Abstract
Diaminopimelic acid is prepared by the submerged aerobic fermentation of a mutant of Escherichia coli which requires lysine for its growth and lacks the enzyme diaminopimelic acid decarboxylase, the fermentation being carried out on a nutrient medium containing a source of lysine, the pH of which is maintained near neutrality. A suitable organism is E. coli ATCC 12408. Neutrality is maintained by the addition of alkali, e.g. sodium hydroxide, potassium hydroxide, or preferably ammonium hydroxide, or urea; towards the end of the fermentation the pH tends to rise, and is adjusted with sulphuric acid. The medium may contain glycerol, mannitol, and/or cornsteep liquor. The fermentation is preferably conducted at about 28 DEG C. for about three days. Specification 820,268 is referred to.ALSO:Diaminopimelic acid may be used as a supplement in the feeding of poultry, where it takes the place of lysine.
