Abstract
A process for the preparation of L-lysine comprises fermenting, with the sole organism employed being a mutant of Escherichia coli which initially requires L-lysine for growth, a nutrient medium containing a source of carbohydrate (e.g. crude molasses or dextrose), a source of trace minerals (e.g., MgSO4) and a source of nitrogen (e.g. ammonium hydrogen phosphate), and initially containing 0,1-0,5 g. of L-lysine per litre, the fermentation being initially conducted under submerged aerobic conditions for 36-60 hours (whereby diamino pimelic acid is accumulated in the fermentation broth), and then, after adjusting the pH to 7,2-8,2 and rupturing the cell wall of the organism, e.g. by treating the mixture with toluene, continuing the reaction for at least 12 additional hours, so converting the diaminopimelic acid to lysine. The second stage is preferably conducted under anaerobic conditions. The lysine may be adsorbed on a strong cation exchange resin and eluted with dilute alkali.
