1492579 Vincamine derivatives SANDOZ Ltd 16 Dec 1974 [18 Dec 1973 6 Feb 1974 8 Feb 1974] 54159/74 Heading C2C Novel compounds I (in which R 1 is Br, F, Cl, OH, or 1-40 alkyl or alkoxy with the proviso that when R 1 is in the 11- or 12-position it is other than -OCH 3 ) are prepared (a) by cyclization of a compound II (in which R 2 is 1-4C alkyl) in the presence of acid; (b) by reaction of a compound III at less than 0‹ C. with a hydrogen halide, followed by hydrolysis or (c) by bromination of vincamine to give the 11-bromo derivative. Compounds II are prepared by reaction of a compound IV with a compound V to give a compound VI reaction of the compound VI with imidazole to give a compound VII treatment of the compound VII with acid to give a compound VIII reaction of the compound VIII with a compound IX to give a compound X Bischler-Napieralski ring closure of the compound X to give a compound XI and reduction of the compound XI. Pharmaceutical compositions exhibiting vigilance-increasing and psychostimulation activity comprise a compound I or a pharmaceutically acceptable acid addition salt thereof together with a suitable excipient and are administered orally or parenterally.
