Abstract
【解決手段】NBD−アミンのN−オキサイドまたはその誘導体を酸化還元指示薬として提供する。還元によって形成されるNBD−アミンは高い蛍光を特徴とし、青色光照射によって非常に容易に励起することができる。

R1およびR2はそれぞれ独立に、R、(CH2CH2O)mR、COR、COORおよびOCORから選択され;
R3は各場合において独立に、NO2、CN、R、OR、OCOR、COOR、SRおよびハロゲンから選択され;
Rは、HまたはC1〜C4アルキルであり、その場合にアルキルはハロゲン、OR、SR、NR2、COOR、CONR2、SO3Rおよびその塩および/またはPO(OR)3およびその塩で置換されていても良く;
mは、1〜20の整数、好ましくは1〜10の整数であり;
nは1、2または3である。]
で表される化合物を含む検出試薬と接触させることを特徴とする方法である。
3mL蛍光キュベットに下記の化合物を加えた(記載の濃度は、キュベット中での最終濃度を指す;NBD−アミンのN−オキサイドは、文献(P.B. Ghosh, M.W. Whitehouse, J. Med. Chem., 11, 305-311 (1968))に従って製造した)。

- 酸化還元反応および蛍光測定によってアナライトを検出する方法であって、前記アナライトを含むサンプルを、蛍光酸化還元指示薬として下記一般式(I)で表される化合物を含む検出試薬と接触させることを特徴とする方法。
R1およびR2はそれぞれ独立に、R、(CH2CH2O)mR、COR、COORおよびOCORから選択され;
R3は各場合において独立に、NO2、CN、R、OR、OCOR、COOR、SRおよびハロゲンから選択され;
Rは、HまたはC1〜C4アルキルであり、その場合にアルキルはハロゲン、OR、SR、NR2、COOR、CONR2、SO3Rおよびその塩および/またはPO(OR)3およびその塩で置換されていても良く;
mは、1〜20の整数であり;
nは1、2または3である。] - R1およびR2が、OHで置換されたC1−2アルキル基である請求項1に記載の方法。
- R3がNO2である請求項1または2に記載の方法。
- 前記酸化還元指示薬(I)が電子を直接受容することができる請求項1〜3のいずれかに記載の方法。
- 前記酸化還元指示薬(I)がメディエータを介して電子を受容することができる請求項1〜3のいずれかに記載の方法。
- 前記アナライトとして酸化可能物質を検出する請求項1〜5のいずれかに記載の方法。
- 前記アナライトを還元または酸化するための1種以上の酵素および場合により補酵素をさらに含む検出試薬を用いる請求項6に記載の方法。
- グルコース、ラクテート、アルコール、ガラクトース、コレステロール、果糖、グリセリン、ピルベート、クレアチニン、アラニン、フェニルアラニン、ロイシン、トリグリセリドまたはHDLコレステロールをアナライトとして検出する請求項6または7に記載の方法。
- グルコースオキシダーゼ、グルコース色素オキシドレダクターゼまたはグルコースデヒドロゲナーゼ/ジアフォラーゼを用いてグルコースを検出する請求項8に記載の方法。
- 酸化還元反応を触媒する酵素または反応をオキシドレダクターゼ反応と組み合わせることができる酵素を前記アナライトとして検出する請求項1〜5のいずれかに記載の方法。
- グルタミン酸−オキサル酢酸トランスアミナーゼ(GOT)、(AST)、グルタミン酸−ピルビン酸トランスアミナーゼ(GPT)、アラニンアミノトランスフェラーゼ(ALT)、乳酸デヒドロゲナーゼ(LDH)またはクレアチンキナーゼ(CK)をアナライトとして検出する請求項10に記載の方法。
- 酸化還元反応および蛍光測定によってアナライトを検出するための試薬であって、下記一般式(I)で表される化合物を含むことを特徴とする試薬。
