(1996年)840〜847ページ)中に、フーリエ変換赤外分光法(FTI
R)によるタンパク質アビジンとビオチンおよびビオチン−脂質の複合体の構造
を解明する研究を記載する。このために、まず重水(D2O)中でアビジンのF TIRスペクトルを撮影した。引き続き、アビジンとビオチンまたはビオチン−
脂質を溶媒としてのD2O緩衝液と混合し、室温で数時間保存し、これにより明 らかに生じるアビジン複合体のできるだけ高い収率が得られるとされる。複合体
から再度FTIRスペクトルを撮影した。アビジンのスペクトルおよびアビジン
複合体のスペクトルから、差スペクトル(Differenzspektren
)を形成した。この論文はアビジン複合体の構造とだけ関係するので、時間従属
スペクトルは撮影されない。記載されたFTIRスペクトルはすべての場合に平
衡状態を反映する。結合した通常水素の振動スペクトルは撮影されないで、−よ
り高い質量のためずれて−結合した重水素の振動スペクトルが撮影される。その
結果、比較的厚いキュベット(50μm)を使用することができる。化合物、た
とえばタンパク質が配位位置で特定の配位子と複合体を形成できるか否かの問題
は、アビジンがビオチンと複合体を形成する事実は既に公知であったので、この
論文においては重要ではない。

時間的範囲内でD2O緩衝液中で撮影した、その際光学的層厚さは50μmであ る。12C=O−シンモイル−キモトリプシンおよび13C=O−シンナモイル
−キモトリプシンのスペクトルから、差スペクトルが形成される。“結論”にお
いて、(非重水素化)水の使用を許容する“ストップドフロー”IR透過キュベ
ットの構成は、1640cmー 1における水の強い吸収が5μmの層厚さを要求す
る(論文中では誤って5mmが記載される)ので不可能である。

制作用を有するか否かがテストされる。テストすべき試薬の数は一般に非常に大
きく;5桁、6桁またはむしろ7桁であってもよいので、迅速な自動的スクリー
ニング法の使用が決定的に経済的に重要である。
光計の高い走査速度および本発明による方法に必要な僅かな需要時間のため、多
数の試薬をテストするための前提条件が創造されている。方法のもう1つの重要
な利点は、普通に使用される標準微量滴定板(Standard−Mikrot
iterplatten)を問題なく使用することができることである。傾向は
再び約384個またはむしろ864個の、再び約100μlないしは約50μl
の僅かな体積を有する細孔を備えるより小さい微量滴定板に向っているので、本
発明による方法および本発明による装置の場合、必要な試料の僅かな量がとくに
重要である。他の重要な利点および方法マススクリーニング法の適性の基礎は、
重水素化水を使用する必要がないことおよび装置は構造的変更なしに多数の調査
に使用することができることである。

める代わりに迂回させて、IRキュベットを通過する流れを止める。これにより
、ポンプは保護され、迅速な応答時間が生じる。さらに、これによりIRキュベ
ットの入口管および出口管が短絡され、このため過圧は迅速に補償され、層厚さ
の変化はIRキュベットのアーチ形のため除去される。

- 【請求項1】 複合体を形成する少なくとも2つの反応物を互いに混合し、
混合物中のまだ反応しない個々の反応物のIRスペクトルを第一の時点で撮影し
、第二の時点で少なくとも1つの他のIRスペクトルで反応物の複合体を把握し
、異なる時点で撮影した2つのIRスペクトルから差スペクトルを形成し、その
差スペクトルがバンド構造を有する反応物を作用物質として選択する、作用物質
の確認方法。 - 【請求項2】 第一の時点として反応物を混合した直後の時点を選択する、
請求項1記載の方法。 - 【請求項3】 反応物の少なくとも1つが低分子量化合物である、請求項1
または2記載の方法。 - 【請求項4】 反応物の少なくとも1つがタンパク質である、請求項1から
3までのいずれか1項記載の方法。 - 【請求項5】 反応物の少なくとも1つがDNS分子である、請求項1から
4までのいずれか1項記載の方法。 - 【請求項6】 第一の時点および第二の時点の間に1ミリ秒から1日までの
範囲の待ち時間が存在する、請求項1から5までのいずれか1項記載の方法。 - 【請求項7】 方法を順次にまたは平行に多数の反応物を用いて実施し、そ
の際反応物は微量滴定板中に存在する、請求項1から6までのいずれか1項記載
の方法。 - 【請求項8】 IRスペクトルの測定を1〜25μmの層厚さで行う、請求
項1から7までのいずれか1項記載の方法。 - 【請求項9】 請求項1記載の方法を実施する装置において、 −第一のポンプ(6)で第一の試料ループ弁(3)が第一の反応物で充填可能で
ありおよび −第二のポンプ(7)で第二の試料ループ弁(4)が少なくとも1つの第二の反
応物で充填可能であり、 −試料ループ弁(3、4)は、有機化合物および試薬が密閉空間中に包含できる
ように互いに接続可能であり、および −有機化合物および試薬は第三のポンプ(8)で混合し、密閉空間からIRキュ
ベット(2)中へ供給させる、作用物質の確認装置。
