Abstract

Tryptophane is prepared by contacting a tryptophane auxotroph of E. coli in a nutrient medium with the compounds indole and serine. The tryptophane auxotroph fermentation inoculum is prepared by fermentation of the microorganism in a nutrient medium containing one or more of indole, tryptophane and anthranilic acid. After adjusting the pH to between 7,4 and 8,5 indole and serine are added and the broth is gently mixed and allowed to stand at 25 to 40 DEG C. for from 8 to 24 hours. Best results are obtained under stationary conditions. The nutrient medium may contain a carbohydrate source such as glucose, mannitol, sorbitol or glycerol of which one or more may be used. One or more organic acids such as acetic, citric, succinic, fumaric and glutamic may be present.
