Abstract
Description
IL−18BPmRNAは、白血球、結腸、小腸、前立腺および特には脾臓細胞において検出される(Novick et al. 1999)。脾臓細胞は、マクロファージ、リンパ球および血漿細胞、さらに循環(circulation)由来の細胞から構成される。
IFNγがIL−18BPmRNAおよびタンパク質を、ケラチノサイト細胞株、結腸癌細胞株、および主には腎臓メサンギウム細胞などの種々の細胞株において誘導されることが以前に報告されている(Muhl et al. 2000)。種々のヒト細胞株および末梢血単核細胞(PBMC)における、IFNγや他のサイトカインによるIL−18BPの誘導を検討した。IFNγは、用量依存的にIL−18BP発現を誘導し(mRNAのモニタリングについては実施例11、ELISAについては実施例12参照)、WISHおよびHepG2細胞において50U/mlのEC50を示した(図2AおよびB)。IL−18BPは、その濃度が24時間と比較して48時間で高かったように(図2A)、明らかにWISH細胞の培養上清に蓄積された。
Il−18BPプロモーター領域を検討するために、まず、転写開始部位を明確に位置付けることが要求される。
IL−18BP遺伝子の上流で、ルシフェラーゼレポーター遺伝子などの異種遺伝子の発現を導くことができる最小DNAフラグメントを見つけるために、ルシフェラーゼ遺伝子に融合された、塩基1の上流のDNA配列に相当する1601bpまでを含み、転写開始部位の下流50bp(配列番号5)を含むベクター、ならびにこのDNAの切断型を有するベクター(図5A)を作出した(実施例15)。1601bp上流DNAを含むベクター(pGL3(1601))でトランスフェクトしたヒトHepG2細胞(ヒト肝細胞癌株)におけるルシフェラーゼ活性は、空のpGL3ベクターで得られるものより10.3±0.9倍高かった。そのような活性は、同じDNAを反対の方向で挿入した(pGL3(−1601))場合には観測されなかった。この結果は、塩基1の上流1601bpDNAが基礎プロモーター活性を有するということを証明した。この1601bpDNAフラグメントの配列決定試験により、それはTATAボックスエレメントを含まず、いくつかのGCリッチドメインを転写開始部位近くの塩基−3から−9、−39から−48および−122から−132に有することが明らかになった。プログラムTFSEARCHによる1601bpDNA配列の分析は、塩基−24から−32にガンマ活性化配列(GAS)を同定した(図1)。さらなる分析により、塩基−57から−69にかかるIRF1、2応答エレメント(IRF−E)および塩基−309から−322および−621から−634の2つのC/EBPβ応答エレメント(C/EBP−E)が明らかになった。
IL−18BPプロモーターの上流領域でIFNγ誘導ルシフェラーゼ発現を促進することができる最小DNAフラグメントを同定するために、前述の実施例の切断DNAベクターを、IFNγの存在下でトランスフェクトされたHepG2細胞において検討した(トランスフェクションについては図5B、実施例16参照)。
IL−18BP発現におけるIRF−1の関与、すなわちIL−18BPのプロモーターにおいて見出されたその結合部位を探るために、IRF−1欠損マウスにおけるIL−18BPの発現を検討した。
IL−18BPプロモーター内の様々な応答エレメントのあいだのタンパク質−DNA相互作用を同定するために、電気泳動度シフトアッセイ(EMSA 実施例18)が用いられた。塩基−33から−75(IRF−Eを含有する)および−8から−55(GASを含有する)に対応するラベルされたdsDNAプローブを、コントロール細胞とIFNγ処理細胞の核抽出物と結合させた。IRF−E含有プローブと核タンパク質の複合体は、IFNγとの1時間の細胞のインキュベーションの後に明らかとなり、最大の応答は、3時間で見られた(図7A、レーン1〜5)。予想されたように、IRF−Eの抗体の添加は、「スーパーシフト」を招き、一方、コントロールの転写1の抗シグナル誘導因子および活性化因子(STAT1)抗体は、効果を持たなかった(データは示さない)。IRF−Eとは対照的に、GAS−含有プローブは、本質的にタンパク質と関係付けられ(図7A、レーン6)、この複合体は3から6時間IFNγによる細胞の誘導の際増強された(レーン7、8)。GASは、IFNγ−誘導STAT1二量体に結合することが予想される。にもかかわらず、その複合体は、STAT1の抗体に影響されず(レーン10)、IFNγ−誘導STAT1二量体はこのGASと無関係であることが示唆された。同様の負の結果は、IFNγでたった15分または30分間処理した核抽出物で得られた(データは示さない)。驚くべきことに、この複合体はC/EBPβの抗体により廃止され(レーン9)、IRF−1の抗体でスーパーシフトした(レーン10)。したがって、GAS−含有DNAプローブは合意C/EBPβEの欠如にもかかわらず、C/EBPβと結合すると思われる。
IL−18BP遺伝子誘導におけるIRF−1およびC/EBPβの役割をさらに研究するために、発現ベクターのトランスフェクションを用いることによるIRF−1およびC/EBPβの過剰発現ののち、IL−18BPamRNAを半定量的RT−PCRにより測定した(実施例14、図7B)。HepG2細胞における各転写因子または両因子の組合せのいずれの過剰発現もIL−18BPmRNAを誘導しなかった。この結果は、付加的なIFNγ−誘導因子がIL−18BP遺伝子の活性化のために要求されるということを示唆した。IFNγによる誘導ののち、いずれかの1つの発現ベクターによる細胞のトランスフェクションは、IFNγ単独と比較して、実際はIL−18BPmRNAを減じた。対照的に、2つの転写因子の共発現は、IFNγによるIL−18BPmRNAの誘導を増加した。この結果は、IRF−1およびC/EBPβが、特定の比率で存在しなければならず、あるいは転写開始複合体の内部に複合体を形成するということを示唆している。IRF−1とC/EBPβとのあいだの可能な相互作用をさらに研究するために、pGL3(1272)、定量のIRF−1発現ベクターおよび可変量のC/EBPβ発現ベクターでの共トランスフェクト細胞によるルシフェラーゼ活性の滴定を行なった。同様に、C/EBPβベクターが一定に保たれ、IRF−1ベクターが可変量でのルシフェラーゼ活性が測定された。両ケースにおいて、ベル型の用量応答曲線が得られ、最適のIL−18BP誘導が、これらの2つの転写因子のあいだの固定モル比を必要とすることが示唆された(図7C)。
−309から−322位および−621から−634位の2つのC/EBPβ部位は、隣接したIRF−Eをもたない。実際、−309から−322位のC/EBPβ部位に対応するプローブのEMSA(実施例18)は、C/EBPβの抗体によりスーパーシフトされる(空矢印)が、IRF−1の抗体によってはスーパーシフトされない遅延バンド(rentarded band)(塗矢印)を示した(図8A)。したがって、この部位は、C/EBPβに結合し、IRF−1とのその複合体には結合しないと推論された。さらに、このバンドは、本質的にC/EBPβを発現し、IRF−1を欠いた非誘導HepG2細胞の核抽出物で生成された。実際、IFNγは、これらの細胞においてC/EBPβの発現を増加させず(図8D)、その結果、遅延バンドの強度を増加させなかった(図8A)。同様の結果は、より遠位のC/EBPβ部位で得られた(データは示さない)。
遠位のエンハンサーの調節的役割を、ヌクレオチド−1081から−1272に対応する192bpのDNAプローブでEMSAにより検討した(実施例18)。コントロールHepG2細胞の核抽出物は、このプローブと複合体を形成した(図8B、塗矢印)。IFNγによる細胞の処理では、複合体はより強く、いくぶんより遅延された。IRF−1、C/EBPβおよびcFosに対する抗体によるこの複合体のスーパーシフトが行なわれた。このうち、抗IRF−1のみがスーパーシフトを導き出した(図8B、空矢印)。ヌクレオチド−1083から−1174に対応する非ラベルdsDNAは、放射線ラベルプローブと競合せず、核タンパク質は、残基−1175から−1272に結合することを示した。IRF−Eは近位の領域でのみ同定されたので、この結果は、遠位のエンハンサーは、おそらく近位のIRF−Eと関係付けられるということを示唆する。
ヒトIL−18BPは、記載されたように二重抗体ELISAにより測定した(Novicket al. 2001)。マウスIL−18BPは、マウスIL−18BPに対するウサギ抗原アフィニティー−精製ポリクローナル抗体およびビオチン化抗体(サイトラブ、イスラエルより入手)を用いて二重抗体ELISAにより測定した。
血清不含培地での処理後、HepG2およびWISH細胞(106)を示した時間で回収し、総RNAはTRI試薬を用いて抽出した。cDNAは、ランダム六量体およびSuperscriptII(インビトロゲン(商標)、リーク(Leek)、オランダ)を用いて、製造業者の使用説明書にしたがって製造した。PCRは、以下のプライマー:ヒトIL−18BP 5′CACGTCGTCACTCTCCTGGおよび5′CGACGTGACGCTGGACAAC;ヒトIRF−1 5′GACCCTGGCTAGAGATGCAGおよび5′GAGCTGCTGAGTCCATCAG;ヒトβアクチン 5′GTGGGGCGCCCCAGGCACCAおよび5′CTCCTTAATGTCACGCACGATTTCを用いて行なった。増幅は、初期変性(92℃、2分)、28サイクルの変性(92℃、45秒)、アニーリング(62℃、1分)および伸張(72℃、1.5分)、ならびに最終伸張(72℃、10分)でなされた。得られたPCR産物をアガロース(1%)ゲル電気泳動で分析した。
5′RACEは、5′RACEシステム(GIBCO BRL)を用いて製造業者の使用説明書にしたがって行なった。要約すると、IFNγ処理WISH細胞からの総RNAを、IL−18BPamRNA(ジーンバンク、受託番号AF110799)のヌクレオチド89〜70に相補的なプライマーを用いて逆転写し(実施例13)、アンカーDNAで新規に合成された末端をテーリングした。つぎに、アンカーDNAに相補的なフォワードプライマーと、IL−18BPamRNAのヌクレオチド31〜11に相補的なネストされたリバースプライマーによりPCRを行なった。ついで、PCR産物をサブクローニングし、DNA配列分析にかけた。
1601bpの全推定IL−18BPaプロモーター領域を、KpnI部位を含むセンスプライマー(S4753.pgl)(5′CTATATGGTACCCACCCTTCCTTTTACTTTTTCC)とNheI部位を含むリバースプライマー(RlexA)(5′TATCGCTAGCCAGTCACACAGGGAGGCAGT)とを用いてゲノムDNAのPCRにより得た。PCR産物をpGEM−T Easyベクター(プロメガ、マジソン、WI)にクローニングし、DNA配列分析により確認した。KpnI−NheIフラグメントをpGEM−T Easyクローンから単離し、急速DNAライゲーションキット(ロッシュ)を用いてpGL3−Basicベクター(プロメガ)にライゲートしてpGL3(1601)を得た。一連の5′−切断レポーター(pGL3(1454)、pGL3(1274)、pGL3(1106)、pGL3(656)、pGL3(280)およびpGL3(122))を同様にして、同一のリバースプライマーとそれぞれ以下のセンスプライマーで製造した。

すべてのPCR産物は、コントロールとして役立てるために逆方向でもpGL3−Basicベクターにクローニングした。
6ウェルプレート(0.5×106/ウェル)においてHepG2またはWISH細胞を、FuGENE6および示したルシフェラーゼレポーターベクター(0.5μg/ウェル)およびpSV40βGAL(0.2μg/ウェル、プロメガ)を用いて製造元の使用説明書にしたがってトランスフェクトした。いくつかのケースでは、共トランスフェクションが、以下の発現ベクター:pcDNA3−IRF−1(0.07〜1.5μg/ウェル、B.Levy博士から提供、テクニオン(Technion)、イスラエル);pcDNA3−C/EBPβ(0.5〜2.5μg/ウェル、D.Zipori博士から提供、ワイズマン インスチチュート オブ サイエンス)16時間後、細胞をIFNγ(100U/ml)、IL−6(150U/ml)、TNFα(10ng/ml)またはそれらの示した組合せのいずれかで、血清不含培地において24時間処理した。ついで細胞を回収し、溶解し、ルシフェラーゼ活性を測定した。すべての結果は、βガラクトシダーゼ活性に対して標準化した。
細胞を氷冷リン酸緩衝生理食塩水(PBS)で3回洗浄し、液体窒素で速やかに凍結した。細胞ペレットを、4包装細胞容量(four packed cell volume)の細胞質緩衝液(10mM Hepes、pH 7.9、10mM NaCl、0.1mM EDTA、5容量%グリセロール、1.5mM MgCl2、1mM ジチオスレイトール(DTT)、0.5mM PMSF、50mM NaF、0.1mM Na3VO4、2mM EGTA、10mM EDTA、10mM Na2MoO4、ロイペプチン、ペプスタチンおよびアプロチニン 各2μg/ml)に再懸濁した。溶解物を遠心分離(3000×g、10分)し、細胞質タンパク質を含む上清を回収した。ペレットを2.5包装細胞容量の核緩衝液(NaClが0.42Mに増加されている以外は細胞質緩衝液と同一)に再懸濁した。核デブリを遠心分離(1,5000×g、20分、4℃)により除去し、上清の部分標本を液体窒素で凍結し、−80℃で保管した。タンパク質濃度はBCAタンパク質アッセイ試薬キット(ピアス、ロックフィールド、USA)によりウシ血清アルブミンを標準として用いて決定した。
選択された応答エレメント(10pmol)に対応するdsオリゴヌクレオチドをポリヌクレオチドキナーゼ(ニュー イングランド バイオラブ(New England Biolabs))により[32P]3 ATPで標識した。核抽出物(5μgタンパク質)を、20μlのEMSA緩衝液(20mM Hepes、pH 7.5、5mM MgCl2、2mM EDTA、5mM DTTおよび5容量%グリセロール)中でポリ(dI−dC)(アマシャム ファルマシア バイオテクノロジー)と共に前インキュベート(15分、0℃)した。ついで標識したプローブ(3×104cpm)を添加し、インキュベーションを室温でさらに30分継続した。スーパーシフトアッセイのために、試料をプローブの添加前に示した抗体とインキュベートした(4μg、1h、0℃)。200倍過剰の野生型と変異型の競合物を、関連プローブと共に添加した。ついで、反応混合物を5%非変性ポリアクリルアミドゲルで電気泳動した。ゲルを真空で乾燥し、−80℃で一晩オートラジオグラフにかけた。
核または細胞質タンパク質抽出物(80μg)を、6μgの示したポリクローナル抗体と共に一晩4℃でインキュベートし、タンパク質Gセファロースビーズ(ファルマシア)で1時間室温で免疫沈降した。次にビーズを、10%DTTを含むSDS−PAGE試料緩衝液で煮沸し、上清を還元条件下でSDS−PAGE(10%アクリルアミド)により分離した。ついでゲルをニトロセルロース膜にブロットし、示した抗体でタンパク質を検出した。免疫複合体は、Super Signal(商標)(ピアス)検出キットにより同定した。
ヒト可溶性LDLRを発現する安定な組換えCHO細胞は、ジヒドロ葉酸還元酵素(DHFR)遺伝子を欠いたCHO−DUKX細胞を2つの発現ベクター:LDLRのN−末端リガンド結合ドメインを含み、アミノ酸残基Asp(+4)で始まりGlu291(+291)までのもの、およびDHFR(DHFRは初期SV40プロモーターによって制御されている)のマウス遺伝子およびIL−18BPプロモーター(配列番号2)によるsLDLR遺伝子およびSV40初期領域の転写末端エレメントを含むpDHFRでの共トランスフェクションにより作出する。トランスフェクションは、リポフェクトアミン(LipofectAmine)(ギブコ BRL)を用いて製造元のプロトコールに従い陽イオンのリポソームにより行なわれた。トランスフェクションの72時間後に、細胞をデオキシおよびリボヌクレオシドを欠き、10%透析FCSを補完した選択培地に移す。DHFR活性を発現する細胞は、トリプシン浸漬ペーパーディスクで細胞をリフティングすることにより単離されるクローンを形成することができる。細胞は増殖し、r−hsLDLR活性に対してスクリーニングした。ついで、トランスフェクトされた細胞をMTXによる遺伝子増幅に付し、そののちサブクローニングし安定な産生クローンを選択する。

Claims (34)
- ヒトIL−18BPプロモーターをコードするDNA配列(配列番号1)、またはそのフラグメントもしくは機能的誘導体であって、該DNA配列またはそのフラグメントの3′末端が配列番号5の5′末端由来の1つ以上のヌクレオチドを含むDNA配列。
- 誘導体が配列中のサイレンサーエレメントに存在する1つ以上のAP1部位で変異されている請求項1記載のDNA配列。
- フラグメントが配列番号2を含有する請求項1記載のDNA配列。
- フラグメントが配列番号3を含有する請求項1記載のDNA配列。
- さらにイントロンを含有する請求項1、2、3または4記載のDNA配列。
- イントロンがIL−18BPの第1イントロンからなる請求項5記載のDNA配列。
- さらにIL−18BPプロモーターに作動可能に連結された遺伝子を含む請求項1、2、3、4、5または6記載のDNA配列。
- 遺伝子がIL−18BPをコードする請求項7記載のDNA配列。
- 遺伝子が異種タンパク質をコードする請求項7記載のDNA配列。
- 異種遺伝子がルシフェラーゼ遺伝子をコードする請求項9記載のDNA配列。
- 異種遺伝子が、インターフェロン−ベータ、TNF、エリスロポエチン、ティッシュプラスミノーゲンアクチベーター、顆粒細胞コロニー刺激因子、超酸化マンガンジスムターゼ、イムノグロブリン、またはそれらのフラグメント、成長ホルモン、FSH、hCG、IL−18、hsLDLRおよびTNF受容体結合タンパク質から選択されるタンパク質をコードする請求項9記載のDNA配列。
- 請求項1、2、3、4、5、6、7、8、9、10または11記載のDNA配列を含有するベクター。
- 請求項12記載のベクターを含有する宿主細胞。
- 哺乳類の細胞である請求項13記載の宿主細胞。
- CHO、WISH、HepG2、Cos、CV−1、HeLa、およびHakat U937細胞から選択される請求項14記載の宿主細胞。
- 請求項13、14または15記載の宿主細胞を培養し、産生された組み換えタンパク質を単離することを含む組み換えタンパク質の製造方法。
- ウイルスゲノムの一部分、目的の遺伝子をコードするDNAフラグメントおよび目的の遺伝子に作動可能に連結された請求項1、2、3、4、5または6記載のヒトIL−18BPプロモーターをコードするDNA配列を含むDNAフラグメントを含有する組み換えウイルスベクター。
- 目的の遺伝子が、インターフェロン−ベータ、TNF、エリスロポエチン、ティッシュプラスミノーゲンアクチベーター、顆粒細胞コロニー刺激因子、超酸化マンガンジスムターゼ、イムノグロブリン、またはそれらのフラグメント、成長ホルモン、FSH、hCG、IL−18、hsLDLRおよびTNF受容体結合タンパク質から選択される請求項17記載の組み換えウイルスベクター。
- ウイルスゲノムの一部分がアデノ随伴ウイルスに属する請求項17記載の組み換えウイルスベクター。
- ウイルスゲノムの一部分がレトロウイルスに属する請求項17記載の組み換えウイルスベクター。
- レトロウイルスがHIV、HFV、MLV、FIVおよびVSVから選択される請求項20記載の組み換えウイルスベクター。
- 目的の遺伝子の細胞特異的発現を制御する方法であって、標的哺乳類細胞を請求項17、18、19、20または21記載のベクターで形質転換すること、および必要とする個体にその細胞を移植すること含む方法。
- 標的細胞が造血幹細胞である請求項22記載の方法。
- 標的細胞が単球である請求項22記載の方法。
- 標的細胞がマクロファージである請求項24記載の方法。
- 目的の遺伝子がHIV感染症に耐性を与えるタンパク質をコードする請求項22、23、24または25記載の方法。
- HIV感染症の治療のための請求項26記載の方法。
- 造血系疾患の治療のための請求項22、23、24または25記載の方法。
- 造血系疾患がSCID、慢性肉芽腫症およびサラセミアから選択される請求項28記載の方法。
- 体組織のIFNγの上昇を示す個体における疾患の治療のための遺伝子治療法であって、有効量の請求項17、18、19、20または21記載のベクターを投与することを含む方法。
- さらにIL−6および/またはTNF−αおよび/またはIRFおよび/またはC/EBPβ因子の投与を含む請求項30記載の方法。
- 請求項1、2、3、4、5、6、7、8、9、10または11記載のDNA配列をコードするDNA配列を宿すトランスジェニックマウス。
