Claims (15)
1. Vektor naznačen time što sadrži
a) najmanje 10 funkcionalnih V gena za ljudski teški lanac pri čemu je najmanje 10 funkcionalnih V gena za ljudski teški lanac u istom redoslijedu u kojem se mogu naći u ljudskoj zametnoj liniji i sastoje se od intervencijskih sekvenci;
b) najmanje jedan D gen ljudskog teškog lanca i najmanje jedan J gen ljudskog teškog lanca;
c) mišji µ pojačivač;
d) mišji preklopni µ element;
e) mišji C gen koji nema CH1 egzon pri čemu je C gen odabran od Cγ1, Cγ2b i/ili Cγ2a i
f) mišju regiju 3' pojačivača.
pri čemu vektor ne sadrži daljnji mišji C gen.
2. Vektor prema zahtjevu 1 naznačen time što navedena mišja regija 3' pojačivača je veličine barem oko 42kb.
3. Vektor prema zahtjevu 2 naznačen time što navedena mišja regija 3' pojačivača 'sadrži jedan ili više ili svih pojačivačkih elemenata koji su odabrani od hs3A, hs1.2, hs3B, hs4, hs5, hs6 i hs7.
4. Vektor prema prethodnom zahtjevu naznačen time što sadrži 10 do oko 44 funkcionalnih ljudskih V gena.
5. Vektor prema bilo kojem prethodnom zahtjevu naznačen time što navedeni vektor je umjetni kromosom kvasca (YAC).
6. Stanica domaćina transgenog miša naznačena time što je transformirana s vektorom prema prethodnom zahtjevu.
7. Stanica domaćina transgenog miša prema zahtjevu 6 naznačena time što navedena stanica je ES stanica.
8. Transgeni miš naznačen time što sadrži vektor prema bilo kojem od zahtjeva 1 do 5 ili stanicu prema zahtjevu 6 ili 7.
9. Transgeni miš prema zahtjevu 8 naznačen time što navedeni miš sadrži jedan ili više nefunkcionalnih endogenih imunoglobulinskih lokusa.
10. Transgeni miš prema zahtjevu 9 naznačen time što navedeni miš sadrži nefunkcionalni endogeni lokus lambda laganog lanca, nefunkcionalni endogeni lokus kapa laganog lanca i/ili nefunkcionalni endogeni lokus teškog lanca.
11. Uporaba transgenog miša prema bilo kojem od zahtjeva 8 do 10 ili stanice prema zahtjevu 6 ili 7 naznačena time što je za proizvodnju HCAb ili VH domena.
12. Uporaba transgenog miša prema bilo kojem od zahtjeva 8 do 10 naznačena time što je za izgradnja biblioteke.
13. Postupak za dobivanje biblioteke koji obuhvaća ex vivo imunizaciju transgenog miša prema bilo kojem od zahtjeva 8 do 10 ili ex vivo tkiva ili stanica transgenog miša prema bilo kojem od zahtjeva 8 do 10.
14. Postupak za dobivanje HCAb ili VH domene koji obuhvaća uvođenje i ekspresiju vektora prema bilo kojem od zahtjeva 1 do 5 u mišu ili u mišjoj stanici domaćinu.
15. Postupak za proizvodnju topive VH vezujuće domene naznačen time što sadrži slijedeće korake:
a) ekspresiju vektora iz bilo kojeg od zahtjeva 1 do 5 u transgenom mišu,
b) izoliranje stanice ili tkiva koje eksprimira HCAb,
c) kloniranje sekvence koja kodira VH domenu iz mRNA dobivene iz izolirane stanice ili tkiva,
d) izgradnja biblioteke iz kloniranih transkripata i
e) izoliranje VH domene.
