Abstract

Lysine racemase, an enzyme capable of racemising D-or L-lysine to DL-lysine, is produced by growing a lysine-racemase-producing strain of Proteus vulganis, P. ammoniae, P. mirabilis, P. americanus, P. para-americanus, P. rettgeri, P. spingidis, P. inconstans, Escherichia anindolica and E. freundii in an aqueous nutrient medium containing sources of carbohydrate, organic nitrogen and inorganic nitrogen at an initial pH between 6,0 and 8,0. The temperature of cultivation is 20 DEG to 40 DEG C. and the process is preferably submerged and the duration may be one day. A suitable medium contains cornsteep liquor, ammonium hydrogen phosphate, crude beet molasses, glycerol and magnesium sulphate. For the production of DL-lysine from D-lysine or L-lysine, the lysine racemase is contacted at pH 7,0 and 9,0 and temperature of 20. to 40 DEG C. with the lysine. After acidification the DL-lysine may be absorbed on a sulphonic acid resin eluted with aqueous alkali and crystallised by known methods. The lysine racemete may be used in the form of (a) the whole broth adjusted to pH 7,0 to 9,0 and preferably covered with toluene to prevent contamination (b) bacterial cells removed from the broth, washed with water and suspended in an aqueous medium at pH 7,0 to 9,0 and (c) the cell free product.
