Claims (1)
- 【特許請求の範囲】 1.p110δをコードする、精製及び単離されたポリヌクレオチド。 2.前記ポリヌクレオチドがDNAである請求の範囲第1項記載のポリヌクレオ チド。 3.前記ポリヌクレオチドが、ゲノミックDNA、cDNA及び化学合成されたDNAよ りなる群から選択される請求の範囲第1項記載のポリヌクレオチド。 4.配列番号:1で示されるDNAを含む請求の範囲第2項記載のポリヌクレオ チド。 5.前記ポリヌクレオチドがRNAである請求の範囲第1項記載のポリヌクレオ チド。 6.請求の範囲第2項記載のDNAを含むベクター。 7.前記DNAが、発現制御DNA配列と作動可能に連結される請求の範囲第6項記 載のベクター。 8.請求の範囲第2項記載のDNAを用いて、安定に形質転換またはトランスフ ェクトされた宿主細胞。 9.脂質キナーゼをコードするポリヌクレオチドであって、 配列番号:1のポリヌクレオチドとストリンジェントなハイブ リダイゼーション条件下でハイブリダイズするポリヌクレオチド。 10.p110δを生産する方法であって、以下の工程すなわち、好適な普通培地に て請求の範囲第8項記載の宿主細胞を生育し、そして該細胞またはその生育培地 から、発現されたポリペプチドを単離する工程を含む方法。 11.配列番号:2のp110δアミノ酸配列を含む、精製及び単離されたポリペプ チド。 12.p110δに特異的な免疫反応性を有する抗体物質。 13.前記抗体がモノクローナル抗体である請求の範囲第12項記載の抗体物質 。 14.請求の範囲第13項記載のモノクローナル抗体を生産するハイブリドーマ 細胞系。 15.ハイブリドーマ細胞系208F(HB12200)。 16.請求の範囲第15項記載のハイブリドーマ細胞系によって生産されるモノ クローナル抗体。 17.前記抗体物質がヒト化抗体である請求の範囲第12項記載の抗体物質。 18.p110δのモジュレーターである化合物を同定する方法で あって、以下の工程すなわち、 a)化合物の非存在下及び存在下に、p110δのキナーゼ活性を定量し、 b)工程(a)で観察されるキナーゼ活性を比較し、ならびに c)該化合物の存在下と非存在下とでキナーゼ活性の差が観察される場合に、 p110δのモジュレーターとして化合物を同定する工程を含む方法。 19.請求の範囲第18項記載の方法によって同定される、 p110δキナーゼ活性のモジュレーター。 20.生物学的試料中のp110δの存在を判定する方法であって、以下の工程すな わち、 a)生物学的試料にp110δ特異抗体を曝し、及び b)生物学的試料中のp110δへの、p110δ特異抗体の結合を検出する工程を含 む方法。 21.配列番号:2で示されるp110δアミノ酸配列の一部またはすべてをコード する、検出可能に標識付けされたポリヌクレオチドを含む診断試薬。 22.p110δと結合パートナーとの間の結合のモジュレーターである化合物の同 定方法であって、以下の工程すなわち、 a)化合物の非存在下及び存在下に、p110δと結合パートナーとの間の結合の レベルを定量し、 b)工程(a)で観察される結合のレベルを比較し、ならびに c)該化合物の存在下と非存在下とで結合の差が観察される場合に、p110δと 結合パートナーとの間の結合のモジュレーターとして化合物を同定する工程を含 む方法。 23.前記結合パートナーがp85である、請求の範囲第22項記載の方法。 24.前記結合パートナーがRasである、請求の範囲第22項記載の方法。 25.前記結合パートナーが成長因子受容体である、請求の範囲第22項記載の 方法。 26.請求の範囲第22項記載の方法によって同定される、 p110δ結合のモジュレーター。
