本発明は、第1に、DARPP-21が関与している神経系のカルシウムシグナル伝達をモジュレートする活性について、薬剤をスクリーニングする方法を提供する。例えば、本発明は、DARPP-21とカルモジュリンとの間のタンパク質-タンパク質結合を阻害または増強させる薬剤をスクリーニングする方法、およびカルモジュリンとのその結合がDARPP-21によって阻害されるかまたは増強される候補カルモジュリン結合タンパク質をスクリーニングする方法を提供する。本発明はまた、DARPP-21関連シグナル伝達経路、例えば、ドーパミン作動性シグナル伝達経路などをモジュレートするための組成物および方法にも関する。例えば、本発明は、そのようなシグナル伝達経路でのDARPP-21のリン酸化状態、タンパク質発現レベル、および/またはタンパク質間相互作用をモジュレートする方法および組成物に関する。本発明はまた、例えば、DARPP-21/カルモジュリン複合体の形成をモジュレートする薬剤を投与することにより、ドーパミン関連疾患を予防、治療、またはその症状を改善する組成物および方法にも関する。
神経伝達物質、ホルモン、薬物、および細胞の脱分極などの種々の刺激への応答としての細胞内カルシウムレベルの上昇は全ての細胞にあまねく見られるものである。神経のカルシウム依存性シグナル伝達は記憶の獲得、てんかん、および脳虚血のような重要かつ様々な各種の生理学的および病理学的プロセスに関与している。
現在では、DARPP-21のリン酸化が中枢神経系におけるカルシウム依存性シグナル伝達をモジュレートすることが判明している。特に、本発明は、部分的には、DARPP-21のリン酸化状態が生理的条件下でDARPP-21とカルシウム/カルモジュリンとの相互作用を調節するとの発明者らによる驚くべき発見に基づいたものである。DARPP-21は以前はARPP-21と呼ばれていたことに注意すべきである;例えば、米国仮特許第60/358,548号を参照せよ。

5. 発明の詳細な説明
本発明は、部分的には発明者らによる、生理的条件下でDARPP-21のリン酸化状態がDARPP-21とカルシウム/カルモジュリンとの相互作用を調節するとの驚くべき発見に基づいたものであり、カルシウム依存性シグナル伝達の調節について薬剤をスクリーニングする方法において用いることができる。これらの知見に基づいて、細胞内カルシウムシグナル伝達をモジュレートし、ニューロン細胞カルシウムシグナル伝達をモジュレートし、さらにドーパミン関連疾患を予防し、治療し、または症状を改善することができる薬剤の標的として、DARPP-21/カルモジュリン複合体の形成が同定されている。
下記の実施例は本発明の原理と利点を説明するためのものである。これらの実施例はいかなる意味においても本発明を限定することを意図したものではない。
リン酸化されたDARPP-21を調製するために、既に報告(Caporasoら, 2000, Neuropharmacology 39:1636-1643)されているように[γ-32P]ATPをPKAの触媒性サブユニットとともに用いることによって、タンパク質DARPP-21をin vitroでリン酸化した。リン酸化の化学量論により、DARPP-21の95%がリン酸化されていることが判った。
免疫染色はサンプル中のカルモジュリンまたはDARPP-21の検出のために次のようにして用いた。カルモジュリンのウエスタンブロット解析は、Van EldikとWolchokが報告している方法(1984, BBRC 124:752-759)にマイナーな改変を加えて行った。タンパク質は、SDS-PAGEで分離し、次いで電気泳動により20分間(カルモジュリン)、または60分間(DARPP-21)かけてPVDF膜に転写した。続いてその膜をリン酸塩緩衝生理食塩液(PBS)で2回すすぎ、0.2%グルタールアルデヒド/ PBSで室温で45分間かけて固定した。次いで、その膜をPBS/0.05% Tween-20中で5% 粉ミルク溶液を用いて1時間ブロックした後、一次抗体とインキュベートした。DARPP-21はウサギポリクローナル#204抗体またはモノクローナル6A抗体を用いて同定した。抗カルモジュリンモノクローナル抗体はUpstate Biotechnology, Inc,(Lake Placid, NY)から入手した。
融合タンパク質の構築および発現のために、DARPP-21フォワードプライマー(5'-AGCGAATTCATGTCTGAGCAAGGAGAACT-3';配列番号1)およびDARPP-21リバースプライマー(5'-ACGGGATCCGGAGAGTCTGATCCTGGTGAC-3';配列番号2)を用いてDARPP-21 cDNAを増幅し、さらに酵母においてDARPP-21/Lex A融合タンパク質を作製した。Alaフォワードプライマー(5'-ACCTGCTCCTGACTTGGCTTTTCTTCTCTCTTG-3';配列番号3)、Aspフォワードプライマー(5'-CAAGAGAGAAGAAAAGACAAGTCAGGAGCAGGT-3';配列番号4)、およびAspリバースプライマー(5'-ACCTGCTCCTGACTTGTCTTTTCTTCTCTCTTG-3';配列番号5)を用いて、Ser55の置換としてそれぞれAlaまたはAspを有するDARPP-21突然変異体を作製した。ここで得られた変異はDNA配列決定によって確認した。Caporasoら(2000, Neuropharmacology 39:1636-1643)が報告しているようにして、DARPP-21のC末端 6×Hisタグ融合タンパク質を作製し、発現させ、精製した。
6.4.1 酵母2-ハイブリッドタンパク質-タンパク質結合アッセイ
タンパク質-タンパク質結合アッセイは、酵母株EGY48においてDupLexAシステム(Origene Technologies, Rockville, MD)を用いる酵母2-ハイブリッドシステムにて行った。LexAとDARPP-21の融合タンパク質を作製するために、DARPP-21 cDNAをpEG202ベイト(餌)ベクターのEcoRI-XhoI部位にサブクローニングした。オリゴdTプライマー増幅ラット脳ライブラリー(Origene Technologies, Rockville, MD)を含んでいるプレイ(捕食)ベクターを、B42 酸性ブロブ(blob)ドメインに結合させ、gal-Iプロモーターによって制御した。
ウシ線条体(2グラム)を10mLのバッファーA(150mM NaCl、20mM Tris-HCl, 7.5、0.5mM CaCl2、1% Triton X-100、およびEDTA不含のプロテアーゼインヒビターカクテル(Roche Diagnostics Corporation, Indianapolis, IN)を含有したもの)中でホモジナイズした。不溶物を100,000 x g で15分間かけて簡易遠心分離することによって除去した。その上清をバッファーAで希釈して最終タンパク質含量を1 mg/mLとした。その抽出物(1mL)を抗カルモジュリン抗体または抗DARPP-21抗体とともに4℃で一晩インキュベートした。プロテインA−Sepharoseスラリー(50% w/vの70μL;Amersham Pharmacia Biotech, Piscataway, NJ)を各サンプルに添加し、それらのサンプルをさらに3時間インキュベートした。ビーズを、300mM NaClを含有する改変バッファーAで3回洗い、結合したタンパク質をSDS-PAGEサンプルバッファーで溶出させた。得られたサンプルを4〜20%勾配SDS Tris/グリシンゲル(Novex)で分画し、上記の第6.2節に記載の免疫染色で分析した。
6.4.3.1 DARPP-21-Sepharoseを用いたアフィニティークロマトグラフィー
精製組換えDARPP-21(2〜5mg)を、2mLのAminolink Sepharose(Pierce Inc., Rockford, IL)に、共有結合により架橋した。凍結線条体抽出物(2g)を前述のバッファーA中でホモジナイズした。ビーズへの非特異的なタンパク質結合を避けるために、線条体抽出物(1mg/mL)は、4mLのSepharose 4Bを含有する模擬カラムを通過させた。その溶出物を半分ずつ2つに分け、第1の半量をDARPP-21を固定化したカラムにアプライし、第2の半量は模擬カラムを通過させた。カラムを150mM NaClと20mM Tris pH7.5で洗い、10mM EGTA/20mM Tris pH7.5; 2mM NaClおよび6M グアニジニウムHClで溶出した。各溶出ステップからのサンプルをSDS-PAGEで分離し、PVDF膜に転写し、抗DARPP-21特異的抗体によってプローブした。
DARPP-21(0.1mg/mLのものを0.5mL)を、バッファーB(100mM NaCl、20mM Tris pH7.5、1mM MgCl2、0.5mM CaCl2、1mM DTT)または0.5mM CaCl2の代わりに1mM EGTAを含有するバッファーC中に入れた0.5mLの1mg/mLカルモジュリン-Sepharose(Amersham Pharmacia Biotech, Piscataway, NJ)にアプライした。ビーズを、数倍量の300mM NaClおよび20mM Tris pH7.5を含有するバッファーDで洗い、結合しているタンパク質を10mM EGTA/20mM Tris pH7.5で溶出させた。
DARPP-21を第6.1節に記載のようにしてリン酸化した。CaMKIおよびカルシニューリンのキナーゼ活性およびホスファターゼ活性のそれぞれに対するin vitro阻害研究は、当業界の標準的な技法に従って行った。
Claims (28)
- 細胞または組織をDARPP-21の生物活性をモジュレートする薬剤と接触させて、シグナル伝達経路をモジュレートすることを含んでなる、細胞または組織内でDARPP-21の生物活性をモジュレートする方法。
- 前記薬剤がDARPP-21のリン酸化状態を変化させるものである、請求項1に記載の方法。
- 前記薬剤がDARPP-21/カルモジュリン複合体の形成を変化させるものである、請求項1に記載の方法。
- ドーパミン関連疾患を予防、治療、またはその症状を改善する方法であって、それらを必要としている被験者にDARPP-21の生物活性をモジュレートする薬剤を投与することを含んでなる、前記方法。
- 前記薬剤がDARPP-21のリン酸化状態を変化させるものである、請求項4に記載の方法。
- 前記薬剤がDARPP-21/カルモジュリン複合体の形成を変化させるものである、請求項4に記載の方法。
- 細胞または組織におけるカルシウムシグナル伝達をモジュレートする活性について薬剤をスクリーニングする方法であって:
(a) 細胞または組織の第1のサンプルにおけるDARPP-21の第1のリン酸化レベルを測定すること;
(b) 細胞または組織の第2のサンプルを、カルシウムシグナル伝達を調節する能力について試験すべき薬剤と接触させること;
(c) 該細胞または組織におけるDARPP-21の第2のリン酸化レベルを測定すること;および
(d) 第1および第2のリン酸化レベルを比較するが、ここでそれらのレベルの差は、該薬剤がDARPP-21リン酸化をモジュレートし、ひいてはカルシウムシグナル伝達をモジュレートすることを示すこと、
を含んでなる前記方法。 - 細胞または組織におけるカルシウムシグナル伝達をモジュレートする活性について薬剤をスクリーニングする方法であって:
(a) 細胞または組織のサンプルを、カルシウムシグナル伝達を調節する能力について試験すべき薬剤と接触させること;
(c) (a)の細胞または組織中におけるDARPP-21のリン酸化レベルを測定すること;および
(d) (c)で測定したレベルを、該薬剤と接触させなかった対応する細胞または組織の対照サンプルでのレベルと比較するが、ここでそれらのレベルの差は、該薬剤がDARPP-21リン酸化をモジュレートし、ひいてはカルシウムシグナル伝達をモジュレートすることを示すこと、
を含んでなる前記方法。 - 該細胞または組織が脳の細胞または組織である、請求項7または8に記載の方法。
- カルモジュリン依存性キナーゼI(CaMKI)またはプロテインホスファターゼ2B(カルシニューリン)の生物活性をモジュレートする能力について、同定された薬剤を試験することをさらに含む、請求項7または8に記載の方法。
- 細胞または組織におけるカルシウムシグナル伝達をモジュレートする能力について試験すべき薬剤を同定する方法であって:
(a) DARPP-21と薬剤を、DARPP-21と結合させるのに十分な時間にわたり接触させること;
(b) 結合していない薬剤を洗って除去すること;および
(c) DARPP-21に結合した薬剤の存在をアッセイし、ここで該薬剤がDARPP-21と結合するならば、次にカルシウムシグナル伝達をモジュレートする能力について試験すべき薬剤が同定されること、
を含んでなる前記方法。 - 同定された薬剤を、DARPP-21、カルモジュリン依存性キナーゼI(CaMKI)、またはプロテインホスファターゼ2B(カルシニューリン)の生物活性をモジュレートする能力について試験することをさらに含んでなる、請求項11に記載の方法。
- 細胞内カルシウムシグナル伝達をモジュレートする活性について薬剤をスクリーニングする方法であって:
(a) 薬剤の不在下でDARPP-21とカルモジュリンを含んだタンパク質-タンパク質結合アッセイを行うこと;
(b) 薬剤の存在下でDARPP-21とカルモジュリンを含んだタンパク質-タンパク質結合アッセイを行うこと;
(c) ステップ(a)でのDARPP-21とカルモジュリンの結合のレベルをステップ(b)での同レベルと比較するが、ここでその結合レベルの差は、該薬剤が細胞内カルシウムシグナル伝達をモジュレートすることを示すこと、
を含んでなる前記方法。 - 細胞内カルシウムシグナル伝達をモジュレートする活性について薬剤をスクリーニングする方法であって:
(a) 薬剤の存在下でDARPP-21とカルモジュリンを含んだタンパク質-タンパク質結合アッセイを行うこと;
(b) ステップ(a)でのDARPP-21とカルモジュリンの結合のレベルを測定すること;および
(c) (c)におけるそのレベルを、該薬剤の不在下で行った対応する対照のタンパク質-タンパク質結合アッセイでの結合のレベルと比較し、ここでその結合レベルの差は該薬剤が細胞内カルシウムシグナル伝達をモジュレートすることを示すこと、
を含んでなる前記方法。 - 同定された薬剤をカルモジュリン依存性キナーゼI(CaMKI)またはプロテインホスファターゼ2B(カルシニューリン)の生物活性をモジュレートする能力について試験することをさらに含んでなる、請求項13または14に記載の方法。
- DARPP-21との結合がカルモジュリンによって阻害または増強される候補DARPP-21結合タンパク質をスクリーニングする方法であって:
(a) カルモジュリンの不在下でDARPP-21と候補タンパク質を含んだタンパク質-タンパク質結合アッセイを行うこと;
(b) カルモジュリンの存在下でDARPP-21と候補ポリペプチドを含んだタンパク質-タンパク質結合アッセイを行うこと;
(c) ステップ(a)でのDARPP-21と候補ポリペプチドの結合をステップ(b)での結合と比較することによって、カルモジュリンがそのタンパク質-タンパク質結合を阻害するか増強するかを決定すること、
を含んでなる前記方法。 - カルモジュリンとのその結合がDARPP-21によって阻害または増強される候補カルモジュリン結合タンパク質をスクリーニングする方法であって:
(a) DARPP-21の不在下でカルモジュリンと候補タンパク質を含んだタンパク質-タンパク質結合アッセイを行うこと;
(b) DARPP-21の存在下でカルモジュリンと候補ポリペプチドを含んだタンパク質-タンパク質結合アッセイを行うこと;
(c) ステップ(a)でのカルモジュリンと候補ポリペプチドの結合をステップ(b)でのそれらの結合と比較することによって、DARPP-21がタンパク質-タンパク質結合を阻害するか増強するかを決定すること、
を含んでなる前記方法。 - 同定された薬剤を、カルモジュリン依存性キナーゼI(CaMKI)またはプロテインホスファターゼ2B(カルシニューリン)の生物活性をモジュレートする能力について試験することをさらに含む、請求項16または17に記載の方法。
- DARPP-21がアミノ酸55位に負電荷を含む、請求項7〜18のいずれか1項に記載の方法。
- DARPP-21がリン酸化されていない、請求項7〜18のいずれか1項に記載の方法。
- DARPP-21がリン酸化されている、請求項7〜18のいずれか1項に記載の方法。
- タンパク質-タンパク質結合アッセイが、酵母における2-ハイブリッドアッセイまたは大腸菌/BCCP 双方向システムを含む、請求項7〜18のいずれか1項に記載の方法。
- タンパク質-タンパク質結合アッセイが、免疫共沈または固定化タンパク質-タンパク質結合アッセイを含む、請求項7〜18のいずれか1項に記載の方法。
- 請求項7〜18のいずれか1項に記載の方法によって同定される、細胞または組織におけるニューロンのカルシウムシグナル伝達をモジュレートするための組成物。
- 同定された薬剤を、カルモジュリン依存性タンパク質の生物活性をモジュレートする能力についてさらに試験することを含む、請求項7、8、11、13、または14のいずれかに記載の方法。
- カルモジュリン依存性タンパク質が酵素である、請求項25に記載の方法。
- 前記酵素がホスホジエステラーゼ、キナーゼ、ホスファターゼ、またはアデニル酸シクラーゼである、請求項26に記載の方法。
