- 【特許請求の範囲】 1.静止した内皮細胞には影響を与えずに、脈管形成を阻害するかあるいは脈管 形成を促進する、脈管形成過程に関与する内皮細胞を含む病状の処置のための医 薬の調製のための、あるいは、脈管形成に関与する内皮細胞を含む病状の診断の ための製品の調製のための、抗イディオタイプ血管内皮細胞成長因子抗体の使用 。 2.KDRレセプターの選択的刺激により脈管形成内皮細胞を含む病状の処置の ための医薬の調製のための抗イディオタイプ血管内皮細胞成長因子抗体の使用。 3.静止した内皮細胞には影響を与えずに、脈管形成を阻害する、脈管形成過程 に関与する内皮細胞を含む病状の処置のための医薬の調製のための、請求項1ま たは2記載の抗イディオタイプ血管内皮細胞成長因子抗体の使用であって、その 抗イディオタイプ抗体が、その機能がタンパク質の翻訳をブロックすることであ るトキシンに結合するか、あるいはその抗イディオタイプ抗体がFabフラグメ ントの形態である、使用。 4.静止した内皮細胞には影響を与えずに、脈管形成を促進する、脈管形成過程 に関与する内皮細胞を含む病状の処置のための医薬の調製のための、請求項1ま たは2記載の抗イディオタイプ血管内皮細胞成長因子抗体の使用。 5.抗体が、サポリンおよびリシンまたは放射活性元素、例えば、ヨウ素−12 5または−131から選択されるトキシンに結合している、請求項3記載の抗イ ディオタイプ血管内皮細胞成長因子抗体の使用。 6.瘢痕形成または卵巣の黄体の成熟過程において血管形成のスピードを増加さ せる、生理学的脈管形成を刺激するための、あるいは血管血栓症の間に虚血にさ れた領域を再潅流するために血管の閉塞性の異常の過程で脈管形成を刺激するた めの医薬の調製のための抗−イディオタイプ血管内皮細胞成長因子抗体の使用。 7.トキシンと結合した抗イディオタイプ抗体または抗イディオタイプ抗体のF abフラグメントの、脈管形成の阻害を必要とする病状、例えば、癌、糖尿病性 網膜症および角膜移植の拒絶、の処置のための医薬の調製のための使用。 8.脈管形成過程に関与する内皮細胞を含む病状の診断のための製品の調製のた めの、請求項1記載の抗イディオタイプ血管内皮細胞成長因子抗体の使用。 9.ヒトKDRレセプターの、あるいはネズミflk-1レセプターのリガンドであ り、かつfltのリガンドではないことを特徴とする、抗イディオタイプ血管内皮 細胞成長因子抗体。 10.以下の性質: −KDRに対して特異的である、 −循環している、 −約23日の、特に約21日の、そしてさらに特定すると22.5日 の半減期を有する、 −200kDaのタンパク質のチロシンにリン酸化を誘導する、 −血管内皮細胞の増殖を誘導する、 −内皮細胞の移動を誘導しない、 −脈管形成を刺激する、 −低血圧を引き起こさない、 −血管透過性に影響を与えない、 を有することを特徴とする、抗イディオタイプ血管内皮細胞成長因子抗体。 11.請求項9記載の抗イディオタイプ抗体のFabフラグメント。 12.請求項9記載の抗イディオタイプ抗体と、特にサポリンおよびリシンから 選ばれるトキシンとの間の複合体、または請求項9記載の抗イディオタイプ抗体 とヨウ素−125または−131のような放射活性元素との間の複合体。 13.以下の方法: −精製したVEGFを動物、特にウサギに注射する、 −血液を抜き取って特定の抗−VEGF IgGを含む精製Igを、 例えば、プロテインAに対するアフィニティークロマトグラフィーで回収し、次 いで、可能な段階で、その特定の抗−VEGF IgGを精製Igから、例えば 、VEGFに対するアフィニティークロマトグラフィーにより精製する、 −上記の精製Igまたは上記の精製抗−VEGF IgGを、VEG Fの注射に用いたのと同じ種の動物に、特にVEGFの注射に用いたのと同じ起 源のウサギの膝窩神経節に注射する、 −血液を抜き取り、全Igを、例えば、プロテインAにより回収し、 次いで、その全Igを2つの免疫吸着に付す: ・抗−VEGF IgGを産生し、抗−アロタイプもしくは アイソタイプ抗体を排除するために用いたウサギの予備免疫 Igを用いて調製したアフィニティーカラムでの免疫吸着、 ・抗イディオタイプを精製するために抗−VEGF IgG を用いて調製したアフィニティーカラムでの免疫吸着 によって得られ得る請求項9記載の抗−イディオタイプ抗体。 14. −精製VEGFを動物、特にウサギに注射する、 −血液を抜き取って特定の抗−VEGF IgGを含む精製Igを、 例えば、プロテインAに対するアフィニティークロマトグラフィーで回収し、次 いで、可能な段階で、その特定の抗−VEGF IgGを精製Igから、例えば 、VEGFに対するアフィニティークロマトグラフィーにより精製する、 −上記の精製Igまたは上記の精製抗−VEGF IgGを、VEG Fの注射に用いたのと同じ種の動物に、特にVEGFの注射に用いたのと同じ起 源のウサギの膝窩神経節に注射する、 −血液を抜き取り、全Igを、例えば、プロテインAにより回収し、 次いで、その全Igを2つの免疫吸着に付す: ・抗−VEGF IgGを産生し、抗−アロタイプもしくは アイソタイプ抗体を排除するために用いたウサギの予備免疫 Igを用いて調製したアフィニティーカラムでの免疫吸着、 ・抗イディオタイプを精製するために抗−VEGF IgG を用いて調製したアフィニティーカラムでの免疫吸着 を特徴とする、請求項9記載の抗イディオタイプ抗体の調製方法。 15.活性物質として、請求項9または10記載の抗イディオタイプ抗体、また は請求項11記載のFabフラグメント、または請求項12記載の複合体を含む ことを特徴とする、医薬組成物。
