遺伝情報の正確な伝達は、細胞、生物、および種の生存において重要である。変異が細胞に対し有害であり得る新しい遺伝子型をもたらし得ることから、遺伝物質を1つの世代から次の世代へ、高い正確さでの伝達を確実にするのに役立つ多くの機構が発達してきた。頻繁に変異をもたらすDNA損傷とは、改変された、失われた、またはミスマッチのヌクレオチドである。多様な酵素経路が、これらの損傷を特異的に修復し得る原核生物システムにおいて記載されてきた。
(1)真核細胞のDNAミスマッチ修復経路において変化があるかどうかを調べる方法であって、以下の工程:

(発明の要旨)
本発明者らは、今回、哺乳類を含む真核生物が、細菌に存在する経路に類似するDNAミスマッチ修復経路を有することを発見した。哺乳動物におけるこのミスマッチ修復経路の欠損または変化は、不安定なDNA反復配列の蓄積をもたらす。そのような表現型は、遺伝性結腸ガンのような多くのガンにおける、疾病状態に高い相関関係を有する。従って、本経路の欠損または変化を発見することは、ガンの素因に対する診察、および特定のガンに対する予後となり得る。
配列番号1は、酵母MSH2遺伝子のヌクレオチド配列である。
配列番号2は、酵母MSH1遺伝子のヌクレオチド配列である。
配列番号3は、酵母MSH2タンパク質のアミノ酸配列である。
配列番号4は、酵母MSH1タンパク質のアミノ酸配列である。

配列番号147/148〜153/154は、それぞれ配列番号62/63〜64/32のプライマーに対して「繰り込まれた(nested)」関係のプライマーのセットであり、実施例9に記載のに記載のような多様なPCRプロトコルにおいて、それぞれ配列番号62/63〜64/32のプライマーとともに使用され得る。
配列番号155は、ヒトMLH1遺伝子、hMLH1のcDNA配列である。
配列番号156は、配列番号155によってコードされるhMlh1タンパク質のアミノ酸配列である。
本発明者らは、今回、哺乳類を含む真核生物が、細菌に存在する経路に類似するDNAミスマッチ修復経路を有することを発見した。ヒトのような哺乳動物におけるこのミスマッチ修復経路の欠損または変化は、不安定なDNA反復配列の蓄積をもたらす。そのような表現型は、遺伝性結腸ガンのような多くのガンにおける疾病状態に高い相関を有する。従って、本経路の欠損または変化を発見することは、ガンの素因に対する診断および特定のガンの予後となり得る。
本明細書に記載の1つの好ましいアッセイは、ミスマッチ修復欠損腫瘍の診断および/または予後を可能にする。本発明の真核生物ヌクレオチド配列、ポリペプチド、および抗体は、以下のような腫瘍の疑いのある病理学上の条件を決定するのに特に有用である:(i)例えば、細菌のミスマッチ修復経路の類似体のメンバーに相同であるヌクレオチド配列の非野生型対立遺伝子を含み、および/または(ii) 細菌のミスマッチ修復経路の類似体のメンバーに相同であるヌクレオチド配列によってコードされるポリペプチド上の、少なくとも1つの抗原決定基を欠き、および/または新しい抗原決定基を含む。
本発明の真核生物ヌクレオチド配列の異なるバージョン(version)、すなわち「対立遺伝子」は、正常宿主細胞における細菌のミスマッチ修復遺伝子に相同である配列のような、内在性ヌクレオチド配列を機能的に置換する能力によって分類され得る。本明細書中で使用されるように、「野生型」対立遺伝子は、宿主細胞に検出可能な有害な効果をもたらさない、正常宿主細胞の内在性ヌクレオチド配列を置換し得る配列として定義される。「非野生型」対立遺伝子または「変化」は、宿主細胞に検出可能な有害な効果をもたらさない、正常宿主細胞の内在性ヌクレオチド配列を置換し得ない真核生物ヌクレオチド配列として定義される。
宿主細胞における、細菌のミスマッチ修復遺伝子に相同である内在性ヌクレオチド配列および/またはそれらがコードするポリペプチドの不活性化
本発明の単離された真核生物ヌクレオチド配列および単離されたポリペプチドによって提供される情報を用いて、例えば、細菌のミスマッチ修復遺伝子に相同な内在性ヌクレオチド配列および/または細菌のミスマッチ修復遺伝子に相同な内在性ヌクレオチド配列によってコードされるポリペプチド産物を、宿主細胞において不活性化し得る(実施例2、実施例6を参照のこと)。
本発明の単離された真核生物ヌクレオチド配列および単離されたポリペプチドは、ヒドロキシルアミンによる処理、変異誘発性細菌株を介しての継代(passage)移行を包含するいくつかの標準的な手順のいずれかによって、変異が誘発され得る。次いで、変異が誘発された配列は上記のように、「野生型」または「非野生型」に分類され得る。
本発明の別の好ましい実施態様は、ガン罹病性の診断にある。本発明の真核生物ヌクレオチド配列、ポリぺプチド、および抗体は、上記のように、経路のメンバーの変化との相関を示すガンに対する罹病性の診断に特に有用である。このようなガンは、上記のように、ミスマッチ修復欠損腫瘍を含むと予想される。
本発明で提供される分子および宿主細胞は、ガンに有効な治療剤を同定するために用いられ得る。特に、本発明の分子および宿主細胞を用いてガンに対して有効な治療剤を同定しうる。ここでガンの発症率は、ミスマッチ修復経路における任意の変化(例えば、細菌のミスマッチ修復遺伝子に相同であるヌクレオチド配列の非野生型対立遺伝子の存在)、および/または、細菌のミスマッチ修復遺伝子に相同であるヌクレオチド配列によってコードされるポリペプチド上の少なくとも1つの抗原決定基の欠陥と相関する。
本明細書中に記載のヌクレオチド配列およびこれらの配列によって発現されるおよびポリペプチドは、例えば、遺伝子治療における薬学的組成物にもまた、使用され得る。1つの例示的な薬学的組成物は、本発明のミスマッチ修復ヌクレオチド配列の治療有効量であり、必要に応じて薬学的に受容可能かつ適合可能なキャリアに含まれる。用語「薬学的に受容可能かつ適合可能なキャリア」は、本明細書中で用いられるように、そしてさらに十分に以下に記載されるように、(i)ヒトまたは他の動物への投与に適した、1つ以上の適合可能な個体または液体の充填希釈剤またはカプセル化物質、および/または(ii)レトロウイルスベクターのようなミスマッチ修復ヌクレオチド配列を標的細胞に送達し得る、システムを意味する。本発明において用語「キャリア」は、従って、有機成分、または無機成分、天然成分または合成成分を示し、これを本発明のミスマッチ修復ヌクレオチド配列およびポリペプチドと組み合わせて適用を容易にする。用語「治療有効量」は、試験される特定の条件に対し、所望の結果を生成するか、または所望の影響を発揮する本発明の薬学的組成物の量である。様々な濃度が、同じ成分を取り込む組成物を調製するのに使用されて、処置される患者の年齢病状の重得度、処置期間、および投与様式における多様性を与え得る。
因子の同定
本発明のヌクレオチド配列およびポリペプチドは、相互作用因子の同定に使用され得、それらの内いくつかはそれ自体が本発明に包含される。すなわち、本発明の種々の真核生物ヌクレオチド配列のポリペプチド産物は、互いによく相互作用し得る。特に、配列番号3のポリペプチドと相互作用するこれらのタンパク質を同定することにより、ミスマッチ修復に作用する他のタンパク質がさらに同定される。酵母は、相互作用因子を遺伝的に同定するための特に強力な系を提供する。遺伝学的方法に加えて、共免疫沈降法およびタンパク質アフィニティークロマトグラフィーのようないくつかの生物化学的方法が、相互作用タンパク質の同定に使用され得る。
本発明の1つの実施態様において、共免疫沈降法は、配列番号3、配列番号4、または配列番号16のポリペプチドのような本発明の単離されたポリペプチドと相互作用するタンパク質を同定するために使用される。共免疫沈降法は、相互作用タンパク質を同定するために有用であると証明された(例えば、KolodziejおよびYoung,Methods Enzymol.194:508,1991,本明細書に参考として援用されている;Pallasら,J.Virol 62:3934,1988, 本明細書に参考として援用されている、を参照のこと)。
あるいは、またはさらに、遺伝学的方法を、本発明のポリペプチドと相互作用するタンパク質を同定するために使用し得る。同定されたタンパク質の少なくともいくつかは、ミスマッチ修復に関与する遺伝子にコードされ、細菌ミスマッチ修復遺伝子に相同であり、そして従ってそれ自身本発明の範囲内にあることが予想される。
本発明のヌクレオチド配列を含有する組換えベクターは、例えば、形質転換、トランスフェクション、感染、エレクトロポレーションなどによって宿主細胞に導入され得る。組換えベクターは、本発明の真核生物ヌクレオチド配列をベクター配列の調節因子(例えば、プロモーター配列、ポリアデニル化シグナルなど)の制御下に置くように設計され得る。このような調節因子は、宿主細胞において、ヌクレオチド転写物および/または本発明の真核生物ヌクレオチド配列にコードされるポリペプチド配列の発現、および/またはプロセシングを導くように機能し得る。
機能的同等物およびユニークフラグメント
本発明は、細菌のミスマッチ修復遺伝子に相同な単離された真核生物ヌクレオチド配列に関係し、従って、単離された真核生物ヌクレオチド配列、機能的同等物、またはこれらの配列のユニークフラグメントが本発明に従って使用され得る。ハイブリダイズ可能なヌクレオチド配列または「プローブ」もまた包含される。さらに、これらの配列にコードされる単離されたポリペプチドおよびこのポリペプチドのユニークフラグメントもまた、本発明に従って使用され得る。
本発明はまた、単離されたヌクレオチド「プローブ」を提供する。このプローブは、細菌のミスマッチ修復遺伝子に相同性のある真核生物標的配列にハイブリダイズし得る。
「抗体」という用語は、モノクローナル抗体、ポリクローナル抗体、および本発明の真核生物ヌクレオチド配列にコードされるポリペプチドに選択的に反応する、組換え核酸技術によって調製された抗体を包含することを意味する。「選択的に反応する」という用語は、細菌のミスマッチ修復遺伝子に相同である真核生物ヌクレオチド配列にコードされるポリペプチドの1以上の抗原決定基に反応するが、他のポリペプチドには反応しない抗体を意図する。抗原決定基は、通常アミノ酸または糖側鎖のような化学的に活性な表面の分子配置(grouping)からなり、特異的な3次元構造特性および特異的電荷特性を有する。抗体は、診断応用または研究目的に使用され得る。
遺伝子−−本明細書で使用されている「遺伝子」という用語は完全なコーディング配列を含有するヌクレオチド配列をいう。一般に、「遺伝子」はまた、コードされたポリペプチドの発現に影響を与える、上流(例えば、プロモーター配列、エンハンサーなど)または下流(転写終結シグナル、ポリアデニル化領域など)のコーディング配列にみられるヌクレオチド配列を含有する。
材料および方法
酵素および化学物質:制限酵素をNew England Biolabs(Beverly,Massachusetts)から得た。T4 DNAリガーゼをTaitら(1980)と類似の方法で調製した。DNAポリメラーゼIのクレノウフラグメントおよびランダムプライムDNA標識キットをBoehringer Mannheim(Indianapolis,Indiana)から得た。TaqDNAポリメラーゼをPerkin Elmer−Cetus(Norwalk,Connecticut)から購入した。シークエナーゼDNA配列決定キットをU.S.Biochemical Corp.(Cleveland,Ohio)から得た。ランダムプライム標識に使用する[a−32P]dATPおよびDNA配列決定に使用される[a−35S]dATPをAmersham(Arlington Heights,Illinois)から得た。
(実施例2:E.coli mutSミスマッチ修復遺伝子の酵母相同体の機能)
酵素および化学物質:化学物質、酵素およびオリゴヌクレオチドは実施例1に記載されている通りである。
MSH2/MSH2生存性実験の増殖プロトコル:最少栄養増殖制限
2つの野生型またはmsh2::TN10LUK半数体を接合させ、そしてシングルコロニー(≧3mm)を富栄養培地(YPD)上で単離した。これらの2倍体コロニーを前胞子形成培地(YPAc)5mlに低細胞密度で接種するために用い、そして増殖を飽和に達するまで続けた。次にその培養物を胞子形成培地で洗浄し、そして胞子形成培地中で24時間インキュベートした。

公知の細菌mutSおよびhexAおよびS.cerevisiae MSHタンパク質の高度に保存された領域をコードするDNAにハイブリダイズする縮重オリゴヌクレオチドを設計した。以下のアミノ酸領域を選択した:プライマー1a.)FATH(F/Y)(非コーディング鎖)5’−CGCGGATCC(G/A)(A/T)A(G/A)TG(G/A/T/C)GT(G/A/T/C)(GC(G/A)AA−3’(配列番号13);プライマー1b.)FTTH(F/Y)(非コーディング鎖)CGCCGATCC(G/A)(A/T)TG(G/A/T/C)GT(G/A/T/C)GT(G/A/T/C)GT(G/A)AA−3’(配列番号14);プライマー1c.)FVTH(FY)(非コーディング鎖)CGCGGATCC(G/A)(A/T)A(G/A)TG(G/A/T/C)GT(G/A/T/C)AC(A/G)AA−3’(配列番号28,およびプライマー2.)TPGNM(コーディング鎖)5’−CTGGATCC AC(G/A/T/C)GG(G/A/T/C)CC(G/A/T/C)AA(T/C)ATG−3’(配列番号12)。それぞれのヌクレオチドの5’端のCGCGATCC配列は、Bluescript SK+ベクターへの増幅産物のクローニングを容易にするため付加されたBamHI制限酵素切断部位である。酵母ゲノムDNAからの公知のミスマッチ修復配列のPCR増幅を、プライマー1a、1bまたは1cとペアにしたプライマー2を用いたPCRの条件を至適化するために使用した。PCRは、10mM Tris(pH 8.3)、50mM KCl,0.1%ゼラチン、各200μMのdGTP/dATP/dTTP/dCTP、1単位のTaqDNAポリメラーゼおよびそれぞれ25pmolの縮重プライマーを含有する50μl容量中で行った。複数の濃度のMgSO4(1mM,3mM,5mM,および10mM)および複数の濃度の酵母ゲノムDNAあるいはヒトcDNA(10ng,100ngおよび1μg)を、それぞれのプライマーペアに対して試験した。cDNAを、mRNA精製キット(Parmacia,SWEDEN)を用いてHPB−ALL細胞(MooreおよびFishel,J.Biol.Chem.265:11108−11117,1990)から調製した。cDNAに対するこれらのの縮重オリゴヌクレオチドを用いた増幅に至適な方法は、35サイクルのa)変性1分、94℃;b)アニーリング2分、45℃;c)ポリメラーゼ反応5分、72℃であることが見い出された。
PCR マッピング: PCRを使用して、プライマー#16388−5’GTTTTTCCTTTCATCCGTTG(配列番号21)および#16389−5’AAACTAGCCAGGTATGG(配列番号22)を用いてDNAのNIGMSマッピングパネル中のMSH2配列を検出した。これらのプライマーは、cDNA配列のヌクレオチド位置2020位に位置するイントロンに含まれるMSH2の推定158bpフラグメントを増幅する。25μlのPCR反応物は、10mM Tris緩衝液pH 8.5、50mM KCl、3mM MgCl2、0.01%ゼラチン、それぞれ50μMのdGTP/dATP/dTTP/dCTP、1.5ユニットのTaq DNAポリメラーゼ、それぞれ5pmoleのプライマーおよびそれぞれ0.5μgのDNAサンプルを含んでいた。PCRMを、a) 変性30秒、94℃;b) アニーリング30秒、55℃, c) 重合1分、72℃の30サイクルを実施し、そして3μlの各反応物を、TAE緩衝液中で1.4%アガロースゲルランを通じての電気泳動により分析した。
ミューテーターアッセイ:プレートアッセイを用いて、野生型E.coli AB1157(F, thr1, leu6, thi1, lacY1, galK4, aral14, xy15, mtl1, proA2, his4, argE3 str31, tsx33, supE44, λ−)におけるrifへの自然変異率を測定した。Bluescript(stratagene, La Jolla, CA)プラスミドの誘導体pDHA11を含有するMsh2huを、Fishelら(J. Mol. Biol. 188:147−157, 1986)に従い、AB1157に形質転換した。アンピシリン耐性形質転換体を選択し、100μg/mlアンピシリン(AMP)および0.5mM IPTGを含有するLB中で飽和するまで増殖させた。この培養物の希釈物を100μg/ml AMPを含むLBプレートにプレートしてpDHA11プラスミドを含有する生存可能細胞の総数を測定した。この希釈物を、100μg/ml AMPに加えて100μg/mlリファンピシン(Sigma, St. Louis, MO)を含むLBプレート上にプレートし、培養物中の自然rif変異体の総数を測定した。変異率を、LeaおよびCoulson(J. Genet. 49:264−285, (1949) J.Genet. 49:264−285)に従い、r0=M(1.24+ln M)を用いて計算した。ここでr0は、奇数個の独立した培養物(通常15)中のrif変異数の中央値であり、Mは、培養物あたりのrif変異の平均数である。Mを既知のr0値から補間法により導き、次いで変異率rを計算するために用いた。ここでr=M/N、Nは、生存細胞の最終平均数である。
いくつかの異なるプローブ(上記のPCR生成クローン22.1およびヒトcDNAクローンを含む)を用いて、L. Kunkeにより提供されたλgt11ヒトゲノムライブラリーをスクリーニングした。任意のヒトゲノムライブラリーがスクリーニングされた。
上記単離したヒトヌクレオチド配列をヒトゲノム中にマッピングした。
異なる細菌種由来のMutS相同体、例えば、S. pneumoniaeのhexAタンパク質のE.coliでの発現は、MutHLSミスマッチ修復経路を妨害し、優性ミスマッチ修復欠損表現型を生じた(Prudhommeら、J.Bacteriol. 173:7196−7203, 1991)。おそらく、S. pneumoniae MutS相同体は、E.coli中のミスマッチした塩基対に結合するが、E.coliミスマッチ修復機構の残りと相互作用し得ず、従って正常なミスマッチ修復を混乱させる。
A. 同定
上記の酵母およびヒト配列の単離により提供される情報は、任意の細菌性ミスマッチ修復遺伝子に相同な任意の他の真核生物のヌクレオチド配列を単離する一般的なプロトコルの開発を可能にする。特に、哺乳動物(例えば、マウス、雌ウシ、ブタ、およびサル)由来のE.coli mutS相同体は、容易に同定され得る。個々の場合において、DNAテンプレートとして、細菌性mutSおよびhexA遺伝子に相同な少なくとも1つの真核生物のヌクレオチド配列を含むことが知られるヌクレオチドライブラリーを用いる反応において、PCR反応条件を最適化することは大事なことであり得る。例えば、酵母ライブラリー(配列番号1または配列番号2を含む)が使用され得る。同様に、ヒト配列番号8または配列番号9を含むライブラリーが使用され得る。記載される手順はまた、細菌性ミスマッチ修復遺伝子ファミリー(例えば、mutH、mutL、hexB、およびmutU(uvrD))の他のメンバーに相同な真核生物ヌクレオチド配列の単離および同定を可能にするように改変され得る。
(手順)
ヒトMSH−2遺伝子(配列番号8に相当)のマップ位置を、マウス相同体(MSH−2mouse: 配列番号10に相当)の位置をマッピングすることによって、より詳細に決定した。これは、配列番号8に相当するヒトMSH−2の高度に保存された領域が、マウス相同体に100%のアミノ酸同一性を有する大きなストレッチを含むこと、およびこの領域のコーディングDNA配列が、ヒトDNA配列と92%同一(配列番号8と配列番号10との比較)であるほぼ100bpの長さのセグメントを含むことに起因し得る。ヒト保存領域に対するプローブ(配列番号15)、およびマウス保存領域に対するプローブ(配列番号10)が、種間マウス交雑の産物から得られたDNAの制限消化のサザンブロット中の単一部位(locus)にハイブリダイズすることを見出された。このことは、制限遺伝子座多型マーカーと比較してヒトMSH−2遺伝子をマッピングすることを可能にした。
種間戻し交雑マウスマッピング:種間戻し交雑子孫を、(CopelandおよびJenkins、前出 1991)に記載のように、(C57BL/6J×M.spretus)F1雌とC57BL/6J雄との交配により作成した。合計205匹のN2マウスを用いて、Hms2部位をマッピングした。DNA単離、制限酵素消化、アガロースゲル電気泳動、サザンブロットトランスファーおよびハイブリダイゼーションを、本質的に(Jenkinsら、J.Virol 43: 26−36, 1982)に記載のように実行した。全てのブロットをZetabindナイロンメンブレン(AMF−Cuno)を用いて調製した。プローブ(360bpヒトcDNAクローン)を、ランダムプライムラベリングキット(Stratagene)を用いて[α−32P]−dCTPで標識した;洗浄を、1.0×SSCP, 0.1% SDS, 65℃の最終ストリンジェンシーまで実施した。
マイクロインジェクションのためのDNAの精製方法は、当業者に周知である。例えば、Hoganら、Manipulating the Mouse Embryo、Cold Sping Harbor Laboratory, Cold Spring Harbor, NY (1986); およびPalmerら、Nature, 300: 611 (1982)を参照のこと。
マイクロインジェクション手順:齧歯類胚のマニピュレーションおよび接合体前核へのDNAのマイクロインジェクションの手順は、当業者に周知である(Hoganら、前出)。魚、両生類卵およびトリについてのマイクロインジェクション手順は、HoudebineおよびChourrout, Experientia, 47: 897−905 (1991)に詳細に記載される。動物組織へのDNA導入についての他の手順は、米国特許第4,945,050号(Sanfordら、1990年7月30日)に記載される。
トランスジェニックマウス:6週齢の雌マウスに、妊娠雌ウマ血清生殖線刺激ホルモン(PMSG; Sigma)の5IU注入(0.1cc, ip)、およびそれに続く48時間後のヒト胎盤性腺刺激ホルモン(hCG; Sigma)の5IU注入(0.1cc, ip)で排卵過度を誘導させる。雌を、hCG注入直後、雄とともに置く。hCG注入の21時間後、交配した雌をCO2窒息または頚部脱臼により屠殺し、そして胚を摘出した卵管から取り出し、0.5%ウシ血清アルブミン(BSA; Sigma)を含むDulbeccoリン酸緩衝生理食塩水(DPSS)中に置く。周囲の卵丘細胞を、ヒアルロニダーゼ(1mg/ml)を用いて除去する。次いで前核胚を洗浄し、インジェクションの時まで、5% CO2, 95%空気の加湿大気とともに37.5℃のインキュベーター中で0.5%BSA含有Earle平衡塩溶液(EBSS)に置いた。
トランスジェニックラット:トランスジェニックラットを作成するための手順は、マウスのトランスジェニック体作成の手順と同様である。Hammerら、Cell, 63:1099−1112 (1990)を参照のこと。30日齢の雌ラットに20IUのPMSG(0.1cc)の皮下注射を与え、48時間後、それぞれの雌を生殖能力を確認された(proven)雄とともに置く。同時に、40〜80日齢の雌を精管切除された雄とともにケージ中に置く。これらは、胚移植のための代理母を提供する。翌朝、雌を膣栓について検査する。精管切除された雄と交配した雌を移植の時までわきに置いておく。交配したドナー雌を屠殺し(CO2窒息)、そしてそれらの卵管を取り出し、0.5% BSAを含むDPSS中に置き、そして胚を回収する。胚の周囲の卵丘細胞を、ヒアルロニダーゼ(1mg/ml)を用いて除去する。次いで胚を洗浄し、マイクロインジェクションの時まで、37.5℃のインキュベーター中の0.5% BSAを含有するEBSS(Earle平衡塩溶液)中に置く。
(DNAのES細胞への導入)
ES細胞の培養方法、およびそれに続くエレクトロポレーション、リン酸カルシウム/DNA沈殿;および直接注入のような方法を用いるDNAのES細胞への導入による遺伝子導入動物の生産は、当業者に公知である。例えば、テトラカルシノーマおよび胚芽茎細胞、実用的アプローチ、E.J.Robertson編、IRLPress(1987)を参照。真核生物ミスマッチ修復遺伝子を含むES細胞の所望のクローンの選択は、いくつかの手段の一つにより行われる。胚芽茎細胞は現在マウスについてのみ手に入るが、本明細書に記載および引用されたような同様な方法および手順が、異なった種からの胚芽茎細胞について、それらが手に入るようになるとき、有効であり得ることが予期される。
胚回収およびES細胞注入
オスのマウスと交配させた、自然の周期のまたは過排卵させたメスのマウスを、ES細胞の移植のための胚を回収するために用いる。マウスを用いるとき、この目的のためにはC57BL165系統を用いることが望ましい。適切な年齢の胚は、うまく交配した後約3.5日で回収される。交配したメスを、CO2による窒息または頸部の脱臼により屠殺し、そして胚を切除した子宮角から流し出し、そしてES細胞を用いる注入のために、ダルベッコの改変必須培地プラス10%子ウシ血清中に置かれる。約10〜20のES細胞を、約20μmの内径を有するガラス製微小針を用いて胚盤胞中へ注入する。
胚の受容性メスへの移入
ランダム周期の成体メスのマウスは、精管切除されたオスと対にされる。Swiss Webster、ICRまたはその他のようなマウス系統をこの目的のために用い得る。受容体のメスは、ES細胞を含む胚盤胞を用いる注入のために必要とされるとき、交配後それらが2.5〜3.5日にあるように交配される。胎児移入のときに、受容体のメスを、体重グラムあたり2.5%アバーティンの0.015mlを腹腔内注射して麻痺する。卵巣を、卵管のすぐ上にある体壁において切開を行うことによって曝し、卵巣および子宮を外面に出す。子宮角内に25ゲージ針を用いて穴が作成され、そこを通って胚盤胞が移入される。移入後、卵巣および子宮を身体内に押し戻し、そして切開を二つの縫合によって閉じる。この手順は、別の移入がなされ得る場合、対向する側で繰り返される。
遺伝子導入マウスおよびラットの同定
尾部サンプル(1−2cm)が三週令の動物から取り出される。DNAを調製し、そしてサザンブロットまたはPCRによって分析し、遺伝子導入始原(F0)動物およびそれらの子孫(F1およびF2)を検出する。このようにして、バクテリアのミスマッチ修復遺伝子の相同体についてトランスジェニックとなった動物が同定される。すべての遺伝子導入動物がミスマッチ修復ポリペプチドを発現するわけではなく、そしてこれらのすべてが実験者によって予期される発現パターンを有するわけではないので、異なった組織におけるポリペプチドの発現に関して、遺伝子導入動物の各系統を特徴づけることが必要である。
げっ歯以外の遺伝子導入動物の生成:げっ歯類でない哺乳動物および他の動物の生成の手順は他によって論議されている。HoudebineおよびChourrout、前出;Purselら、Science 244:1281−1288(1989);およびSimmsら、Bio/Technology、6:179−183(1988)参照。
他の遺伝子導入生物の同定:生物は、DNA抽出のためにその生物のサンプルを採取し、そして目的の遺伝子に相補的なプローブを用いたハイブリダイゼイション分析により潜在的にトランスジェニックであると同定される。あるいは、生物から抽出されたDNAを、目的の遺伝子に相補的なPCRプライマーを用いるPCR分析に供し得る。
マウスのゲノムクローンを、ヒトのミスマッチ修復遺伝子を有するマウスのD3系統からゲノムライブラリーをスクリーニングすることによって単離する。重複リフトが、確立されたプロトコル(Sambrook,J.ら、The Cloning Manual、Cold Spring Harbor Press、N.Y.)によって放射標識されたプローブを用いてハイブリダイズされる。両リフト上の陽性信号に対応するプラークを、プラーク密度を減少させた連続的スクリーニングのラウンドによって単離および精製する。単離されたクローンの有効性は、ヌクレオチド配列決定によって確認される。バクテリアのミスマッチ修復遺伝子の真核生物相同体を不活性化するための多くの可能なプロトコルの一つが、以下に提示される。

ゲノムの配列決定
hMSH2のcDNA配列は、本明細書中では配列番号45で示され、そして受託番号U03911または受託番号U04045でGenBankにおいて見い出され得る。本発明者らは、hMSH2 cDNA配列のこれらの異なる目録において幾分変動があり、それはヒト集団内の多型現象;遺伝子コードの縮重;および/または配列決定結果の編集および解釈の間のわずかな編集誤りから生じ得ることを注記する。
ガン罹病性(susceptibility)を被験体に与える(すなわち、変異を有しない被験体がガンになる可能性よりも高い、ガンになる可能性を与える)変異は、その被験体の組織のいたるところに存在すること(すなわち、腫瘍組織に制限されない)および/または被験体の両親の少なくとも一方の生殖細胞株に存在することが予想される。腫瘍組織はまた、さらに別のミスマッチ修復遺伝子の変異をも含み得、この変異は、被験体の他の組織には存在せず、そして遺伝しなかったが、その腫瘍の発生には関与した(および/または必要であった)。(以下および、例えば、Parsonsら、Cell 75:1227−1236、1993を参照)。そのような腫瘍特異的変異の同定もまた価値があり、そして以下でさらに記載される。
(ii)コドン265位〜314位(エキソン5)をメッセンジャーRNA(mRNA)から除く、推定スプライシングの欠損;および
(iii)アルギニン残基に代えてのストップコドンの置換を生じる、コドン406位(ヌクレオチド1216位)でのCからTへのトランジション
本発明者らが提供した情報に基づき、当業者は、ガン罹病性と相関する別のhMSH2変異を容易に同定し得る。
(i)ヌクレオチド131位でのCからTへのトランジション;このタンパク質の高度に保存された領域であるエキソン2中(図4を参照)。
(ii)エキソン19における2179位〜2182位の4個のヌクレオチドの欠失、これは、フレームシフトを生じ続いて新たなストップコドンを生じる;
(iii)エキソン19における2266位の後(コドン755位と756位との間)に4個のヌクレオチドの挿入、これは、オープンリーディングフレームのフレームシフトおよび伸張を生じる;および
(iv)1038位の後から始まる371個のヌクレオチドの欠失、これは、フレームシフトを生じ続いて新たなストップコドンを生じると報告されている。この変異は、hMLH1エキソン12の欠失を反映するらしく、そしてエキソンスキッピングを生じるスプライシングの欠損を示し得る。
本明細書中に記載されたように、被験体においてガン罹病性を診断する際に有用であることに加えて、細菌ミスマッチ修復遺伝子に相同なヌクレオチド配列は、特に、ミスマッチ修復欠損腫瘍の同定および特徴付けにおける使用について価値があり得る。このような同定および特徴付けは、価値あるものである。なぜなら、ミスマッチ修復欠損腫瘍が、特定の治療養生法により良く応答しているからである。例えば、ミスマッチ修復欠損腫瘍は、特に、他の治療薬剤と併用して投与される場合、DNA損傷薬剤に対して敏感であり得る。
(i)コドン639位(ヌクレオチド1915位)におけるCからTへのトランジション、これはヒスチジンに代えてのチロシンの置換を生じる;および
(ii)コドン663位(ヌクレオチド位置1987位)におけるA残基に代えてTGジヌクレオチドの置換、これは、フレームシフトを生じ、36ヌクレオチド下流で終結コドンを生じる。
(i)コドン252位(ヌクレオチド位置755位)でのCからAへのトランスバージョン、これはSer残基をストップコドンで置換する。この研究において、この腫瘍組織は野生型hMLH1対立遺伝子を含まなかった。
ガン罹病性の研究のためならびに腫瘍の同定および特徴付けのためには、「変異」を「多形性」と区別することが重要である。「変異」は、遺伝子の「非野生型対立遺伝子」を生じる。遺伝子の非野生型対立遺伝子は、通常細胞内で機能しない転写物および/またはタンパク質産物を生じる(上記の定義を参照)。「変異」は、挿入、欠失、置換、および再配列を含む、ヌクレオチド配列におけるいかなる変化でもあり得る。
(i)配列番号45に示すhMSH2 cDNA配列の位置399でのCまたはT。
Leachら、上述、もまた以下のhMSH2多型性を同定している:
(i)エキソン13の6bp上流のポリピリミジン領域におけるCからTへの転位(このエキソンは配列番号45に示すhMSH2 cDNA配列のヌクレオチド位置2006で始まる)。本発明者らは、同一のCからTへの転位がFishelら、上述、によって同定され、そしてこの変化が真に発現しない(silent)多型性でない可能性があることに注目する。すなわち、この変化は、実際、弱い変異であり得、例えば、細胞がさらなるミスマッチ修復欠陥を含まなければ、この影響は明らかではない(または有意ではない)。例えば、他のミスマッチ修復遺伝子変異(特にhMSH2遺伝子変異)は、細胞においてより劇的な表現型を有し得、またこのCからTへの遷移(例えば、この遷移は他の変異の浸透度を増加させ得る)を有する。
hMSH2遺伝子をバクテリア細胞(特に、E.coli細胞)に導入することは、優性変異誘発表現型(Fishelら、1993、上述)を生じる。類似の優性変異誘発表現型が、S.pneumonae MutS相同体であるHexAが、E.coli中で発現されるときに観察されている(Prudhommeら、J.Bacteriol.173:7196−203,1991を参照のこと)。この効果の適当な説明は、異種MutS相同体(例えば、HexAまたはhMsh2)がE.coli細胞中のミスマッチ塩基対に結合し得るが、E.coliミスマッチ修復システムの他の成分と(すなわち、MutL、MutH、などと)生産的に相互作用せず、それ故それらが結合するミスマッチ塩基対の修復を妨げることである。
特定のhMSH2配列変化がhMSH2遺伝子または遺伝子産物の機能に対して有する影響をアッセイし、そしてそれによりこれらの配列変化を「変異」あるいは「多型性」として分類する他の方法は、コードされたhMSH2タンパク質がミスマッチ塩基対に結合する能力をアッセイすることである。
タンパク質−タンパク質相互作用アッセイがまた、ミスマッチ修復遺伝子における配列改変を分析するため、およびそれらを変異または多型性として分類するために使用され得る。E.coliにおいて、MutLタンパク質は、MutSがミスマッチを含むDNAに結合しているときに観察されるフットプリントのサイズを増加する。MutLは、ミスマッチに結合したMutSタンパク質と、近くのDam部位に結合したMutHタンパク質との間の架橋として作用するようである。
多型性を変異と区別する他の可能な方法は、検出可能な表現型がミスマッチ修復遺伝子の制御下にあるアッセイシステムを利用することである。すなわち、特定の挙動が機能的ミスマッチ修復遺伝子を必要とし、そしてその挙動における変化が検出可能である任意のシステムが、異なるミスマッチ修復遺伝子対立遺伝子を「変異体」あるいは「多型性」として分類するために使用され得る。
1)カナバニン耐性に対する前進変異(R.A.G.Reenanら、Genetics 132,963−973,1992;R.A.G.Reenanら、Genetics 132,975−985,1992を参照のこと);
2)LYS2におけるフレームシフト変異の復帰(L.Newら、Mol.Gen.Genet.239,97−108,1993を参照のこと);および
