Abstract
Description
本発明は、生物学の分野に関し、さらに具体的には、本発明の主題は、Mycobacterium tuberculosisに起因する感染症を、Mycobacterium africanum、Mycobacterium canetti、Mycobacterium microti、Mycobacterium bovis、Mycobacterium bovis BCGに起因する感染症から識別することができるヌクレオチド配列の同定である。本発明の主題は、これらの配列の発現産物により問題の配列を検出する方法、およびこれらの方法を行うためのキットでもある。最後に、本発明の主題は、新規なワクチンである。
結核の病原菌であるMycobacterium tuberculosisの発見以来1世紀以上の研究がなされているにも拘わらず、この疾患はヒトの死亡率の主要な原因の一つのままである。M.tuberculosisにより年間3百万人が死亡すると予測されており(Snider, 1989 Rev. Inf. Dis.S335)、毎年新たな罹患患者数は増加しており、8.8百万人と推定されている。これらの大半は発展途上国の人々であるが、この疾患は西欧諸国においてホームレス人口の増加、AIDS流行の影響、世界的な人口移動の変化、および旅行様式により新たな重要性を呈しつつある。

予備ハイブリダイゼーションおよびハイブリダイゼーションを、
− 5X SSPE(1X SSPEは3M NaCl,30mMクエン酸三ナトリウムである)、
− 2X Denhardt溶液、
− 0.5%(重量/容量)ドデシル硫酸ナトリウム(SDS)、
− 100μg ml−lサケ精子DNA、
を含む混合物中で65℃で行う。洗浄は、
− 実験室温度(約21〜25℃)で、2X SSPEおよび0.1% SDSの存在下にて10分間2回洗浄、および
− 65℃で1X SSPEおよび0.1% SDSの存在下にて15分間
行う。

をデザイしている。ハイブリダイゼーション実験用の特異的な500pbのプローブを得るために、TbD1に含まれる断片のPCR増幅をプラスミドX229をマトリックスとして用いて実現することができる。TbD1に含まれる約500bpの断片の増幅は、下記のプライマー:

を用いて行うことができる。PCR条件は下記の通りである:95℃で1分間変性した後、35サイクルの増幅[95℃で30秒間、58℃で1分間]、次いで72℃で4分間伸長。
− 配列番号1に含まれ且つ配列番号6のタンパク質mmpL6をコードする、配列番号5の配列のmmpL6遺伝子、
− 配列番号4のTbD1に含まれ且つ配列番号8の配列のmmpL6タンパク質の切り詰め形態をコードする配列番号7の配列のmmpL6遺伝子の切り詰め形態、
− 配列番号1に含まれ且つ配列番号10の配列のmmpS6遺伝子をコードする、配列番号9のmmpS6遺伝子、

生物学的試料中のMycobacterium africanum、Mycobacterium canettii、Mycobacterium microti、Mycobacterium bovis、Mycobacterium bovis BCG

1.1. 細菌株:
100個の結核菌群株は、30カ国で単離された46種のM. tuberculosis株、14種のM. africanum株、5カ国に由来する28種のM. bovis株、2種のM. bovis BCGワクチン株(PasteurおよびJapan)、5種のM. microti株、および5種のM. canettii株から構成された。これらの株はヒトおよび動物供給源から単離し、多中心研究(8)で用いた60株を包含する広い多様性を示すように選択した。M. africanum株はWadsworth Center, New York State Department of Health, Albany, New Yorkのコレクションから得て、M. bovis単離物の大部分はUniversity of Zaragoza, スペインのコレクションから入手した。4種類のM. canettii株は、Institut Pasteur, Paris, フランスのカルチャーコレクションからのものである。株は、リファレンスタイピング法、すなわちIS6110制限断片長多型(RFLP)タイピングおよびスポリゴタイピングによって徹底的に特性決定した。M. tuberculosis H37Rv、M. tuberculosis H37Ra、M. tuberculosis CDC1551、M. bovis AF2122/97、M. microti OV254、およびM. canettii CIPT 140010059が、リファレンス株として包含された。DNAは、上記の方法で調製した(10)。
M. tuberculosisとM. bovisのゲノムの予備的比較には、ウェブサイトhttp://genolist.pasteur.fr/TubercuList/およびhttp://www.sanger.ac.uk/Projects/M_bovis/、並びに社内データベースを用いた。プライマーデザインには、RDおよびRvD領域内部またはこれに隣接する配列を、同じウェブサイトから得た。プライマーは、リファレンス株における約500塩基対断片を増幅するプライマー3ウェブサイトhttp://www-genome.wi.mit.edu/cgi-bin/primer/primer3_www.cgiを用いてデザインした(表1)。
反応は96穴プレートで行い、反応当たり1.25μlの10xPCR緩衝液(600mM Tris HCl pH8.8、20mM MgCl2、170mM (NH4)2SO4、100mM β−メルカプトエタノール)、1.25μlの20mMヌクレオチドミックス、50nMのそれぞれのプライマー、1〜10ngの鋳型DNA、10%DMSO、0.2単位のTaqポリメラーゼ(Gibco-BRL)を含み、滅菌蒸留水を加えて12.5μlとした。熱循環を、PTC−100増幅装置(MJ Inc.)で、95℃で90秒間の初期変性段階の後、95℃で30秒間、58度で1分間および72℃で4分間の35サイクルを行った。
PCR産物は、上記のように、表1に挙げたプライマーを用いて得た。
プライマー除去については、6μlのPCR産物をシュリンプアルカリホスファターゼ(USB) 1単位、エキソヌクレアーゼI(USB)10単位、および5x緩衝液(200mM Tris HCl pH8.8、5mM MgCl2)2μlと共に37℃で15分間インキュベーションした後、80℃で15分間インキュベーションした。この反応混合物にBig Dyeシークエンシングミックス(Applied Biosystems)2μl、プライマー2μl(2μM)、および5x緩衝液(5mM MgCl2、200mM Tris HCl pH8.8)3μlを加え、35サイクル(96℃で30秒間;56℃で15秒間;60℃で4分間)をサーモサイクラー(MJ- research Inc., Watertown, MA)で行った。DNAを76%エタノール80μlを用いて沈澱させ、70%エタノールで濯いで、乾燥した。反応物をホルムアミド/EDTA緩衝液2μlに溶解し、変性して、48cmの4%ポリアクリルアミドゲルに装填し、電気泳動を377自動化DNAシークエンサー(Applied Biosystems)で10〜12時間行った。あるいは、反応物を0.3mM EDTA緩衝液に溶解し、3700 DNAシークエンサー(Applied Biosystems)で自動化シークエンシングを行った。反応物は、一般に500〜700bpの明確な配列を生じた。
遺伝子mmpS6およびmmpL6を含む祖先M. tuberculosis株No.74(参考文献8)からのTbD1領域の配列を、EMBLに受領番号AJ426486で寄託した。BCGにおいてRD4、RD7、RD8、RD9およびRD10に接している配列は、それぞれ受領番号AJ003103、AJ007301、AJ131210、Y18604、およびAJ132559で入手可能である。
結核菌のゲノムにおける挿入−欠失事象から生じる20可変領域の分布を、全部で100株のMycobacterium tuberculosis、M. africanum、M. canettii、M. microti、およびM. bovisで評価した。この方法は、これらの多型の大部分は結核菌群の異なる株では独立して起きないが、共通の祖先株における古い不可逆的遺伝事象から生じる。M. tuberculosisの特異的欠失(TbD1)の有無に基づいて、M. tuberculosis株を祖先および「近代」株に分類し、後者がBeijing、HaarlemおよびAfrican M. tuberculosis群のような主要な流行病の代表的なものを構成することができる。更に、FD9によって反映されるDNAの連続的喪失とそれに続く他の欠失を同定して、進化系統をTbD1が起こる前に現在のM. tuberculosis株の祖先から分岐したM. africanum、M. microtiおよびM. bovisによって表わした。これらの知見は、ヒト結核の病原体であるM. tuberculosisがウシの疾患の病原体であるM. bovisから進化したというしばしば提出される仮説と矛盾する。これらの欠失領域を全く欠いていないM. canettiiおよび先祖のM. tuberculosis株は、M. africanum->M. bovis系統がM. tuberculosis系統から分離する前に存在した結核菌の直接の子孫であると思われる。これは、結核菌の共通の先祖がM. tuberculosisまたはM. canettiiに似ており、既に十分にヒトの病原体となっていたことを示唆している。
46のM. tuberculosis、14のM. africanum、5のM. canettii、5のM. microti、28のM. bovisおよび2のBCG株中の20の可変領域(表1)についてのPCRスクリーニング分析では、既知のRDsおよびRvDsに対して内部のオリゴヌクレオチド、並びにこれらの領域に隣接するオリゴヌクレオチド(表1)を用いた。この方法は、内部プライマーを用いて得たPCRアンプリコンは隣接プライマー対を有する適当な大きさのアンプリコンの非存在と相関し、逆もまた同じであるので、上部で信頼性の高い大きなデータセットを生じた。
欠失分析による46のM. tuberculosis株の検討によって、ほとんどのRD領域が試験を行った総てのM. tuberculosis株に存在することが明らかになった(表2)。M. tuberculosis H37Rvの2種類のプロファージphiRv1およびphiRv2に対応する領域RD3およびRD11(4)、挿入配列IS1532を含むRD6、およびIS6110のコピーが隣接しているRD5(5)のみが、幾つかの株に含まれていなかった。M. tuberculosis株はRD1、RD2、RD4、RD7、RD8、RD9、RD10、RD12、RD13およびRD14に関して高度に保存され且つこれらのRDはM. tuberculosis株をそれらの地理学的起源およびそれらのタイピング特性から独立して結核菌群のある種の他の菌から区別できる領域を表すことを示唆するので、これは重要な観察である。
M. canettiiは、通常はアフリカ出身またはアフリカに関連した患者から単離されるM. tuberculosisの極めてまれなスムーズ変異体である。M. canettii株はMycobacterium群の他の菌と同一の16S rRNA配列を共有するが、ある種のハウス・キーピング遺伝子における多型、IS1081コピー数、コロニー形態、および細胞壁の脂質含量など多くの点で異なっている(15,16)。従って、本発明者らはM. canettiiにおいて、プロファージ(phiRv1,phiRv2)を除く総てのRD、RvD、およびTbD1領域が存在することを見出したことに驚愕した。対照的に、本発明者らは、RD12と部分的に重複した5個総てのM. canettii株から本質的に存在しない領域(RDcan)を同定した(図4)。
本明細書で検討したM. africanumと呼ばれる単離物は西および東アフリカに起源がある。11株はシエラレオーネ、ナイジェリアおよびギニアで単離され、2株はウガンダで単離された。1株は、オランダに由来する。
M. microti株は、1930年代にハタネズミから単離され(17)、更に最近では免疫抑制患者から単離された(18)。これらの株は同一の特徴的なスポリゴタイプを有することを特徴とするが、それらのIS6110プロフィールは異なっている。ハタネズミおよびヒト単離物はいずれも領域RD7、RD8、RD9およびRD10、並びにM. microtiから特異的に欠失した領域(RDmic)を欠いていた。RDmicは、M. microti Bacterial Artificial Chromosome(BAC)ライブラリー由来のクローンを用いるM. microtiの詳細な比較ゲノム学的検討によって明らかにされた(19)。RDmicは、部分的にBCG由来のRD1と重複している(データは示さず)。更に、ハタネズミ単離物はRD5領域の一部を欠いているが、この領域はヒト単離物には存在する。M. microtiにおけるRD5の接合領域はBCGのものとは異なっているので(データは示さず)、RD5は進化マーカーとして用いなかった。
M. bovisは、ヒトを含む多くの哺乳動物種に感染する極めて大きな宿主スペクトルを有する。RDおよびRvD領域についてスクリーニングしたM. bovis株のコレクションは、3個のヤギ単離物、3個のアザラシ単離物、2個のオリックス単離物、および一層多数のIS6110のコピーを提供するヒト由来の2個のM. bovis株の他に、2個のBCG株と一般的にウシ由来のIS6110の1または2コピーを特徴とする18個の「古典的」(classical) M. bovis株からなっていた。
3.1. ヒト結核の起源
多年の間、ヒト結核は動物の病原体をヒト宿主に適合させることによってウシ疾患から進化したと考えられていた(3)。この仮説は、M. tuberculosisの特性がほぼ全くヒト病原体のみであり、一方M. bovisはずっと広い宿主範囲を有することに基づいている。しかしながら、この研究からの結果はM. tuberculosisと比較してM. bovisが多くの欠失を行ってきたことを明白に示している。これは、ウシから単離された「古典的」M. bovis株であるM. bovis AF2122/97のほぼ完全なゲノム配列の予備分析によって確かめられ、M. bovisに特異的に閉じ込められた新たな遺伝子クラスターを示さなかった。これは、M. bovisのゲノムがM. tuberculosisのゲノムより小さいことを示している(24)。M. bovisは、M. tuberculosis単離物の先祖から分岐したM. africanum(RD9)、M. microti(RD7、RD8、RD9、RD10)およびM. bovis(RD4、RD5、RD7、RD8、RD9、RD10、RD12、RD13)(25)によって表される別系統の最後の菌であることが妥当と思われる。DNAの連続的喪失は、クローン伸長およびある種の新たな宿主における更に順当な病原体の出現の一員となる可能性がある。
ハウスキーピング遺伝子で配列が高度に保存されるので、Sreevatsan et al.は、以前に結核菌が15,000〜20,000年以前に主要な隘路にであったと仮定した(2)。試験を行った総てのTbD1欠失株でのTbD1接合配列の保存が単一クローンに由来することを示唆しているので、TbD1欠失は、今日の結核症例の大半を説明する「近代的」M. tuberculosis株は確実にこのような隘路を経験した後に世界中に広がったことを完全に示している。
本発明者らの研究は、広汎な結核菌の可変領域の多様性および保存の全体像を提供する。様々な宿主および国由来の100株の欠失分析により、ほとんどがアフリカ起源の幾つかの進化的に「古い」(old) M. canettii、M. tuberculosisおよびM. africanum株、並びに「近代的」M. tuberculosis株であって、北京、ハールレムおよびアフリカのような主要な流行クラスターからの典型的なものなどを同定した。特異的突然変異の分子タイピングおよび分析と共同した欠失分析の使用は、結核菌の進化の研究および進化マーカーの同定のための極めて強力な方法であることが示された。更に実際的な観点では、これらの領域、主としてRD9およびTbD1であるが、RD1、RD2、RD4、RD7、RD8、RD10、RD12、およびRD13も、M. tuberculosis群の菌を迅速且つ明確に同定するための強力な診断手段の発展のための極めて興味深い候補である(32)。この発生学的な区別の方法を用いて、M. tuberculosis群の混乱することが多い伝統的な分類を厳密に確定した亜種とすることができる。
M. tuberculosis群の菌は異常なほど高い保存度を共有しており、市販の核酸プローブおよび増幅分析ではこれらの生物を区別することはできない。その上、従来の同定法は曖昧で、扱い難く、生物の増殖が遅いため時間がかかることが多い。

生物学的試料中のMycobacterium africanum、Mycobacterium canetti、Mycobacterium microti、Mycobacterium bovis、Mycobacterium bovis BCGと
を区別して検出し同定する方法であって、
a) 分析を行う生物学的試料を請求項25または26に記載の少なくとも1対のプライマーと接触させ、この試料に含まれるDNAを、適当な場合には、予めハイブリダイゼーションに利用できるようにし、
b) マイコバクテリウムのDNAを増幅し、
c) DNA断片の増幅を可視化する
工程を含んでなる、方法。 - 遺伝子katGのコドン463に配列CTGを有し且つゲノムにはIS6110配列を全く挿入していないかまたはごく僅かしか挿入していないMycobacterium tuberculosis株を除くMycobacterium tuberculosisと、
生物学的試料中のMycobacterium africanum、Mycobacterium canetti、Mycobacterium microti、Mycobacterium bovis、Mycobacterium bovis BCGと
を区別して検出し同定するためのキットであって、下記の因子:
a) 請求項25または26に記載の少なくとも1対のプライマー、
b) DNA増幅反応を行うのに必要な試薬、
c) 必要に応じて、増幅した断片の配列および/または大きさを明らかにしまたは比較することができる必要な成分
を含んでなる、キット。 - Mycobacterium tuberculosis、Mycobacterium africanum、Mycobacterium canettii、Mycobacterium microti、Mycobacterium bovisまたはMycobacterium bovis BCGからのDNA配列を増幅するための、請求項25または26に記載の少なくとも1対のプライマーの使用。
- Mycobacterium tuberculosis、Mycobacterium africanum、Mycobacterium canettii、Mycobacterium microti、Mycobacterium bovisまたはMycobacterium bovis BCGからのDNA配列を増幅するための、請求項2、3、4、5、6、7、8、9、10、11、12、13または14のいずれか一項に記載の少なくとも1対のプライマーまたは少なくとも1個の核酸断片の使用。
- 請求項6、7または8のいずれか一項に記載の核酸断片の全部または一部の発現産物。
- 遺伝子katGのコドン463に配列CTGを有し且つゲノムにはIS6110配列を全く挿入していないかまたはごく僅かしか挿入していないMycobacterium tuberculosis株を除くMycobacterium tuberculosisに対して特異的な抗体と、
生物学的試料中のMycobacterium africanum、Mycobacterium canetti、Mycobacterium microti、Mycobacterium bovis、Mycobacterium bovis BCGに対して特異的な抗体と
をイン・ビトロで区別して検出する方法であって、
a) 生物学的試料を請求項31に記載の少なくとも1個の産物と接触させ、
b) 形成した抗原−抗体複合体を検出する
工程を含んでなる、方法。 - Mycobacterium bovis BCGを用いる予防接種、M. bovis、M. caraettii、M. microti、M. africanum、または遺伝子katGのコドン463に配列CTGを有し且つゲノムにはIS6110配列を全く挿入していないかまたはごく僅かしか挿入していないM. tuberculosis株による感染症と、哺乳類の遺伝子katGのコドン463に配列CTGを有し且つゲノムにはIS6110配列を全く挿入していないかまたはごく僅かしか挿入していないMycobacterium tuberculosis株を除くMycobacterium tuberculosisによる感染症とをイン・ビトロで区別して検出する方法であって、
a) 細胞、より詳しくは上記哺乳類の免疫系の細胞、更に詳しくはT細胞を調製し、
b) 工程a)の生物学的試料を、請求項31に記載の少なくとも1個の産物と共にインキュベーションし、
c) 上記産物に対する哺乳類の予備増感を示す細胞反応、特に細胞増殖および/またはγ−インターフェロンのようなタンパク質の合成を検出する
工程を含んでなる、方法。 - Mycobacterium bovis BCGを用いる予防接種、M. bovis、M. caraettii、M. microti、M. africanumと、哺乳類における遺伝子katGのコドン463に配列CTGを有し且つゲノムにはIS6110配列を全く挿入していないかまたはごく僅かしか挿入していないM. tuberculosis株による感染症と、哺乳類の遺伝子katGのコドン463に配列CTGを有し且つゲノムにはIS6110配列を全く挿入していないかまたはごく僅かしか挿入していない株を除くM. tuberculosisによる感染症とをイン・ビトロで区別して診断するためのキットであって、
a) 請求項31に記載の産物、
b) 適当な場合には、免疫反応に適する培地を構成するための試薬、
c) 免疫反応によって産生した抗原−抗体複合体を検出することができる試薬、
d) 適当な場合には、上記産物によって認識された抗体を含まない参照用生物学的試料(ネガティブコントロール)、
e) 適当な場合には、上記産物によって認識された抗体の予定量を含む参照用生物学的試料(ポジティブコントロール)
を含んでなる、キット。 - 請求項31に記載の産物を特異的に認識することができる、モノクローナルまたはポリクローナル抗体、そのキメラ断片またはキメラ抗体。
- 遺伝子katGのコドン463に配列CTGを有し且つゲノムにはIS6110配列を全く挿入していないかまたはごく僅かしか挿入していない株を除くMycobacterium tuberculosisの抗原と、生物学的試料中のMycobacterium africanum、Mycobacterium canetti、Mycobacterium microti、Mycobacterium bovis、Mycobacterium bovis BCGまたは遺伝子katGのコドン463に配列CTGを有し且つゲノムにはIS6110配列を全く挿入していないかまたはごく僅かしか挿入していないMycobacterium tuberculosisの抗原の存在を区別して検出する方法であって、
a) 生物学的試料を請求項35に記載の抗体と接触させ、
b) 形成した抗原−抗体複合体を検出する
工程を含んでなる、方法。 - 遺伝子katGのコドン463に配列CTGを有し且つゲノムにはIS6110配列を全く挿入していないかまたはごく僅かしか挿入していない株を除くMycobacterium tuberculosisの抗原と、生物学的試料中のMycobacterium africanum、Mycobacterium canetti、Mycobacterium microti、Mycobacterium bovis、Mycobacterium bovis BCGまたは遺伝子katGのコドン463に配列CTGを有し且つゲノムにはIS6110配列を全く挿入していないかまたはごく僅かしか挿入していないMycobacterium tuberculosisの抗原の存在を区別して検出するためのキットであって、
a) 請求項35に記載の抗体、
b) 免疫反応に適する培地を構成するための試薬、
c) 免疫反応によって産生した抗原−抗体複合体を検出することができる試薬
の工程を含んでなる、キット。 - 少なくとも1個の請求項31に記載の産物を含んでなる、免疫原性組成物。
- 少なくとも1個の請求項31に記載の産物を、薬学上適合性のビヒクル、および適当な場合には、1種類以上の適当な免疫アジュバントと組み合わせて含んでなる、ワクチン。
- 生物学的試料中の遺伝子katGのコドン463に配列CTGを有し且つゲノムにはIS6110配列を全く挿入していないかまたはごく僅かしか挿入していないMycobacterium tuberculosis株を除くMycobacterium tuberculosisを検出し同定するイン・ビトロ法であって、
a) 分析を行う生物学的試料からDNAを単離しまたは生物学的試料のRNAからcDNAを産生し、
b) 上記生物学的試料に含まれるマイコバクテリウムの核酸配列を検出し、
c) 請求項6、7または8のいずれか一項に記載の核酸断片の存在または非存在について分析を行う
工程を含んでなる、イン・ビトロ法。 - 生物学的試料中の遺伝子katGのコドン463に配列CTGを有し且つゲノムにはIS6110配列を全く挿入していないかまたはごく僅かしか挿入していないMycobacterium tuberculosis株を除くMycobacterium tuberculosisを検出し同定するイン・ビトロ法であって、
a) 分析を行う生物学的試料を、請求項1〜14、17または18のいずれか一項に記載の核酸から選択される、更に好ましくは配列番号13、配列番号14、配列番号15、配列番号16、配列番号17、配列番号18を含んでなる群から選択されるプライマーから選択される、少なくとも1対のプライマーと接触させ、試料に含まれるDNAを、適当な場合には、予めハイブリダイゼーションを行うことができるようにしておき、
b) マイコバクテリウムのDNAを増幅し、
c) DNA断片の増幅を可視化する
工程を含んでなる、イン・ビトロ法。 - 生物学的試料において、遺伝子katGのコドン463に配列CTGを有し且つゲノムにはIS6110配列を全く挿入していないかまたはごく僅かしか挿入していないMycobacterium tuberculosis株を除くMycobacterium tuberculosisを検出し同定するためのキットであって、下記の因子:
a) 請求項1〜14、17または18のいずれか一項に記載の核酸から選択される、更に好ましくは配列番号13、配列番号14、配列番号15、配列番号16、配列番号17、配列番号18を含んでなる群から選択される、少なくとも1対のプライマー、
b) DNA増幅反応を行うのに要する試薬、
c) 必要に応じて、増幅した断片の配列および/または大きさを明らかにしまたは比較することができる必要な成分
を含んでなる、キット。 - 生物学的試料において、遺伝子katGのコドン463に配列CTGを有し且つゲノムにはIS6110配列を全く挿入していないかまたはごく僅かしか挿入していないMycobacterium tuberculosis株を除くMycobacterium tuberculosisに対して特異的な抗体をイン・ビトロで検出する方法であって、
a) 生物学的試料を請求項31に記載の少なくとも1個の産物と接触させ、
b) 形成した抗原−抗体複合体を検出する
工程を含んでなる、方法。 - Mycobacterium群のMycobacterium株を識別するための遺伝子マーカーとしてのTbD1の使用。
- Mycobacterium群のMycobacterium株を識別するための遺伝子マーカーとしてのmmpL6551多型の使用。
- RD1、RD2、RD3、RD4、RD5、RD6、RD7、RD8、RD9、RD10、RD11、RD13、RD14、RvD1、RvD2、RvD3、RvD4、RvD5、katG463、gyrA95、oxyR'285、pncA57、mmpL6551、Mycobacterium株をMycobacterium群と区別するためのM. canettiiの特異的挿入因子から選択される少なくとも1個の遺伝子マーカーに関連した請求項44に記載の遺伝子マーカーの使用。
- 生物学的試料中のMycobacteria群からマイコバクテリアを検出して同定するためのイン・ビトロ法であって、
c) 請求項6、7または8のいずれか一項に記載の配列の核酸断片の存在または非存在について分析し、
d) RD1、RD2、RD3、RD4、RD5、RD6、RD7、RD8、RD9、RD10、RD11、RD13、RD14、RvD1、RvD2、RvD3、RvD4、RvD5、katG463、gyrA95、oxyR'285、pncA57、mmpL6551、M. canettiiの特異的挿入因子から選択される少なくとも1個の追加遺伝子マーカーを分析する
工程を含んでなる、イン・ビトロ法。 - 2個の追加マーカー、好ましくはRD4およびRD9を用いる、請求項47に記載のイン・ビトロ法。
- 3個の追加マーカー、好ましくはRD4、RD9およびRD12を用いる、請求項47に記載のイン・ビトロ法。
- 分析を配列ハイブリダイゼーション、核酸増幅、抗原−抗体複合体から選択される手法によって行う、請求項47に記載の方法。
- 生物学的試料中のMycobacteriun群からマイコバクテリアを検出し同定するためのキットであって、下記の因子:
a) 請求項1〜14、17または18のいずれか一項に記載の核酸から選択される、更に好ましくは配列番号13、配列番号14、配列番号15、配列番号16、配列番号17、配列番号18を含んでなる群から選択されるプライマーから選択される少なくとも1対のプライマー、
b) RD1、RD2、RD3、RD4、RD5、RD6、RD7、RD8、RD9、RD10、RD11、RD13、RD14、RvD1、RvD2、RvD3、RvD4、RvD5、katG463、gyrA95、oxyR'285、pncA57、mmpL6551、M. canettiiの特異的挿入因子から選択される遺伝子マーカーに特異的な少なくとも1対のプライマー、
c) DNA増幅反応を行うのに要する試薬、
d) 必要に応じて、増幅した断片の配列および/または大きさを明らかにしまたは比較することができる必要な成分
を含んでなる、キット。 - 下記の因子を含んでなる、請求項51に記載のキット:
a) 請求項1〜14、17または18のいずれか一項に記載の核酸から選択される、更に好ましくは配列番号13、配列番号14、配列番号15、配列番号16、配列番号17、配列番号18を含んでなる群から選択されるプライマーから選択される少なくとも1対のプライマー、
b) 遺伝子マーカーRD4に特異的な1対のプライマー、
c) 遺伝子マーカーRD9に特異的な1対のプライマー、
d) DNA増幅反応を行うのに要する試薬、
e) 必要に応じて、増幅した断片の配列および/または大きさを明らかにしまたは比較することができる必要な成分。 - 配列番号22の配列のポリペプチドを含んでなる、免疫原性組成物。
- 配列番号22の配列のポリペプチドを、薬学上適合性のビヒクルと、適当な場合には、1種類以上の適当な免疫アジュバントと組み合わせて含んでなる、ワクチン。
- Mycobacterium群のMycobacterium株を識別するための、RD1、RD2、RD3、RD4、RD5、RD6、RD7、RD8、RD9、RD10、RD11、RD13、RD14、RvD1、RvD2、RvD3、RvD4、RvD5、TbD1、katG463、gyrA95、oxyR'285、pncA57、M. canettiiの特異的挿入因子から選択される少なくとも1個の遺伝子マーカーに関連した、請求項45に記載の遺伝子マーカーの使用。
- M. canettiiの株に特異的に存在し且つMycobacterium群の他の総ての構成員には存在せず、配列番号19の399位〜2378位の配列を有する、核酸。
- Mycobacterium群のMycobacterium株を識別するための遺伝子マーカーとしての、請求項53に記載の核酸の使用。
- ストリンジェント条件下で、RD1、RD4、RD9およびTbD1遺伝子マーカーの少なくとも8〜20個のヌクレオチドとハイブリダイゼーションすることができる少なくともポリヌクレオチド配列を含んでなる、マイコバクテリア感染症を同定するための試薬。
- 同一免疫原性分子またはその断片に対して生じた抗体または免疫血清と反応することができるRD1、RD4、RD9およびTbD1遺伝子マーカーのそれぞれによってコードされる少なくとも1個のポリペプチドを含んでなる、マイコバクテリア感染症を同定するための試薬。
