Claims (12)
1. Postupak za dekontaminaciju biološkog materijala od virusnih, prionskih i / ili bakterijskih onečišćenja, koji obuhvaća centrifugiranje troslojnog tekućeg sustava koji sadrži:
(a) gornji sloj koji sadrži vodenu otopinu biološkog materijala za dekontaminaciju;
(b) međusloj koji sadrži jedno ili više organskih otapala;
(c) donji sloj koji sadrži inaktivirajuću otopinu za navedena onečišćenja,
gdje navedeni slojevi imaju različitu gustoću, redoslijedom: (a) <(b) <(c), i naznačeni time što se sloj (b) ne miješa sa slojem (a).
2. Postupak prema zahtjevu 1, naznačen time što slojevi (a) i (b) imaju razliku u gustoći od najmanje 0.025 g / ml.
3. Postupak prema zahtjevima 1-2, naznačen time što slojevi (a) i (b) imaju razliku u gustoći između 0.030 i 0.3 g / ml.
4. Postupak prema zahtjevima 1-3, naznačen time što je sloj (b) mješavina halogeniranog alifatskog organskog otapala s aromatičnim organskim otapalom, u volumetrijskim omjerima između 1: 0.5 i 1: 4, poželjno između 1: 0.8 i 1:3.
5. Postupak prema zahtjevima 1-4, naznačen time što je halogenirano alifatsko organsko otapalo odabrano između metil klorida, metilen klorida, kloroforma i ugljičnog tetraklorida, i aromatsko organsko otapalo je odabrano između toluena, benzena, C2-4 alkil benzena, ksilena i njihovih derivata.
6. Postupak prema zahtjevima 1-5, naznačen time što se srednji sloj (b) djelomično miješa sa slojem (c).
7. Postupak prema zahtjevima 1-6, naznačen time što je sloj (c) otopina uree koja ima koncentraciju od najmanje 6M, ili otopina hidroksida alkalnog ili zemno-alkalnog metala s koncentracijom od najmanje 0.5 M.
8. Postupak prema zahtjevima 1-7, naznačen time što je sloj (b) mješavina kloroforma i toluena i / ili sloj (c) sadrži jedan ili više: ditiotreitola, uree, natrijevog bromida, natrijevog klorida, natrijevog jodida i sredstva koje navedenoj mješavini daje pH između 8 i 11.
9. Postupak prema zahtjevima 1-8, naznačen time što biološki materijal pripada jednoj ili više klasa aminokiselina, proteina, enzima, hormona.
10. Postupak prema zahtjevima 1-9, naznačen time što je biološki materijal odabran između FSH, LH, hCG, testosterona, progesterona, estradiola, T3 i / ili T4 hormona štitnjače, inzulina, glukagona, gastrina, somatostatina, somatotropina, hormona rasta, prolaktina, renina, angiotenzinogena, angiotenzina, gonadotropina, kortizola, adrenokortikotropnog hormona (ACTH), modificirane aminokiseline, L-DOPA, dopamina, epinefrina, norepinefrina, serotonina, histamina, GABA i njihovih derivata.
11. Postupak prema zahtjevima 1-10, gdje je zadovoljen jedan ili više sljedećih uvjeta:
- sloj (a) sadrži: navedeni biološki materijal u koncentraciji između 1 i 100 mg / ml, pufersko sredstvo koje sloju (a) daje pH između 6.5 i 8.0;
- sloj (b) je mješavina kloroforma i toluena u volumetrijskim omjerima između 1: 0.5 i 1:4;
- sloj (c) je otopina uree s molarnošću između 7M i 9M, koja sadrži ditiotreitol, NaBr, i pufersko sredstvo koje sloju (c) daje pH između 8.0 i 11.0.
12. Postupak prema zahtjevu 11, gdje je zadovoljen jedan ili više sljedećih uvjeta:
- sloj (a) sadrži: navedeni biološki materijal u koncentraciji između 10 i 50 mg / ml, pufersko sredstvo koje sloju (a) daje pH između 7 i 7.5;
- sloj (b) je mješavina kloroforma i toluena u volumetrijskim omjerima između 1: 0.8 i 1: 3;
- sloj (c) je 8M otopina uree, koja sadrži ditiotreitol, NaBr i pufersko sredstvo koje sloju (c) daje pH između 8.0 i 11.0.
