Abstract
Ristocetin, ristocetin A and ristocetin B, are purified by adding an aromatic aldehyde to an aqueous solution of the said antibiotic in order to precipitate the compound of ristocetin with the aldehyde. The aqueous solution, which may be a clarified broth, is preferably adjusted to pH 7.0 to 8.1 and especially 7.5 to 8.1 by the addition of alkali such as sodium bicarbonate or sodium hydroxide prior to the addition of the aldehyde which is preferably salicyl-aldehyde or cinnamaldehyde. The compound contains three aromatic aldehyde groups per mol. of ristocetin and in the case of the salicyl-aldehyde-ristocetin A is soluble in polyethylene glycol, propylene glycol, aqueous acetone, aqueous methanol, aqueous ethanol, pyridine and formamide and isoluble in amyl acetate, ethyl acetate, benzene and chloroform. It is non-irritating to tissue in 50% aqueous methanol. Ristocetin is recovered from the aldehyde compound by acidification, e.g. to pH 2.0 and (a) extracting the freed aromatic aldehyde with a solvent such as chloroform, or (b) precipitating the ristocetin salt, e.g. the sulphate by the addition of a non-solvent such as acetone.
