上有効な量の、条件(i)および(ii)を満たす若しくは満たさない式1、2、3、または4の少なくとも一つの化合物か、あるいは条件(i)および(ii)を満たす若しくは満たさない式1、2、3、または4の少なくとも一つの化合物を製薬上許容される希釈剤、補助剤、または賦形剤と一緒に含む製薬組成物を、前記哺乳類へ投与することを含む、哺乳類における鉄過剰症の治療方法。
- 前記鉄過剰症がβ−サラセミア(thalassaemia)である、請求項22の方法。
- 細胞増殖を阻害する方法であって、有効な量の、条件(i)および(ii)を満たす若しくは満たさない式1、2、3、または4の少なくとも一つの化合物またはその金属イオン錯体か、あるいは条件(i)および(ii)を満たす若しくは満たさない式1、2、3、または4の少なくとも一つの化合物またはその金属イオン錯体を製薬上許容される希釈剤、補助剤、または賦形剤と一緒に含む製薬組成物と、細胞を接触させることを含む方法。
- 哺乳類において細胞増殖を阻害する方法であって、有効な量の、条件(i)および(ii)を満たす若しくは満たさない式1、2、3、または4の少なくとも一つの化合物またはその金属イオン錯体か、あるいは条件(i)および(ii)を満たす若しくは満たさない式1、2、3、または4の少なくとも一つの化合物またはその金属イオン錯体を製薬上許容される希釈剤、補助剤、または賦形剤と一緒に含む製薬組成物と、哺乳類細胞を接触させることを含む方法。
- 哺乳類において増殖性疾患を治療する方法であって、治療上有効な量の、条件(i)および(ii)を満たす若しくは満たさない式1、2、3、または4の少なくとも一つの化合物またはその金属イオン錯体か、あるいは条件(i)および(ii)を満たす若しくは満たさない式1、2、3、または4の少なくとも一つの化合物またはその金属イオン錯体を製薬上許容される希釈剤、補助剤、または賦形剤と一緒に含む製薬組成物を、前記哺乳類へ投与することを含む方法。
- 前記増殖性疾患が、血管新生依存性疾患、細胞増殖性疾患、および癌から選ばれる、請求項26の方法。
- 前記癌が、悪性腫瘍か、または良性腫瘍である、請求項27の方法。
- 前記癌が、固形腫瘍か、または非固形腫瘍である、請求項27の方法。
- 細胞においてアポトーシスを誘導する方法であって、前記細胞を、有効な量の、条件(i)および(ii)を満たす若しくは満たさない式1、2、3、または4の少なくとも一つの化合物またはその金属イオン錯体か、あるいは条件(i)および(ii)を満たす若しくは満たさない式1、2、3、または4の少なくとも一つの化合物またはその金属イオン錯体を製薬上許容される希釈剤、補助剤、または賦形剤と一緒に含む製薬組成物と接触させることを含む方法。
- 細胞増殖を阻害し、かつ細胞においてアポトーシスを誘導する方法であって、前記細胞を、有効な量の、条件(i)および(ii)を満たす若しくは満たさない式1、2、3、または4の少なくとも一つの化合物またはその金属イオン錯体か、あるいは条件(i)および(ii)を満たす若しくは満たさない式1、2、3、または4の少なくとも一つの化合物またはその金属イオン錯体を製薬上許容される希釈剤、補助剤、または賦形剤と一緒に含む製薬組成物と接触させることを含む方法。

- 哺乳類においてアポトーシスを誘導しかつ細胞増殖を阻害する方法であって、前記哺乳類に、治療上有効な量の、条件(i)および(ii)を満たす若しくは満たさない式1、2、3、または4の少なくとも一つの化合物またはその金属イオン錯体か、あるいは条件(i)および(ii)を満たす若しくは満たさない式1、2、3、または4の少なくとも一つの化合物またはその金属イオン錯体を製薬上許容される希釈剤、補助剤、または賦形剤と一緒に含む製薬組成物を投与することを含む方法。
- 前記金属イオンが、鉄、銅、亜鉛、パラジウム、白金、およびガリウムから選ばれる遷移金属イオンである、請求項24〜33のいずれか1項の方法。
- 前記金属イオンが、鉄(II)および鉄(III)から選ばれる、請求項24〜34のいずれか1項の方法。
