Abstract
【解決手段】 下記の遺伝子構成物を含む非哺乳動物宿主細胞:
パピローマウイルスL1キャプシッドタンパク質をコードするパピローマウイルスL1立体配座コード配列を組換えベクター中に含む遺伝子構成物であって、前記構成物が、前記パピローマウイルスL1キャプシッドタンパク質を含むキャプソメア構造の自己組み立てにより天然のパピローマウイルスに対する高力価中和抗体を誘導する立体配座エピトープを有するキャプソメア構造の宿主細胞内での発現を指示し、但し、前記組換えベクターが非哺乳動物真核細胞用ベクターであり且つ前記細胞が非哺乳動物真核宿主細胞であり、同宿主細胞は前記立体配座エピトープを有するキャプソメア構造を産生することを特徴とする、パピローマウイルスL1キャプシッドタンパク質をコードするパピローマウイルスL1立体配座コード配列を含む遺伝子構成物。

rd,L.,et al.,1963)。これら2つのタンパク質の配列は互いに全く関連がない。キャプシッドにおけるL2の機能および位置は明らかになっていない。しかしながら、免疫学的データからL2のほとんどはL1に対して内部にあるのではないかと思われる。
u,J.,et al(1991)および(1992)は、HPV L1およびL2遺伝子と、HPV E3/E4遺伝子を組み合わせたHPV L1およびL2遺伝子とをワクシニアウイルスベクターにクローニングし、この組み換えワクシニアウイルスでCV−1哺乳類細胞を感染することによってウイルス様の粒子(virus-like particle)を産生した。これらの研究はZhouによって解釈され、上皮細胞におけるHPV16のL1およびL2タンパク質の発現はビリオン型の粒子の組立てを可能にするために必要で十分なものであるということが確立された。L1およびL2タンパク質を発現する二重組み換えワクシニアウイルスを感染させた細胞では、HPVキャプソメアの不完全に組み立てられたアレイであると思われる核中に小さな(40nm)ウイルスの粒子認められた。L1タンパク質のみあるいはL2タンパク質のみを発現させても、ウイルス様粒子は産生されなかった。また、L1およびL2遺伝子を含有する単一組み換えワクシニアウイルスを二重に感染させた細胞でも粒子を産生させることはできなかった。中和活性は全く報告されなかった。
et al.(1993)は、ヒト細胞においてHPV1からL1が発現されると、L1は自己組み立てされてウイルス様の粒子になると報告した。中和抗体についての研究は行われなかった。
パピローマウイルスL2コード化配列を有することができる。
本発明の他の態様によれば、キャプソメア構造またはその一部に組み立てられる、組み換えパピローマウイルスキャプシッドタンパク質を産生するための方法であって、(1)L1立体配座キャプシッドタンパク質をコード化するパピローマウイルス遺伝子を転移ベクターにクローニングするステップであって、前記遺伝子のオープンリーディングフレームは前記ベクターの制御下にある前記ステップと、(2)組み換えベクターを宿主細胞に転移するステップであって、クローン化したパピローマウイルス遺伝子はパピローマウイルスキャプシッドタンパク質を発現する前記ステップと、(3)宿主細胞からパピローマウイルスキャプシッドタンパク質を含有するキャプソメア構造を単離するステップと、を含む方法が提供される。好ましい実施態様において、クローン化したパピローマウイルス遺伝子は本質的に立体配座L1コード化配列からなり、発現タンパク質は組み立てられて本質的にL1キャプシッドタンパク質からなるキャプソメア構造になる。他の好ましい実施態様において、上述した方法のクローニングするステップは、L2キャプシッドタンパク質をコード化するパピローマウイルス遺伝子をクローニングすることをさらに含み、それによって前記L1およびL2タンパク質を同時に宿主細胞に発現させる、前記宿主細胞が哺乳類細胞である時に前記転移ベクターはワクシニアウイルスではないという条件で、単離されたキャプソメア構造はL1およびL2キャプシッドタンパク質を有する。立体配座L1コード化配列は、ウシ、サル、ヒトパピローマウイルスからクローンすることができる。好ましい実施態様によれば、立体配座L1コード化配列は野生型HPV16パピローマウイルスからクローニングされる。特に好ましい実施態様において、立体配座L1コード化配列は配列番号2である。また、好ましい実施態様において、転移ベクターはバキュロウイルスに基づく形質転換ベクターであり、パピローマウイルス遺伝子は昆虫細胞において活性であるプロモーターの制御下にある。従って、この実施態様では、組み換えバキュロウイルスDNAはSf−9昆虫細胞にトランスフェクションされる。
本発明のさらに他の態様によれば、本発明の方法によって産生される、本質的にパピローマウイルスL1キャプシッドタンパク質からなるウイルスキャプソメア構造またはその蛋白質が提供される。また、ウイルスキャプソメア構造は本発明の方法により生成される本質的にパピローマウイルスL1およびL2キャプシッドタンパク質からなるものであってもよい。特に好ましい実施態様において、ウイルスキャプソメア構造は、野生型ウイルスからクローニングされたHPV16 L1DNAの発現産生物であるパピローマウイルスL1キャプシッドタンパク質を含む。
本発明の方法によれば、野生型HPV16パピローマウイルスゲノムからのL1オープンリーディングフレーム(ORF)を使用することで、ワクチンの使用に適したスケールでの調製量のウイルス様粒子の産生を特に促進できる。
本においてパピローマウイルスを検出するための方法であって、前記パピローマウイルスのキャプシッドの立体配座エピトープの1つまたは複数に対する特異性を有する抗体と標本とを接触させるステップであって、抗体は検出可能なシグナルを発生させる標識を有し、あるいは検出可能に標識された試薬に結合されるステップと、抗体をパピローマウイルスに結合できるようにするステップと、検出可能なラベルによって標本中に存在するパピローマウイルスの存在を決定するステップと、を含む方法が提供される。
HPV16 L1 DNA源は、例えばZhouら(1991)が報告した研究で開示された通り、プロトタイプクローン、GenBank Accession No.K02718で、子宮頚部癌より単離されている(Seedprfら、1985)。我々は、野生型HPV16ゲノム由来のL1は、単一点での突然変異によりプロトタイプゲノムとは異なるが、BPV1またはBPV L1/L2でみられる効果と同様の効果により、自己組み立てされてウイルス様粒子を形成する。プロトタイプHPVゲノム由来のL1をL2と共に使用した場合に見られる自己組み立てと比較すると、野生型ゲノム由来のL1は少なくとも100倍以上の効率で自己組み立てされる。
分析の結果、プロトタイプ株の報告されているL1の塩基配列の内、下記の通り2箇所にエラーが検出された。
パピローマウイルス感染異形成病変より単離したクローン16PATと、悪性子宮頚部癌より単離したプロトタイプHPV16は両者ともnt6242-6244の位置でヒスチジンをコード化しているのに対し、良性のパピローマウイルス感染病変より単離したクローン2及び11は(他の多くのパピローマウイルスと同様に)同位置でアスパラギン酸をコード化している。
る。きわめて近縁のHPVタイプの間では、この場所にアスパラギン酸があることが効率的な自己組み立てを得るのに必要で、アスパラギン酸をヒスチジンに置き換えるとキャプシッドタンパク質からこの能力が失われる。悪性関連ウイルスにおいてキャプシッド組み立てが失われることは、中和抗体産生に必要な立体配座エピトープの喪失と関連しており、またこれはパピローマウイルスが免疫制御を逃れるための免疫原性の低下と関係していると思われる。
本発明の方法により、任意のパピローマウイルスの組換えキャプシッド粒子も調製することができる。L1またはL2キャプシッドタンパク質のいずれか一方のみ、またはL1及びL2キャプシッドタンパク質の両者からなる粒子を調製することができる。L1/L2キャプシッドタンパク質粒子は天然のパピローマウイルスビリオンの組成とより一層密接に関係するが、おそらくL2のほとんどはキャプシッド構造内でL1よりも内部にあるため、L2はL1ほど免疫に関して重要とは思われない。L2が存在しなくてもL1は自身で自己組み立てされることが出来るが、自己組み立てされたL1/L2キャプシッドタンパク質粒子はさらに天然のパピローマウイルスビリオンの組成と密接に関係している。従って、L1のみを含む粒子の方がより単純であり、L1/L2を含む粒子の方が一層真正の構造を有していると思われる。自己組み立てされたL1及びL1/L2粒子は両者とも高力価の中和抗体を誘導し、従ってワクチン産生に適している。野生型HPVゲノムにより発現されるL1キャプシッドタンパク質を含む粒子は、L1のみの場合でもL1/L2のいずれの場合でも、特に望ましい。
パク質産生は新しい昆虫細胞を組換えバキュロウイルスに感染させることよって行う。従って、L1キャプシッドタンパク質及びL1及びL2キャプシッドタンパク質は、実施例2に記載されている通り組換えバキュロウイルスを感染させた昆虫細胞内で発現される。
異種細胞内で合成された自己組み立て主要キャプシッドタンパク質に基づくサブユニットワクチンはヒトB型肝炎を含む多くの病原ウイルスによる感染の予防に有効であることが明らかにされている(Stevens.C.ら、1987)。非感染性またはホルマリン不活性化BPVはワクチンとして有効であるがBPV形質転換細胞は有効ではないことを明らかにする研究により、初期の遺伝子産物よりもウイルスキャプシドタンパク質は免疫応答を誘引することが示唆されている。他の論文のデータからも、細菌から抽出されたL1タンパク質は中和抗体の力価が低いにもかかわらず一部免疫応答を誘引することが示唆されている。従って、昆虫細胞内で発現してウイルス様粒子に組み立てられるBPV L1について家兎中の中和抗体を誘導する能力を調べた。2種類の調製物について試験を行った。それらは、L1組換えまたは野生型感染Sf-9細胞の全細胞抽出物と種々の遠心分離及び硫安沈澱により単離した部分的精製粒子である。1次接種後、家兎に隔週毎に2回追加接種を行った。
の産生には必要ないことが明かである。細菌により誘導されたL1の中和免疫原性が弱いことが報告されたが、これはコンフォメーションが適切ではなく、また組み立てされてビリオン様構造物を形成しないと思われることが原因である。また、L1の多数の電気泳動的変異体がビリオン内で検出された(Larsen.P.ら、1987)。これら修飾物のいくつかはおそらく細菌により誘導されたL1内には存在しないと思われるが、これは中和抗体の産生を促進すると思われる。
パピローマウイルスビリオンタンパク質に対するヒト免疫応答に関する公知の血清検査では、原則として細菌で誘導したL1およびL2キャプシッドタンパク質を利用しており、その結果は他のHPV感染症の測定(Jenison,S.et al.,1990)とはあまりよい相関関係にはない。上記のBPVパピローマウイルス免疫検査により、細菌より抽出されたパピローマウイルスビリオンタンパク質は、タイプ特異的で多くの中和抗体により認識される立体配座依存性エピトープを提示しないことが示唆されている。主に線状エピトープを認識するこのようなアッセイと比較すると、自己組み立てされるL1粒子を用いた血清検査は、ヒトの集団において抗HPVビリオン免疫性の程度をさらに正確に測定すると思われる。従ってここに開示された組換えL1キャプシドタンパク質は立体配座エピトープを提示しており、いくつかのタイプの結合アッセイにおいてパピローマウイルスに対する中和抗体を与える免疫性を検出するための特異性の高い診断薬として使用できる。その操作方法は、一般に抗原抗体対中のキャプシッドタンパク質と特異的に結
合する体液中の抗体を検出する手段を提供する固相あるいは液相アッセイとして行われる。循環抗体を評価するための当業者間で公知の手順の例として、二重抗体ラジオイムノアッセイおよび酵素イムノアッセイなどの液相アッセイや、ストリップラジオイムノアッセイ、ウエスタンブロッティングおよびGallo et al.に付与された米国特許第4,520,113号酵素結合免疫吸着アッセイ(ELISA)、Skold et al.に付与された米国特許第5,039,607号に開示されているimmunochromatographicアッセイなどが挙げられる。抗体の検出用として好ましいELISA法は、Harlow E.およびLane,D.著、Antibodies:A Laboratory Manual, Cold Spring Harbor,NY,1988,pp.563〜578に開示されているようなものである。
また、本発明のキャプシッドタンパク質を免疫原として使用し、周知の方法によって多クローン性あるいは単一クローン性抗体を得ることもできる。これらのパピローマウイルス特異抗体は、特に抗体のクラスや種に特異的な第2の標識抗体と組み合わせて、免疫組織化学などの様々な周知のアッセイ手順に基づいて、体の組織や体液の標本中におけるキャプシッドタンパク質の存在の検出に診断的に利用することもできる。
BPV1 DNAのクローニングしたプロトタイプ(Chen,E.,et al.,1982),GenBank Assession No.X02346またはHPV16 DNA(Seedorf,K.,et al.,1985)GenBank Association No.K02718、あるいは野生型HPV16 DNA(配列番号2)をテンプレートとして使用して、完全長のL1あるいはL1およびL2のオープンリーディングフレーム(ORF)をPCRによって増幅した。単一の制限部位をオリゴヌクレオチドプライマーに組み込んだ(各配列において、6番目〜11のヌクレオチド)。

昆虫細胞中でのL1タンパク質またはL1/L2タンパク質の発現
wt AcMNPV(wt)またはAcBPV−L1(B−L1)、AcHPV16−L1(16−L1)またはAcHPV16−L1(16−L1)およびAcHPV16−L2(16−L2)組換えウイルスによって、Sf−9細胞を疑似感染(疑似)させるかあるいは10の感染多重度で感染させた。72時間後、Laemmli緩衝液において煮沸することによって細胞を溶解し、10%ゲル内でライゼートをSDS−PAGEにかけた。タンパク質を0.25%クーマシーブルーで染色するか、免疫ブロッティングしてBPV L1mAb AU−1(Nakai,Y.,et al.,1986)またはHPV16 L1 mAb CAMVIR−1(McLean,C.,et al.,1990)、125I標識Fab抗マウスIgG(Amersham)でプローブした。Pは、ポリヘドリンタンパク質を示す。抗BPV L1mAbは予測した55kdのタンパク質を認識した。抗HPV 16L1mAbは予測した58kdのタンパク質を強く染色すると共に、上述したように分子量がこれよりも小さい5つのバンドも淡く染色した。また、上述したように、この抗HPV16 L1は、BPV L1タンパク質と若干交差反応した。
抗血清の製造
家兎に対し、凍結/溶解を1サイクル行い20×ダウンス ホモジェナイズ処理をすることにより調製した全細胞Sf−9溶解物(3×107 細胞)(家兎#1,2および8)、
または分画遠心分離および35%硫安沈澱により部分精製したL1タンパク質200μg(#3、4、6および7)を、完全フロイントアジュバント中に添加したものを皮下より注入して免疫し、次に不完全フロイントアジュバント中に同様の調製物を添加したものを用いて2週間間隔で2回追加免疫を行った。
粒子の精製及び透過型電子顕微鏡による解析
Sf−9細胞(2×106 /ml)500mlにAcBPV−L1またはAcHPV16−L1またはAcHPV16−L1/L2(16−L1/L2)組換え体バキュロウイルスを感染させた。72時間後、回収した細胞をPBS中で60秒間超音波処理した。低速清澄化後、溶解物を40%(wt/vol)ショ糖/PBSクッションで、110,000gで2.5時間遠心分離を行った(SW−28)。再懸濁したペレットを141,000g、室温で20時間、10〜40%(wt/wt)CsCl/PBS勾配中で平衡になるまで遠心分離を行った。分画を底から回収してSDS−PAGEによる分析を行った。免疫反応性分画をPBSに対して透析し、セントリコン30(Millipore)限外濾過により濃縮し、(HPV16−L1の場合)エアーヒュージ(Beckman)でA−100ローター中で10分間30psiで遠心分離してペレットを形成させた。BPV1ビリオンを記述通り(Cowsert,L.ら、1987)ウシ疣贅(A.B.Jenson氏より供与)より精製した。精製した粒子をカーボンコートTEMグリットに吸着させ、1%酢酸ウラニルで染色してフィリップ電子顕微鏡EM400Tにより倍率36,000倍で観察した。電子顕微鏡観察により結果が得られ、上記で考察したとおりである。
BPV1中和アッセイ
第2追加免疫3週間後に得られた血清の系列希釈物を約500フォーカス形成単位のBPV1ウイルスと共に30分間インキュベートし、ウイルスをC127細胞に1時間吸着させて、これらの細胞を3週間培養した(Dvoretzky,I.ら、1980)。フォーカスを0.5%メチレンブルー/0.25%カルボルフクシン/メタノール液で染色した。結果は、フォーカスの数を評価して得た。考察は以下に行う。抗−AcBPV−L1は家兎#1より得られ、抗−wt AcMNPVは家兎#8より得られた(表1)。1:400に希釈した前免疫血清を標準として使用した。抗−AcBPV−L1は希釈率1:400または1:600で実質的にフォーカスを消失したが、抗wt AcMNPVは希釈率1:400または1:600でフォーカスの数が上昇したように思われた。1:00希釈時の「nrs」と表記した正常家兎血清のネガティブコントロールを抗BPV−1ビリオンの標準として使用したところ、抗BPV−1ビリオンは希釈率1:400または1:600で実質的にフォーカスを消失したと思われた。抗BPV−1ビリオンは以前の研究(Nakai,Y.ら、1986)で、野生のBPVビリオンに対して励起された。最後に、ダコは変性BPVビリオンに対して励起された市販の家兎抗血清(Dako Corp.,Santa Barbara, California)である。この血清は、見かけ上抗体を含まないコントロールよりも大量のフォーカスを発生させた。ネガティブコントロールとして、ウイルスを含まない検査では、実質的にフォーカスを発生しなかった。
BPV1に対する血清中和力価
実施例5と同様にしてアッセイを行った。家兎#1、2および8に粗全細胞Sf−9溶解物を接種し、家兎#3、4、6および7には部分精製したL1タンパク質(表1)を接種した。家兎#6および7を、1%SDS中で煮沸変性させたL1タンパク質調製物で免疫した。各家兎とも、2回目の追加免疫後3〜6週間後に少なくとも2回採血して検査を行ったところ、同一の抗体力価であることが認められた。各家兎の免疫前血清の力価は、20より小さく、最も低い希釈率で検査した。

Claims (22)
- 下記の遺伝子構成物を含む非哺乳動物宿主細胞:
パピローマウイルスL1キャプシッドタンパク質をコードするパピローマウイルスL1立体配座コード配列を組換えベクター中に含む遺伝子構成物であって、前記構成物が、前記パピローマウイルスL1キャプシッドタンパク質を含むキャプソメア構造の自己組み立てにより天然のパピローマウイルスに対する高力価中和抗体を誘導する立体配座エピトープを有するキャプソメア構造の宿主細胞内での発現を指示し、但し、前記組換えベクターが非哺乳動物真核細胞用ベクターであり且つ前記細胞が非哺乳動物真核宿主細胞であり、同宿主細胞は前記立体配座エピトープを有するキャプソメア構造を産生することを特徴とする、パピローマウイルスL1キャプシッドタンパク質をコードするパピローマウイルスL1立体配座コード配列を含む遺伝子構成物。 - 前記パピローマウイルスL1キャプシッドタンパク質は野生型である請求項1記載の非哺乳動物宿主細胞。
- 前記パピローマウイルスL1キャプシッドタンパク質は、配列番号1に示す変異型HPV16 L1タンパク質のアミノ酸配列の202位に相当する位置に、ヒスチジン残基以外のアミノ酸残基を有する請求項2に記載の非哺乳動物宿主細胞。
- 前記パピローマウイルスL1キャプシッドタンパク質は、前記202位に相当する位置を除いて、配列番号2に示すアミノ酸配列、又は同アミノ酸配列において、1若しくは数個のアミノ酸の置換、欠失、挿入又は付加を含むアミノ酸配列を有することを特徴とする請求項3に記載の細胞。
- 天然のパピローマウイルスに対する高力価の中和抗体を誘導する立体配座エピトープを有するキャプソメア構造を製造する方法であって、該方法は、該キャプソメア構造の発現を指示する遺伝子構成物のための条件を準備することを含み、該遺伝子構成物はパピローマウイルスL1キャプシッドタンパク質をコードするパピローマウイルスL1立体配座コード配列を組換えベクター中に含む遺伝子構成物であって、該パピローマウイルスL1キャプシッドタンパク質を含む該キャプソメア構造の自己組み立てによる天然のパピローマウイルスに対する高力価中和抗体を誘導する立体配座エピトープを有する該キャプソメア構造の宿主細胞内での発現を指示し、但し、該組換えベクターが非哺乳動物真核細胞用ベクターであり且つ該細胞が非哺乳動物真核宿主細胞である当該方法。
- 前記パピローマウイルスL1キャプシッドタンパク質は野生型である請求項5に記載の方法。
- 前記パピローマウイルスL1キャプシッドタンパク質は、配列番号1に示す変異型HPV16 L1タンパク質のアミノ酸配列の202位に相当する位置に、ヒスチジン残基以外のアミノ酸残基を有する請求項6に記載の方法。
- 前記パピローマウイルスL1キャプシッドタンパク質は、前記202位に相当する位置を除いて、配列番号2に示すアミノ酸配列、又は同アミノ酸配列において、1若しくは数個のアミノ酸の置換、欠失、挿入又は付加を含むアミノ酸配列を有することを特徴とする請求項7に記載の方法。
- 前記のキャプソメア構造を単離するステップを更に含む、請求項5〜8のいずれか一項に記載の方法。
- 前記の組換えベクターが昆虫用ベクターであり且つ前記の宿主細胞が昆虫細胞である、請求項5〜9のいずれか一項に記載の方法。
- 前記の昆虫用ベクターが、バキュロウイルスベクターである、請求項10に記載の方法。
- .前記のバキュロウイルスベクターが、昆虫宿主細胞の組換えバキュロウイルスDNAと野生型バキュロウイルスDNAの同時トランスフェクションにより形成される、請求項11に記載の方法。
- 前記の組換えベクターが酵母細胞用ベクターであり且つ前記の宿主細胞が酵母宿主細胞である、請求項5〜9のいずれか一項に記載の方法。
- 前記の配列が、ヒトパピローマウイルスL1遺伝子、ウシパピロー
マウイルスL1遺伝子又はサルパピローマウイルスL1遺伝子から導かれる、請求項5〜13のいずれか一項に記載の方法。 - 前記のキャプソメア構造が、パピローマウイルスL2キャプシッドタンパク質の同時発現を必要としない、請求項5〜14のいずれか一項に記載の方法。
- 前記のキャプソメア構造が、更に、パピローマウイルスL2キャプシッドタンパク質を含む、請求項5〜13のいずれか一項に記載の方法。
- 前記の配列が、前記のパピローマウイルスL1キャプシッドタンパク質と前記のパピローマウイルスL2キャプシッドタンパク質を二重にコードする、請求項16に記載の方法。
- 免疫化投与スケジュールに従って投与した場合に少なくとも約103のパピローマウイルス中和抗体力価を誘導するのに十分な有効な免疫原濃度のキャプソメア構造を含む単位投与量のワクチンであって、該キャプソメア構造が、下記の遺伝子構成物の指示の下に発現された天然のパピローマウイルスに対する高力価中和抗体を誘導する立体配座エピトープを有する当該ワクチン:
パピローマウイルスL1キャプシッドタンパク質をコードするパピローマウイルスL1立体配座コード配列を組換えベクター中に含む遺伝子構成物であって、前記構成物は、前記パピローマウイルスL1キャプシッドタンパク質を含むキャプソメア構造の自己組み立てによる天然のパピローマウイルスに対する高力価中和抗体を誘導する立体配座エピトープを有するキャプソメア構造の宿主細胞内での発現を指示し、但し、前記組換えベクターが非哺乳動物真核細胞用ベクターであり且つ前記細胞が非哺乳動物真核宿主細胞である遺伝子構成物。 - 患者のパピローマウイルス感染に対する液性免疫を検出する方法であって、下記:
(a)有効な抗体検出量のキャプソメア構造を用意し;
(b)該キャプソメア構造をパピローマウイルス感染について試験すべき患者からの体液試料と接触させ、該試料中に含まれるパピローマウイルス抗体をそれに結合させて抗原抗体複合体を形成させ;そして
(c)該試料中の該パピローマウイルス抗体の存在を該複合体の形成を測定することにより測定する
ことを含み、上記のキャプソメア構造が、下記の遺伝子構成物の指示の下に発現された天然のパピローマウイルスに対する高力価中和抗体を誘導する立体配座エピトープを有する当該方法:
パピローマウイルスL1キャプシッドタンパク質をコードするパピローマウイルスL1立体配座コード配列を組換えベクター中に含む遺伝子構成物であって、前記構成物は、前記パピローマウイルスL1キャプシッドタンパク質を含むキャプソメア構造の自己組み立てによる天然のパピローマウイルスに対する高力価中和抗体を誘導する立体配座エピトープを有するキャプソメア構造の宿主細胞内での発現を指示し、但し、前記組換えベクターが非哺乳動物真核細胞用ベクターであり且つ前記細胞が非哺乳動物真核宿主細胞である遺伝子構成物。 - パピローマウイルス感染の疑いのある患者からの試料において該ウイルスを検出する方法であって、該方法は、下記:
(a)キャプソメア構造に対して高めた有効なパピローマウイルス検出量の抗体を用意し;
(b)該試料を該抗体と接触させ、該試料に含まれるパピローマウイルスをそれに結合させて抗原抗体複合体を形成させ;そして
(c)該試料中の該パピローマウイルスの存在を該複合体の形成を測定することにより測定する
ことを含み、上記のキャプソメア構造が、下記の遺伝子構成物の指示の下に発現された天然のパピローマウイルスに対する高力価中和抗体を誘導する立体配座エピトープを有する当該方法:
パピローマウイルスL1キャプシッドタンパク質をコードするパピローマウイルスL1立体配座コード配列を組換えベクター中に含む遺伝子構成物であって、前記構成物は、前記パピローマウイルスL1キャプシッドタンパク質を含むキャプソメア構造の自己組み立てによる天然のパピローマウイルスに対する高力価中和抗体を誘導する立体配座エピトープを有するキャプソメア構造の宿主細胞内での発現を指示し、但し、前記組換えベクターが非哺乳動物真核細胞用ベクターであり且つ前記細胞が非哺乳動物真核宿主細胞である遺伝子構成物。 - 患者におけるパピローマウイルス感染を検出するための、即座に使えるようにパッケージされた診断用キットであって、第1のコンパートメントに有効な抗体検出量のキャプソメア構造を含み且つ第2のコンパートメントに該キャプソメア構造と患者から得られた試料中のパピローマウイルス抗体との結合を検出する材料を含み、上記のキャプソメア構造が、下記の遺伝子構成物の指示の下に発現された天然のパピローマウイルスに対する高力価中和抗体を誘導する立体配座エピトープを有する当該キット:
パピローマウイルスL1キャプシッドタンパク質をコードするパピローマウイルスL1立体配座コード配列を組換えベクター中に含む遺伝子構成物であって、前記構成物は、前記パピローマウイルスL1キャプシッドタンパク質を含むキャプソメア構造の自己組み立てによる天然のパピローマウイルスに対する高力価中和抗体を誘導する立体配座エピトープを有するキャプソメア構造の宿主細胞内での発現を指示し、但し、前記組換えベクターが非哺乳動物真核細胞用ベクターであり且つ前記細胞が非哺乳動物真核宿主細胞である遺伝子構成物。 - 患者におけるパピローマウイルス感染を検出するための、即座に使えるようにパッケージされた診断用キットであって、第1のコンパートメントに有効なパピローマウイルス検出量のキャプソメア構造に対して高めた抗体を含み且つ第2のコンパートメントに該抗体と患者から得られた試料中のパピローマウイルスとの結合を検出する材料を含み、上記のキャプソメア構造が、下記の遺伝子構成物の指示の下に発現された天然のパピローマウイルスに対する高力価中和抗体を誘導する立体配座エピトープを有する当該キット:
パピローマウイルスL1キャプシッドタンパク質をコードするパピローマウイルスL1立体配座コード配列を組換えベクター中に含む遺伝子構成物であって、前記構成物は、前記パピローマウイルスL1キャプシッドタンパク質を含むキャプソメア構造の自己組み立てによる天然のパピローマウイルスに対する高力価中和抗体を誘導する立体配座エピトープを有するキャプソメア構造の宿主細胞内での発現を指示し、但し、前記組換えベクターが非哺乳動物真核細胞用ベクターであり且つ前記細胞が非哺乳動物真核宿主細胞である遺伝子構成物。
