Abstract
Description

(発明の分野)
本発明は、一般的に癌の検出および治療に関する。本発明は、スフィンゴ脂質の代謝に関するポリペプチドをコードするポリヌクレオチド、ポリペプチド、ならびにこのようなポリペプチドの発現および/または活性を調節する薬剤により特に関する。このような薬剤は、例えば、癌(例えば、乳癌、結腸癌、子宮癌、胃癌、卵巣癌、肺癌、腎臓癌および直腸癌)の診断および/または処置、筋肉発達欠陥および心筋症の診断および処置、ならびに遺伝知覚神経障害1型およびスフィンゴリピドーシスの診断および処置に使用され得る。本発明は、スフィンゴ脂質代謝に関するポリヌクレオチドおよび/またはポリペプチドの発現および/または活性を調節する薬剤をスクリーニングする方法にさらに関する。
乳癌は、米国および世界中で女性にとって重要な健康の問題である。進歩は、この疾患の検出および処置についてなされたが、乳癌は現在も、癌の最も共通の形態であり、そして米国の女性にとって、癌の死亡を導く第二の原因である。米国黒人女性と15〜54歳の女性との間で、乳癌は、癌死亡の主要な原因である。米国で8人に1人の女性は、乳癌を発生し、その危険性は、1950〜1990年の間に52%増加した。1994年には、182,000人の新しいケースの女性乳癌が、診断され、そして46,000人の女性が、この疾患により死亡することが推定される。
上に記したように、本発明は、一般的に癌の検出および治療に関連する。本発明は、より具体的には、スフィンゴ脂質、ポリペプチドの代謝および/または信号伝達に関するポリペプチドをコードするポリヌクレオチド、ならびにこのようなポリペプチドの発現および/または活性を調節する薬剤、および/またはスフィンゴ脂質中間体のレベルを変化させる薬剤に関連する。このような薬剤は、例えば、乳房、結腸、子宮、胃、卵巣、肺、腎臓および直腸の癌のような癌の診断および/または処置、筋肉発達障害および心筋症の診断および処置、ならびに1型遺伝性感覚ニューロパシーおよびスフィンゴリピドーシスの診断および処置に使用され得る。本発明は、スフィンゴ脂質の代謝および/または信号伝達に関する構成要素および中間体を調節する薬剤のスクリーニングの方法に、さらに関連する。
本発明は、スフィンゴ脂質の代謝および/またはシグナル伝達を調節する薬剤のための組成物および方法(スクリーニングアッセイを含む)を提供し(その薬剤は、スフィンゴ脂質中間体のレベル、および/またはスフィンゴ脂質の代謝経路および/またはシグナル伝達経路の一つ以上の構成要素の活性に対して影響を有する。)、かつさらに、上記薬剤のスクリーニングのための方法を提供する。本発明の薬剤は、癌および変化したスフィンゴ脂質の代謝および/またはシグナル伝達に関連した他の疾患の検出、診断、および治療における有用性を持つ。
スフィンゴ脂質の代謝経路および/またはシグナル伝達経路のどの要素も、本発明の状況に含まれる。そういうものとして、スフィンゴ脂質の代謝経路および/またはシグナル伝達経路の構成要素としては、SPL、SK、セラミダーゼ、S−1−PP、セリンパルミトイルトランスフェラーゼ(SPT)、3−ケトジヒドロスフィンゴシンレダクターゼ、セラミドシンターゼ、スフィンゴシンデサチュラーゼ、セラミドキナーゼ、ホスホエタノールアミンシチジリルトランスフェラーゼ、CDP−エタノールアミンホスホトランスフェラーゼ、酸性スフィンゴミエリナーゼ、スフィンゴミエリンシンターゼ、中性スフィンゴミエリナーゼ、オキソスフィナニンレダクターゼ、およびグルコシルセラミドシンターゼといった、これらの経路に関与する酵素(及び上記酵素をコードするポリヌクレオチド)が挙げられるが、それらに限定されない。スフィンゴ脂質の代謝経路および/またはシグナル伝達経路の構成要素は、細胞内レセプターまたは細胞表面のレセプター、および、EDGレセプター(例えば、EDG1、EDG3、EDG5、EDG6、EDG8)およびCFTRのような、上記レセプターをコードするポリヌクレオチドを、さらに含む。

本発明に従って使用する薬剤は、人工的または天然由来のどちらでも、本明細書にさらに詳細に記載される、スフィンゴ脂質の代謝および/またはシグナル伝達を調節する、任意の組成物、化合物、基質、分子、物質、生成物などとして定義される。スフィンゴ脂質の代謝および/またはシグナル伝達を調節する薬剤は、少なくとも一つのスフィンゴ脂質中間体のレベル、または、スフィンゴ脂質の代謝および/またはシグナル伝達の少なくとも一つの要素の活性を変化させる(例えば、統計学的に有意な様式で増加または減少する)薬剤である。レベルまたは活性の変化は、単離された要素、または、中間体または要素を含む宿主細胞または動物が、その薬剤と接触しない単離された要素、中間体または要素を含む宿主細胞または動物と比較して、その薬剤と接触する場合に、任意の統計学的に顕著な変化(例えば、一つ以上のスフィンゴ脂質中間体のレベル、または、本明細書に記載のスフィンゴ脂質の代謝および/またはシグナル伝達の一つ以上の要素の活性における、増加または減少)を含む。このように、一つの実施形態において、調節は、本明細書に記載のスフィンゴ脂質の代謝および/またはシグナル伝達に関与するタンパク質をコードするポリヌクレオチドにおける、変化されるレベル(例えば、減少または増加)を含む。ポリヌクレオチドのレベル(すなわち、遺伝子発現)を検出する多数の方法が、当該分野において公知であり、本発明の文脈において有用である。図示される方法は、Ausubelら(1993 Current Protocols in Molecular Biologly,Greene Publ.Assoc.Inc.&John Wiley&Sons,Inc.,Boston,MA);Sambrookら(1989 Molecular Cloning, 第2版,Cold Spring Harbor Laboratory,Plainview,NY);Maniatisら(1982 Molecular Cloning,Cold Spring Harbor Laboratory,Plainview,NY)および他の場所に記載される。
スフィンゴ脂質の代謝および/またはシグナル伝達の成分の調節を検出するための多数のアッセイが、当該分野で利用可能である。例示的なアッセイは、例えば、実施例の節で記載されるように、本明細書中でさらに記載される。
他の局面において、本発明は、癌を診断するため、および/あるいは癌を発症する危険性があるか、または平均よりも高いかもしくは低い転移の危険性がある個体を同定するための、方法およびキットを提供する。本発明の文脈において、特定のヒト腫瘍細胞は、変更されたSK発現およびSPL発現を含むことが見出されている。特に、SPL発現の減少は、実施例においてさらに記載されるように、同じ個体由来の対応する正常組織と比較して、特定の腫瘍組織において観察された。さらに、SK発現の増大は、同じ個体由来の対応する正常組織と比較して、多数の腫瘍組織において観察された。換言すると、このようなポリヌクレオチドまたはこれらのポリヌクレオチドによってコードされるタンパク質は、患者における癌の存在または非存在を示すためのマーカーとして使用され得る。

SPL、SPTおよび他の変異体表現型の病因を同定するための種々の方法は、当業者に公知であり、そしてまた、本明細書中に提供される。本発明は、sPL遺伝子、SK遺伝子、SPT遺伝子およびS−1−Pホスファターゼ遺伝子を欠損するか、または過剰発現する変異体Drosophila melanogasterを提供する。特定の実施形態において、異常なスフィンゴ脂質代謝は、本発明の変異体Drosophila melanogasterの表現型である。さらなる実施形態において、異常なスフィンゴ脂質代謝は、変異体ハエにおいて発生プログラムに影響する。特定の実施形態において、本発明の変異体Drosophila melanogasterおよび/またはトランスジェニックDrosophila melanogasterは、1つ以上の腫瘍を発達させる。腫瘍は、当業者に公知の種々の技術を使用して同定され得、そして測定され得る。特に例示的な技術は、例えば、De Lorenzo,C.,B.M.MechlerおよびP.J.Bryant.1999.Cancer Metastasis Rev.18:295−311.;Watson,K.,R.JusticeおよびP.Bryant.1994.J Cell Sci 補遺18:19−33.;Gateff,E.およびH.Schneiderman.1967..Amer Zool.7:760.;Gateff,E.1994.Int J Dev Biol.4:565−590または本明細書中に引用されるいずれかの参考文献に記載される。
本明細書中に記載されるような、スフィンゴ脂質の代謝および/またはシグナル伝達の成分および/またはスフィンゴ脂質中間体を調節する薬剤は、スフィンゴ脂質の代謝および/またはシグナル伝達の変更に関連した任意の疾患の検出、診断および処置に有用である。例示的な疾患としては、種々の癌(例えば、乳癌、結腸癌、子宮癌、胃癌、卵巣癌、肺癌、腎臓癌および直腸癌)、筋肉の発達欠陥から生じる疾患、心筋症、および遺伝性感覚ニューロパシー1型およびスフィンゴリピドーシスが挙げられるがこれらに限定されない。従って、本発明の組成物は、癌に罹患した個体における癌の発生、転移、または癌の発生と転移との両方を阻害するために用いられ得る。
0〜24時間胚を、調製し、標準的な技術(Rubin Manual)で固定し、そして指定される一次抗体により染色するか、またはTUNELベースの染色法(In situ cell death detection kit,Roche 1 684 795)を使用してアポトーシスについてアッセイした。フルオレセインの取り込みを、Leica DMIRBE落射蛍光顕微鏡および倒立型Leica TCS−NT共焦点レーザー走査型顕微鏡にて評価した。
ry+PZ P因子の正確な切除は、Δ2−3トランスポザーゼ対立遺伝子を、挿入株BL−11393に導入する操作により行った。その後の世代では、トランスポザーゼを取り除き、そして二番染色体を、CyOでバランスした。P因子を欠いたこれらのハエの子孫は、ry+の消失の評価により選択した。ホモ接合型株を作製し、飛行能力の回復についてアッセイし、そして回復が見られた場合に、PCRにより正確な切除を評価した。Sply05091対立遺伝子およびlacek05305対立遺伝子に対するホモ接合株を、減数分裂組み換えにより作製した。Sply05091およびlacek05305の変異を、トランスに導入し、次の世代でバランスした。lacek05305対立遺伝子を保有するハエを、w+の存在により選別した。Sply05091の存在を、PCRにより検証した。

(Drosophila melanogaster(Sply)からのSPL cDNAの単離および特徴付け)
Drosophila melanogaster SPL cDNAおよびゲノムの配列を見出すために、D.melanogasterゲノムデータベースを、ヒトSPL cDNAと顕著な相同性を呈示する配列について検索した。Drosophila melanogasterゲノムデータベース(http://flybase.bio.indiana.edu)を、マウス(登録番号AAH26135;配列番号:6に示されるアミノ酸配列)およびヒト(登録番号XP_166113;配列番号:18に示されるアミノ酸配列)のSPL配列を使用して、予想されるタンパク質について検索した。DNA相同性検索は、Berkeley Drosophila Genome Projectウェブサイトを通じて、BLAST検索プログラム(http://www.ncbi.nlm.nih.gov)を使用して行った。予想されるSPL遺伝子に相当する、1つの算出された遺伝子(CG8946)を、同定した。続いて、2つの予想されるスプライシング改変体に相当するオープンリーディングフレームを含む、2つのEST(LP04413,配列番号:27に示されるcDNAおよびGH13783,配列番号:26に開示されるcDNA)を、同定した。その2つのクローンは、選択的な5’エクソン使用に基づいて予想され、異なる細胞下での位置で発現され得る。予想されるDrosophila melanogasterSPLは、2番染色体の右腕上、53F8−12の位置に位置する。Drosophila melanogasterSPLのコード領域に対するcDNA配列は、配列番号:15に示され、配列番号:16に示されるSPLタンパク質をコードする。配列番号:16に示されるDrosophilaSPL予想タンパク質の配列は、ヒト、マウスおよび酵母のSPLタンパク質配列に対し、それぞれ49%、49%および43%の同一性を持つ。これらのクローンが、機能的なSPL遺伝子を含むかどうかを評価するために、これらを、酵母発現ベクターである、pYES2(その中では、遺伝子発現が、ガラクトース誘導性プロモーターにより駆動される)の中に再びクローニングした。LP04413(配列番号:に示されるポリヌクレオチド配列)に含まれるオープンリーディングフレームを、プライマー、LPEcoRI5=5’−TGGAATTCGATGCGTCCGTTCTCCGGCAGC−3’およびLPXhoI3’=5’−CTCCTCGAGTCTATTTCTGGCTGGGAGT−3’、を使用して増幅し、酵母発現ベクターである、pYES2の中に、EcoRIおよびXhoI制限酵素部位にて、クローニングした。この構築物を、酢酸リチウム法(Ito,H.,Fukuda,Y.,Murata,K.およびKimura,A.(1983).J Bacteriol 153,163−8.)を使用してdpl1Δ株に形質転換した。これらの構築物を、単一の内因性Saccharomyces cerevisiae SPL遺伝子が欠損しているdpl1Δ株(Saba,J.D.,Nara,F.,Bielawska,A.,Garrett,S.およびHannun,Y.A.(1997).J Biol Chem 272,26087−26090.)に形質転換した。dpl1Δ株は、LCBPを異化する能力がなく、そして、低濃度のD−エリトロ−スフィンゴシンを含む倍地上で増殖し得ない。
(SPLトランスポゾン変異体D.melanogasterの作製および特徴付け)
本明細書に記載の実験に使用されるDrosophila melanogasterのストックは、以下の通りである:野生型Canton−S(BL−1),Sply05091(BL−11393),lace2(BL−3156)およびlacek05305(BL−12176)株、Bloomington Drosophila Stock Center(Indiana University,Bloomington,IN)より入手。一般的なハエの管理を、記載されるように行った(Sullivan,W.,Ashburner,M.およびHawley,R.S.(2000).Drosophila protocols.Cold Spring Harbor,N.Y.:Cold Spring Harbor Laboratory Press)。Berkeley Drosophila Genome Projectの遺伝子破壊計画(Spradling,A.C.,Stern,D.M.,Kiss,I.,Roote,J.,Laverty,T.およびRubin,G.M.(1995).P転移性因子を使用しての遺伝子破壊:ショウジョウバエゲノムプロジェクトの不可欠な一要素。Proc Natl Acad Sci USA 92,10824−30.)からの、Splyオープンリーディングフレームの中にトランスポゾンを持つハエ(Sply05091と命名)を、同定した。そのトランスポゾンは、LP04413の長い方の転写産物(配列番号:27に示されるcDNA)の開始部位に比べて+269ヌクレオチドに位置する。
(P因子挿入Sply変異体Drosophila melanogasterのさらなる特徴付け)
Drosophila melanogasterのスフィンゴ脂質は、C14およびC16スフィンゴシンおよびジヒドロスフィンゴシン LCBを含む(実施例4を参照のこと)。野生型および変異体のハエの抽出物を、内因性のDrosophila melanogasterLCBPをインビトロで分解する能力について比較した。Sply05091変異体の抽出物は、内因性LCBPを分解しなかったが、一方、野生型ハエの抽出物は、内因性Drosophila melanogasterLCBPを分解し、Sply遺伝子産物は、この生物体におけるLCBP分解のために重要であることを呈示した。
Sply05091の飛行障害の原因をさらに追求するため、成虫変異体を、胸部領域を通じて切片化し、筋肉を、光学顕微鏡で検査した。この研究は、飛行に必要なDLMを含む筋肉繊維の数の減少を呈示した。野生型ハエの胸部には、6対の対称的な線維を不変的に含む一方で、Sply05091ホモ接合体は、線維の消失、非対称性、および残りの繊維の肥大の一般的なパターンを呈示した。DLM線維の定量的分析により、野生型でのヘミソラックスあたり6本から、変異体でのヘミソラックスあたり平均4.15本への減少が、呈示された(表1)。偏光照射によるヘミソラックスの顕微鏡分析により、異常な筋肉の立体配置が確認された一方、筋肉挿入は、影響を受けないことが、呈示された。
そのDLM欠損の起源を決定するため、成虫筋肉細胞の超微細構造ならびに幼虫および胚の筋肉発生を調べた。野生型および変異体の初期胚の初期筋肉繊維の筋芽細胞核における、Dmef2の発現の調査は、筋肉の組織化において明確な変化を呈示しなかった。従って、筋芽細胞は、変異体の胚期体節において、原体節から正確な部位へと首尾良く移動するようである。同様に、野生型および変異体の0〜24時間胚におけるミオシン重鎖の発現の分析は、野生型に比べて発生中の変異体の筋肉の、全体としての変化は呈示せず、筋肉細胞融合が、損なわれないことを呈示した。
ホモ接合型Sply05091の交配の結果から得られる子の数は、野生型の交配で観測される数のおよそ10%であった。この子孫の減少は、減少した産卵ならびに/または胚および幼虫の減少した生存の結果であり得た。産卵の分析により、その変異体の生殖能力が、コントロールハエのおよそ1/3であることが、呈示された(表1)。この結果は、雄および/または雌の減少した稔性の結果であり得た。これらの可能性を区別するために、雄および雌の両方のSply05091ホモ接合体を、野生型ハエと交配し、そして産卵を、野生型対およびホモ接合型変異体対に比較して測定した。産んだ卵の数は、野生型の相手との雄および雌の両方の変異体ハエの交配において、顕著に減少し(データは示さず)、生殖能力へのその効果が、性別特異的でないことが、呈示された。さらに、Sply05091ホモ接合型の雄および雌の間での交配は、野生型ハエにおける80%の生存率に比べて、33.5%の総体的な生存性(卵から成虫期まで)を持つ子孫の結果となった。Sply05091変異体の致死性は、幼虫期の間で高く(46%、野生型における3%に対して比較)、大部分の幼虫の死は、一齢幼虫期(first larval instar)の間に起こった。蛹化の間では、より穏やかな効果が、観測され(致死率22%、野生型における1%に対して比較)、胚発生の間に、生存における明確な違いは、何もなかった。Sply05091変異体胚を、インサイチュTUNELアッセイにより試験し、アポトーシスのパターンを、野生型のパターンに対して比較した。Sply05091変異体胚は、野生型コントロールに比べて、特に発生中の生殖器成虫原基に近い後部極の特定の領域で、アポトーシスのはっきりとした増大を呈示した。
(Drosophila melanogasterにおけるスフィンゴ脂質類の特徴付け)
理論に束縛されずに、SPL変異体Drosophila melanogasterの表現形は、発生中におけるS−1−Pの異常なレベルにより引き起こされると、仮定される。さらに、理論に束縛されずに、リン酸化スフィンゴイドベース類は、細胞増殖、移動および、このモデル生物体における腫瘍形成および通常の発生プロセスの両方のために必要な他の事象を調節するために重要である、というのが、本発明者らの仮定である。それゆえ、Drosophila melanogasterにおけるスフィンゴ脂質類の特徴付けを、決定した。
野生型(Canton S)全ハエ抽出物を、機械的破砕により調製した。脂質を、二相式抽出により単離し、本質的にCaliganらに記載されるように、蛍光性分子o−フタルアルデヒド(pthalaldehyde)で誘導体化した(Caligan,T.B.,K.Peters,J.Ou,E.Wang,J.SabaおよびA.H.Merrill,Jr.2000.A high−performance liquid chromatographic method to measure sphingosine 1−phosphate and related compounds from sphingosine kinase assays and other biological samples.Analytical Biochemistry.281:36−44)(その全体が本明細書中に参考として援用される))。誘導体化された脂質抽出物を、C18ODSカラム(LUNA4.6×250mm)および移動相MeOH/H2O/1M TBAP 82:17:0.9,pH4.8を使用してHPLCにより分離した。標準物質は、市販のC10、C12、C14、C16、C18およびC20スフィンゴシン、ならびにこれらの標準物質のリン酸化体を含み、これらを、スフィンゴシン標準物質の、主要酵母スフィンゴシンキナーゼ、LCB4を過剰発現する酵母株からの抽出物とインキュベーションすることにより調製した。
Drosophila melanogaster抽出物は、定めた条件下でC14スフィンゴシンおよびC14スフィンゴシン−1−リン酸(S−1−P)標準物質と共に共移動するスフィンゴ脂質類のみを含んだ。C14スフィンゴシンおよびC14S−1−P標準物質と共に共移動するハエ抽出物におけるピークの同一性を検証するため、抽出物および標準物質を、4つの異なる移動相緩衝液にて比較した。C14スフィンゴシン標準物質と共に共移動するピークを、4つ全ての条件下でC14スフィンゴシンであると同定した(表2)。しかしながら、C14S−1−Pであると同定されたピークは、電荷における違いを利用した条件下で(MeOH/10mM KP/1M TBAP,pH7.2 81:18:1)、C14S−1−P標準物質からのわずかな違いを呈示した。
(Sply05091変異の遺伝学的復帰は、正常な筋肉立体配置を回復させる)
Splyがその変異株の半致死性、産卵機能欠損および飛行筋肉の表現型を媒介することにおける重要性を確認するために、Sply05091遺伝子座でのトランスポゾンを、転移させた。Sply05091変異の遺伝学的復帰は、活性化トランスポザーゼの導入後のSply05091ホモ接合体で正常な筋肉形状を回復させる。トランスポゾンの正確な切除を、引き続いて、PCRおよびDNA配列分析により確定した。ホモ接合型復帰変異株(Sply14a)を、材料および方法に記載の通りに産生し、SplyのmRNAが野生型と等しいレベルで発現されることを見出した。Sply14aは、その筋肉線維形態欠損の回復を示し、飛行機能が、大部分回復した(表1)。加えて、その復帰変異体胚のアポトーシスは、Sply変異体に比べて減少した。TUNEL陽性細胞のその特異的クラスターの出現は、12〜15ステージ胚において、Canton−S、Sply14aおよびSply05091でそれぞれ、<1%(n=197)、48%(n=160)、72%(n=324)であった。表現型の復帰は、復帰変異体の抽出物におけるLCBおよびLCBPのレベルの正常化と相関した(表1)。
(Sply05091の筋肉欠損は、スフィンゴ脂質中間体の減少により抑制される)
そのSply05091の筋肉の表現型が、LCBPの蓄積によって引き起こされる可能性を調査するため、スフィンゴシンキナーゼのひとつの阻害剤である、D,L−スレオ−DHSを、変異体および野生型ハエの増殖培地に導入した。ハエを、その補充培地上で増殖させ、F2子孫を、調査した。野生型ハエを、10μM D,L−スレオ−DHSを補充した培地上で増殖させた場合、有害な効果は、何も観察されなかった。この培地上で増殖させたSply05091変異体は、飛行機能におけるわずかだが有意な改善を示した。その飛行改善が、LCBPレベルの回復と一致するかどうかを決定するため、LCB/LCBPレベルを、D,L−スレオ−DHS上で増殖させた変異体およびコントロールで分析した。スフィンゴシンキナーゼ阻害剤の存在下で増殖させたSply05091ホモ接合体におけるLCBPレベルは、およそ20%減少した(表1)。同様に、野生型ハエにおけるLCBPレベルは、正常レベルの20%に減少した。
(Sply発現の損失は、laceヌル表現型を抑制する)
2つのlacek05305ハイポモルフ対立遺伝子の遺伝形質は、ほとんど完全に致死であり、その一方で、ヘテロ接合型な対立遺伝子の組み合わせ(lacek05305/lace2)は、高頻度で生存するが、眼、剛毛および翅の異常へと導く重篤な発生上の表現型を示すハエを、産生することを報告した(Adachi−Yamada,T.,Gotoh,T.,Sugimura,I.,Tateno,M.,Nishida,Y.,Onuki,T.およびDate,H.(1999).Mol Cell Biol 19,7276−7286.)。本発明者らは、lace変異体の表現型が、スフィンゴ脂質中間体の減少したレベルに起因する、と予測した。さらに、本発明者らは、lace変異体におけるスフィンゴ脂質異化の阻害により、餌を通じて得られる、少量のスフィンゴ脂質の十分な蓄積が、重要なスフィンゴ脂質中間体の欠如により誘導される発生上の欠損を改善させ得ると推論した。この可能性を調査するため、Sply05091およびlacek05305の対立遺伝子両方についてホモ接合型のDrosophila melanogaster株を、産生した。明らかに、Sply05091対立遺伝子の存在により、laceホモ接合体の回復が、独立した取り合わせ(assortment)により、予期されていたものの9%から39%に増加した。さらに、Sply05091の導入により、結果として産生されるハエにおいて、眼、剛毛および翅その表現型は、完全に抑制された。従って、スフィンゴ脂質中間体は、この株において、lace2/lacek05305ヘテロ接合体に比べて引き続き増加し、lace2/lacek05305ヘテロ接合体は、比較のために十分な生存性を伴う、使用可能な唯一のlaceの変異体である(表1)。
(癌におけるヒトSPLおよびSKの発現パターン)
SKおよび/またはSPLの発現が、ヒト腫瘍において変化するかを決定するため、本発明者らは、240より多数のcDNA対を含む癌プロファイル化アレイを利用した。これらのcDNA対は、同一患者からの腫瘍組織および相当する正常組織を示す。一人の患者に由来する組織対の利用により、ヒトからヒトへの多様性が原因となり得る、腫瘍組織における遺伝子発現と正常組織における遺伝子発現との間の違いにより、結果の解釈は混同されないはずである。加えて、各ブロットを、ローディングのために、4つの別々のハウスキーピング遺伝子を使用して正規化した。
(スフィンゴ脂質代謝に関与するDrosophila melanogasterの遺伝子の単離)
スフィンゴ脂質代謝に関与するDrosophila melanogasterの遺伝子を、同定した。配列番号21に示されるヒトSKタンパク質配列を、プローブとして使用して、本発明者らは、ハエSKタンパク質を潜在的にコードし得る2つの相同的なDrosophila melanogasterの配列を同定した。これらの2つのDrosophila melanogasterSKタンパク質を、配列番号19および20に示し、図1に示す。CG1747遺伝子に関する注釈FBan0001747は、FlyBaseアクセス番号FBgn0030300を有し、X染色体腕上に位置し、2020ヌクレオチドの長さの転写単位を有する。この遺伝子は、転写物CT5088を有する。この遺伝子の機能は、保存された脂質キナーゼドメインに基づき、酵素/ジアシルグリセロールキナーゼとして分類されている。CG2159遺伝子に関する注釈FBan0002159は、FlyBaseアクセス番号FBgn0035391を有し、3L染色体腕上に位置し、4431ヌクレオチドの長さの転写単位を有する。この遺伝子は、転写物CT2650を有する。これらの2つの配列は、いまだクローニングされておらず、機能的データおよびESTの両方とも、入手不能である(DSK1747アミノ酸配列は、配列番号19に示される;DSK2159アミノ酸配列は、配列番号20に示される)。
(スフィンゴ脂質代謝に関与する2つのDrosophila melanogasterSK遺伝子の特徴付け)
Drosophila melanogasterにおける潜在的なSK遺伝子を同定するため、Drosophila melanogasterゲノムデータベースにて、tBLASTn調査を使用して、ヒトSK cDNAに対して顕著な相同性を示す配列を検索した。このことによって、上記のように、2つの候補SKが同定された。CG1747遺伝子は、X染色体上に位置し、保存された脂質キナーゼドメインに基づき、酵素/ジアシルグリセロールキナーゼとして分類されている。CG2159遺伝子は、3L染色体腕上に位置し、4431ヌクレオチドの長さの転写単位を有する。これらの2つの遺伝子座に相当するESTを入手し、それらが無傷であることを、配列決定および制限酵素分析によって確認した(SK1およびSK2の全長アミノ酸の配列を、配列番号28および29に示す;SK1およびSK2についての全長cDNAを、配列番号24および25に示す)。それらのCG1747およびCG2159 cDNAクローンを、それから、ガラクトース誘導型プロモーターの調節下の、酵母発現ベクターpYES2の中に再クローニングした(Rubin, G,Hong L,Brokstein P,Evans−Holm M,Frise E,Stapleton MおよびHarvey D,2000.A Drosophila complementary DNA resource,Science 287:2222−2224.)。これらの構築物を、JSK392酵母株(Kim,S,Fyrst HおよびSaba J,2000.Genetics 156:1519−1529.)の中に形質転換した。JSK392において、内因性のSPL、SKおよびS−1−PP遺伝子(それぞれ、DPL1、LCB4およびYSR2)が、欠失されている。この株は、LCBP合成の非存在下で生存し得るが、この背景下における機能的SK遺伝子の発現は、分解され得ないLCBPの重篤な蓄積が原因となり、致死となる。CG1747およびCG2159の発現を、この背景下においてガラクトース存在下で誘導する場合、酵母の増殖は、まったく起きず、これらのcDNAが、機能的なSK酵素をコードすることを示す。最後に、CG2159のトランスポゾン変異体を入手し、この遺伝子座からの発現の欠如を示した(Rosemann,R,Johnson E,Rodesch C,Bjerke M,Nagoshi RおよびGeyer P,1995.Drosophila melanogaster,Genetics 141:1061−l074.)。この変異体(Sphk2KG05894)を、以前に記述されるように、P因子をCG2159の5’UTRへ挿入することにより、作製した。予備調査により、この変異体が、Sply変異体で見られたのと同様に、DLMにおける軽度の欠損を有することが、呈示される。

lace遺伝子は、Drosophilaセリンパルミトイルトランスフェラーゼの1つのサブユニットをコードする。2つのlacek05305ヌル対立遺伝子の遺伝形質は、一様に致死であると報告され、一方、これらの実験で使用されたヘテロ接合型の対立遺伝子の組み合わせである、lacek05305/lace2は、重篤な発生上の表現型および生存可能な子孫の割合の低さをもたらす(Adachi−Yamada,T.,T.Gotoh,I.Sugimura,M.Tateno,Y.Nishida,T.Onuki,およびH.Date.1999.Mol.Cell.Biol.19:7276−7286.)。2つの予想されるスフィンゴシンキナーゼ(SK)遺伝子のうちの1つのヌル対立遺伝子についてホモ接合型のDrosophila株も、これらの調査に利用した。この変異体(Sphk2KG05894)を、以前に記述されるように、P因子をCG2159の5’UTRへ挿入することにより、作製した。この遺伝子の産物は、酵母のSK変異体を機能的に補完する。野生型Canton−S(BL−1)、lace2(BL−3156)、lacek05305(BL−12176)株およびSpnkKG05894(BL−14133)株は、Bloomington Drosophila Stock Center(Indiana University,Bloomington,IN)より入手した。
25mgの凍結したインタクトなハエ材料を含むサンプルを、7mlのPotter Elvehjemホモジナイザーに入れた。250から500pmolのそれぞれのLCBを含む、20μlの内部LCB基準物質混合物(Matreya Inc.,Pleasant Gap,PA)を、さらに加えた。ハエを、2mlの氷冷メタノール/水、1:1(v/v)の中で、ゆるいペストルでその後きついペストルで、それが滑らかに動くまでホモジェナイズした。抽出物を、氷上でチップソニケーター(3×20秒)によりさらにホモジェナイズし、その後ガラス管へと移し、1500×gで10分間遠心分離した。上清を、回収し、スピードバック(speed vac)の中で乾燥させた。抽出物を、0.1Mの水酸化アンモニウムを含むメタノール200μlの中で再懸濁し、その後ボルテックスし、水槽ソニケーションをし、さらに、37℃で1時間インキュベーションし、エステル化アシル鎖の加水分解を行った。加水分解の後、そのサンプルを、室温まで冷やし、スピードバックの中で乾燥させ、さらに、500μlの、0.1%の氷酢酸を含むメタノール/水、2:3(v/v)(溶媒A)の中で再懸濁させた。
Strata C18−E固相抽出カラム(50mg/ml)(Phenomenex,Torrance,CA)を、最初に200μlのメタノールにて湿らせ、その後1mlの溶媒Aにて平衡化した。溶媒Aの中のハエ抽出物またはLCB標準物質を、平衡化Strata C18−Eカラムにアプライし、その後1mlの溶媒Aにて洗浄した。そのカラムの2回目の洗浄を、600μlのメタノールの添加により行った。LCBを、600μlのメタノール:10mM酢酸アンモニウム、9:1(v/v)によりそのカラムから溶出させ、さらにスピードバックの中で乾燥させた。
LCBを、以前に記述されるように、オルト−フタルアルデヒド(OPA)(Sigma St.Louis,MO)で誘導化した(Caligan,T.B.,K.Peters J.Ou,E.Wang,J.Saba,およびA.H.Jr.Merrill.2000.Anal.Biochem.281:36−44.)。そのOPA−誘導化LCBを、逆相カラム(Luna RP−18,3μ,4.6×75mm)(Phenomenex,Torrance,CA)上で、メタノール:10mM酢酸アンモニウム、pH5.2、82:18(v/v)の移動相により、分離した。流速は、1ml/分であった。使用したHPLC系は、125種類の溶媒モジュールを備えるBeckman System Goldであった。その蛍光性LCBを検出し、Spectra−Physics蛍光検出器(SP 8410)を使用して定量した。
成虫Sphk2KG05894ハエからStrata C18−Eカラムにより精製された脂質抽出物またはC14So標準物質を、10μlの試料の直接注入の後、Micromass Quattro LCZ装置上で分析した。移動相は、0.1パーセントの蟻酸を含む80パーセントメタノールであった。流速は、0.2ml/分であった。LCBの構造的確証は、陽性電気噴射イオン化(ESI+)質量分析により得られた。LCBを、記載されるように、構造的に別個のイオン断片の前駆イオン走査により検出した(Sullards,M.C.,およびA.H.Jr.Merrill.2001.Sci.STKE.67:1−11.)。そのキャピラリーへの3.5kVの印加により、その噴射およびその衝突により誘導される分解スペクトルを、20Vのコーン電圧で開始し、アルゴンを衝突気体として用いて、15eVの衝突エネルギーで記録した。
(スフィンゴイドベースのHPLC分離およびスフィンゴイドベースの固相抽出)
HPLC法を、14から18の炭素数を持つLCBの分離のために開発した。成虫ハエからのメタノール/水粗製脂質抽出物の初めのHPLC分離は、蛍光物質の高程度の混在により、複雑であった。その結果、HPLC分析に先立ち、Strata C18−Eカラムを使用する固相抽出工程を導入した。メタノールを、溶出溶媒として採用した場合、LCB標準物質の回収は、2パーセントより少なかった。Strata C18−EカラムからのLCB標準物質のこの不十分な回収は、10容積%の10mM酢酸アンモニウム溶液をメタノール溶出溶媒に添加することにより、完全に回復した。この溶出系の採用により、C14およびC16スフィンゴイドベース標準物質について、60から95パーセントの範囲での回復が得られた(表3)。
メタノール/10mM酢酸アンモニウム 9:1(v/v)を用いるStrata C18−Eカラムからの成体ハエ脂質の溶出は、なお、望ましくない有意なバックグラウンド蛍光を伴うHPLCスペクトルを生じた。このバックグラウンドを、メタノール/10mM酢酸アンモニウム9:1(v/v)を用いる溶出の前にメタノールを添加することによって最小にした(詳細については、材料および方法を参照のこと)。3つの異なる系統の成体ハエを分析した。野生型ハエの脂質プロフィールを、スフィンゴシンキナーゼ(Sphk2)変異体およびセリンパルミトイルトランスフェラーゼ(SPT,lace)変異体の脂質プロフィールと比較した(上記の材料および方法を参照のこと)。このSphk2変異体は、LCBをリン酸化する能力の減少を示すことが推測され、その結果、LCBレベルの増加を示すはずである。対照的に、ハイポモルフlace変異体は、スフィンゴ脂質新規生合成の第1段階を欠損しており、LCBレベルの減少またはLCBの完全な欠如を示すと推測される。C14 So標準物、C16 So標準物、C16 Sa標準物と同じ保持時間を示す3つのピークを、野生型ハエ抽出物において同定した。さらに、C14 SoとC16 Soとの間の保持時間で溶出する主要ピークを、同定した。上記4つのピークは、Sphk2変異体において増加し、lace変異体において減少した。これは、これらのピークがLCBを示す可能性と一致した。C18 LCB標準物に対応する保持時間で溶出したピークは、観察されなかった。
LCBを、陽イオン電子スプレー質量分析(ESI+)を使用して、その衝突誘導性乖離のパターンおよび前駆イオンスキャンのパターンを介して同定し得る。その独特の分子構造に基づいて、代表的分解産物が、2つの水分子の欠失から生じる。m/z 208(C14 So−2水分子)の前駆イオンスペクトルは、m/z244(C14So)およびm/z 226(C14So−1水分子)として、親を示す。DrosophilaにおけるC14 Saの存在を確認するために、本発明者らは、ESI+によるStrata C18−Eカラム精製脂質抽出物を分析した。Sphk2変異体からの脂質抽出物を、この分析のために選択した。なぜなら、この脂質抽出物は、増加したLCBレベルを示したからである。最初に、本発明者らは、内因性C14 Soの存在を探索した。m/z 208の前駆イオンスペクトルは、この抽出物においてC14 So(m/z 244)を同定した。その後、本発明者らは、C14 Saの存在を探索した。m/z 210の前駆イオンスペクトルは、内因性C14 Sa(m/z 246)を同定した。さらに、m/z 236およびm/z 238の前駆イオンスキャンは、このハエ抽出物において内因性C16 Soおよび内因性C16 Saを同定した。m/z 264およびm/z 266の前駆イオンスキャンは、このハエ抽出物においてC18 LCBを同定しなかった。このことは、上記HPLC分析から得られた結果を支持する。
内因性Drosophila LCBを、Strata C18−Eカラム精製抽出物(規定量のC14 So標準物、C16 So標準物およびC16 Sa標準物を添加しているもの、または添加していないもののいずれか)のHPLC分離を実施することによって、定量した。分離後に、各蛍光LCBピークについて得られた積分した面積(表4)の比較を行った。興味深いことに、lace変異体ハエおよびSphk2変異体ハエは、各系統からの脂質抽出物の分析により測定した場合、LCB総量およびLCB比較の両方において、野生型ハエと明らかに異なった。野生型LCBの総量は、ハエ全体100mgにつき、約1.5nmolであった。Sphk2変異体は、野生型ハエと比較して、LCB総量において、3.3倍増加を示し、lace変異体は、2.5分の1の減少を示した。C14 Soは、野生型ハエにおいて、LCB総量のうちの約42%を占め、一方、C14 Saは、約47%を占めた。従って、C14 So対C14 Saのモル比は、約1:1であった。Sphk2変異体において、対応する値は、26%C14 Soおよび67%C14 Saであり、モル比約1:2.5を生じた。一方、lace変異体において、対応する値は、16%C14 Soおよび81%C14 Saであり、モル比約1:5を生じた。

遺伝子研究によって、スフィンゴ脂質中間体の役割は、発生プロセスに関係付けられている。しかし、発生の間にわたるこれらの分子の定量は、実施されていない。スフィンゴ脂質中間体の潜在的役割についての生化学的基礎が存在するか否かを調査するために、本発明者らは、種々のDrosophila発生段階における内因性C14 LCBを評価した(表5)。C14 LCBの総量は、野生型の発生全体にわたって全く一定のままであった。しかし、初期胚からさなぎまでの発生進行は、C14 So対C14 Saのモル比における7倍の増加に関係があった。
(変化したスフィンゴ脂質代謝を有するDrosophila melanogaster系統における肉眼および顕微鏡による腫瘍発症の分析)
いくつかのDrosophila melanogasterスフィンゴ脂質代謝変異体は、腫瘍(特に、S−1−Pの有意な蓄積を示す腫瘍)を発症し得る。lgl変異体およびdlg変異体と関連する以前に特徴付けられたDrosophila melanogaster腫瘍は、脊椎動物における新生物増殖に関連する特徴(宿主に対する致死性、反復移植能、浸潤性、最終分化の欠如、インビボおよび組織培養での迅速な増殖、ならびに細胞間の相互作用および連絡の欠損を含む)を保有する。変異体において生じる肉眼または顕微鏡により可視可能な腫瘍におけるこれらの特徴の各々の評価を、実施する。成虫原基の過剰増殖を、明白な腫瘍がない場合でさえ、すべての変異体において探索する。致死性変異は、初期発生段階の間における腫瘍形成の存在を示唆し得る。変態の間に正常な分化シグナルに対して、成虫原基腫瘍細胞または成虫原基過剰増殖が応答する能力を、変異体成虫原基を野生型幼生に移植し、その運命を追跡することによって、評価した。インビトロおよび成体雌の腹において多数の転移世代の間、腫瘍または過剰増殖成虫原基が連続培養される能力を、評価する。腫瘍浸潤性および転移能力の証拠を、組織学的分析を使用して上皮鞘に囲まれた組織中への浸潤の存在または非存在を決定することによって、測定した。
(変化したスフィンゴ脂質代謝を有するDrosophila melanogaster系統における肉眼および顕微鏡による解剖学的欠損の評価)
特定の例示的実験方法が、本明細書中(例えば、実施例1の前の上記の節)に記載される。本発明をさらに規定するために使用され得るさらなる例示的技術(例えば、Drosophila melanogasterの肉眼および顕微鏡による解剖学的欠損の評価のための一般的技術)が、当該分野で公知であり、本明細書中にさらに記載される。各系統の寿命を測定し、野生型ハエと比較して、生存能に対する影響を評価する。気管系および筋肉系の肉眼による解剖学を、偏光を用いて幼生において観察し、そして光学顕微鏡下で標準厚切片を観察することにより成体において観察して、気管もしくは筋肉の数、サイズ、位置または適切な付着に影響を与える明白な欠損を同定する。光学顕微鏡法のための調製は、グリセロール/エタノール溶液中にハエを貯蔵すること、その後、70%エタノールによって不要な構造物を解体すること、切片化のために標本を再水和し、そして封入もしくは包埋することを包含する。可視化を、明視野、DIC照明、位相差、または暗視野照明下で実施する。IFMの電子顕微鏡分析を、成体、幼生およびさなぎに対して実施する。初期Drosophila melanogaster胚は、蛍光二次抗体またはいくつかの場合においては一次抗体を使用して検出される多数の高度に特異的な抗体のいずれかを使用する構造分析をしやすい。多数の段階的胚を、正常ストックおよび変異体ストックから収集し得、そして蛍光分析のために調製し得る。このプロセスは、漂白剤溶液を使用して脱塩素化、ビテリン膜を透過性にすること、(メタノールを用いない)固定およびビテリン膜除去、染色、洗浄、および封入を包含する。lace変異体は、翅成虫原基の異常なアポトーシスを示すので、本発明者らは、すべての変異体における成虫原基の構造および外転におけるなんらかの変化を調査することを計画する。成虫原基の解体を、生理食塩水溶液中で実施する。この解体は、第三齢の幼生を半分に引き裂くこと、体壁を反転すること、および前部原基または後部原基の塊を体から摘み取ることを包含する。原基を、インサイチュハイブリダイゼーション研究のために体壁に付けたままにしても良いし、または原基を取り出して、他の研究のために固定してもよい。さなぎの成虫原基をまた、一晩固定した後に角質を除去することによって単離し得る。
(スフィンゴ脂質と相同性を有する合理的に設計された一群の化学物質を、SPL変異体子孫に対して飛行を回復する能力をスクリーニングすることによる、SPL変異体が飛行不能であることの薬理学的サプレッサの同定)
Sply変異体Drosophila melanogasterが飛行不能であることの薬理学的サプレッサを、スフィンゴ脂質と相同性を有する合理的に設計された一群の化学物質を、Sply変異体Drosophila melanogasterに対して飛行を回復する能力をスクリーニングすることによって、同定する。変異体SPLハエを、ビヒクルコントロールまたはμM濃度のインヒビターのいずれかを補充した培地において、18℃で増殖させる。種々のインヒビターが飛行を回復させる能力を、標準的スコア付け法(本明細書の他の場所にもまた記載される)を使用して、測定する(Vigoreauxら、1993、J Cell Biol 121:587〜598もまた、参照のこと)。その後、興味深い化合物の効力および生化学的特徴を、1)精製SK、SPL、およびスフィンゴ脂質代謝に関与する他の酵素(例えば、セリンパルミトイルトランスフェラーゼ、セラミドシンターゼ、スフィンゴシンデサチュラーゼ、セラミダーゼ、セラミドキナーゼ、ホスホエタノールアミンシチジルイルトランスフェラーゼ、CDP−エタノールアミンホスホトランスフェラーゼ、酸性スフィンゴミエリナーゼおよび中性スフィンゴミエリナーゼ)に対するそのインヒビターのIC50を決定することによって、定量する。そのインヒビターのIC50をまた、組換えヒトSK、SPL、および上記に列挙したスフィンゴ脂質代謝に関与する他の酵素に対しても決定し、2)古典的Michaelis−Menten法を使用して阻害の速度論を分析し、そして3)その阻害の可逆性を決定する。合成アナログを、このスクリーニングのために生成する。潜在的SKインヒビターとしてのスクリーニングのために、1−アリール−2−ジメチルアミノプロパン−1,3−ジオール誘導体のライブラリーを、合成する。4つのジアステレオマー(DまたはL、erythro−、またはthreo−)を、このライブラリーの各メンバーについて合成する。このライブラリーにおいて、その脂肪酸アミド基を、2つのN−メチル基で置換し、極性置換基および芳香族置換基において同様の変化を生じる。この合成計画は、出発物質として、周知のGarnerアルデヒド(図2における1を参照のこと)を利用する。なぜなら、1は、いずれかのエナンチオマー形態において容易に利用可能であるからである。金属、溶媒、および添加Lewis酸触媒の効果をまとめる、1への有機金属添加についての最近の精緻な概説が、刊行されており、それは、erythro産物が、ほとんどの反応において通常は支持されることを示す。従って、出発物質として1のD−エナンチオマーまたはL−エナンチオマーを選択することによって、各ライブラリーメンバーの純粋なerythro立体異性体を調製する。対応するthreoアナログ(4a〜c、図2)を構築するための新規かつ柔軟な経路を、PDMPアナログを生成するための方法論の簡単な拡張法を使用して実行する。親化合物である4aは、すでに公知であり、容易に入手可能である。このストラテジーは、特定のスルフィド添加剤およびホスフィン添加剤の存在下での、アリール金属化合物(Aryl−Met)の1へのsyn選択的添加に依存する。このerythro合成経路およびthreo合成経路の両方とも、一級炭素原子における置換変化形を調製するために改変する。7a〜cの調製のための代表的合成手順を、図3に示す。広範囲の窒素求核剤、酸素求核剤、および炭素求核剤が、5a〜cのようなメシレートと反応して、ジメチル化PDMPアナログおよびホモログの新規ライブラリーを提供し得た。
(SKの阻害が細胞生存を付与するように考案された酵母スクリーニングにおいてヒトSKを阻害する能力を試験することによる、候補薬物のさらなる評価)
実施例14に記載されるハエスクリーニングにおいて同定した化合物を、酵母モデルにおいてヒトSKを阻害する能力についてさらに評価する。ヒトSKの活性をブロックする化合物は、酵母系統dpl1ysr2lcb4(Gal1,10p:ヒトSPHK1)に対して、ガラクトース上での増殖を回復するはずである。この系統は、S−1−Pも内因性酵母S−1−Pアナログも代謝し得ない。それは、SPLホスファターゼをコードする遺伝子およびS−1−Pホスファターゼをコードする遺伝子を欠失していることに起因する。内因性酵母SK遺伝子であるLCB4もまた欠失しており、その結果、内因性S−1−Pアナログが、基準条件下で生成される。この系統は、ガラクトース誘導性プロモーターの調節下にヒトSPHK1遺伝子を含むプラスミドで形質転換されており、その結果、ガラクトース存在下での活性SKの発現は、この系統に対して致死性である。ヒトSKの阻害は、ガラクトース誘導性致死性から細胞を保護し、問題となるその化合物の効力を確認する。この系統におけるヒトSPHK1遺伝子の活性は、2つの方法により確認されている。第1の方法は、ガラクトース存在下で(制限期間の間)増殖した細胞の全抽出物におけるSK活性のHPLC分析であり、第2の方法は、ガラクトース含有プレート上でこの系統がコロニーを形成不能であることである。
(トランスジェニックDrosophila melanogaster発現ヒトSPLおよびヒトSPL−GFP融合物の生成)
ヒトSPL(配列番号23に示されるcDNA;配列番号18に示されるアミノ酸配列)およびヒトSPL−GFP融合タンパク質を過剰発現するトランスジェニックDrosophila melanogasterを、本明細書中で記載される標準技術を使用して生成した。これらのトランスジーンを、野生型Canton−S(BL−1)、およびSply05091(BL−11393)、変異体ハエバックグラウンド中に導入した。

- スフィンゴ脂質の代謝を調節する薬剤を同定するための方法であって、該方法は、以下の工程:

請求項1、20および24に記載の方法のいずれか1つによって同定された、薬剤。 - 生理学的に受容可能な賦形剤と組み合わせて、請求項25に記載の薬剤を含む、組成物。

- 前記薬剤が、スフィンゴシンキナーゼ活性を示す、請求項27に記載の組成物。
- スフィンゴシン−1−リン酸リアーゼ(SPL)ポリペプチドを調節するための方法であって、該方法は、
配列番号15に示されるヌクレオチド配列を含むポリヌクレオチドに作動可能に連結されたプロモーターを含む核酸構築物で形質転換またはトランスフェクトした宿主細胞を培養する工程;および
スフィンゴシン−1−リン酸リアーゼポリペプチドを回収する工程、
を包含する、方法。 - スフィンゴシン−1−リン酸リアーゼ活性を調節する薬剤を同定するための方法であって、該方法は、以下:
(a)候補薬剤を、以下:
(i)配列番号16に示されるアミノ酸配列;および
(ii)配列番号16に示されるアミノ酸配列に対して少なくとも90%の同一性を有するアミノ酸配列、
からなる群より選択されるアミノ酸配列を含む、単離されたポリペプチドと接触させる工程であって、
ここで、該ポリペプチドは、スフィンゴシン−1−リン酸リアーゼ活性を有し、そして該接触させる工程は、該候補薬剤が該ポリペプチドと相互作用することを可能にするのに十分な条件下および時間にわたって、実行される、工程;ならびに
(b)該候補薬剤の存在下での、該ポリペプチドによるスフィンゴシン−1−リン酸またはそのスフィンゴシン−1−リン酸誘導体の分解を、該候補薬剤の非存在下での、該ポリペプチドによるスフィンゴシン−1−リン酸またはそのスフィンゴシン−1−リン酸誘導体の分解に対して決定し、そこから、スフィンゴシン−1−リン酸リアーゼ活性を調節する薬剤を同定する工程、
を包含する、方法。 - スフィンゴシン−1−リン酸リアーゼ活性を調節する薬剤を同定するための方法であって、該方法は、以下:
(a)候補薬剤を、以下:
(iii)配列番号16に示されるアミノ酸配列;および
(iv)配列番号16に示されるアミノ酸配列に対して少なくとも90%の同一性を有するアミノ酸配列、
からなる群より選択されるアミノ酸配列を含むポリペプチドを発現する細胞を含む生物学的サンプルと接触させる工程であって、
ここで、該ポリペプチドは、スフィンゴシン−1−リン酸リアーゼ活性を有し、そして該接触させる工程は、該候補薬剤が該ポリペプチドと相互作用することを可能にするのに十分な条件下および時間にわたって、実行される、工程;ならびに
(b)該候補薬剤の存在下での、該ポリペプチドによるスフィンゴシン−1−リン酸またはそのスフィンゴシン−1−リン酸誘導体の分解を、該候補薬剤の非存在下での、該ポリペプチドによるスフィンゴシン−1−リン酸またはそのスフィンゴシン−1−リン酸誘導体の分解に対して決定し、そこから、スフィンゴシン−1−リン酸リアーゼ活性を調節する薬剤を同定する工程、
を包含する、方法。 - 前記決定する工程が、前記細胞由来の抽出物のインビトロアッセイを含む、請求項31に記載の方法。
- 生理学的に受容可能なキャリアと組み合わせて、ポリペプチドのスフィンゴシン−1−リン酸リアーゼ活性を調節する薬剤を含む組成物であって、該ポリペプチドは、配列番号16に示される配列を含む、組成物。
- 前記薬剤が、ポリヌクレオチドを含む、請求項33に記載の組成物。
- 請求項33に記載の組成物であって、前記薬剤が、配列番号16に示される配列を含むスフィンゴシンリン酸リアーゼ(SPL)ポリペプチドに特異的に結合する抗体またはその抗原結合フラグメントを含み、ここで、該抗体が、スフィンゴシン−1−リン酸を分解する該SPLポリペプチドの能力を増大させる、組成物。
- 癌細胞の増殖を阻害するための方法であって、該方法は、配列番号16に示される配列を含むポリペプチドのスフィンゴシン−1−リン酸リアーゼ活性を増大させる薬剤と、該癌細胞とを接触させる工程を包含する、方法。
- 前記薬剤が、内因性スフィンゴシン−1−リン酸リアーゼ遺伝子の発現を増大させる、請求項36に記載の方法。
- 前記癌細胞が、乳癌細胞である、請求項36に記載の方法。
- 哺乳動物における癌の発症、転移、または癌および転移の両方の発症を阻害するための方法であって、該方法は、配列番号16に示される配列を含むポリペプチドのスフィンゴシン−1−リン酸リアーゼ活性を増大させる薬剤を、該哺乳動物に投与する工程を包含する、方法。
- 前記薬剤が内因性スフィンゴシン−1−リン酸リアーゼ遺伝子の発現を増大させる、請求項39に記載の方法。
- 前記薬剤が、標的化成分に連結されている、請求項40に記載の方法。
- 前記標的化成分が、抗腫瘍抗体である、請求項41に記載の方法。
- 前記標的化成分が、エストロゲンレセプターに結合する、請求項42に記載の方法。
- 前記哺乳動物が乳癌に罹患している、請求項39に記載の方法。
- 患者における癌の存在を決定するための方法であって、該方法は、以下:
(a)少なくとも1つのポリヌクレオチドを含み、かつ癌を有することが疑われる患者から得られた第一の生物学的サンプルを、配列番号23に示される核酸配列を含むポリヌクレオチドに特異的な少なくとも1つのオリゴヌクレオチドと接触させる工程;
(b)該第一のサンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量を検出する工程;ならびに
(d)該第一のサンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量を、癌を有さないことが既知の正常なコントロール被験体から得られた第二の生物学的サンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量と比較する工程であって、
ここで、該第二のサンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量と比較して、該第一の生物学的サンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量における統計的に有意な増加は、該患者における癌の存在を示す、
方法。 - 変更されたスフィンゴ脂質代謝に関連する疾患を診断するための方法であって、該方法は、以下:
(a)少なくとも1つのポリヌクレオチドを含み、かつ該変更されたスフィンゴ脂質代謝に関連する疾患を有することが疑われる患者から得られた第一の生物学的サンプルを、配列番号23に示される核酸配列を含むポリヌクレオチドに特異的な少なくとも1つのオリゴヌクレオチドと接触させる工程;
(b)該第一のサンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量を検出する工程;ならびに
(d)該第一のサンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量を、変更されたスフィンゴ脂質代謝に関連する疾患を有さないことが既知の正常なコントロール被験体から得られた第二の生物学的サンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量と比較する工程であって、
ここで、該第二のサンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量と比較して、該第一の生物学的サンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量における統計的に有意な増加は、該患者における変更されたスフィンゴ脂質代謝に関連する疾患の存在を示す、
方法。 - 患者における癌の存在を決定するための方法であって、該方法は、以下:

(d)該第一のサンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量を、癌を有さないことが既知の正常なコントロール被験体から得られた第二の生物学的サンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量と比較する工程であって、

(b)該第一のサンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量を検出する工程;ならびに
(d)該第一のサンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量を、変更されたスフィンゴ脂質代謝に関連する疾患を有さないことが既知の正常なコントロール被験体から得られた第二の生物学的サンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量と比較する工程であって、
ここで、該第二のサンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量と比較して、該第一の生物学的サンプル中のポリヌクレオチドにハイブリダイズするオリゴヌクレオチドの量における統計的に有意な減少は、該患者における変更されたスフィンゴ脂質代謝に関連する疾患の存在を示す、
方法。 - 患者において、変更されたスフィンゴ脂質代謝に関連する疾患を処置するための方法であって、請求項1、20、24および30のいずれか1項に記載の方法に従って同定された薬剤を該患者に投与する工程を包含する、方法。
- 前記疾患が、結腸癌、乳癌、子宮癌、胃癌、卵巣癌、肺癌、腎臓癌、直腸の腺癌、遺伝性感覚ニューロパシー1型、およびスフィンゴリピドーシス(sphingolipidoses)の任意の1つからなる群より選択される、請求項49に記載の方法。
