細胞
6mM L−グルタミンを補充した10%熱不活化ウシ胎児血清(FCS、Life Technologies、Eggenstein、Germany)および抗生物質(100 IU/mlペニシリンおよび100μg/mlストレプトマイシン; Life Technologies)を含有するRPMI−1640培地(Life Technologies、Eggenstein、Germany)1mlあたり0.1〜0.5×106個の細胞密度で、NK細胞株YTを培養した。過渡的プラスチック接着性(transient plastic adherent)NK細胞は、健康なヒトボランティアのバフィーコートに由来するものである。フィコール分離した末梢血単核細胞(PBMC)をrIL−2(100 IU/ml、Chiron、Frankfurt、Germany)中で12時間培養した。修正されたVujanovic(Vujanovic ら、(1993) Cell. Immunol. 151、133)の方法にしたがって、単球が枯渇した末梢血リンパ球の接着選択を行った後、細胞を、Hsp70ペプチドTKD(2μg/ml)を補充したRPMI−1640培地中で3日間培養した。
ウシ血清アルブミン(BSA、1 mg/ml、Sigma-Aldrich、Steinheim、Germany)、1mg/mlの凍結乾燥した(lyophylize)組換えヒトHsp70タンパク質(Stressgen、British Columbia、Canada)または2mg/mlのHsp70ペプチドTKD(TKDNNLLGRFELSG、aa450-463、Bachem、Bubendorf、Switzerland)を、均衡化したAminoLinkアガロースビーズ(Pierce、Rockford、USA)(2ml)、還元剤NaCNBH3とともに6時間インキュベートした。BSA、Hsp70タンパク質およびHsp70ペプチドTKDの結合能は、95%であった。未結合の物質をトリス緩衝液で集中的に洗浄することによって除去し、未反応基をクエンチングした後、細胞溶解物をBSA、Hsp70タンパク質およびHsp70ペプチドTKD結合カラムに1時間適用した。
膜の精製は、低調の、プロテアーゼインヒビターPMSFを含有するEDTAフリーの緩衝液中で、50×106個の細胞のダウンスホモジナイゼーションで行い、続いて1,000gで5分間および100,000gで4℃で60分間、連続的に遠心分離を行った。膜を含有するペレットを50mMトリス緩衝液(0.5%、NP40、pH7.6)中2mlの0.3M NaClに再懸濁した。
脱脂乳(0.1%)中でのブロッキングおよびmAbを用いたグランザイムB 2C5(IgG2a、Becton Dickinson、Heidelberg、Germany)のインキュベーション(4℃、5時間)の後、ウェスタンブロットを洗浄し、二次マウス抗IgG HRP Ab(Dianova、Hamburg、Germany)とともに1時間インキュベートした。ECLキット(Amersham Bioscience)を用いて5秒間タンパク質の検出を行った。
Hsp70タンパク質およびHsp70ペプチドTKDが沈降した32kDaタンパク質バンドをクーマシーブルー染色ゲルから切り出し、トリプシンで消化し、逆相ZIPチップ(Millipore、Eschborn、Germany)を用いて脱塩した。そのサンプルを4−ヒドロキシ−α―シアノけい皮酸中に包埋し、Perseptive Voyager DePro MALDI-TOF(Matrix Assisted Laser Desorption Ionisation?Time Of Flight)マススペクトロメーターを用いて、反射モードでペプチド質量を測定した。ピークリスト(peaklist)をm/zソフトウェア(Proteometrics)を用いて編集し、ピーク選択に用いた。得られたペプチドマスフィンガープリントについて、Profound検索エンジン(Proteometrics)を用いて重複していないNCBIタンパク質データペースを検索した。グランザイムBは、100%の確率、>95%の 信頼度で同定された。
細胞(0.5×106)を10分間パラホルムアルデヒド中(PBS中1%PFA)に固定し、BSA(0.5%)、NaN3(0.1%)およびサポニン(0.1%)を含有するPBS中、透過処理した。次いで、透過処理した細胞を、グランザイムB−フィコエリトリン結合モノクローナル抗体HC2−PE(IgG1;Holzel Diagnostika、Cologne、Germany)、またはアイソタイプが一致するIgG1対照抗体のいずれかとともに、4℃で1時間、暗所でインキュベートした。細胞を洗浄後、FACSCalibur機器(Becton Dickinson、Heidelberg、Germany)で分析した。
カンプトテシン(4mg/ml、Sigma、Munich、Germany)の貯蔵液をDMSOで希釈し、4℃で暗所にて保存した。グランザイムB(6ng/ml、Holzel Diagnostics、Cologne、Germany)溶液を使用直前に新鮮に調製した。指数関数的に成長する細胞(0.5〜1.5×106/ml)を最終濃度4μg/mlのカンプトテシンで、または、精製された、酵素的に活性なグランザイムB(10ng/ml、1μg/ml、2μg/ml、4μg/ml)(Shiら、(2000) Methods in Enzymology 322、125)を用いて、10分間および30分間、4℃または37℃でインキュベートした。RPMI−1640培地中で洗浄後、結合と取り込みを、透過していないおよび透過した腫瘍細胞中において、フローサイトメトリーおよび蛍光顕微鏡検査法を用いて、40倍対物レンズと標準フィルターの備え付けられているAxioscop 25走査顕微鏡(Zeiss、Jena、Germany)を用いて測定した。画像を、ソフトウェアAxiovison (Zeiss Vison、Jena、Germany)を用いて乗法的シェイディング補正によって処理した。HC2−PE抗体を用いることによってグランザイムBを赤で視覚化した。
アポトーシスアッセイ
アネキシンV−FITC染色:簡単に、細胞を、5mM CaCl2を含有するHepes緩衝液で2回洗浄し、アネキシンV−FlTC(Roche)を用いて室温で10分間インキュベートした。アネキシンV−FITC陽性染色細胞を、FACSCaliburフローサイトメーター(Becton Dickinson、Heidelberg、Germany)で測定した。
腫瘍細胞株CX+/CX−およびColo+/Colo−を用いて、20:1〜2:1の間の異なるエフェクタ:標的細胞比(E:T)で、4時間コインキュベートした後、非刺激NK細胞(NK d0)とTKD刺激NK細胞(NK d3)とでグランザイムBの放出を比較した。検出には、グランザイムB ELISPOTキット(#552572、BD、Heidelberg、Germany)を用いた。簡単に、96ウェルELISPOTプレート(MAIPN45、Millipore)に4℃で一晩、捕捉抗体を塗布し、10%FCSを含有するRPMI−1640培養培地でブロックし、前記のようにして、腫瘍細胞とエフェクタ細胞を用いて37℃で4時間インキュベートした。脱イオン水および洗浄緩衝液AおよびBで洗浄後、ビオチン化抗グランザイムB抗体を2時間添加した(2μg/ml)。別の2つの洗浄ステップ後、新鮮に調製されたアビジン−西洋ワサビペルオキシダーゼ(2h)および基質溶液(インキュベート時間:25分)を添加することによって、グランザイムBを視覚化した。ImmunoSpot Series I Analyzerを用いて、スポットを自動的に計数した。
NK細胞媒介性細胞毒性を、12時間の51Cr放射性同位体アッセイによって測定した。標的細胞として、結腸カルシノーマサブラインCX+およびCX−を用いた。ブロッキング研究には、最終濃度5μg/1×106細胞で、mAb C92F3B1およびアイソタイプが一致する対照抗体(IgG1)を用いた。抗体とともに4℃で30分、CX+およびCX−標的細胞をインキュベートした後、細胞を51Crで標識し、MacDonald (MacDonaldら、(1974) J. Exp. Med. 140,718)に記載のようにして、細胞毒性アッセイを行った。特異的溶解のパーセンテージは、[(実験的放出−自発的放出)/(最大放出−自発的放出)]×100から計算した。
パートナータンパク質を、固定化されたヒトHsp70タンパク質(1mg)上で、または、NK細胞との相互作用を媒介することがわかっているHsp70の細胞外エピトープを含有する、14アミノ酸ペプチド、造られた(coined)TKD(TKDNNLLGRFELSG、aa450-463、2 mg)上で、アフィニティクロマトグラフィーによって同定した。このペプチドは、Hsp70特異的抗体(Welch and Suhan (1986) J. Cell. Biol. 103、2035)のエピトープ (Reinekeら、(1996) Immunobiol. 196、96)として予め同定されており、生育可能な腫瘍細胞上の膜結合Hsp70を特異的に検出するものである(Multhoffら、(1995) Int. J. Cancer 61,272; Multhoff ら、(1995) Blood 86, 1374)。NK細胞株YTの細胞溶解物(Drexlerら、(2000) Leukemia 14,777)を、固定化Hsp70またはTKDペプチドカラム上にて分画した。カラムに結合した材料を、3M塩化ナトリウムで5つの画分内に溶出した。対照として、YT細胞溶解物を担体またはBSA結合カラムに塗布した。さらに、非NK細胞株(K562)の溶解物をTKD結合アフィニティカラム上に分画した。溶出した画分をSDS−PAGE(10%)によって分離し、銀染色によって視覚化した。見かけの分子量が32kDaの、強いタンパク質バンドが、Hsp70タンパク質(Hsp70)およびHsp70ペプチド(TKD)カラム(図1A、YT)に由来するYT細胞溶出物の画分2(F2)および3(F3)に観察された。このバンドは、未結合セファローズカラム(データは示さず)やBSA結合カラムの溶出物には観察されず、また、K562細胞溶解物が載ったTKDアフィニティカラムからの溶出物にも観察されなかった(図1A)。Hsp70タンパク質カラムによっても同じ結果が得られた(データは示さず)。並行して、画分3(F3)に由来するHsp70ペプチドTKDおよびHsp70タンパク質の溶出物をSDS−PAGEによって分離し、クーマシーブルーで染色した。Hsp70ペプチドカラムのF3に由来する32kDaタンパク質バンドを切り出し、トリプシンで消化した(図1B)。得られたペプチドをMALDI-TOFペプチドマスフィンガープリンティング法で分析した。トリプシンペプチドの配列は、グランザイムBと100%のホモロジーを示し、推定Z値は1.89であり、グランザイムBについて95%を超える確率を示すものであった(図1B)。グランザイムBと32kDaタンパク質バンドとの同一性を、グランザイムB特異的抗体2C5(IgG2a)を用いたウェスタンブロット分析によってさらに確認した。Hsp70タンパク質(Hsp70)およびHsp70ペプチド(TKD)カラムから得られたYT細胞溶出物の両方には、強い32kDaグランザイムBタンパク質バンドが現れた(図1C)。グランザイムBは、Hsp70またはK562溶解物が載ったTKDアフィニティカラムに溶出した画分には検出されなかった(図1C)。グランザイムBに対するフィコエリトリン(PE)結合グランザイムB抗体(IgG1)を用いたフローサイトメトリーによって、YT細胞中においては、細胞質のグランザイムBの陽性染色が示されたが、K562では示されなかった(図1D)。これらの所見は、我々のこれまでの結果を裏付けるものである。要約すれば、これらのデータは、グランザイムBがHsp70の潜在的なパートナータンパク質であることを示している。グランザイムBは、TKDと名づけたHsp70のC末端領域と相互作用する可能性がある。
上記所見は、膜結合Hsp70によってグランザイムBが細胞質ゾルに特異的に結合し、またそこに入ることを可能とするかどうかという問題を提起している。したがって、パーフォリンを含まない、精製グランザイムBを、差異的(differential)Hsp70膜発現を示す腫瘍サブラインCX+/CX−およびColo+/Colo−とともにコインキュベートした。未処理のCX+およびColo+細胞(対照)の光学顕微鏡検査法(light microscopy)による分析を4℃と37℃で行い、各パネルの上の列に示している(図2A)。4℃と37℃で行った、細胞の対応する免疫蛍光顕微鏡検査法を、下に示している(対照)。最初、どの細胞においても、細胞表面上でも、細胞質内でも、グランザイムB染色は認められなかった。しかしながら、該細胞を精製グランザイムB(grB)とともに4℃で15分間インキュベートした後、典型的な膜表面染色を示すリング状の蛍光発光が、HsP70膜陽性CX+およびColo+腫瘍サブラインにおいて検出された(図2A,左のパネル)。CX+およびColo+腫瘍サブライン中の細胞質の染色パターンによって測定されるように、30分間のインキュベート中、4℃から37℃へ温度シフトを行った結果、グランザイムBの取り込みが認められた(図2A、右のパネル)。対照的に、Hsp70膜陰性対応物CX−およびColo−は、4℃でのグランザイムB細胞表面結合も、または37℃での取り込みも認められなかった(データは示さず)。透過した細胞のフローサイトメトリー分析から、細胞を1μg/mlグランザイムBとともに37℃で30分間コインキュベートした場合、Hsp70膜陽性CX+およびColo+において選択的に、グランザイムBのピークがかすかに右にシフトするが、CX−/Colo−腫瘍サブラインにおいてはしないことが明らかになった(図2B、上のグラフ)。2μg/mlおよび4μg/mlのグランザイムBを用いたコインキュベーション後、Hsp70の膜陽性腫瘍細胞(CX+/Colo+)中において、グランザイムB取り込みの用量依存的増加が検出された(図2B、下のグラフ)。しかしながら、最も高い濃度である4μg/mlにおいても、グランザイムBは、Hsp−70陰性腫瘍細胞(CX−/Colo−)と比較して、Hsp70膜陽性によってより顕著に内面化した。Hsp70によって形成された潜在的イオンチャネルは、Hsp70膜陽性腫瘍細胞における、グランザイムBの選択的取り込みのメカニズムにおいて役割を果たすかもしれない。実際に、Hsp70膜陽性(CX+)腫瘍サブラインの精製リン脂質に由来する小胞の取り込み後、特定のイオン電導経路(ion conductance pathway)が観察された。これは、Hsp70膜陰性(CX−)腫瘍から得られた細胞小胞においては認められなかった(データは示さず)。これらの結果に基づいて、グランザイムBのHsp70膜陽性腫瘍細胞への選択的取り込みを容易にするイオンチャネル活性について推定することができるかもしれない。
上記所見は、精製グランザイムBは、細胞表面にHsp70を提示する腫瘍細胞のアポトーシスを誘発することができるかどうかという問題を提起している。同一のMHCクラスI発現(Multhoffら、(1997) J. Immunol. 158,4341)を示す、Hsp70膜陽性(CX+/Colo+)および膜陰性(CX−/Colo−)結腸カルシノーマ細胞を、単離された、酵素的に活性なグランザイムB(6ng/ml)とともに4時間、12時間および24時間インキュベートした(Shiら、(2000) Methods in Enzymology 322、125)。アポトーシスの陽性対照として、全ての細胞タイプをトポイソメラーゼインヒビターカンプトテシンとともに、4μg/mlの濃度でインキュベートした。FACS(FACSCalibur、Becton Dickinson、Heidelberg、Germany)、DAPI染色およびミトコンドリア性チトクロームc放出によって測定される、アネキシンV−FITC染色によってアポトーシスを測定した。カンプトテシン(cam)またはグランザイムB(grB)とともに4時間インキュベートしたCX+およびCX−細胞においては、アポトーシスは検出されなかった(図3A)。カンプトテシンとともにインキュベートしたCX+およびColo+ならびにCX−およびColo−を、カンプトテシンとともに12時間および24時間インキュベートした後、アポトーシスを起こさせた。結腸カルシノーマサブラインCX+/CX−は、膵臓カルシノーマサブラインColo+/Colo−と比較して、カンプトテシン媒介性の細胞死からより保護さていれた。
未処理(対照)、またはカンプトテシン(4μg/ml)もしくはグランザイムB(6ng/ml)で24時間処理した後のCX+およびCX−腫瘍細胞中のヒトチトクロームcの定量的測定。データは、4つの独立した実験における中間値±標準偏差で表している。対照とは有意に異なる値(p<0.05)に*を付している。
我々の所見の生理学的役割を、未処置およびHsp70で刺激したヒトNK細胞を用いた機能的アッセイにおいて試験した。以前からNK細胞を10〜50μg/mlの濃度のHsp70タンパク質とともに、または等価のHsp70ペプチド濃度(0.2〜2.0μg/ml)とともにインキュベートすることによって、NK細胞のHsp70膜陽性腫瘍標的細胞に対する細胞溶解活性が増加することがわかっていた。同時に、C型レクチンレセプターCD94を活性化するキラー細胞の発現はアップレギュレートされた(Multhoffら、(1999) Exp. Hematology 27,1627; Grossら、(2002) submitted)。Hsp70は、抗体ブロッキング研究によって結論付けられているように(Multhoffら、(1997) J. Immunol. 158,4341; Multhoffら、(1995) Blood 86,1374)、NK細胞の腫瘍選択的認識構造として機能するにもかかわらず、NK細胞毒性のメカニズムは、依然として不明である。可能なメカニズムを解明するために、NK細胞をHsp70ペプチドTKD(2μg/ml)とともに3日間インキュベートした。3つの独立した実験において証明されたように、有意に上昇した細胞内グランザイムB発現が観察された。対照的に、グランザイムB発現は、Hsp70ペプチドTKDで処理したCD3陽性T細胞においては増加しなかった。Hsp70膜陽性CX+およびHsp70膜陰性CX−とともにコインキュベートしたHsp70ペプチド活性化NK細胞の光顕微鏡検査による分析を図4Aに示す。Hsp70膜陽性CX+およびHsp70膜陰性CX−腫瘍細胞(0.1×106個/ml)を24ウェルプレートにて2日間、複製培養した。両腫瘍細胞株の増殖率は、同じ細胞数(0.3×106個/ml)であることが測定されているように類似していた。上のパネルのCX+およびCX−腫瘍細胞は、NK細胞不存在下で培養し、下のパネルの腫瘍細胞は、Hsp70ペプチドTKD(2μg/ml、3日)で刺激されたNK細胞とともに12時間共培養した。CX+細胞コロニーのほぼ100%がNK細胞を有するクラスター中で認められ、CX+腫瘍細胞の生存能力は減少していると思われる。対照的に、CX−腫瘍細胞およびNK細胞はクラスターには認められなかった。Hsp70膜陰性CX−腫瘍細胞は、NK細胞を引き付けなかった。NK細胞との接触後、CX−腫瘍細胞の細胞生存能力は、CX+腫瘍細胞と比較して、あまり影響を受けなかった。各グラフの右下の角の挿入図は、1つの代表的な細胞コロニーを倍率2.5倍で示した図である。

- ナチュラルキラー(NK)細胞中のグランザイムBの発現を誘導または促進する方法であって、NK細胞を、
(a)Hsp70タンパク質;

(c)アミノ酸配列TKDNNLLGRFELSGを含む、(ポリ)ペプチド;または、
(d)(a)、(b)および/または(c)の組み合わせ

- 前記腫瘍細胞は、胃、腸、結腸直腸、肺、膵臓、乳房、産婦人科系、頭頚部の腫瘍、皮膚の腫瘍(例えば、メラノーマ)、ニューロン性腫瘍、白血病およびリンパ腫からなる群から選択される、請求項12〜15、21もしくは22のいずれか1項に記載の使用、または請求項16〜22のいずれか1項に記載の方法。
- 前記ウイルス感染は、HIVまたは肝炎ウイルスによる感染である、請求項12〜15、21または22に記載の使用、または請求項16〜22のいずれか1項に記載の方法。
