Claims (1)
- 【特許請求の範囲】 1.細胞サンプルを採取し、 そのサンプル内の細胞核を識別するためにサンプルを染色し、 デジタルCCDカメラおよびプログラム可能なスライドステージを含むタイプ のデジタル顕微鏡により細胞サンプルの映像を獲得し、 映像の焦点を結び、 その映像中の対象物を識別し、 各対象物について1組の特徴値を計算し、 特徴値を解析して、各対象物が悪性腫瘍に関連した変化を有する細胞核である かどうかを決定するステップを含んでいる細胞サンプル中の悪性腫瘍に関連した 変化の検出方法。 2.細胞映像中の各対象物は、強度値をそれぞれ有する画素のグループを含んで おり、映像中の対象物を識別するステップは、 映像中の全ての画素のヒストグラムを計算し、 映像中の背景画素の平均強度を決定し、 映像中の対象物画素の平均強度を決定し、 背景画素の平均強度と対象物画素の平均強度との間にあるしきい値を決定し、 しきい値より大きい強度を有する全ての画素を背景として識別し、しきい値以 下の強度を有する全ての画素を対象物として識別するステップをさらに含んでい る請求項1記載の方法。 3.各対象物は、背景から対象物を分離するエッジを有しており、細胞映像中の 対象物を識別するステップは、 対象物のエッジを拡張して環状リングの外側エッジを限定し、対象物のエッジ を破壊して環状リングの内側エッジを限定することによって対象物を包囲する環 状リングを生成し、 その環状リンクにおける各画素のグラジエント値を計算し、 その環状リングが対象物を囲む単一の画素チェーンを含むまで、低いグラジエ ント値を有する画素を環状リングから除去するステップを実行することによって 対象物のエッジを識別するステップをさらに含んでいる請求項1記載の方法。 4.1組の特徴値中の1以上の特徴を計算するステップは、 特徴を計算する前に、対象物の本当のエッジを拡張または縮小するステップを さらに含んでいる請求項3記載の方法。 5.映像は、コンピュータシステムのメモリに記憶され、 異なるステージ位置において一連の映像を獲得することによって各対象物の局 部的焦点を調節し、 各対象物について、対象物の焦点が最良である映像を選択し、 その最良の焦点の各対象物の映像をコンピュータのメモリに重書きするステッ プをさらに含んでいる請求項1記載の方法。 6.デジタル顕微鏡中の空白スライドから得られたテスト映像を読込み、 テスト映像の各画素の強度値を細胞サンプルの映像中の対応した画素から減算 し、 テスト映像中の画素の平均強度を決定することによって映像のデジタル顕微鏡 の光源の照明強度の変化を補償し、 細胞サンプルから得られた映像の各画素に平均強度を加算するステップをさら に含んでいる請求項5記載の方法。 7.対象物の近くの画素のグループに対する平均画素強度を決定し、 対象物の近くの画素のグループに対する平均画素強度を対象物に含まれている 各画素から減算することによって対象物の周囲の局部吸光度を補償するステップ をさらに含んでいる請求項6記載の方法。 8.対象物の近くの画素のグループは、対象物の面積より少し大きい面積を有す る正方形の境界線によって限定される請求項7記載の方法。 9.各対象物について面積、形状および光密度を計算することによって映像から 人工物を除去し、 2,000平方ミクロンより大きい面積を有する対象物、ならびに4より大き い形状または球形度と、1cより大きい光密度を有する対象物とを映像から除去 するステップをさらに含んでいる請求項1記載の方法。 10.悪性腫瘍に関連した変化を有する細胞核を悪性腫瘍に関連した変化を有し ない核から分離するために使用される特徴は、 (1)面積 (2)平均半径 (3)OD分散 (4)OD非対称性(skewness) (5)範囲平均 (6)OD最大値 (7)光スポットの密度 (8)低DNA面積 (9)高DNA面積 (10)低DNA量 (11)高DNA量 (12)高平均距離 (13)中/高平均距離 (14)相関 (15)均質性 (16)エントロピー (17)フラクタルディメンション (18)DNAインデクス (19)ラン0パーセント (20)ラン45パーセント (21)ラン90パーセント (22)ラン135パーセント (23)中間調レベル0 (24)中間調レベル45 (25)中間調レベル90 (26)中間調レベル135 (27)ラン長0 (28)ラン長45 (29)ラン長90 (30)ラン長135 を含むグループから選択される請求項1記載の方法。 11.患者から見掛け上正常な細胞のサンプルを採取し、 (1)サンプル中の細胞の核を染色し、 (2)デジタル顕微鏡で細胞の映像を獲得して、その映像をコンピュータシ ステムに記録し、 (3)記録された細胞の映像を解析して、核を識別し、 (4)サンプル中で見出だされた各核について1組の特徴値を計算し、その 特徴値から、その核が悪性腫瘍に関連した変化を示しているかどうかを決定する ことによってサンプル中の細胞が悪性腫瘍に関連した変化を示しているかどうか を決定し、 悪性腫瘍に関連した変化を示しているサンプル中の核の総数を決定して、その 個数から、患者が癌になるかどうかを予測するステップを含んでいる患者の癌の 進行を予測する方法。
