Abstract
Pripravak za tvorbu kosti koji je dobiven postupkom za pripremu pripravka za tvorbu kosti korištenjem smjese osteoblasta i bio-matrice, naznačen time da se sastoji od: uzgoja/izrastanja izoliranih osteoblasta i njihovih prekursorskih stanica u alfa-MEM (minimalni esencijalni medij, iz alfa izmjene koštanog tkiva) za pripremu suspenzije osteoblasta; imiješanja rezultirajuće suspenzije osteoblasta sa bio-matricom za pripremu osteoblastnog terapijskog sredstva, pri čemu je bio-matrica mješavina kolagena i hidroksiapatita, pri čemu je kolagen dodan u iznosu od 67 µg/mL do 20 mg/mL u suspenziju osteoblasta, a hidroksiapatit se dodaje se u iznosu od 30 µg/mL do 3,4 mg/mL u suspenziju osteoblasta. Patent sadrži još 4 patentna zahtjeva.
Claims (6)
1. Pripravak za tvorbu kosti koji je dobiven postupkom za pripremu pripravka za tvorbu kosti korištenjem smjese osteoblasta i bio-matrice, naznačen time da se sastoji od:
uzgoja/izrastanja izoliranih osteoblasta i njihovih prekursorskih stanica u alfa-MEM (minimalni esencijalni medij, iz alfa izmjene koštanog tkiva) za pripremu suspenzije osteoblasta; i
miješanja rezultirajuće suspenzije osteoblasta sa bio-matricom za pripremu osteoblastnog terapijskog sredstva,
pri čemu je bio-matrica mješavina kolagena i hidroksiapatita,
pri čemu je kolagen dodan u iznosu od 67 µg/mL do 20 mg/mL u suspenziju osteoblasta, a hidroksiapatit se dodaje se u iznosu od 30 µg/mL do 3,4 mg/mL u suspenziju osteoblasta.
2. Pripravak za tvorbu kosti prema zahtjevu 1, naznačen time da korak pripreme terapijskog sredstva nadalje uključuje miješanje otopine izmiješanih osteoblasta, u kojoj je izmiješana bio-matrica u suspenziju osteoblasta, sa koagulacijskim sredstvom.
3. Pripravak za tvorbu kosti prema zahtjevu 2, naznačen time da korak miješanja uključuje:
4. miješanje izmiješane otopine osteoblasta sa 10 do 100 IU/mL trombina kao koagulacijskog sredstva; te miješanje nastale izmiješane otopine koja sadrži u njoj izmiješan trombin sa 20 do 100 mg/mL fibrinogena kao koagulacijskog sredstva.
5. Pripravak za tvorbu kosti prema zahtjevu 1, naznačen time da je kolagen neutraliziran do neutralnog pH uz dodavanje neutralizacijske otopine prije miješanja sa suspenzijom osteoblasta.
6. Pripravak za tvorbu kosti prema zahtjevu 3, naznačen time da
prije dodavanja u smjesu osteoblasta i bio-matrice, liofilizirani trombin kao trombin je otopljen u tekućini alfa-MEM u koju su dodani fosfatni (PO43-) ioni kao mineralna komponenta kosti, te se dodaje u dvostrukoj količini od količine trombina sadržanog u mediju kulture u koju se dodaju fosfatni (PO43-) ioni; te
prije dodavanja u rezultirajuću otopinu smjese osteoblasta, liofilizirani fibrinogen kao fibrinogen je otopljen u tekućini alfa-MEM u koju su dodani kalcijevi (Ca2+) ioni kao mineralna komponenta kosti, te se dodaje u dvostrukoj količini od količine fibrinogena sadržanog u mediju kulture u koju se dodaju kalcijevi (Ca2+) ioni.
