Description
Domanda di brevetto per invenzione industriale dal titolo:
”Derivati dell’acido retinoico e processo per la loro preparazione”
DESCRIZIONE
La presente invenzione riguarda nuovi derivati dell’acido retinoico, un processo per la loro preparazione ed il loro uso cosmetico.
Il fenomeno dell’invecchiamento risulta notevole soprattutto a livello della pelle, dove si ha un aumento delle rughe, aumento dell’inspessimento, rigidità, secchezza, cosa che potrebbe anche essere attribuita all’esposizione della pelle alla luce solare per prolungati periodi di tempo. I sintomi causati dall’esposizione alla luce solare vengono definiti come fotoinvecchiamento e si basano su modifiche a livello dell’epidermide e del derma. È dimostrato che il foto-invecchiamento può essere rallentato applicando sulla pelle una crema contenente sostanze quali tretinoina, retinolo e derivati, alfa-idrossi acidi (AHAs).
La tretinoina è un composto liposolubile ed instabile nel corpo vivente, inoltre è irritante sulla pelle, quindi potrebbe causare alcuni effetti collaterali se applicata direttamente sulla pelle, come ad esempio secchezza, ferite e raschiature. Il retinolo, al contrario, può essere utilizzato, ma essendo instabile a luce, calore, perossidi ed ossigeno, può comportare costi aggiuntivi per la presenza dei vari stabilizzanti.
I derivati dell’acido retinoico si sono dimostrati molto più stabili e con miglior effetti, in particolare i derivati con la nicotinamide, o vitamina PP, molecola dotata di molteplici attività: normalizza il contenuto di coenzima nicotinamide a livello del derma, stimola la produzione di collagene, incrementa le proprietà di barriera della pelle normalizzando la produzione di sfingolipidi epidermici (ceramidi in particolare), stimolazione dei cheratinociti epidermici basali formando uno strato corneo funzionale e riduzione della pigmentazione. I derivati nicotinamidici dell’acido retinoico quindi, grazie all’effetto sinergico, possono essere impiegati per il trattamento del foto-invecchiamento e per l’iperpigmentazione della pelle. Costituiscono pertanto oggetto della presente invenzione, composti di formula (I)
dove
X e Y, uguali o diversi tra loro, sono NH, N-alchil, N-aril o un atomo di ossigeno;
A è un gruppo C1-8alchilidene, lineare o ramificato;
e loro sali.
In particolare, costituiscono oggetto preferito della presente invenzione i seguenti composti
I composti di formula (I) sono utili in campo cosmetico e medico, in particolare per la prevenzione e/o il trattamento del foto-invecchiamento della pelle e dell’acne.
Costituiscono pertanto un ulteriore oggetto della presente invenzione composizioni cosmetiche contenenti un composto di formula (I) per la prevenzione o il trattamento del foto-invecchiamento della pelle come pure composizioni cosmetiche o mediche contenenti un composto di formula (I) per la prevenzione o il trattamento dell’acne.
Le composizioni oggetto della presente invenzione vengono preparate secondo tecniche convenzionali e contengono un veicolante adatto per l’uso cosmetico o farmaceutico.
Costituisce ulteriore oggetto della presente invenzione un processo per la sintesi di composti di formula (I) comprendente la reazione di condensazione di composti di formula (II)
dove A, X ed Y hanno i significati sopra riportati;
con acido retinoico di formula (III)
in presenza di un agente condensante, eventualmente di una base e in un solvente aprotico a dare composti di formula (I).
I composti di formula (II) sono noti e preparati secondo le tecniche note nell’arte.
Nel processo oggetto della presente invenzione, adatti agenti condensanti sono N,N’-carbonildiimidazolo, N,N’-dicicloesilcarbodiimmide, 4,6-tri-npropil-2,4,6-triosso-1,3,5,2,4,6-triossa-trifosforinano, preferibilmente viene utilizzato 4,6-tri-n-propil-2,4,6-triosso-1,3,5,2,4,6-triossa-trifosforinano. La base è scelta tra trietilammina, diisopropiletilammina, N,N-dimetilamminopiridina, nicotinato di metile; preferibilmente trietilammina e N,N-dimetilamminopiridina catalitica.
Adatti solventi aprotici sono acetonitrile ed esteri; preferibilmente viene utilizzato acetato di etile.
Costituisce una forma preferita del processo oggetto della presente invenzione, la reazione di condensazione del composto di formula (IIa)
con acido retinoico di formula (III) in presenza di 4,6-tri-n-propil-2,4,6-triosso-1,3,5,2,4,6-triossa-trifosforinano, trietilammina e N,N-dimetilamminopiridina catalitica, in acetato di etile.
Sebbene l’invenzione sia stata descritta nei suoi aspetti caratteristici, modifiche ed equivalenti che sono evidenti all’esperto del ramo sono incluse nella seguente invenzione.
La presente invenzione sarà ora illustrata per mezzo di alcuni esempi, che non devono essere visti come limitanti la portata dell’invenzione.
ESEMPIO 1
Sintesi di 2-(nicotinammido)-etil retinoato (Ia).
In un pallone di reazione sono stati caricati 0,6 g di idrossietilnicotinammide (3,6 mmol), 5 ml di acetato di etile, 1,08 g di acido retinoico (3,6 mmol), 0,8 g di trietilammina (7,92 mmol), N,N-dimetilamminopiridina catalitica e sotto agitazione, a circa 25°C, è stato aggiunto 2,52 g di una soluzione di 4,6-trin-propil-2,4,6-triosso-1,3,5,2,4,6-triossa-trifosforinano al 50% in acetato di etile (3,96 mmol). La miscela di reazione è stata mantenuta in queste condizioni per quindici ore e a reazione terminata sono stati aggiunti 5 ml di acqua. La fase organica è stata lavata con sodio idrossido 1N (1 x 5 ml) e acido cloridrico 1N (1 x 5 ml). Le fase organica è stata ridotta a residuo mediante distillazione sottovuoto, che è stato purificato mediante cromatografia su colonna (eluente: cloruro di metilene:metanolo 97,5:2,5), a dare 1 g di 2-(nicotinammido)-etil retinoato come solido.
<1>H-NMR (CDCl3, 300 MHz): δ 8,99 (d, 1H), 8,68 (dd, 1H), 8,11 (dt, 1H), 7,35 (dd, 1H), 7,10 (m, 1H), 7,00 (dd, 1H), 6,25 (m, 2H), 6,16 (m, 2H), 5,75 (s, 1H), 4,33 (t, 2H), 3,75 (m, 2H), 2,32 (s, 3H), 1,99 (m, 4H), 1,68 (s, 3H), 1,50 (m, 3H), 1,43 (m, 2H), 0,98 (s, 6H).
ESEMPIO 2
Sintesi di 2,2-dimetilpropil-retinil nicotinato (Ib)
In un pallone di reazione sono stati caricati 0,6 g di 3-idrossi-2,2-dimetilpropil-nicotinato (3,6 mmol), 5 ml di acetato di etile, 1,08 g di acido retinoico (3,6 mmol), 0,8 g di trietilammina (7,92 mmol), N,N-dimetilamminopiridina catalitica e sotto agitazione, a circa 25°C, è stato aggiunto 2,52 g di una soluzione di 4,6-tri-n-propil-2,4,6-triosso-1,3,5,2,4,6-triossa-trifosforinano al 50% in acetato di etile (3,96 mmol). ESEMPIO 3
Sintesi di 2-(amminoetil)-retinil nicotinato (Ic)
In un pallone di reazione sono stati caricati 0,6 g di 2-amminoetilnicotinammide (3,6 mmol), 5 ml di acetato di etile, 1,08 g di acido retinoico (3,6 mmol), 0,8 g di trietilammina (7,92 mmol), N,N-dimetilamminopiridina catalitica e sotto agitazione, a circa 25°C, è stato aggiunto 2,52 g di una soluzione di 4,6-tri-n-propil-2,4,6-triosso-1,3,5,2,4,6-triossa-trifosforinano al 50% in acetato di etile (3,96 mmol). Claims (9)
- RIVENDICAZIONI 1) Un composto di formula (I)
- dove X e Y, uguali o diversi tra loro, are NH, N-alchil, N-aril o un atomo di ossigeno; A è un gruppo C1-8alchilidene, lineare o ramificato; e loro sali. 2) Un composto secondo rivendicazione 1 di formula
- 3) Un processo per la sintesi di composti secondo la rivendicazione 1 comprendente la reazione di condensazione di composti di formula (II)dove A, X ed Y hanno i significati sopra riportati; con acido retinoico di formula (III)in presenza di un agente condensante, eventualmente di una base e in un solvente aprotico.
- 4) Un processo secondo rivendicazione 3 in cui l’agente condensante è scelto tra N,N’-carbonildiimidazolo, N,N’-dicicloesilcarbodiimmide, 4,6-trin-propil-2,4,6-triosso-1,3,5,2,4,6-triossa-trifosforinano, preferibilmente 4,6-tri-n-propil-2,4,6-triosso-1,3,5,2,4,6-triossa-trifosforinano.
- 5) Un processo secondo rivendicazione 3 in cui la base è scelta tra trietilammina, diisopropiletilammina, N,N-dimetilamminopiridina, nicotinato di metile; preferibilmente trietilammina e N,N-dimetilamminopiridina catalitica.
- 6) Un processo secondo rivendicazione 3 in cui il solvente aprotico è scelto tra acetonitrile ed esteri; preferibilmente acetato di etile.
- 7) Composizioni cosmetiche contenenti un composto secondo la rivendicazione 1 per la prevenzione o il trattamento del foto-invecchiamento della pelle.
- 8) Composizioni cosmetiche o mediche contenenti un composto secondo la rivendicazione 1 per la prevenzione o il trattamento dell’acne.
- 9) Uso cosmetico o medico di un composto secondo la rivendicazione 1.
