Abstract
Description

4mLのCH3CN中のジアステレオマースルファミダイト13a(97mg、0.22mmol)の溶液を氷浴中で冷却し、そして、連続して1.1当量のNaIO4(51mg)、触媒量のRuO2・H2O(約1mg)および2.4mLのH2Oで処理した。攪拌の5分後、氷浴を取り除き、その後反応を20分間継続した。反応混合物を10mLのEtOAc中に希釈し、その後10mLの飽和NaHCO3溶液で洗浄した。水層を、2×10mLのEtOAcで抽出し、その後合わせた有機相を10mLのブラインで洗浄し、その後通常のワークアップを行った。粗生成物をシリカゲルクロマトグラフィー(ヘキサン中の30% EtOAc)により精製し、環状スルファミデート14aを、淡黄色固体(90mg、89%)として得た:mp143.5〜145EC(EtOAc.ヘキサン);1H NMR(CDCl3)δ1.29(3H,s),1.51(9H,s),3.77(3H,s),3.80(3H,s),4.16(1H,d,J=8.8),4.73(1H,d,J=8.8),5.98(1H,s),6.82〜6.89(4H,m),7.38〜7.44(4H)。分析(C23H29NO7S)C,H,N。
14aを得るために使用した同じ反応条件を、42mgの13b(0.18mmol)に適用して、14b(42mg、94%)を白色固体として得た:mp54〜55℃(EtOAc/ヘキサン);1H NMR(CDCl3)δ1.50(9H,s),1.52(3H,s),2.93(3H,s),4.22(1H,d,J=8.8),4.88(1H,d,J=8.8)。分析(C9H17NO5S)C,H,N。
CH3CN(4mL)中の環状スルファミデート14a(130mg、0.28mmol)の溶液に、3当量のテトラブチルアンモニウムフッ化物(THF中に1.0M)を添加し、生じた溶液を室温で一晩攪拌した。反応混合物を減圧下で、濃縮し、残留物を5mLの3N HClで、85℃で1時間処理した。冷却後、水溶液を5mLのエーテルで洗浄し、次いで、6N NaOHを用いてpH7にした。減圧下で溶媒を除去し、生じた白色固体を、9:1 CH3OH:Et3Nに溶解した。この溶液に、2当量部分の(Boc)2O(122mg)を添加し、その後、反応混合物を一晩、室温で攪拌した。ワークアップおよび精製を前に記載したように行い、アミノニトリル経路により得た生成物と同じ1H NMRスペクトルを有する生成物5a(25mg、40%)を得た。
14aから[18F]5aおよび14bから[18F]5bを調製するために、同じ条件を使用した。350μLの[18O]H2O中に150〜200mCiの、キャリアを添加していない[18F]HF(20μA、10〜15分ボンバードメント(bombardment)、1.7Ci/nモルの理論上の比活性)を含むウィートンバイアルに、CH3CN中に10mgのK222 Kryptofixおよび1mgのK2CO3を含む溶液を1mL添加し、その後、115ECで、アルゴンガスフロー下で、溶媒を取り除き、その後、さらに1mLのCH3CNを添加し、その後アルゴンフローでエバポレーションを行った。この乾燥を全部で3回繰り返し、残留H2Oを取り除いた。1mLの脱水CH3CN中の環状スルファミデート前駆体14aまたは14bの1〜2mg部分をバイアルに添加し、その後反応混合物を85℃で20分間加熱した。溶媒をアルゴンガスフロー下で115℃で除去し、その後、中間生成物を0.5mLの6N HClで、85℃で10分間処理した。放射標識したアミノ酸の溶液を1〜2mLのH2Oに希釈し、2つのアルミナ N SepPaks(登録商標)および1つのC−18 SepPak(登録商標)を有するシリーズ中の7×120mmのイオン抑制樹脂(Bio Rad AG11A8 50〜100メッシュ)のカラムを通して、H2Oに抽出した。放射性活性を有する溶出画分を、直接げっ歯類の研究に使用した。放射標識された生成物の同一性を、放射分析TLCを用いて可視化された放射性生成物のRf-を、ニンヒドリン色素で可視化した標準19F化合物のRf(Rf=0.6、Alltech 0.25mm RP Chiralplate、20:5:5 CH3CN:H2O:MeOH)と比較することにより確認した。全ての放射性合成において、放射測定のTLC分析に存在する唯一のピークは、5aまたは5bに一致し、生成物の放射性化学純度は、99%を上回る。単離された放射性化学的な収率は、用量キャリブレーターを使用して決定された(Capintec CRC−712M)。
9L神経膠肉腫細胞を最初に、加湿インキュベーター条件下(37℃、5%CO2/95%空気)でDulbecco改変Eagle培地(DMEM)を含むTフラスコに、単層として増殖させた。増殖培地は、10%ウシ胎仔血清および抗生物質(10,000ユニット/ml ペニシリンおよび10mg/mlストレプトマイシン)を補充した。増殖培地は、1週間に3回取り替え、細胞が1週間の時間でコンフルエンシーに到達するように、細胞を継代した。
全ての動物実験を、人道的な条件下で行い、そして、エモリー大学Institutional Animal Use and Care Committee(IUCAC)により承認された。ラット9L神経膠肉腫細胞を、雄性フィッシャーラットの脳に、以前記載されたように、移植した[Shoup,TMら(1999)J.Nucl.Med.40:331〜338]。簡単にいうと、定位ヘッドホルダーに置いた、麻酔したラットに、中線の3mm右側の位置および頭蓋外版に対して5mmの深さで、ブレグマから1mm前方に、4×104個のラット9L神経膠肉腫細胞の懸濁液(mLあたり1×107個)を注射した。腫瘍細胞の逆流を最小限にするために、注射は、2分の期間にわたって行い、針を1分の期間をかけて引き抜いた。傷ついた穴および頭皮の切開を閉じ、その後、手順からの回復後、動物をそれらの元のコロニーに戻した。頭蓋内の腫瘍が発生し、このことにより、腫瘍を有するラットにおいて、体重の減少、無気力および猫背の姿勢を生じ、その動物を、移植後17〜19日で使用した。30匹の腫瘍細胞を移植した動物の内、25匹が、解剖により肉眼で見える腫瘍が発生し、本実験に使用された。
げっ歯類中で、[18F]5aおよび[18F]5bを別個に評価するために、同じ手順を使用した。0.3mLの滅菌水中の約85μCiの[18F]5aまたは[18F]5bの静脈内への注射の後、放射性活性の組織分布を、16匹の健常な雄性フィッシャー344ラット(200〜250g)で決定した。動物には、実験の前に適宜に飼料および水を与えた。0.1mL/100gの1:1 ケタミン(500mg/mL):キシラジン(20mg/mL)溶液の筋肉内への注入により誘導された麻酔の後、放射標識されたアミノ酸をラットに、尾静脈カテーテルにより注入した。4匹のラットの群を、用量の注入の5分後、30分後、60分後および120分後に、屠殺した。動物を解剖し、そして、選択した組織を秤量し、Packard Cobra II 自動γ計測器中で用量標準に従って計測した。未処理の計測により、減少を補正し、勘定は、組織のgあたりの全注入用量の百分率として、標準化した(%ID/g)。各時間点での組織における活性の取り込みの比較は、一元配置ANOVA(GraphPad Prismソフトウェア・パッケージ)を使用して解析した。
