Claims (1)
- 【特許請求の範囲】 1.アミノ酸残基の少なくとも約30%がグリシンであり、アミノ酸残基の少な くとも約10%がプロリンまたはヒドロキシプロリンである、反復するアミノ酸 トリマーの鎖を含む合成コラーゲン様物質において、該アミノ酸鎖における、少 なくとも1部での、ペプトイド残基の取り込みおよび該鎖からのペプトイド残基 含有トリプルヘリックスの形成を含む改良。 2.該アミノ酸鎖において少なくとも10%のペプトイド残基を含む、請求項1 記載の合成コラーゲン様物質。 3. Gly-Xp-Pro; Gly-Pro-Yp; Gly-Pro-Hyp;および Gly-Pro-Pro; またはこれらの組合わせ、 からなる群から選択される少なくとも3つのトリペプチドまたはジペプチド−ペ プトイド残基トリマーの少なくとも1つのポリペプチド鎖を含んでいる合成コラ ーゲン物質であって、ここで、XpおよびYpは、ペプトイド残基であり、該コラー ゲンがらせん二次構造およびトリプルヘリックスコンホメーションを含む三次構 造を有している3つのポリペプチド鎖で並んでいる、物質。 4.Gly-Xp-ProおよびGly-Pro-Yp;またはそれらの組合わせからなる群から選択 される反復するペプチド−ペプトイド残基トリマーの少なくとも1つの鎖を含ん でいる請求項3記載の合成コラーゲン物質。 5.反復するペプチド−ペプトイド残基トリマーのペプトイド残基がN−置換グ リシンである請求項4記載の合成コラーゲン物質。 6.該ペプトイド残基が、グリシン、バリン、ロイシン、イソロイシン、グルタ ミン、リシン、フェニルアラニンおよびアスパラギン酸のN−置換ペプトイド同 質異性からなる群から選択される、請求項5記載の合成コラーゲン物質。 7.該ペプトイドが、N−イソブチルグリシン(Nleu)、N−メチルグリシン(Sar );N−イソプロピルグリシン(Nval);およびN−sec−ブチルグリシン(Nile )からなる群から選択される請求項6記載の合成コラーゲン物質。 8.反復するペプチド−ペプトイド残基トリマーが、式Gly-Pro-NleuまたはGly- Nleu-Proのものである請求項7記載の合成コラーゲン物質。 9.式 (Z)n [式中、Zは、 (Gly-Xp-Pro)j; (Gly-Pro-Yp)k; (式中、XpおよびYpは、ペプトイド残基である) からなる群から選択されるジペプチド−ペプトイドトリマーである] で示される合成コラーゲンアミノ酸鎖であって、場合により該鎖は、 (Gly-Pro-Pro)l;および (Gly-Pro-HyP)m からなる群から選択される少なくとも1つのトリペプチドを含み、そして該アミ ノ酸鎖におけるj、k、l、およびmの合計はnであり、nは>3である、合成コラー ゲンアミノ酸鎖。 10.式 X-(Z)n-Y で示される合成コラーゲンアミノ酸鎖であって、 (Gly-Xp-Pro)j; (Gly-Pro-Yp)k; (式中、XpおよびYpは、ペプトイド残基である)、 からなる群から選択される、少なくとも1つのジペプチド−ペプトイド残基トリ マーZ、および場合により、 (Gly-Pro-Pro)l;および (Gly-Pro-Hyp)m; からなる群より選択される少なくとも1つのトリペプチドを含み、そして該アミ ノ酸鎖において、j、k、l、およびmの合計はnであり、nは>3である;合成コラ ーゲンアミノ酸鎖。 (式中、Xは、直鎖、分岐鎖、飽和または不飽和脂肪酸、芳香族カルボン酸、お よびアラールキルカルボン酸からなる群から選択され、アミド結合によりポリペ プチドのN−末端アミノ酸に結合しており、かつ Yは、アンモニア、直鎖、分岐鎖、飽和または不飽和脂肪族アミン、芳香族アミ ン、およびアラールキルアミンからなる群から選択され、Yは、アミド結合によ りポリペプチドのC−末端に結合している) 11.Xが、アセチル基であり、Yが、アミド、ビピリシンおよびマレイミドから なる群から選択される、請求項10記載の合成コラーゲンアミノ酸鎖。 12.式 TP-[A-(Gly-Xp-Pro)n-NH2]3またはTP-[A-(Gly-Pro-Yp)n-NH2]3 (式中TPは、3つの規則正しくスペースをおく官能性基を有するテンプレート分 子であって、各該基は反復するペプチド−ペプトイド残基鎖に共有結合しており 、該テンプレート分子は該鎖のトリプルヘリックス折りたたみを誘導することが でき; Aは、随意の2官能性スペーサー分子であり; XpおよびYpは、ペプトイド残基であり; nは、>3である) またはこれらの組合せ; で示される反復するペプチド−ペプトイド残基トリマーのテンプレート結合アミ ノ酸鎖集合体。 13.式 TP-[A-(Gly-Pro-Hyp)n-NH2]3 (式中、TPは、3つの規則正しくスペースをおく官能性基を有するテンプレート 分子であって、各該基は、反復するトリペプチドの鎖に共有結合されており、該 テンプレート分子は該鎖のトリプルヘリックス折りたたみを誘導することができ 、 Aは、随意の2官能性のスペーサー分子であり、 nは、>3である) で示される反復するトリペプチドのテンプレート結合ポリマー。 14.テンプレート結合アミノ酸鎖集合体であって、 3つの規則正しくスペースをおいた官能性基を有しているテンプレート分子、TP ;官能性末端基を有するC1〜C6アルキル鎖を含む随意のスペーサー、A; (Gly-Xp-Pro)j; (Gly-Pro-Yp)k; (Gly-Pro-Pro)l;および (Gly-Pro-HyP)m (式中、XpおよびYpは、ペプトイド残基である); からなる群から選択されるトリペプチドおよびペプチド−ペプトイド残基トリマ ーを含み、そして、j、k、l、およびmの合計=nであり、ここで、nは>3である 、3アミノ酸鎖; を含み、該鎖は同じかまたは異なるアミノ酸配列を有し、直接的にまたは該スペ ーサー、Aを介して間接的に該テンプレート分子の官能性基に共有結合し、それ によって該テンプレート分子は該鎖のトリプルヘリックス折りたたみを誘導する 、テンプレート結合アミノ酸鎖集合体。 15.TPがケンプトリ酸(KTA)(cis,cis−1,3,5−トリメチルシクロヘキ サン−1,3,5−トリカルボン酸)、またはトリメシン酸(TMA)(1,3,5 −ベンゼントリカルボン酸)である、請求項12記載のテンプレート結合集合体 。 16.「A」が、Gly-(NH-CH2-COO-)および6−アミノヘキサン酸(NH2-(CH2)5- COO-)からなる群から選択されるスペーサーである、請求項12記載のテンプレ ート結合集合体。 17.XpおよびYpが、各々、グリシン、バリン、ロイシン、イソロイシン、グル タミン、リシン、フェニルアラニンおよびアスパラギン酸に関連するN−置換ペ プトイドからなる群から選択されるペプトイド残基である、請求項12記載のテ ンプレート結合集合体。 18.該ペプトイド残基が、N−イソブチルグリシン(Nleu)、N−メチルグリシ ン(Sar);N−イソプロピルグリシン(Nval);およびN-sec-ブチルグリシン( Nile)からなる群から選択される請求項17記載のテンプレート結合集合体。 19.式 KTA-(Gly-(Gly-Pro-Yp)n-NH2)3; KTA-(Gly-(Gly-Xp-Hyp)n-NH2)3; KTA-(Aha-(Gly-Pro-Yp)n-NH2)3;または KTA-(Aha-(Gly-Xp-Hyp)n-NH2)3 (式中、YpおよびXpはペプトイド残基であり; KTAは、ケンプトリ酸(cis,cis−1,3,5−トリメチルシクロヘキサン−1, 3,5−トリカルボン酸)であり; Ahaは、6−アミノヘキサン酸(-NH2-(CH2)5-COO-)であり;そして、 nは、>1である) で示されるテンプレート結合アミノ酸鎖集合体。 20.式 TMA-(Gly-(Gly-Pro-Yp)n-NH2)3;または TMA-(Gly-(Gly-Xp-Hyp)n-NH2)3; (式中、XpおよびYpはペプトイド残基であり; TMAは、トリメシン酸(1,3,5−ベンゼントリカルボン酸)であり;そして 、 nは、>1である) で示されるテンプレート結合アミノ酸鎖集合体。 21.Xpがグリシン、バリン、ロイシン、イソロイシン、グルタミン、リシン、 フェニルアラニンおよびアスパラギン酸に関連するN−置換ペプトイドからなる 群から選択されるペプトイド残基である、請求項12記載のテンプレート結合ア ミノ酸鎖集合体。 22.該ペプトイド残基が、N−イソブチルグリシン(Nleu)、N−メチルグリシ ン(Sar);N−イソプロピルグリシン(Nval);およびN−sec−ブチルグリシン (Nile)からなる群から選択される、請求項21記載のテンプレート結合集合体 。 23.XpがNleuである、請求項22記載のテンプレート結合集合体。 24.3アミノ酸鎖のトリプルヘリックス折りたたみを誘導するための方法であ って、 (a)1つの該アミノ酸鎖のN−末端を、ヘリックス誘導テンプレート分子の3 つの官能性基の各々に、直接的にかまたは2官能性スペーサー分子を介してかの い ずれかで、共有的に付着させる工程;および (b)テンプレートに集合しているアミノ酸鎖が安定なトリプルヘリックスコン ホメーションをとることを可能にする工程、 を含む方法。 25.該アミノ酸鎖を樹脂支持体上の固相セグメント縮合法により集合させる予 備的工程を含んでいる、請求項24記載の方法。 26.工程(a)が、樹脂に結合した集合させられたアミノ酸鎖のN末端を該テ ンプレートの官能性基に付着させることを含んでいる、請求項25記載の方法。 27.工程(a)が、集合した遊離のアミノ酸鎖のN末端を溶液中の該テンプレ ートの官能性基に付着させることを含んでいる、請求項24記載の方法。 28.各該アミノ酸鎖が Gly-Xp-Pro; Gly-Pro-Yp; Gly-Pro-Hyp;および Gly-Pro-Pro またはこれらの組合せ (式中、XpおよびYpは、ペプトイド残基である) からなる群から選択される反復するトリマー配列を含んでいる、請求項24記載 の方法。 29.該アミノ酸鎖が、式 (Gly-Xp-Pro)n;または (Gly-Pro-Yp)n またはこれらの組合せ (式中、XpおよびYpはペプトイド残基であり、nは>1である) で示される反復するペプチド−ペプトイド残基トリマーを含んでいる、請求項2 4記載の方法。 30.ポリペプチド鎖が反復するトリペプチドを含む、請求項24記載の方法。 31.該ポリペプチド鎖が、Gly-Pro-HypおよびGly-Hyp-Proまたはこれらの組合 せからなる群から選択される反復するトリペプチドを含んでいる、請求項 30記載の方法。 32.請求項24記載の方法により調製されるヘリックスにコンホメーションを 形成しているトリプレックスアミノ酸鎖を含んでいる合成コラーゲン材料。 33. Gly-Xp-Pro; Gly-Pro-Yp; Gly-Pro-Hyp;および Gly-Pro-Pro; (式中、XpおよびYpは、グリシン、バリン、ロイシン、イソロイシン、グルタミ ン、リシン、フェニルアラニンおよびアスパラギン酸のN−置換ペプトイド同質 異性からなる群から選択される) からなる群から選択される少なくとも1つのトリペプチドを含んでいる、単離さ れ、精製されている化合物。 34.XpおよびYpがN−イソブチルグリシン(Nleu)、N−メチルグリシン(Sar) ;N−イソプロピルグリシン(Nval);およびN−sec−ブチルグリシン(Nile )からなる群から選択される請求項33の化合物。
