Abstract
【解決手段】 検体中の標的核酸を検出する方法であって、
(a)プライマー対を用いて検体中の核酸をPCR反応により増幅させ、ここで、前記プライマー対は、前記標的核酸中に存在するジンクフィンガー蛋白質認識配列がPCR反応により増幅されるよう設計されており、そして
(b)ジンクフィンガー蛋白質を用いて工程(a)で得られたPCR増幅産物を検出する
の各工程を含む方法が開示される。ジンクフィンガー蛋白質としては、例えば配列5’−GCGTGGGCG−3’を認識するZif268を用いることができる。

Description
(a)プライマー対を用いて検体中の核酸をPCR反応により増幅させ、ここで、該プライマー対は、該標的核酸中に存在するジンクフィンガー蛋白質認識配列がPCR反応により増幅されるよう設計されており、そして、
(b)ジンクフィンガー蛋白質を用いて工程(a)で得られたPCR増幅産物を検出する、
の各工程を含むことを特徴とする。
ジンクフィンガー蛋白質としてはZif268の変異体であるZifMを用いた。ZifMは、Zif268にSphI、BsiWI部位を導入したものであり、アミノ酸配列および標的認識配列はZif268と同じである。
標的および対照の二本鎖DNAの配列は以下のものを用いた(Bio=ビオチン、下線はジンクフィンガー蛋白質認識配列)。

まず、BLASTにより、S.typhimuriumのゲノム中からZif268結合部位であるgcgtgggcgの9塩基の配列を検索した。S.typhimuriumのゲノム配列は先の報告にしたがった(GenBank受託番号:AE008878,AE006468;McClelland et al., Nature 413(6858), 852-856 (2001))。次に、この9塩基配列の5’側および3’側のそれぞれ20塩基をプライマー領域として含む連続する49塩基配列についてBLASTで検索し、他の細菌が類似した配列を有するか否かを調べた。さらに、その遺伝子の比較対照となる他の遺伝子配列の多さ、進化速度、水平移動能の有無、などを基準として、標的とすべき遺伝子を選択した。
Zif268が結合する連続する9塩基対の両端20bpの配列をプライマー結合部位としてプライマーを設計した。

Claims (13)
- 検体中の標的核酸を検出する方法であって、
(a)プライマー対を用いて検体中の核酸をPCR反応により増幅させ、ここで、前記プライマー対は、前記標的核酸中に存在するジンクフィンガー蛋白質認識配列がPCR反応により増幅されるよう設計されており、そして
(b)ジンクフィンガー蛋白質を用いて工程(a)で得られたPCR増幅産物を検出する
の各工程を含む方法。 - ジンクフィンガー蛋白質が検出可能な標識により標識されている、請求項1記載の方法。
- ジンクフィンガー蛋白質が担体上に固定化されている、請求項1または2に記載の方法。
- ジンクフィンガー蛋白質がファージ上に提示される、請求項1記載の方法。
- ジンクフィンガー蛋白質がZif268であり、ジンクフィンガー蛋白質認識配列が5’−GCGTGGGCG−3’である、請求項1−4のいずれかに記載の方法。
- 前記標的核酸がSalmonella typhimuriumのgyrB遺伝子である、請求項1−5のいずれかに記載の方法。
- 検体中の標的核酸を検出するためのキットであって、前記標的核酸中に存在するジンクフィンガー蛋白質認識配列がPCR反応により増幅されるよう設計されたプライマー対を含むキット。
- 前記ジンクフィンガー蛋白質認識配列に結合しうるジンクフィンガー蛋白質をさらに含む、請求項7記載のキット。
- ジンクフィンガー蛋白質がファージ上に提示された形で提供される、請求項8に記載のキット。
- ジンクフィンガー蛋白質が担体上に固定化されている、請求項8記載のキット。
- 前記ジンクフィンガー蛋白質認識配列に結合しうるジンクフィンガー蛋白質をコードする遺伝子をさらに含む、請求項7記載のキット。
- ジンクフィンガー蛋白質がZif268であり、ジンクフィンガー蛋白質認識配列が5’−GCGTGGGCG−3’である、請求項7−11のいずれかに記載のキット。
- 前記標的核酸がSalmonella typhimuriumのgyrB遺伝子である、請求項7−11のいずれかに記載のキット。
