Abstract
Description
しかしながら、該明細書に記載されている技術によっては、今日までのところ、いずれの観点においても、特に使用開始時におけるプラスチック製品の含浸処理による殺菌性の観点において、満足すべき結果は得られていない。
本発明が解決しようとする課題は、満足すべき抗菌活性を発揮するプラスチック製品の製造方法を提供することである。
(A)中間製品を形成させ、
(B)該中間製品の少なくとも1つの構成部材を抗菌性金属コロイドを用いて処理し、次いで
(C)抗菌性金属の易溶性塩または難溶性塩を添加する。
1つの好ましい態様においては、大きさが約1mm3のポリウレタンペレットをポリマー材料として使用する。中間製品の別の構成材料は、担体として機能する硫酸バリウムである。約3〜10重量%(または、所望によりこれよりも多量)のナノシルバーを硫酸バリウムに沈着させた。中間製品はさらに約0.5〜1重量%の硫酸銀またはリン酸銀(特に発生期のリン酸銀)を含有する。中間製品の構成材料を混合し、押出成形法によって加工する。
図1〜図3は、抗菌活性に関連する実験の結果を示す。微生物としては、いずれの場合もスタフィロコッカス・エピデルミジス(Staphylococcus epidermidis)ATCC14990を使用した(出発時点での微生物数:5×107CFU/ml)。
図2に示す実験においては、0.8%のナノシルバーおよび1.0%の硫酸銀を使用した。
図3に示す実験においては、0.8%のナノシルバーを使用し、硫酸銀は使用しなかった。
A:銀コロイドの調製
AgNO3 p.a. 1.0g(5.88mmol)を蒸留水100mlに溶解させ、得られた溶液を25%のアンモニア水1.0ml(14.71mmol)と混合した。銀コロイドを調製するために、アセトアルデヒド258.7mg(5.88mmol;330μl)を蒸留水50mlに溶解させた溶液を上記の溶液中へ40℃において30分間かけてゆっくりと滴下した。
上記の実施例1に記載のようにしてゆっくりと滴下する操作の終了から10分間経過後、ポリウレタンペレット[テコタン(Tecothane)TT−1085A]約50gを添加し、該ペレットを銀コロイドで被覆するために、40℃で2時間激しく攪拌した後、さらに室温で3時間激しく攪拌した。適当な孔径を有する溝付きフィルターを用いる迅速濾過によって銀コロイドを分別させ、ペレットを濾液で洗浄し、湿潤状態で蒸発用ボート内へ移した。ポリマーに付着しなかって過剰の銀コロイドを除去した後、ペレットを70℃で10時間乾燥させた。
A:銀コロイドの硫酸バリウム上への吸着
蒸留水(50℃)360ml中へゼラチン0.6gを溶解させた後、さらにAgNO36.0gを溶解させた。得られた溶液中へ25%のアンモニア水7.8mlを添加した。50℃において激しく攪拌しながら、蒸留水120mlに無水グルコース3.18gを溶解させた溶液を上記の溶液中へゆっくりと添加した。グルコース溶液の約半分を滴下した時点で、既に形成された銀コロイド中へ激しく攪拌しながら硫酸バリウム100gを添加した後、グルコース溶液の添加を続行した。グルコース溶液の添加が終了した後、得られた懸濁液をタービンスターラーで50℃においてさらに2時間攪拌し、次いで70℃で3時間攪拌した。
乾燥後に粉砕した硫酸バリウムを2.5〜5重量%の微粉末状硫酸銀と混合し、この2成分を十分に混和させた。
被覆処理に付した硫酸バリウム/硫酸銀混合物20重量%をポリウレタンペレット77.6重量%および他の非被覆物(例えば、二酸化チタン)2.4重量%と十分に混合させ、得られた混合物を加工処理(例えば、押出成形)に付した。
A:銀コロイドの硫酸バリウムへの吸着
AgNO3(18g)を蒸留水(50℃)1080mlに溶解させ、得られた溶液に硫酸バリウム200gを添加した。得られた懸濁液を約20分間激しく攪拌した後、25%アンモニア水溶液23.4mlと混合させた。
温度を同温に保持して攪拌を続行しながら、無水グルコースを9.6g含有する溶液をゆっくりと滴下した。グルコースの添加が終了後、上記の実施例2のAに記載の場合と同様の処理をおこなうことによって乾燥した硫酸バリウム微粉末を得た。
硫酸銀の混合は実施例2のBに記載の場合と同様にしておこなった。
実施例2の場合と同様して、硫酸バリウムと硫酸銀との混合物を他の構成成分と混合した後、加工処理に付した。
本発明によるプラスチックの抗菌活性は、種々の微生物を含有するトリプカーゼ(trypcase)−大豆ブイヨン栄養溶液と共に該プラスチックの試料を37℃でインキュベートすることによって測定した。
1.スタフィロコッカス・エピデルミジス:ATCC14990。
2.スタフィロコッカス・エピデルミジス:カテーテルに関連する敗血症患者から臨床的に単離した直後のもの。
3.スタフィロコッカス・アウレウス:ATCC25923。
4.エシェリシア・コリ:カテーテルに関連する敗血症患者から臨床的に単離した直後のもの。
5.シュードモナス・アエルギノサ:カテーテルに関連する敗血症患者から臨床的に単離した直後のもの。
ポリウレタン[テコフレックス(Tecoflex)社製]を使用した実質上全ての移植可能な中枢静脈用カテーテルを製造した。ナノシルバー(粒径:3〜5nm)0.8〜1.3重量%および種々の濃度(0.25%、0.5%、0.75%または1.0%)の硫酸銀と共にポリウレタンを共押出加工処理に付した。外径が1.6mmの押出物を製造した。これらの押出物から長さが1mmのペレットを切り取った。10個のペレットの表面積は約1cm2であり、50個のペレットの表面積は5cm2であった。
生理的塩水中に上記の微生物を5×107CFU/mlまたは109CFU/mlの量で含有する懸濁液中へ、表面積が1cm2または5cm2のプラスチックペレットを導入した。これらの試験片を120回転/分の速度で振盪させた。各々の試験においては、試験の開始時(微生物の初期量)並びに6時間後、12時間後、18時間後、24時間後、36時間後、および48時間後に1ループの試料(2μl)を取出して寒天(ミュラー−ヒントン寒天)上に移し、37℃で24時間インキュベートした。次いで寒天上の微生物の数をコロニーの計数によって測定した。
全ての試験は3回繰返した。以下に示すいずれのデータも3回の対応する試験の平均値である。
ケイ酸ジルコニウム含有担体の実験
硫酸バリウム担体を、第1の一連の実験においては20重量%のケイ酸ジルコニウムと混合し、また、第2の一連の実験においては20重量%のナノシルバーおよび20重量%のケイ酸ジルコニウムと混合した。得られた混合物を種々の量の微生物と混合し、該微生物の増殖を48時間にわたって記録した。
硫酸バリウム担体上に坦持させたナノシルバーと発生期のリン酸銀を用いたときの抗菌活性試験(Ag:3.6%;リン酸銀:5%)
硫酸バリウム上へのコロイド状銀の吸着および発生期の超微細リン酸銀の調製について説明する。
硝酸銀(14.45g)を蒸留水(50℃)(360ml)に溶解させ、得られた溶液中への硫酸バリウム(100g)を激しく攪拌しながら添加した。得られた懸濁液を約20分間攪拌した後、25%アンモニア水溶液(19.3ml)を添加した。

硝酸銀(9g)を50℃まで加温した蒸留水(360ml)に溶解させ、得られた溶液中へ激しく攪拌しながら硫酸バリウム(100g)を添加し、攪拌を20分間続行した後、25%アンモニア溶液を添加した。
次いで、温度を同温に維持しながら、グルコース1水和物(5.25g)を蒸留水(182ml)に溶解させた溶液をゆっくりと添加した。グルコースの添加終了後、得られた懸濁液を50℃においてさらに2〜4時間攪拌し、次いで70℃で1〜3時間攪拌を続行した。
反応が完結した後、水性相から固体を分離させ、該固体を、電解質が検出されなくなるまで超純水を用いて繰返して洗浄した。洗浄した固体を乾燥室内において70℃〜80℃で乾燥させた後、一次粒径まで粉砕させた。
所望量(1〜5重量%)の超純度リン酸銀を上記のAで得られた固体へ添加し、この2成分を十分に混合した。実施例4に記載のような試験をおこなったところ、実施例7に記載した結果に類似した良好な結果が得られた。
Claims (17)
- 下記の工程(A)〜(C)を含む抗菌性プラスチック製品の製造方法:
(A)中間製品を形成させ、
(B)中間製品の少なくとも1つの構成成分を抗菌性金属コロイドで処理し、次いで
(C)抗菌性金属の易溶性塩または難溶性塩を添加する。 - 難溶性塩が下記の塩から成る群から選択される塩である請求項1記載の方法:銀塩、亜鉛塩、銅塩、セリウム塩、ジルコニウム塩、ビスマス塩、白金塩および/または金塩。
- 金属塩が硫酸銀および/またはリン酸銀を含有する請求項2記載の方法。
- 金属塩の含有量が、中間製品の全重量に基づいて0.1〜1.0重量%である請求項3記載の方法。
- 金属塩を、銀/銅比が約2:1になる割合で存在させる請求項2記載の方法。
- 中間製品が1種もしくは複数種のポリマー材料を含有する請求項1から5いずれかに記載の方法。
- 中間製品がポリウレタンを含有する請求項6記載の方法。
- 中間製品が添加剤をさらに含有する請求項1から7いずれかに記載の方法。
- 添加剤が有機粒子または無機粒子を含有する請求項8記載の方法。
- 有機粒子および/または無機粒子が下記の物質から成る群から選択される物質の粒子である請求項9記載の方法:硫酸バリウム、硫酸カルシウム、硫酸ストロンチウム、二酸化チタン、酸化アルミニウム、二酸化ケイ素、ゼオライト、フッ化カルシウム(CaF2)、マイカ、タルク、熱分解法シリカ、カルシウムヒドロキシルアパタイト、カオリンおよび/またはマイクロセルロース。
- 無機粒子が硫酸バリウムおよび/または熱分解法シリカを含有する請求項10記載の方法。
- ポリマー材料および無機粒子を金属コロイドで処理する請求項1から11いずれかに記載の方法。
- 無機粒子を金属コロイドで処理する請求項1から11いずれかに記載の方法。
- 金属コロイドがコロイド状銀を含有する請求項1から13いずれかに記載の方法。
