Abstract
Description
本発明は、光熱、光化学物質及び光変調(photomodulatory)の新規の組み合わせを用いた、或いは、発色団又は他の光活性型発色団(chromophore)、天然由来薬品又は美容食品を、毛包、毛球、毛髪膨張部(hair bulge)、毛髪幹細胞(hair stem cell)又は周囲の組織に適用し、且つ、電磁放射線を曝露することによる、毛髪成長を刺激するためのシステム及び方法に関する。
ヒトの皮膚において、毛髪の成長を減弱し且つ消失させる試みに関して種々の公知の技術が存在する。局所的なミノキシジル(minoxidil)や市販製品である「ロガイン(Rogaine)」などの数少ないこれら公知の技術は、科学的に証明されており、効果を有する物質として広く受け入れられている。しかしながら、これらの効力は多いに変化する。
本発明は、毛髪成長を刺激する方法に関し、この中で、毛髪成長構造は、390nm〜1600nmの範囲の主放射波長を有する電磁放射線源に曝露される。定義すると、この主放射波長は、電磁放射線源により放射される初期波長であって、つまり、他の波長よりも大きな強度の波長が放射される。毛髪成長構造の光刺激(photostimulating)は、臨床的に効果的な期間で、且つ、臨床的に効果的な光強度にて電磁放射線源に、毛髪成長構造を曝露することを保持することにより実行される。臨床的に効果的な期間及び強度は、さらに、本発明の記述及び例において詳細に述べられ、且つ、これらには、単一の電磁放射線源に由来する単一パルス、単一の電磁放射線源に由来する多重パルス、多重的な電磁放射線源に由来する多重パルス、多重的な電子放射線源に由来する単一パルス、多重電磁放射線源に由来する同時パルスや、これらの組み合わせを含んでいる。
本発明の好適実施例に関する詳細な記載は、添付した図面に関する参照文にて行われる。
以下の詳細な記載は、本発明を実行する最良に意図された様式である。この記述は、限定的な意味合いで行われているわけではなく、本発明の一般的原則を示す目的にのみ行われている。本発明の範囲は、添付した請求項により最良に規定されている。
エネルギー密度−−このエネルギー密度は、放射中送達されるエネルギー量に対応し、且つ、エネルギー強度及び光強度として参照されもする。最適な「投与量」は、パルス保持時間及び波長により影響を受け、従って、これらは、相互作用され、且つ、パルスの保持時間は、非常に重要であり、−一般に高エネルギーは、阻害を生み出し、且つ、低いエネルギーは刺激を生み出す。
、酸素発生剤、ポリミキシン、プロシアニジンB2、プロシアニンジンC1、藻、酵母抽出物、銅ペプチド、オクチルブチエート、唐辛子、チョウセンニンジン、ニアシナミド、醤油、ソイイソフラボン、甘草並びにゲネステインである。
毛髪成長刺激
はげ頭の3人の男性は、本発明の非研磨方法に従った処置を受ける前及び後の毛髪成長の刺激に関して試験された。毛髪のカウントは、訓練を受けた医療従事者による主観的な表毛を利用してその頭皮にて行われた。LED処置は、患者の皮膚の標的領域を、250m秒のパルス幅、90パルスにて250m秒のパルス間隔のLED光に指向することを含む。12週間に渡り、顔全体に、590nmの多重色調のLEDにて、1.05〜2.05μWの範囲の強度にて8回処置を行った。±5〜15nmのバンド幅を有するLEDは、575〜605nmの範囲の波長の光を発生する。さらに、この処置は、皮膚の温度を、熱的障害の閾値以下に保持する。毛髪計測における平均的な向上性を、表1に示した。
毛髪成長刺激−連続波処置
前頭部の毛髪が消失した一人の女性に関して、上述の例2の工程に従って毛髪成長刺激を検討した。専門的な採点者による計測は、全体で200秒で、1.05〜2.05μWの強度で、単一の連続波パルスにて、590nmの多重色調のLEDにて処置する前後において、患者の頭皮にて行われた。12週間に渡り、等間隔にて、患者の前頭部及び後頭部にて、8回処理を行った。
毛髪成長刺激に関する非研磨毛髪治療
パルス処理
ナローバンドで、1.05〜2.05μWのエネルギー出力、20m秒のパルス保持時間(各パルスの長さ)、パルス間隔(各パルスの時間)が100m秒で、多重色調の590nmのLEDにて、180パルスで、ヒトの皮膚を曝露した。この処置は、重篤なパターンにて毛髪が消失している6人の男性グループの顔全体に、12週間に渡って8回繰り返された。処置される前と後の皮膚に関する写真を観察して盲検の専門的な採点者のチームにより測定された毛髪成長量は、表4に示した。
毛髪成長刺激に関する非研磨毛髪治療
連続波処理
1.0〜2.0μWのエネルギー出力で、ナローバンドで、多重色調の590nmのLEDの200秒のマイクロ波にヒトの皮膚を曝露した。この処置は、はげ頭の男性被検者の頭皮全体に、12週間にわたり8回繰り返された。処置される前と後の皮膚に関する写真を観察して盲検の専門的な採点者のチームにより測定された毛髪成長量は、表4に示した。

パルスレーザーダイオード
また、本発明の使用に適しているのは、レーザーダイオードである。典型的なパルス保持時間は、パルス処理に関しては約100m秒から1秒であり、連続波処置に関しては、約1〜30秒である。このレーザーダイオードに関する適した制御出力は、約10mW〜約1Wの範囲を含み、好適なのは、200mW〜800mWである。市販され利用可能な400〜1000nmの波長を有するレーザーダイオードを用いてもよい。この例に関して、ヒトの頭皮は、2.0μWのエネルギー出力で、810nmのレーザーダイオードにて90パルスを曝露される。100m秒のパルス間隔を使用した。この処置は、はげ頭の3人の男性の全頭皮に対して12週間にわたって6回繰り返された。毛髪成長量は、表6に示した。
毛髪成長刺激−パルス処置
本発明の非研磨LILT(「低強度光治療」)に指向された患者の写真の前後を観察する盲検の専門的な採点者のチームは、こめかみにおける前面の毛髪線の退行の全体的な向上を評価した。
例9では、10mW/cm2の出力で、940nmのダイオードレーザーにて、対象に対して、50m秒のパルス、250m秒のパルス間隔にて曝露することを除いて、例8と同一の条件にて行われた。12週間に渡って6回処理して、同様の結果を得た。その機構は、非サーマルフォトアクティベーションである。
例10は、対象に対して、2600nW/cm2、10cmのビーム径で、810nmのダイオードレーザーを、100m秒のパルス間隔で、60、100m秒のパルスを曝露することを除いて、例9と同一の条件にて行われた。12週間に渡って6回処理することにより同様の結果を得た。その作用機構は、非サーマルフォトアクティベーションである。
例11は、2mW/cm2、940nmのダイオードレーザーを対象に対して、100秒、連続波にて曝露することを除いては、例10と同一の条件で行われた。12週間に渡って4回処理することで同様の結果を得た。非サーマルフォトアクティベーション方法を用いた。
例12は、2.5J/cm2の出力、595nmの閃光灯パルスダイレーザーを、20m秒パルスにて、4週間に渡って等間隔にて対象に曝露することを除いて、例11と同一の条件にて行われた。16週間に渡って4回処理することで、同様の結果を得た。非研磨フォトサーマル方法を用いた。
例13は、例12と同一の条件にて、毛髪成長再生に関して行われた。6.0J/cm2、595nmの閃光灯パルスダイレーザーを、単一の40m秒のパルスにて、4週間等間隔にて対象に曝露した。20週間に渡った5回の処置を行った。毛髪密度は、42%増加し、実際の毛髪カウントは、18%増加した。その機構は、サーマル非研磨方法である。
例14は、毛髪成長刺激に関して、例13と同一の条件にて行われた。150mW/cm2、10cmのビーム径の532nmのNd:YAGレーザーを、単一の最小限に重なり合った30m秒のパルスにて、4週間に渡って等間隔に患者に曝露した。20週間にわたって5回処理した。毛髪カウントは、28%増加した。この方法は、サーマル非研磨技術である。
例21は、毛髪成長刺激に関して、5人のはげ頭の男性対象に対するのと同一の条件にて行われた。590nmのLEDにて、50m秒のパルス、150m秒のオフタイム、90パルスを用いた。1週間間隔で8回処理し、最終的な評価は12週間目に行った。毛髪成長刺激に加えて、例10と同様の複数の他の有意な変化を示し、これには、灰色の毛髪の色調の明らかな「回復」及び毛根が明らかに太くなることを含んでいる。
例16は、毛髪成長刺激に関して同一の条件にて行われた。対象は、頭皮の病気を持たない、20〜40歳の、毛髪欠損のパターンを有する男性である。対象に、2.2mW/cm2の出力、200m秒のパルス、200m秒のオフ時間、50個のパルスにて、644nmのLEDを曝露した。24週間にわたって6回処理した。毛髪視聴の外観における増加は、22%であった。
例17は、頭皮に斑点状の円形脱毛を有する女性対象にて行われた。150mW/cm2の出力、10cmのビーム径の940nmのダイオードレーザーは、患者の影響を受ける領域における皮膚を、4分間の連続光にて曝露した。処置は、18週間に渡って3週間隔で行った。この円形脱毛領域は、26%減少し、スポット状の毛髪成長が、残存する脱毛部分にて観察された。
例18は、毛髪再成長を刺激する目的にて、化学療法による急性の毛髪脱落に対して行われた。1.5mW/cm2、100m秒のオン時間、100m秒のオフ時間の60個のパルスの632nmのLEDアレイを、7×10インチの方形領域の皮膚に曝露した。処置は、毛髪成長の回復が達成されるまで、2週間毎に繰り返した。回復時間は、毛髪脱落の重篤度及び報告された化学療法のアイライニング(eyelining)と同様に、その他の不確定のファクターに依存する。
完全なヒトの毛嚢の単一層を有する細胞培養のシリーズは、595nmのパルスダイレーザー及び590nmのLEDにて実行された処置の有効性の差を示すべく、比較実験にて検討された。このLEDは、2mW/cm2のエネルギー強度、100m秒間隔の50m秒のパルスであった。非サーマルフォトモデュレーション処置は、50パルスの上述のLEDを使用した。この595nmのパルスダイレーザーは、フォトサーマル処置に関して、2.5J/cm2のエネルギー強度の単一パルス及び0.5m秒のパルス長さを用いた。処置後5日間の毛根成長率の解析は、コントロールに対して有意な変化を示さなかった。毛嚢に処理されたフォトサーマルダイレーザーは、コントロールに相対してもウコン成長率において12%の減少を示した。本発明のフォトサーマルフォトモデュレーション処置にて処理された毛嚢は、毛幹及びコントロールに対する成長率において22%の増加を示した。
Claims (16)
- 毛髪成長構造を、約390nm以上約1600nm以下の主放射波長を有する電磁気放射源に曝露し;且つ
医療的に効果的な保持時間及び医療的な効果を有する光強度にて、前記毛髪成長構造の前記の曝露を、前記電磁気放射源へと保持することにより、前記毛髪成長構造をフォトスティミュレートする;
ことを有する毛髪成長を刺激する方法。 - 前記毛髪成長構造の前記曝露の前に、前記毛髪成長構造及び該構造の近傍の皮膚に、フォトモデュレーション促進剤を適用することをさらに有することを特徴とする請求項1に記載の方法。
- 前記フォトモデュレーション促進剤は、ハイドロキノン、コージ酸、成長因子、エキナシア、抗生物質、抗カビ剤、抗ウィルス剤、ブリーチ剤、塩水誘導体、酵素、触媒、抗老化物質、インスリン、ミネラル、毛髪成長刺激物質、毛髪成長阻害物質、ダイ、天然又は合成メラニン、プロリン、ヒドロキシプロリン、麻酔物質、クロロフィル、銅クロロフィル、葉緑体、カロテノイド、バクテリオクロロフィル、フィコビリン、カロテン、キサントフィル、アントシアニン、フォスフォリパーゼA2の阻害剤及びS−アデノシルメチオニンの阻害剤を包含する毛髪成長阻害剤であって、該阻害剤の例は、限定しないが、甘草、リコカロンA、ゲネステイン、ソイイソフラボン、ビタミンD、豆乳、NF−κBの阻害剤、b3−ARアジポサイト受容体、レプチン、イミキノイド、ウルシオール、局所又は全身性の免疫調節剤、スルフヒドリル化合物、フリーラジカルスカベンジャー、抗アンドロゲン剤、スルフォン、複素環エステル、複素環アミド並びにこれら薬剤の、誘導体、アナログコンジュゲート、天然又は合成の代謝阻害剤並びにこれら薬剤の遺伝子改変物又は免疫学的コンジュゲートの少なくとも一つからなる群から選択された活性物質を含むことを特徴とする請求項2に記載の方法。
- 前記フォトモデュレーション促進剤を前記電磁気放射線源に曝露する前に、前記フォトモデュレーション促進剤を、透過促進工程に指向することをさらに有することを特徴とする請求項3に記載の方法。
- 前記透過促進工程は、酵素剥離、マイクロダーム研磨、溶剤除去、テープ除去、スクラビング、レーザーアブレーション、レーザー揮発、化学的剥離、電気刺激、高ピーク出力及び短パルス保持時間でのレーザー処置、超音波並びにこれらの組み合わせからなる群から選択された工程を有することを特徴とする請求項4に記載の方法。
- 前記浸透促進工程は、前記フォトモデュレーション促進剤を超音波に曝露することを特徴とする請求項5に記載の方法。
- 前記浸透促進工程は、前記毛髪成長構造及び/又は前記の該構造の近傍の皮膚を電気刺激に曝露することを有することを特徴とする請求項5に記載の方法。
