Abstract
Description
法、更に、本発明の方法により得られた生成物の医薬的使用に関する。
ストレスを中和することで知られている。シャペロンの内もっとも解析されてい
るのは、分子量が70kDaである熱ショックタンパク質(HSP)群である(Multhoffら 、Cell Stress & Chaperones 1 (3) (1996)、167)。これらのタンパク質は、進 化過程において高率で保存されている。それらは、細胞内の折り畳まれていない
ポリペプチド又は間違って折り畳まれたポリペプチドへ結合し、それらを安定化
し、その結果それらの重合を阻害するか、又はトランスメンブラン転位を可能に
する。Hsp70は、核内、細胞質ゾル内及び特定の腫瘍細胞の細胞表面に位置する 。細胞内でのそのシャペロンとしての役割以外に、Hsp70ファミリーのメンバー は、免疫系の刺激、例えば、病原体の存在による炎症過程に関する役割、及び、
インビボ及びインビトロ細胞内抗腫瘍免疫反応に関する役割を担っているようで
ある。従って、熱ショックタンパク質の治療的使用の範囲は、従来技術において
示されている。WO 97/10000には、熱ショックタンパク質及び非共役結合でこの タンパク質と結合している外因抗原分子(ペプチド)からなる複合体の使用が、腫
瘍及び感染症の予防及び治療に関して記載されている。複合体に熱ショックタン
パク質と共に存在する抗原は、腫瘍細胞から採取される。それらは、同様の特徴
、即ち、共に免疫反応を誘発する。Multhoffら、Biol. Chem. 379 (1998)、295-
300は、NK細胞を含む非-MHC-制限エフェクター細胞の認識におけるHsp70を含むH
SPsの役割を挙げている。特に、NK細胞は、腫瘍細胞表面に存在するHsp70分子を
認識し、次いで腫瘍細胞を溶解することが見い出された。癌治療へのもう一つの
研究は、Tamuraとその共同研究者(Tamuralら、Science 278 (1997)、117-123)に
より紹介されている。彼らは、自己移植腫瘍から由来する腫瘍を有するマウスを
熱ショック調製物を用いて効果的に治療することが可能であることを証明した。
しかし、自己移植ではない腫瘍又は正常組織からの調製物は、腫瘍の退行を導く
ことはなかった(Blachereら、J. Exp. Med. 186 (1997) 1315-1322)。Tamuraら による研究においては、HSPsは、詳細に渡って識別されていない多くのペプチド
と共に複合体を形成する。これらをまとめると、従来技術は、ペプチドと複合体
を形成するか及び/又は腫瘍細胞のような細胞の表面に存在する場合、HSP分子 の免疫活性を示すと言える。従って、熱ショックタンパク質の潜在力が、様々な
疾患と戦うために基本的に認識されてはいるが、治療における有効な使用は、出
発材料(腫瘍材料)の分量と同様に、特定の複合体又は細胞調製物の調製に通常依
存している。これらの複合体又は細胞調製物の普遍的又は患者を選ばない使用を
想像することは難しい。

それらは、本発明によって使用されるHsp70又は上記フラグメント若しくは誘導 体により活性化することができることが予め必要となる。本発明によれば、単離
されたNK細胞を使用することができる。更に、NK細胞を含む抹消単核血液細胞(P
BMC)のような細胞混合物を使用することもできる。
同性」という用語は、2つのアミノ酸配列が整列される時に少なくとも70%のア
ミノ酸が同一であり、一方の整列された(aligned)アミノ酸配列はHsp70のC末 端領域の1つであることを意味する。更に、70%の同一アミノ酸を有するが、整
列された時にギャップ(gaps)によって、Hsp70のC末端参照配列と異なる配列も
含む。これらのギャップは、本発明によって使用される相同分子において、又は
参照分子において発生し得る。通常、コンピューターにより行われる比較による
整列は、従来技術において、そのような整列を行うことができるプログラムとし

最も驚くべきことであると思われる。例えば、1998年6月(この明細書においては
、専門家の意見として参照されている Srivastavaら、Immunity 8 (1998)、657-
665参照)では、単離された熱ショックタンパク質は、CTL誘導のような免疫的効 果を持たず( Blachereら、 J. Exp. Med. 186 (1997)、1315-1322)、かつ、数種
類の癌に対しては保護免疫を誘発することができない(UdonoとSrivastava、J. I
mmunol. 152 (1994)、5398-5403)と依然として推察されていた。本発明を用いて
、患者に左右されずにNK細胞のインビトロ又はインビボ活性化を誘発することが
可能になり、これは、様々な疾患、例えば、腫瘍疾患に対して効果的に使用する
ことができる。更に、単離された熱ショックタンパク質の使用は、活性化過程の
標準化を改善することを可能にする。本発明は、量についての制限なくHCPを生 成することを可能にするが、HSPペプチド複合体の患者特異的調製物の場合、HSP
量は腫瘍の大きさにより制限される。
発明の結果を導くことは驚くべきことである。特に、C末端がNK細胞に認識され 、かつ、その活性化を導く同一の三次元構造を明らかに有することは予測できな
かった。NK細胞活性化におけるHsp70 C末端フラグメントの使用の可能性は、更 に、組換え体生成が、全タンパク質の組換え体プレゼンテーション(presentati
on)と比較して収率が増加するという利点を有する。

ーター、lacI、lacZ、T3、T7、gpt等である。真核プロモーターは、例えば、CMV
イミディエットアーリープロモーター(CMV immediate early promoter)、HSV プロモーター、チミジンキナーゼプロモーター、SV40プロモーター、レトロウイ
ルス由来LTRs及びマウスメタロチオニンIプロモーターである。原核又は真核細
胞における発現のための様々な発現ベクター、例えば、真核生物pKK223-3 (Phar
macia Fine Chemicals、Uppsala、Sweden)又はGEM1 (Promega Biotec、Madison 、WI、USA)、pSV2CAT、pOG44及び原核生物pQE70、pQE60、pBluescript SK等につ
いては既に記載されている。プロモーター以外のこれらのベクターは、転写を更
に向上する因子、例えば、いわゆる転写エンハンサーも含むことができる。例と
しては、SV40エンハンサー、ポリオーマエンハンサー、サイトメガロウイルス
アーリープロモーターエンハンサー及びアデノウイルスエンハンサーを挙げるこ
とができる。従って、組換えタンパク質は、例えば、上記ベクターが使用される
場合、様々な原核及び真核ホスト細胞において発現される。そのようなホスト細
胞の例は、細菌細胞(例えば、大腸菌、ストレプトミセス、バシラス、ネズミチ

医薬的アジュバントのもう一つの好ましい態様においては、Hsp70タンパク質は 、ヒトHsp70のC末端フラグメント(アミノ酸384-561)又は本発明の効果を有す るもう一つのHsp70の対応する領域又はヒトHsp70のC末端フラグメント(アミノ 酸454-460)を含む。本発明によれば、7アミノ酸(NLLGRFE)からなるこの最小
配列を有するフラグメントは、抗体により阻害され、その結果NK活性化は妨げら
れるか又は停止することが驚くべきことに示された。この実験はMulthoffら、J.
Immunol. 158 (1997)、4341-4350に記載されているように行われた。Amersham から入手可能な抗体RPN 1197が使用された。前記7アミノ酸は、天然隣接(natura
lly flanking)Hsp70配列又は他のアミノ酸により隣接させる(flank)ことができ る。前記7アミノ酸は、その天然における状態に留まることが好ましい。他の隣 接する(flanking)アミノ酸が使用される場合、前記7アミノ酸が自然に有する三 次元状態が保存されることが好ましい。相同性が少なくとも70%に保たれる限り
、前記7アミノ酸内で、更に、アミノ酸交換を行うことができる。しかし、これ らの交換は、458の位置にあるアルギニンをリシンへ交換することを含まない。 この交換は、構造における変化を導く。従って、構造の変化を導く7アミノ酸領 域における交換は、求められる活性化の性質を有する場合にのみ本発明に含まれ
る。

/5 % FKSを用いて非特異的結合サイトをブロッキングした後、1:2及び2:1の比 でPBS/1 % FKS中に懸濁されたT及びNK細胞の混合物は、1時間室温で培養フラス コ内においてインキュベートされた。粘着性でない細胞は、インキュベイション
後、上清から採取された。粘着細胞は、氷冷PBS/10 % FKS溶液を用いた一連の洗
浄段階により採取された。一回の穏やかな洗浄段階を使用して、やや粘着性であ
る細胞を除去し、一方、高度粘着性細胞は更なる激しい洗浄段階により採取され
た。それぞれの段階において得られた細胞集団は、別々にカウントされ、フロー
サイトメトリー及び表現型により特徴づけられた。

メンバーである))が入った平らな底を有する96ウェルマイクロタイタープレート
(Greiner, Nurtingen、ドイツ)に入れられた。Hsp70タンパク質の様々な濃度(1
−200 μmg/ml)を試験することで、最終濃度100 μg/mlが刺激のためには理想 的であることが分かった。IL-2 (100 IU)に対する増殖活性は、更なる参照とし て、並行して決定された。24時間又は48時間のインキュベーション後、細胞は、 3 H-チミジン(1 μCi/ウェル)により標識され、全吸収は37℃で18時間のインキュ
ベーションの後、流体シンチレーションカウンター(Beckmann Instruments、Mu
nich、ドイツ)を用いて決定された。更に、内部参照(internal control)として
、増殖容量は同じドナーから採取したTリンパ球から決定された。投与量段階的 上昇実験(dose escalation examinations)における濃度1-200μg/mlの様々なHsp
70タンパク質/フラグメントの使用は、増殖容量の最大刺激は、100 μg/ml Hsp
70タンパク質により達成されることができることを示した。単離されたNK及びT 細胞の増殖活性は、rHsp70、DnaK、 Hsc70、 rHsp70-C term. 又は rHsp70homC
(アミノ酸384-562)を用いてインビトロ刺激された後に試験された。図1Aに見ら
れるように、rHsp70によるNK細胞の増殖は、顕著に刺激された。Hspのカルボキ シ末端領域による及びアミノ酸384-561を有するC末端ドメインにおいてHsp70と9
4%の相同性を有するrHsp70homCによる刺激も可能である。それに対して、DnaK 及びHsc70はNK細胞増殖を刺激しなかった。熱変性されたrHsp70は、その増殖に 対する刺激性を完全に失った。
sp70、 Hsc70及びrHsp70homC並びに熱変性されたrHsp70は、T細胞の増殖容量へ の効果を示さなかった(図1B)。

り選択的に刺激されることができた。
機能的分析により、Hsp70によるプラズマ膜発現はNK細胞に仲介される溶解に対 する感受性増加と関連していることが示された。NK細胞により仲介されたこの溶
解は、Hsp70のカルボキシ末端領域(アミノ酸504-617)に向けられたモノクローナ
ル抗体及び抗体RPN1197と共に腫瘍細胞系を予めインキュベートすることにより ブロックすることができる(1, 4)。次に自家由来のHsp70を発現する(CX+)及びHs
p70を発現しない(CX-)腫瘍細胞についてNK細胞の細胞溶解活性における組換えHs
p70タンパク質(rHsp70)の影響、が分析された。濃度5 μg/mlでHsp70タンパク質
と4日間予めインキュベートされた高度精製されたNK細胞を用いた細胞毒性試験 の結果は、図2A及びBにまとめられている。実験的なセットアップは、下記のと おりであった:細胞毒性活性刺激のためにNK細胞を10 μg/mlのrHsp70と共にイ ンキュベートした。刺激は、4日後に繰り返された。

特異的溶解のパーセントは、下記のように計算された:[(実験的放出−自然発生
的放出)−最大放出−自然発生的放出]×100。Cr-51の実験的放出は、全ての実験
において15%未満であった。
ベーションした場合、増殖及びHsp70発現腫瘍細胞(CX+)に対するNK細胞の細胞溶
解活性の両方が、刺激されることを示すことが可能であった。それに対して、rH
sp70により処理されていない同じドナーから採取されたNK細胞は、10日後にこの
反応性を失った(データは示されていない)。rHsp70により刺激されていないNK細
胞の溶解活性は、rHsp70により処理されかつ刺激されたNK細胞と比較して低く、
かつ、Hsp70を発現する及び発現しない腫瘍細胞の溶解においては、溶解性の顕 著な相違は見ることができなかった。
