Abstract
Description
本発明は、一般に、核酸分野に関し、より詳細には、gp120に特異的に結合するアプタマーまたはアプタマー組成物を使用したHIVの治療または予防のための組成物および方法に関する。
アプタマーは、古典的ワトソン−クリック塩基対合以外の相互作用によって分子に対する特異的結合親和性を有する核酸分子である。
本発明の新規の態様は、目的の標的の特定の所望の高次構造への変化を促進する核酸を単離するためのSELEX(「アゴニストSELEX」)の使用である。好ましい実施形態では、目的の標的はgp120であり、所望の高次構造の変化は、例えば、gp120を誘導することによって感染時に存在する中間構造を予想して「固定する」有効な中和抗体応答を誘発するものである。目的の標的は、細胞表面受容体であり、所望の高次構造の変化は、細胞内シグナル伝達経路またはウイルス表面分子のサブユニットを誘発するものであり、所望の高次構造の変化は、ウイルスの一部としてその天然の構造中にサブユニットが固定されたものであり得る。
(定義)
本明細書中で使用される、「アプタマー」は、古典的ワトソン−クリック塩基対合以外の相互作用によって分子に対する特異的結合親和性を有する核酸分子である。
gp120に対するアプタマーの適切な作製方法は、一般に、図1に示す「試験管内進化法」(「SELEXTM」)と呼ばれるプロセスを使用する。SELEXTMプロセスは、標的分子に高特異性で結合する核酸分子のin vitro進化(evolution)法であり、例えば、1990年6月11日提出の米国特許出願番号07/536,428号(現在は放棄)、「核酸リガンド」というタイトルの米国特許第5,475,096号、および「核酸リガンド」というタイトルの米国特許第5,270,163号(WO91/19813号も参照のこと)に記載されている。各SELEXによって同定された核酸リガンドは、所与の標的化合物または分子の特異的リガンドである。SELEXTMプロセスは、核酸が種々の二次元および三次元構造の形成に十分な能力ならびに実質的に任意の化合物(単量体または重合体のいずれでも)とのリガンドとして作用する(特異的結合対を形成する)ためのその単量体内で利用可能な十分な化学的多用途性を有するという固有の洞察に基づく。任意のサイズまたは組成の分子を、標的として使用することができる。

標的およびエフェクターの結合特性の両方を有する分子が出発プールに存在する確率が低い場合、選択方法(1)を以下のように修正することができる。最初の標的結合およびエフェクター依存性の両方の選択の代わりに、in vitro選択を使用して、2つの個別の分子プール(一方はエフェクターに高親和性を有し、他方は標的に高親和性を有する)を単離することができる。2つのプール内のサブドメインを、各分子がエフェクター結合モチーフの1つのコピーおよび標的結合モチーフの1つのコピーを含む分子のキメラプールを作製するように操作することができる。前述のように、このキメラプールを、エフェクター依存性の標的結合選択に供する。
図4は、治療目的のアプタマーの作製に典型的に必要な工程を示す。プロセスは、およそ以下の4つの段階と見なすことができる:(i)および(ii)アプタマー同定、(iii)アプタマーの最小化、および(iv)安定性についてのアプタマーの至適化。
一旦sp120BaL結合のナイーブプールが首尾よく富化されると、アゴニスト(または活性)ベースの選択ストラテジー(アゴニストSELEX)を行った。CCR5ペプチドのみに結合することができるRNA分子を除去するために、4×1014〜4×1015個のナイーブRNAプール分子の、100μl結合反応物を含む選択緩衝液を含むニュートラビジンコーティング96ウェルプレート(Pierce)に固定した配列:
SELEXにより、典型的には、70〜90ヌクレオチド長のRNA分子が得られる。アプタマー長の最小化により、アプタマー候補の化学合成が容易になり、別の非結合構造の排除によってアプタマー−リガンド複合体の親和性を増大させることができる。一旦各アプタマーが元のプールから同定されると、高親和性結合に必要な最小配列エレメントを、以下の2つの並行アプローチによって同定することができる:(1)部分的アルカリ化水分かによる短縮分析、および(2)ドーピング再選択(方法は、Fitzwater & Polisky,1996に概説されている)。
核酸は、血清中でエンドヌクレアーゼと5’→3’および3’→5’エキソヌクレアーゼとの組み合わせによって分解される。適切な化学修飾(または本明細書中に開示されているもの)は、各活性を遮断する(Pieken et al.,1991;Cummins et al.,1995;Jellinek et al.,1995;Dougan et al.,2000)。簡単に述べれば、転写反応における選択時の2’−フルオロピリミジンの組み込みおよび2’−O−メチルプリンの選択後付加によってエンドヌクレアーゼ分解からアプタマーが保護され、3’−3’チミジンキャップでの末端修飾により、エキソヌクレアーゼに対する有意な耐性を得ることができる。
さらなるラウンドによりgp120に結合したプールRNA画分または上で詳述した他の複合体をさらに増加させない時点まで選択が到達した場合、プールしたテンプレートDNAをTOPO TAクローニングシステム(Invitrogen)を使用してクローン化する。各クローンを配列決定する。以下に概説の技術を使用して、固有のクローンを所望の特性についてスクリーニングする。
アプタマーをgp120に共有結合する場合、有効なワクチン中でのアプタマー活性が増強される。その5’末端にポリエチレングリコール(PEG)スペーサーを使用して抗gp120アプタマーを合成して、第一級アミン部分の末端に約20〜200Å(オングストローム)のスペーサーを有するアプタマーが得られる。スペーサーの長さおよび「水のような」性質により、非結合2片系(uncoupled 2−piece system)で認められるものと同一の様式でアプタマーがgp120に結合可能である。標準的な変異誘発技術によって、gp120のN末端nおよびC末端ならびに非中和表面中の一連の単一システイン変異を作製する。次いで、Pierceから利用可能なヘテロ二官能性架橋剤(スルホ−LC−SPDP(スルホスクシンイミジル6−[3’−(2−ピリジルジチオ)−プロピオンアミド]ヘキサノエート))を使用して、アミン末端化アプタマーをgp120上の遊離チオールに共有結合させる。次いで、アプタマー/gp120抱合体を、上記のCCR5ペプチドまたはCCR5発現細胞に結合する能力についてスクリーニングする。複数のgp120 BaL変異体およびアプタマースペーサーの長さの試験により、生化学活性に至適な配置が同定される。
in vitro機能アッセイで活性が証明されたアプタマーをgp120に共有結合させ、複合体を中和抗体応答を誘導する能力についてアッセイした。免疫原を、最終濃度がそれぞれ50〜100μg/mlおよび100μg/mlのサポニンベースのQS21アジュバントを使用して処方する(Evans et al.,2001 およびMcGaughey et al.,2003)。少なくとも6組の免疫化試験を並行して行うことができる。モルモットに、i)至適化アプタマー/gp120抱合体、ii)混合配列(非機能的)アプタマー/gp120抱合体、iii)共有結合抱合を含まないアプタマー/gp120複合体、iv)共有結合抱合を含まない混合配列アプタマー/gp120複合体、v)gp120のみ、またはvi)アジュバントのみのいずれかで免疫化する。したがって、免疫源としてのgp120のみと比較したアプタマー、抱合、および核酸の効果を評価する。免疫応答を誘発するために、種々のアプタマー複合体をモルモットに注射する。各試験のために、3匹の動物に0.05ml(50〜100μg)のワクチンを皮下注射する。3週間間隔で2回の追加免疫化を行い、免疫化から10〜28日後に動物を交配する。gp120に対する血清抗体を、gp120 ELISAによって最初に定量する(Moore et al.,1989)。
1サイクルHIV−1感染アッセイにおいて、HIV−1 LTR駆動β−ガラクトシダーゼおよび非蛍光蛍光発生基質(5−クロロメチルフルオレセインジ−β−D−ガラクトピラノシド(CMFDG)(Molecular Probes))で一過性にトランスフェクトしたU87.CD4.CCR5細胞(NIH AIDS Research Reagent Databaseから利用可能)(Bjornal et al.,1997およびRichman et al.,2003)を使用して中和アッセイを行う。HIV感染により、フロレセイン産生によって検出することができるβ−ガラクトシダーゼ遺伝子の発現をトランス活性化するTatが発現される。各希釈物を三連で試験する。非特異的中和のコントロールとして前免疫血清も試験する。HIV−1 BaL株は、Advanced Bioscience Laboratories(Gaithersburg,MD)の1サイクル感染性アッセイに利用可能である。20Uのインターロイキン−2を含む50μlのRPMI完全培地(Hoffman−LaRoche)中のウイルス(50〜100、50%組織培養感染用量)を、同体積の連続希釈熱変性血清(56℃で35分間)と室温で10分間予備インキュベートする。次いで、この混合物を、トランスフェクトしたU87.CD4.CCR5細胞と共に96ウェルの平底プレートにて37℃で48時間インキュベートして感染およびβ−ガラクトシダーゼの産生サイクルを1回行う。次いで、蛍光発生基質CMFDGの添加およびPackard Fusion蛍光プレートリーダーにおけるフルオレセイン蛍光の定量によって、β−ガラクトシダーゼ産生を測定することができる。各希釈物を、三連で試験する。非特異的中和のコントロールとして前免疫血清も使用する。
アプタマーの血清安定性を、記載のようにin vitroでアッセイする(Green et al.,1995)。簡単に述べれば、2nMの5’−32P末端標識アプタマーをヒト血清中にて37℃でインキュベートする。特定の時点で87%ホルムアミドの添加によって反応を停止させ、変性PAGEによって変性率を分析する。
1つの実施形態では、gp120特異的結合アプタマーの選択を、スペーサー(例えば、PEGスペーサー)の末端で結合する(オリゴヌクレオチド)プローブを捕捉するためのRNAプールの連結によって容易にすることができる。プローブ−スペーサーを、gp120に対する公知の遺伝子座特異性を有するモノクローナル抗体に結合させるかまたはgp120に直接結合させる。この様式では、gp120の高次構造シフトを誘導することができる低親和性アプタマーをより容易に同定することができる。1つの実施形態では、当該分野で公知のリンカーおよび連結方法による抗体もしくはフラグメント上のグリコシル残基への連結化学によって、プローブ−スペーサーを、gp120特異的結合モノクローナル抗体もしくはそのフラグメントに連結させる。1つの実施形態では、当該分野で公知の前記リンカーおよび連結化学を使用して、プローブ−スペーサーを、gp120上のグリコシル残基への連結によってgp120と直接連結させる。
Claims (41)
- gp120およびgp120と結合してgp120の高次構造のシフトを誘導し、それにより体液性免疫応答を誘発することができる、膜結合受容体に対するエピトープが曝露される核酸アプタマーを含む組成物。
- 前記膜結合受容体に対するエピトープがCCR5に対するエピトープである、請求項1に記載の組成物。
- 前記膜結合受容体に対するエピトープがCXCR4に対するエピトープである、請求項1に記載の組成物。
- 前記核酸アプタマーが、配列番号9〜配列番号225から選択される、請求項1、請求項2、または請求項3に記載の組成物。
- gp120およびgp120と結合してgp120の高次構造のシフトを誘導し、それにより体液性免疫応答を誘発することができる、膜結合受容体に対するエピトープが曝露される核酸アプタマーを含む組成物を被験体に投与する工程を含む、被験体へのHIV感染症ワクチンの接種方法。
- 前記膜結合受容体に対するエピトープがCCR5に対するエピトープである、請求項5に記載の方法。
- 前記膜結合受容体に対するエピトープがCXCR4に対するエピトープである、請求項5に記載の方法。
- 前記核酸アプタマーが、配列番号9〜配列番号225から選択される、請求項5、請求項6、または請求項7に記載の方法。
- gp120およびgp120と結合してgp120の高次構造の変化を誘導し、それにより体液性免疫応答を誘発することができる、膜結合受容体に対するエピトープが曝露される核酸アプタマーを含む組成物を投与する工程を含む、gp120に特異的な中和抗体の産生方法。
- 前記膜結合受容体に対するエピトープがCCR5に対するエピトープである、請求項9に記載の方法。
- 前記膜結合受容体に対するエピトープがCXCR4に対するエピトープである、請求項9に記載の方法。
- 前記核酸アプタマーが、配列番号9〜配列番号225から選択される、請求項9、請求項10、または請求項11に記載の方法。
- 前記中和抗体が産生される、請求項9に記載の方法。
- gp120およびgp120と結合してgp120の高次構造の変化を誘導し、それにより体液性免疫応答を誘発することができる、膜結合受容体に対するエピトープが曝露される核酸アプタマーを含む組成物を投与する工程を含む、HIV感染症の処置方法。
- 前記膜結合受容体に対するエピトープがCCR5に対するエピトープである、請求項14に記載の方法。
- 前記膜結合受容体に対するエピトープがCXCR4に対するエピトープである、請求項14に記載の方法。
- 前記核酸アプタマーが、配列番号9〜配列番号225から選択される、請求項14、請求項15、または請求項16に記載の方法。
- 配列番号9〜配列番号225から選択されるgp120に結合するアプタマー。
- 配列番号9〜配列番号225から選択されるgp120に結合するアプタマーを含む組成物。
- 前記アプタマーが配列番号9〜配列番号225から選択される、gp120と複合体化したgp120に結合するアプタマー。
- 治療有効量の配列番号9〜配列番号225から選択されるgp120に結合するアプタマーを投与する工程を含む、被験体におけるHIV感染症の処置方法。
- gp120と複合体化したgp120に結合する治療有効量のアプタマーを投与する工程であって、前記アプタマーが配列番号9〜配列番号225から選択される工程を含む、被験体におけるHIV感染症の処置方法。
- gp120と複合体化したgp120に結合する予防有効量のアプタマーを投与する工程であって、該アプタマーが配列番号9〜配列番号225から選択される工程を含む、被験体におけるHIV感染症の予防方法。
- gp120と複合体化したgp120に結合するアプタマーを投与する工程であって、該アプタマーが配列番号9〜配列番号225から選択される工程を含む、HIV gp120に対する特異性を有する中和抗体の産生方法。
- gp120と複合体化したgp120に結合するアプタマーを投与する工程であって、該アプタマーが配列番号9〜配列番号225から選択される工程を含む、gp120に特異的な中和抗体が得られる、被験体の体液性免疫応答を誘発する方法。
- 以下:
(a)第1級アミン部分で終わる約20Åから約200Åまでのスペーサーを含むアプタマーを合成する工程;
(b)gp120のN末端、C末端、および/または非中和表面中のシステイン変異上に単一の遊離チオールを生成する工程;
(c)架橋剤を使用して該アプタマーをgp120上の該遊離チオールと共有結合させる工程;ならびに
(d)gp120エピトープへの結合についてアプタマーをスクリーニングする工程
を含む、gp120と結合してgp120の高次構造の変化を誘導し、それにより体液性免疫応答を誘発することができるエピトープが膜結合受容体に曝露されるアプタマーの同定方法。 - 以下:
(a)第1級アミン部分で終わる約20Åから約200Åまでのスペーサーを含むアプタマーを合成する工程;
(b)gp120に結合する抗体または抗体フラグメント中のシステイン変異上に単一の遊離チオールを生成する工程;
(c)架橋剤を使用して該アプタマーを該抗体または抗体フラグメント上の該遊離チオールと共有結合させる工程;ならびに
(d)gp120エピトープへの結合についてアプタマーをスクリーニングする工程
を含む、gp120と結合して前記gp120の高次構造の変化を誘導し、それにより体液性免疫応答を誘発することができるエピトープが膜結合受容体に曝露されるアプタマーの同定方法。 - 前記スペーサーがポリエチレングリコールである、請求項26または請求項27に記載の方法。
- 前記架橋剤がヘテロ二官能性架橋剤である、請求項26または請求項27に記載の方法。
- 前記ヘテロ二官能性架橋剤が、スルホ−LC−SPDP(スルホスクシニミジル6−[3’−(2−ピリジルジチオ)−プロピオンアミド]ヘキサノエート)である、請求項29に記載の方法。
- 前記gp120エピトープが、CCR5およびCXCR4から選択される、請求項30に記載の方法。
- 前記スクリーニングをgp120 BaL変異体を使用して行い、該スクリーニングをCCR5ペプチドまたは全長CCR5受容体を使用して行う、請求項26または請求項27に記載の方法。
- 第2のリガンドを認識する第2の結合ドメインに結合した第1のリガンドを認識する第1の結合ドメインを含むアプタマーにおいて、該第2の結合ドメインによる第2のリガンドの結合が、該第1の結合ドメインによる該第1のリガンドの結合によって調節される、アプタマー。
- 前記第1のリガンド結合ドメインがアロステリックエフェクター分子と特異的に相互作用し、前記第2のリガンド結合ドメインが該アロステリックエフェクター分子の標的と特異的に相互作用する、請求項33に記載のアプタマー。
- 前記アロステリックエフェクター分子が、gp120であり、前記標的がCCR5受容体である、請求項34に記載のアプタマー。
- 前記第2の結合ドメインによる前記第2のリガンドの結合が、前記第1の結合ドメインによる前記第1のリガンドの結合によって活性化される、請求項33に記載のアプタマー。
- 前記第2の結合ドメインによる前記第2のリガンドの結合が、前記第1の結合ドメインによる前記第1のリガンドの結合によって活性化される、請求項34に記載のアプタマー。
- 前記第2の結合ドメインによる前記第2のリガンドの結合が、前記第1の結合ドメインによる前記第1のリガンドの結合によって抑制される、請求項33に記載のアプタマー。
- 前記第2の結合ドメインによる前記第2のリガンドの結合が、前記第1の結合ドメインによる前記第1のリガンドの結合によって抑制される、請求項34に記載のアプタマー。
- SELEXを使用して、それぞれ第1および第2のリガンドに結合する第1および第2のアプタマーを単離する工程、該第1および第2のアプタマーの結合ドメインを含む分子の種々の配列プールを操作する工程、ならびに該プールから調節されたアプタマーを選択する工程であって、該第2の結合ドメインによる第2のリガンドの結合が該第1の結合ドメインによる第1のリガンドの結合によって調節される工程を含む、調節されたアプタマーを選択する方法。
- 治療有効量のCCR5受容体アプタマーを投与する工程であって、該CCR5受容体アプタマーがgp120エフェクターに結合し、該gp120エフェクターによって活性化されて該CCR5受容体に結合する工程と、細胞へのgp120の結合を防止する工程を含む、被験体におけるHIVの処置方法。
