Abstract
Description
の類似体に関する。 発明の背景 骨粗鬆症は老人特に女性の老人の疾病の主要な要因である。最近、ヒト副甲状
腺ホルモン(hPTH)およびある種の類似体が骨粗鬆症の治療に有効な、骨生長の
刺激因子であることが明らかにされた。骨粗鬆症は、総骨量の減少および骨の脆
弱性を増加させる進行性疾病である。骨粗鬆症はしばしば負荷のかかった骨の自
発性骨折および、運動抑制の特徴である物理的および精神的衰退を生じさせる。 閉経後骨粗鬆症はエストロゲンの消失によって引きおこされる。エストロゲン
の消失は、古い骨の再吸収と新しい骨の形成とのバランスの悪さの増加を伴う骨
のターンオーバーの十年単位の加速を引き起こす。このことは負荷のかかった骨
の薄化、空隙率の増加、および、小柱欠乏(trabecular depletion)を生じさせる
。骨粗鬆症はまた、甲状腺機能亢進症、副甲状腺機能亢進症、クッシング病、あ
る種のステロイド剤の使用とも関連している。歴史的にはこの治療剤は食餌性カ
ルシウム増加、エストロゲン治療、およびビタミン投与量増加と関連しているが
、主として破骨細胞による骨再吸収を阻害する抗吸収剤のような薬剤と関連して
いる。
レベル制御因子である。PTHはポリペプチドであり、合成ポリペプチドは、Erick
sonとMerrifieldによるThe Proteins, Neurathら編集、Aademic Press, New Yor
k, 1976, 第257頁、によって開示され、Hodgesら(1988)、Peptide Research 1,
19またはAtherton, E.とSheppard, R.C. Solid Phase Peptide Synthesis, IRL
Press, Oxfordによって改変された方法によって調製することができる。 血清カルシウムが正常レベルよりも下がった場合、副甲状腺はPTHを放出し、
骨カルシウムの再吸収増加、腸からのカルシウム吸収の増加、および、尿細管に
おける新生尿からのカルシウムの腎再吸収の増加によりカルシウムレベルが増加
する。低濃度のPTHの連続インフュージョンは骨からカルシウムを除くことが出
来るが、同じ低濃度で、断続的に注射した場合は、実際に骨生長を促進すること
ができる。
tro細胞アッセイにおいて最初の27〜34アミノ酸がアデニリルシクラーゼを刺激
するという点で最も効果的であることが主張されている。Rosenblatt、米国特許
第4,771,124号には、hPTH類似体の特性が開示されており、Trp23がフェニルアラ
ニン、ロイシン、ノルロイシン、バリン、チロシン、β-ナフチルアラニン、ま
たはα-ナフチルアラニンに置換されたhPTH類似体がアンタゴニストして記載さ
れている。これらの改変hPTH類似体は、アミノ末端の2および6個のアミノ酸も
除去されており、その結果、骨粗鬆症を治療するために使用された場合にアゴニ
スト活性の大部分が欠損している。これらの類似体はPTHおよびPTH-関連ペプチ
ドの阻害因子として設計された。これらの類似体はいくつかの腫瘍と関連した高
カルシウム血症の治療にも有用であろうと主張された。
アラニン、アスパラギン、アスパラギン酸、システイン、グルタミン、グルタミ
ン酸、グリシン、ヒスチジン、イソロイシン、ロイシン、メチオニン、フェニル
アラニン、プロリン、セリン、スレオニン、トリプトファン、チロシン、または
バリンを含む種々のアミノ酸への置換を含むhPTH類似体が記載されている。これ
らは、血圧および平滑筋に対する影響が最小限であり、骨粗鬆症の治療に有効で
あると主張されているが、動物またはヒト試験からのデータの支持は無かった。 hPTH類似体を開示する、注目される他の文献はCodonとMorzeのPCT出願公開WO9
8/51324、Chorevらの米国特許第5,747,456号、Rosenblattらの米国特許第5,717,
062号であり、これらは13−17および26−30環化を含む一環または二環構造を開
示する。
ゼ(AC)およびGq-タンパク質活性化ホスホリパーゼCβ、の活性化を介して機能す
る。後者は膜結合タンパク質キナーゼCs(PKC)活性の刺激を生じさせる。ラッ
ト骨肉腫(RCO)細胞株において、PKC活性はPTHの29〜32番残基を必要とすること
が示されている(Jouishommeら、(1994) J. Bone Mineral Res.9 ,(1179-1189)
)。骨生長の増加、すなわち、骨粗鬆症の治療に有効な効果、はAC活性を増加さ
せるペプチド配列の能力と共役していることが確立されてきた。天然のh(PTH)-(
1-34)配列はこれらの活性の全てを有していることが示されている。このh(PTH)-

国特許第5,556,940号): Ser Val Ser Glu Ile Gln Leu Met His Asn Leu Gly Lys His Leu Asn Ser Met Glu Arg Val Glu Trp Leu Arg Lys Lys Leu Gln Asp Val-NH2 (配列番号2) B 上記分子Bは記載したアミド末端の変わりに遊離のカルボキシル末端を有して
いても良い。 本発明の目的は、より大きな代謝安定性、増大した骨修復活性、増大したAC活
性を有し、臨床的副作用が最小限である、新規なPTH類似体を提供することであ
る。
製薬的に許容できる塩が提供され、前記類似体は以下のアミノ酸配列を有する:

チルアミンおよびメチルアミンのような有機塩基の塩が含まれる。 本発明の別の特徴によれば、環化は22番残基と26番残基のような選択したアミ
ノ酸対間の側鎖のカップリングを含むラクタム形成によって行われる。他の型の
環化は、例えばCys含有類似体のCys22-Cys26間ジスルフィド架橋の形成も考慮す
ることが出来る。 種々のアミノ酸の置換も効果的であることが分かっている。Lys27はLeuまたは
種々の他の天然の疎水性アミノ酸または極性アミノ酸に置換しても良い。他の要
因はその残基がどれくらい良好にレセプターに適合するかである。AlaはLeuほど
疎水性ではない。
水性相互作用をする。例えば、Lysは疎水性部分がPTHレセプターの疎水性残基ま
たは残基の部分と相互作用し、その側鎖のNH2基は溶媒中に露呈されたままであ
るようにフォールディングすることが可能である。好ましいLeu置換以外のLysに
関する置換は、オルニチンおよびシトルリンのような極性残基および、アラニン
、ノルロイシン、イソロイシンおよびチロシンまたは側鎖に2〜10個の炭素原
子を有する直鎖若しくは分枝のα-アミノ脂肪酸が含まれ、脂肪鎖の末端に極性
基または荷電基を有する類似体も含まれる。極性基または荷電基の例には、アミ
ノ、カルボキシル、アセトアミド、グアニドおよびウイレドが含まれる。Leu27
は最も良好な置換のようであるが、位置27の他の多くの置換がほぼ完全な活性を
維持し、所望の特性、たとえばタンパク質分解安定性または水可溶性の増加、を
有し得る。Ile、ノルロイシン、Metおよびオルニチンが最も活性であると予想さ
れる。
ーゼ刺激活性を有し(Neugebauer, W., Barbier, J.R., Sung, W.L., Whitfield

同化活性は少なくともN-末端31残基を必要とし(Whitefield, J.F., Morley, P.
, Willick, G.E., Ross, V., Barbier, J.R., Isaacs, R.J.およびOhannessian,

-30)NH2によって可能であるが、(1-31)類似体によるよりも高い用量でのみ可能

one hPTH(1-30)NH2", Calc. Tiss. Int., 65: 143-147)。従って、我々は、更
に短い類似体hPTH(1-28)およびhPTH(1-29)も弱い同化活性しか有しないであろう
と予想した。hPTH(1-28)NH2の活性の低さは以下の実施例に示されている。
基および26番残基環のラクタム形成はhPTH(1-34)に匹敵するレベルのアデニルシ
クラーゼ刺激活性をもたらし、hPTH(1-34)に匹敵する同化(骨生長-刺激)活性
をもたらす証拠を持っている。 NMR研究により、CDによって高度にヘリックスを形成することが見いだされて
いるモデルペプチドであっても多くの非ヘリックスコンフォメーションをとって
いることが明らかになった。従って、PTHのようなレセプター結合ペプチドホル
モンの構造は溶液中のその遊離構造からは信頼性高く推定することができない。
ペプチドに利用可能な多数のコンフォメーション状態を制限するためにペプチド
ホルモンの束縛された類似体が使用されて来た。hPTHの配列を調べたところ、17
-29番残基内にα-ヘリックスを安定化または不安定化することが出来る3つの可
能な塩橋が明らかになった。それらは、Glu22とLys26、およびLys28とAsp30の間
、これらはいずれもα-ヘリックスを安定化させると予想されるものであり、お
よび、α-ヘリックスを不安定化させると予想されるLys27とAsp30の間である。
能なコンフォメーションを制限するであろう。さらに、これらのラクタムのうち
の2つ、Glu22-Lys26およびLys26-Asp30はα-ヘリックス構造を安定化させると
予想され、α-ヘリックスの両親媒性部分の極性表面に位置している。3つめ、L
ys27-Asp30は少なくとも部分的にはα-ヘリックスを不安定化させると予想され
、21-31番残基が結合している両親媒性ヘリックスの疎水表面上にある。Lysまた
はOrnが位置25のArgを置換し、Gln29がGluまたはAspによって置換される場合に
は、位置25と29間のラクタムとしての環化も起こり得る。 Lys27のLeuによる置換は、より疎水性の残基を両親媒性ヘリックスの疎水性表
面に生じさせる。このことは、ROS細胞株におけるアデニルシクラーゼ刺激活性
の増加を生じさせる。上述したのとは別の他の置換も類似体に同様なまたは増加
した活性を生じさせるであろうことは当業者に理解できるであろう。
学的活性を増加させ、分子のこの領域をタンパク質分解から保護することが予測
される。C-末端にアミドが存在することは、ある種のペプチダーゼがそのペプチ
ドを加水分解し得るとはいえ、ペプチドをエキソタンパク質分解から更に保護す
るという意味で好ましい(Leslie F.M.とGoldstein, A. (1982) Neuropeptides
2, 185-196)。 また、他のアミノ酸置換も有用に行い得ることが分かっている。具体的には、
日本特許公報61-24598に従って、我々は位置8および18の酸化感受性Met残基を
天然に存在する疎水性残基Nleに置換した。Nagataらに与えられた米国特許第5,3
93,869によるように、Leu、Ile、Val、PheおよびTrpのような他の疎水性残基も
有用であることが予想される。 リバースラクタム(reverse lactam)も考慮される。たとえば、22-26交換、た
とえば、Lys22-Glu26交換の有効性を示した。従って、他のラクタム、たとえば1
3-17ラクタム間にも同様な交換を有効に作製できることが期待される。
ではいくつかの置換hPTH類似体がかなりのAC活性を有すること、タンパク質分解
性攻撃に対して増強された安定性を有するかもしれないことが報告されており、
特に以下の類似体が報告されている。 1.Ser-1からAib(α-アミノイソ酪酸)への置換 2.Lys-27からGlnへの置換(2.5倍のAC活性を有すると報告されている) 3.第14番、15番、16番、17番残基のLysへの部分的または全部の置換は、8倍
にいたる活性の大きな増加が報告されている(これは水可溶性の増加によるもの
かもしれない。In vivoでは、これらはトリプシン様酵素に対して、より不安定
であると予想される)。このテトラペプチド(14-17番残基)は少なくとも一つ
の水可溶性アミノ酸が存在するようにクレームされている。可能性としては、Hi
s-14またはLys-14;Leu-15、Lys-15またはArg-15;Asn-16、Orn-16、Hci-16、As
p-16、Arg-16、Lys-16、D-Dlys-16、Ser-16またはGly-16;およびSer-17、Lys-1
7、Asp-17、Arg-17、またはGlu-17が含まれる。我々のラクタムは、特に位置27
にLeuまたは他の疎水性アミノ酸を有するラクタムは幾分不溶性になり溶解が困
難になり得るので、我々のラクタムにおいて同じ置換が有効であると予想される
。 4.プロテアーゼの攻撃を最小限にするためのArg25からHisへの置換 当業者であれば、環式1-28hPTHが好ましいかもしれないが本明細書に提示した
データから環式1-29断片も効果的であることが予測できるであろう。特に、文献
には追加のアミノ酸の存在がホルモンの生物学的特性に影響を与えるという証拠
はない。特に、我々の先行する米国特許出願第08/904,760号に含まれる1-30から
1-34の確認的データを見ればそうである。この出願の開示は引用により本明細書
に含まれるものとする。

、22番および26番残基間のラクタム環化、および、13および17番残基間のラクタ
ム環化が含まれる。例として、2つの別個の環化、Glu22-Lys26およびLys13-Glu
17を有し、好ましくはLeu27置換を有するhPTH分子が挙げられる。そのような類
似体はAC刺激においてより活性でないとしても、たとえば経口デリバリーにおい
て好ましいであろう。 本発明の類似体は以下に記載する既知の方法によって調製することができ、骨
生長を刺激するため、骨修復のため、および、たとえば骨粗鬆症の治療および通
常の骨折のような種々の状況における骨治癒の促進のために使用することができ
る。

る)によって開発された固相ペプチド合成技術は副甲状腺ホルモンのようなポリ
ペプチドの合成のために広く使用されかつ成功をおさめている。このストラテジ
ーは、ペプチドのカルボキシ末端アミノ酸を固相支持体に結合させることに基づ
いている。次に高収率で続く他のアミノ酸が付加される。N-末端α-アミノ基は
、固相からペプチドが除去されずに保護基が除去され得るように保護される。こ
こで用いる化学は、Fmocアプローチと称されるMerrifieldの原法の改変を含む。
Fmoc(フルオレニルメトキシカルボニル)基は温和なアルカリ条件で除去するこ
とができ、アルカリ安定側鎖保護基および支持体への結合に影響を与えない。こ
の技術は、E. AthertonとR.C. Sheppard, "Solid Phase Peptide Synthesis: a
Practical Approach", IRL Press, New York, N.Y.に記載されている。この文献
は引用により本明細書に取り込まれるものとする。
局所的、または直腸内または吸入または経口的デリバリーによって投与すること
ができる。本類似体は、1用量単位あたり約1〜約300mgを通常のビヒクル、賦
形剤、ビアンダー保存剤、安定化剤、色素、許容される医薬的実施例に必要なそ
の他の試薬と共に常法により非経口投与用形態に製剤化することが出来る。 非経口投与のためには、極細の30-ゲージ注射針を用いた1〜2mlの無痛皮下
注射が、疾病の重篤度に応じて毎日僅かに1回、1〜2年間必要であろう。注射
する材料は本発明の一つを水性、等張の滅菌溶液または懸濁液(場合によりフェ
ノールのような保存剤、またはエチレンジアミン四酢酸(EDTA)のような可溶化剤
を含んでもよい)中に含むであろう。使用できる許容可能なビヒクルおよび溶媒
には、水、温和に酸性化した水、リンゲル液および等張塩化ナトリウム溶液が含
まれる。加えて、滅菌した不揮発性油が溶媒又は懸濁媒体として慣習的に使用さ
れる。合成モノグリセリド、ジグリセリド、(オレイン酸のような)脂肪酸は注
射可能物の調製に不揮発性油としての用途が見いだせる。
コールのような非侵襲性賦形剤と混合することにより坐薬の形態で調製すること
ができる。 局所的使用のためには、本発明の類似体は軟膏、ゼリー、溶液、懸濁液、また
は皮膚接着性膏薬の形態で調製することができる。 吸入については、たとえば、PCT出願公開公報WO94/07514に記載された手段で
達成することができる。この文献は引用により本明細書に取り込まれるものとす
る。 毎日の用量は0.05mg/kg体重、すなわち、70kgの人では3.5mgを越えてはならず
、具体的な化合物の活性、年齢、体重、性別および治療する患者の状態に依存す
る。 単一投与量を作製するために担体材料と合わされる活性成分の量は治療される
患者および投与の特定のモードに依存するであろうことは言うまでもない。

に記載されたように合成した。環化すべき残基Glu-22およびLys-26はそれぞれア
リルエステルおよびアロック誘導体として直交的(orthogonally)に保護した。 環化は第17番残基の付加後に行った。産物をC18シリカカラム(Vydac)のクロ
マトグラフィーで0.1%トリフルオロ酢酸水溶液中のアセトニトリル勾配(1%/
分)で溶出し、97%より高度に精製した。観察された分子量を以下の表1に示す
。

Further Definition of the Protein Kinase C Activation Domain of the Para
thyroid Hormone. J. Bone Miner. Res. 1994, 9, 943-949に従って、24穴プレ

ち、OVX後、第8週の終わりまで)行う予防プロトコルを用いた。 実験の終わりに、大腿骨およびL5脊椎骨を麻酔した動物から取り出し洗浄した
。脊椎骨および大腿骨の遠位半分から骨端を除いた部分を酢酸緩衝化10%ホルマ
リンで1週間固定し、5%トリクロロ酢酸中で9〜12日間撹拌することによって
ミネラル除去し、脱水し、清澄化し、パラフィンワックス中に包埋した。脱水し
、清澄化し、パラフィン包埋したL5椎体の8μm厚連続横断切片、およびパラフ
ィン包埋した大腿骨の遠位半分の10μm厚縦断切片をライカRM-2035ミクロトーム
で切り出し、55℃にて2分間サンダーソン迅速骨染色液(Surgipath Medical Indu
stries社、Winnipeg, MB, Canada)で染色した。
約80の面積[μm2]/周囲長[μm] 測定値)を測定することによって評価した。小
柱の面積および周囲長はImaging Research社(St. Catherine's, ON, Canada)
のM4画像化システムで測定した。これらの解析に用いたソフトウエアは、Imagin
g Research社のモルフォメトリー第3.01.7版である。全ての組織形態測定は遠位

ys13-Glu17, Glu22-Lys26)hPTH(1-31)NH2の小柱生長に対する刺激能 ラットをOVX処置(OVX)または擬似OVX処置(擬似)し、OVX後第9週の終わりと
第15週の終わりの間にビヒクルの皮下注射を毎日行った。注射量は5nmol/体重
100gとした。OVX処置ラットには毎日以下を皮下注射で与えた:ビヒクル(OVX)、
hPTH(1-31)NH2、5nmol(直鎖)、[Leu27]シクロ(Glu22-Lys26)hPTH(1-28)NH2、
5nmol(環式)、または、 [Glu17,Leu27]シクロ(Lys13-Glu17, Glu22-Lys26)
hPTH(1-31)NH2、5nmol(二環式)。大腿骨を脱ミネラルし、サンダーソン迅速骨
染色で染色した。図8に見られるように、残存小柱の厚さを増加させることにつ
いて二環式類似体[Glu17,Leu27]シクロ(Lys13-Glu17, Glu22-Lys26)hPTH(1-31)
NH2はその単環式対応物である[Leu27]シクロ(Glu22-Lys26)hPTH(1-31)NH2よりも
良好であり、どちらも直線状分子であるhPTH(1-31)NH2よりも顕著に良好であっ
た。この改善は、相対AC刺激活性とは相関しておらず、タンパク質分解攻撃に対
するより大きな安定性および注射部位からのより大きな輸送され易さの結果であ
ろうと考えられる。 hPTH(1-28)NH2、hPTH(1-29)NH2およびhPTH(1-30)NH2の二環式類似体も直線状
または単環形態のものよりも改善された生物学的活性を有することが予測される
