Claims (14)

1. Upotreba supstrata za pojačavanje fluorescencije jedne ili više fluorescentnih molekula, naznačena time što supstrat sadrži čvrsti polimerni nosač koji ima više međusobno razdvojenih udubljenja, te što je čvrsti nosač barem djelomično obložen srebrom ili legurama koje sadrže srebro, pri čemu su udubljenja međusobno udaljena 0,2 µm do 2,5 µm i duboka 0,1 µm do 5 µm, te pri čemu je sloj metala na čvrstom nosaču debeo 10 nm do 60 nm.
2. Upotreba u skladu s patentnim zahtjevom 1, naznačena time što udubljenja u čvrstom nosaču imaju duljinu i širinu, pri čemu je omjer između duljine i širine 2:1 do 1:2, osobito 1:1.
3. Upotreba u skladu s patentnim zahtjevom 1 ili 2, naznačena time što udubljenja u čvrstom nosaču imaju duljinu i širinu, pri čemu su duljina i širina udubljenja 0,1 µm do 2 µm.
4. Upotreba u skladu s bilo kojim od patentnih zahtjeva 1 do 3, naznačena time što udubljenja imaju uglavnom okrugao oblik.
5. Upotreba u skladu s bilo kojim od patentnih zahtjeva 1 do 4, naznačena time što čvrsti nosač sadrži najmanje jedan materijal kojeg se bira iz skupine koju čine termoplastični polimeri i polikondenzati.
6. Upotreba u skladu s patentnim zahtjevom 5, naznačena time što materijal čvrstog polimernog nosača sadrži organske i/ili anorganske aditive i/ili punila, pri čemu se navedene po mogućnosti bira iz skupine koju čine TiO2, staklo, ugljik, obojeni pigmenti, lipidi i voskovi.
7. Upotreba u skladu s bilo kojim od patentnih zahtjeva 1 do 6, naznačena time što je supstrat dio kapilarne cijevi, mikrotitarske ploče, mikrofluidičkog čipa, analitičke trake, nosača za fluorescentnu mikroskopiju, senzorskog polja ili polja optičkih detektora.
8. Upotreba u skladu s bilo kojim od patentnih zahtjeva 1 do 7, naznačena time što metalna obloga na površini supstrata barem djelomično sadrži molekule za izravno i/ili posredno vezanje fluorescentnih molekula.
9. Postupak određivanja ili kvantificiranja najmanje jednog analita u uzorku, koji se sastoji od ovih koraka: a) izbornog izravnog ili posrednog obilježavanja najmanje jednog analita najmanje jednim fluoroforom, b) primjene najmanje jednog obilježenog analita iz koraka a) ili fluorescentnog analita na supstrat u skladu s bilo kojim od patentnih zahtjeva 1 do 8, c) pobuđivanja najmanje jednog fluorofora ozračivanjem supstrata svjetlom odgovarajuće valne duljine, te d) mjerenja fluorescencije radi određivanja prisutnosti ili kvantificiranja najmanje jednog analita u uzorku.
10. Postupak u skladu s patentnim zahtjevom 9, naznačen time što najmanje jedan fluorofor ima valnu duljinu pobude u rasponu od 360 do 780 nm, po mogućnosti od 490 do 680 nm.
11. Postupak u skladu s patentnim zahtjevom 9 ili 10, naznačen time što najmanje jedan fluorofor ima valnu duljinu emisije u rasponu od 410 do 800 nm, po mogućnosti od 510 do 710 nm.
12. Postupak u skladu s bilo kojim od patentnih zahtjeva 9 do 11, naznačen time što se najmanje jedan fluorofor bira iz skupine koju čine metoksikumarin, aminokumarin, Cy2, Alexa Fluor 488, fluorescein-izotiocijanat (FITC), Alexa Fluor 430, Alexa Fluor 532, Cy3, Alexa Fluor 555, 5-TAMRA, Alexa Fluor 546, fikoeritrin (PE), tetrametilrodamin-izotiocijanat (TRITC), Cy3.5, rodamin, Alexa Fluor 568, Alexa Fluor 594, Alexa Fluor 633, Alexa Fluor 647, Cy5, Alexa Fluor 660, Cy5.5, Alexa Fluor 680 i Cy7, po mogućnosti iz skupine koju čine fluorescein-izotiocijanat (FITC), Cy3, fikoeritrin (PE), tetrametilrodamin-izotiocijanat (TRITC), Cy5 i Alexa Fluor 680.
13. Postupak u skladu s bilo kojim od patentnih zahtjeva 9 do 12, naznačen time što se posredno obilježavanje analita najmanje jednim fluoroforom provodi uz pomoć molekule obilježene fluoroforom koja se veže na analit.
14. Postupak u skladu s patentnim zahtjevom 13, naznačen time što se molekulu koja se veže na analit bira iz skupine koju čine protutijela, fragmenti protutijela, po mogućnosti fragmenti Fab, F(ab)’2 ili scFv, nukleinske kiseline, enzimi, lipidi, virusne čestice, aptameri i njihove kombinacije.
