Claims (1)
- 【特許請求の範囲】 1.標的となる一本鎖核酸配列と核酸捕獲部分の交雑を促進する方法であって 、 a) 末端塩基及び隣接する一本鎖領域を備えた二本鎖領域を持つ単分子 核酸捕獲部分を提供し、この二本鎖領域の末端塩基は二重鎖結合リガンドの結合 部位の全て又は部分であり、この一本鎖領域は前記標的核酸と交雑することが可 能であり、 b) 前記標的一本鎖核酸と、前記核酸捕獲部分と、二重鎖結合リガンド とを含む反応混合物を、前記標的一本鎖核酸と前記核酸捕獲部分が交雑できる条 件で形成し、 c) 前記二重鎖結合リガンドが形成された分子間二重鎖を結合するよう に、前記標的一本鎖核酸を前記核酸捕獲部分と交雑させる段階からなり、 よって、前記一本鎖核酸と前記核酸捕獲部分の交雑が促進される、方法。 2.前記標的一本鎖核酸と前記核酸捕獲部分の一本鎖領域によって形成される 二重鎖の第1塩基対は、前記二重鎖結合リガンドの結合部位の一部を符号化する 、請求項1に記載の方法。 3.前記核酸捕獲部分は核酸ヘアピンを含む、請求項1に記載の方法。 4.前記核酸捕獲部分は固体の支持体に結合される、請求項1に記載の方法。 5.前記核酸捕獲部分の末端塩基と前記標的一本鎖核酸の末端塩基との間の塩 基が重なった状態は前記分子間二重鎖の安定性を増加させる、請求項1に記載の 方法。 6.標的となる一本鎖核酸配列を検出する方法であって、 a) A−B−C−D構造を含む核酸捕獲部分を提供し、 Aが核酸配列であり、 B及びDが、互いに交雑して分子内二重鎖を形成する能力が有る核酸配列 で、 Cは、リンカであり、 Aは、標的一本鎖核酸配列に対して実質的に相補的であり、 b) 前記標的一本鎖核酸と前記核酸捕獲部分とを含む反応混合物を、前 記標的一本鎖核酸と前記核酸捕獲部分が交雑できる条件で形成し、 c)前記標的核酸と前記核酸捕獲部分との交雑が存在するか、存在しない かを検出する段階からなり、 よって、前記標的一本鎖核酸が検出される、方法。 7.Dの末端塩基は二重鎖結合リガンドの結合部位の全て又は部分を含む、請 求項6に記載の方法。 8.前記反応混合物は、二重鎖結合リガンドを更に含む、請求項6に記載の方 法。 9.前記核酸捕獲部分は不溶性の支持体上に固定される、請求項6に記載の方 法。 10.前記二重鎖結合リガンドは如何なる二重鎖も共有結合的に変更しない、 請求項8に記載の方法。 11.前記核酸捕獲部分の末端塩基と前記標的一本鎖核酸の末端塩基との間の 塩基が重なった状態は前記分子間二重鎖の安定性を増加させる、請求項6に記載 の方法。 12.標的となる一本鎖核酸配列を検出する方法であって、 a) A−B−C−D構造を含む核酸捕獲部分を提供し、 Aが核酸配列であり、 B及びDが、互いに交雑して分子内二重鎖を形成する能力がある核酸配列 で、 Cは、リンカであり、 Aは、標的一本鎖核酸配列に対して実質的に相補的であり、 b) 前記標的一本鎖核酸と前記核酸捕獲部分とを含む反応混合物を、前 記標的一本鎖核酸と前記核酸捕獲部分が交雑して、分子間二重鎖を形成できる条 件で形成し、この分子間二重鎖は二重鎖結合リガンドの結合部位を含み、 c)前記標的核酸と前記核酸捕獲部分との交雑が存在するか、存在しない かを検出する段階からなり、 よって、前記標的一本鎖核酸が検出される、方法。 13.前記標的一本鎖核酸と前記核酸捕獲部分との交雑は、ニックのある二重 鎖を形成する、請求項12に記載の方法。 14.前記二重鎖結合リガンドの結合部位はニックを含ない、請求項12に記 載の方法。 15.前記反応混合物は、二重鎖結合リガンドを更に含む、請求項12に記載 の方法。 16.前記二重鎖結合リガンドは配列特異的である、請求項15に記載の方法 。 17.前記二重鎖結合リガンドは如何なる二重鎖も共有結合的に変更しない、 請求項15に記載の方法。 18.前記核酸捕獲部分の末端塩基と前記標的一本鎖核酸の末端塩基との間の 塩基が重なった状態は前記分子間二重鎖の安定性を増加させる、請求項12に記 載の方法。 19.A−B−C−D構造を含む核酸捕獲部分であって、 Aが核酸配列であり、 B及びDが、互いに交雑して分子内二重鎖を形成する能力がある核酸配列 で、 Cは、リンカであり、 前記核酸捕獲部分は固体の支持体に結合されている、核酸捕獲部分。 20.Aは標的核酸配列に対して実質的に相補的である、請求項19に記載の 核酸捕獲部分。 21.前記核酸捕獲部分の末端塩基と前記標的一本鎖核酸の末端塩基との間の 塩基が重なった状態は分子間二重鎖の安定性を増加させる、請求項19に記載の 核酸捕獲部分。 22.前記標的核酸配列と前記核酸捕獲部分との交雑により形成される二重鎖 が、所定の二重鎖結合リガンドに対する結合部位を含むように前記核酸捕獲部分 を選択する、請求項19に記載の核酸捕獲部分。 23.標的となる一本鎖核酸配列と核酸捕獲部分の交雑を促進する方法であっ て、 a) 二本鎖領域及び隣接する一本鎖領域を持つ単分子核酸捕獲部分を提 供し、この一本鎖領域は前記標的核酸と交雑する能力があり、 b) 前記標的一本鎖核酸と、前記核酸捕獲部分と、二重鎖結合リガンド とを含む反応混合物を、前記標的一本鎖核酸と前記核酸捕獲部分が交雑できる条 件で形成し、 c) 分子間二重鎖を形成するように、前記標的一本鎖核酸を前記核酸捕 獲部分と交雑させ、この分子間二重鎖は二重鎖結合リガンドの結合部位を含み、 それにより二重鎖結合リガンドが二重鎖結合リガンドの結合部位に結合する、段 階からなり、 よって、前記標的一本鎖核酸配列と前記核酸捕獲部分の交雑が促進される 、方法。 24.前記核酸捕獲部分は、核酸ヘアピンを含む、請求項23に記載の方法。 25.前記核酸捕獲部分は固体の支持体に結合される、請求項23に記載の方 法。 26.前記二重鎖結合リガンドは如何なる二重鎖も共有結合的に変更しない、 請求項23に記載の方法。 27.前記二重鎖結合リガンドの結合部位はニックを含ない、請求項23に記 載の方法。 28.前記核酸捕獲部分の末端塩基と前記標的一本鎖核酸の末端塩基との間の 塩基が重なった状態は前記分子間二重鎖の安定性を増加させる、請求項23に記 載の方法。 29.反応混合物であって、 固定されたヘアピンと、 不溶性の支持体と、 標的核酸と、を含む反応混合物。 30.前記標的核酸配列の前記ヘアピンとの交雑により形成される二重鎖が、 所定の二重鎖結合リガンドに対する結合部位を含むように、前記ヘアピンが選択 される、請求項29に記載の反応混合物。 31.前記反応混合物は二重鎖結合リガンドを更に含む、請求項29に記載の 反応混合物。 32.前記二重鎖結合リガンドは、配列特異的である、請求項30に記載の反 応混合物。 33.標的核酸の検出用のキットであって、 固定されたヘアピンと、 不溶性の支持体と、 核酸標準物質とを含むキット。 34.二重鎖結合リガンドを更に含む、請求項33に記載のキット。
