Claims (1)
- 【特許請求の範囲】 1. 場合によって医薬上許容し得る組成物中の、0B受容体タンパク質を含む OB受容体タンパク質製剤であって、該OB受容体タンパク質が、配列番号1の アミノ酸配列の一部又は全部を有し且つ天然型OB受容体タンパク質の生物学的 性質を1種以上有することを特徴とするOB受容体タンパク質製剤。 2. 場合によって医薬上許容し得る組成物中の、OB受容体タンパク質を含む OB受容体タンパク質製剤であって、該OB受容体タンパク質のアミノ酸配列が 、(配列番号1として記載されている)以下のアミノ酸配列: (a) 1−896; (b) 22−896(場合により、N−末端メチオニン残基を含む); (c) 23−896(場合により、N−末端メチオニン残基を含む); (d) 29−896(場合により、N−末端メチオニン残基を含む); (e) 1−839; (f) 22−839(場合により、N−末端メチオニン残基を含む); (g) 29−839(場合により、N−末端メチオニン残基を含む); (h) 1−841; (i) 22−841(場合により、N−末端メチオニン残基を含む); (j) 23−841(場合により、N−末端メチオニン残基を含む); (k) 29−841(場合により、N−末端メチオニン残基を含む); (l) 1−891; (m) 22−891(場合により、N−末端メチオニン残基を含む); (n) 23−891(場合により、N−末端メチオニン残基を含む); (o) 29−891(場合により、N−末端メチオニン残基を含む); (p) さらに、OB受容体B(配列番号3)又はC(配列番 号5)の892位で始まるC−末端アミノ酸を有するサブパート(l)〜(o) のアミノ酸配列;及び (q) サブパート(a)〜(p)のいずれかの化学修飾誘導体; から選択されることを特徴とするOB受容体タンパク質製剤。 3. 前記OB受容体タンパク質がさらに、OB受容体タンパク質D(配列番号 7)の892位で始まるC−末端アミノ酸をさらに有するサブパート(l)〜( o)のOB受容体タンパク質から選択される、請求項2に記載のOB受容体タン パク質製剤。 4. 前記OB受容体タンパク質がさらに、799位で始まる置換C−末端アミ ノ酸、GKFTIL(配列番号13)をさらに有するサブパート(l)〜(o) のOB受容体タンパク質から選択される、請求項2に記載のOB受容体タンパク 質製剤。 5. 前記OB受容体タンパク質の細胞外ドメインが修飾されており、該修飾が : (a) ランダムコイルドメインの全て又は一部の欠失; (b) 最初のセリンを別のアミノ酸で置換することによる一方又は両方の“W SXWS”ボックスの修飾; (c) 最後のセリンを別のアミノ酸で置換することによる一方又は両方の“W SXWS”ボックスの修飾; (d) 最初のトリプトファンを別のアミノ酸で置換することによる一方又は両 方の“WSXWS”ボックスの修飾; から選択される、請求項1から4のいずれか一項に記載のOB受容体タンパク質 製剤。 6. 請求項1から5のいずれか一項に記載のOB受容体タンパク質をコードす るDNA分子であって、 (a)配列番号2、4、6、8、11、12及び14に記載のDNA配列; (b) サブパート(a)のDNAに選択的にハイブリダイズするDNA;及び (c) 遺伝暗号の縮重が無ければ、サブパート(a)又は(b)のDNAにハ イブリダイズするであろうDNA; からなる群から選択されることを特徴とする上記DNA分子。 7. 請求項6のDNAを含む生物学的に機能性のウイルス又はプラスミドベク ター。 8. 請求項7のベクターを含む原核又は真核宿主細胞。 9. 内因性OB受容体タンパク質の発現を促進するように修 飾された宿主細胞。 10. 単離されたヒト宿主細胞である、請求項9に記載の宿主細胞。 11. OB受容体タンパク質を産生させる方法であって、適当な条件下に、請 求項8、9又は10のいずれか一項に記載の宿主細胞を培養するステップ、産生 されたOB受容体を得るステップ、及び場合によって、該OB受容体を含む医薬 組成物を製造するステップからなる方法。 12. 肥満症、糖尿病、高脂血症、及び高コレステロール血症から選択される 治療疾患に罹患している患者を治療する方法であって、請求項1から5のいずれ か一項に記載されているか又は請求項11に記載の方法によって製造されたOB 受容体タンパク質を含むOB受容体タンパク質製剤を治療量投与することからな る方法。 13. 単に美容目的で体重を減少させるか又は体重を維持するために患者を治 療する方法であって、請求項1から5のいずれか一項に記載されているか又は請 求項11に記載の方法によって製造されたOB受容体タンパク質を含むOB受容 体製剤を有効量投与することからなる方法。 14. 肥満症、糖尿病、高脂血症、又は高コレステロール血症の治療用薬剤を 製造するための、請求項1から5のいずれか一項に記載されているか又は請求項 11の方法で製造されたOB受容体タンパク質の使用。 15. 場合によって医薬上許容し得る組成物中の、OBタンパク質部分及びO B受容体タンパク質部分を含むOBタンパク質/OB受容体タンパク質複合製剤 であって、 (a) 前記OB受容体タンパク質が請求項1及び2のいずれか一項に記載のも のから選択され; (b) 前記OBタンパク質部分が: (i) 天然型OBタンパク質;及び (ii) 非天然型OBタンパク質又はその類似体若しくは誘導体 から選択されるOBタンパク質/OB受容体タンパク質複合製剤。 16. 前記OB受容体タンパク質が、請求項3、4及び5のいずれか一項に記 載のものから選択される、請求項15のOBタンパク質/OB受容体タンパク質 複合製剤。 17. 肥満症、糖尿病、高脂血症及び高コレステロール血症 から選択される治療疾患に罹患している患者を治療する方法であって、請求項1 5又は16のOBタンパク質/OB受容体タンパク質複合製剤を治療量投与する ことからなる方法。 18. 前記OBタンパク質/OB受容体タンパク質複合製剤が、OBタンパク 質を発現する第1の細胞集団と、OB受容体タンパク質を発現する第2の細胞集 団を患者に投与することによりイン ビボで形成される、請求項17に記載の方 法。 19. 単に美容目的で体重を減少させるか又は体重を維持するために患者を治 療する方法であって、請求項1から5のいずれか一項に記載されているか又は請 求項11に記載の方法により製造されたOB受容体タンパク質部分を含むOBタ ンパク質/OB受容体タンパク質複合製剤を治療量投与することからなる方法。 20. 肥満症、糖尿病、高脂血症、高コレステロール血症の治療用薬剤を製造 するための、請求項15又は16に記載のOBタンパク質/OB受容体タンパク 質複合製剤の使用。
